Abstract
本发明公开了一种负载纳豆激酶的血栓通络药物及其制备方法。该药物的组分包括:纳豆激酶、白果、杏仁、丝瓜藤、当归、铁皮石斛、淀粉、维生素E、天麻、皇菇1‑4份、磷脂酰丝氨酸、有机硒和香茅草秸秆。将白果、杏仁、丝瓜藤、当归、铁皮石斛、天麻、香茅草秸秆投入球磨机中磨碎得细粉;向细粉内加入去离子水,文火煎煮,过滤得第一混合液;将皇菇粉碎,再加入去离子水煎煮过滤第二混合液;将第一混合液、第二混合液、维生素E、磷脂酰丝氨酸、有机硒、淀粉混合,磁力搅拌后,喷雾干燥,即可。本发明负载纳豆激酶的血栓通络药物具有活血化瘀、滋补肝肾、增强抵抗力的功效,主治血栓,且成分多为食物,副作用小,成本低,减轻患者的经济负担。

技术领域
本发明涉及一种血栓治疗药物,具体涉及一种负载纳豆激酶的血栓通络药物及其制备方法。
背景技术
血栓是血流在心血管系统血管内面剥落处或修补处的表面所形成的小块,主要是由不溶性纤维蛋白,沉积的血小板,积聚的白细胞和陷入的红细胞组成,尤为常见为脑血栓。
纳豆是由纳豆菌发酵大豆而制成的功能性食品。大量研究表明,纳豆中除富含氨基酸、有机酸、寡糖外,还包括多种生理活性物质,如纳豆激酶可溶解血栓,主要是有效溶解不溶性。但传统的纳豆产品中纳豆激酶的活性较低,持续时间短,溶栓效果有效。
申请号201310677438.9 ,名称为一种治疗脑血栓的重要,具体公开了该中药由以下重量份数的原料药制成:荆芥9-15份、桂枝5-10份、牛膝5-15份、赤芍6-12份、乳香5-15份、艾叶10-15份、青黛10-15份、穿山甲3-9份、刘寄奴10-15份、柏子仁6-15份、滑石10-20份、蜈蚣3-5份。虽然该发明药物能从根本上整体调整治疗脑血栓,达到恢复健康的目的,制备工艺简单,本发明药物,全方共奏补气祛邪,滋养肝肾,破血逐瘀,活血通络,疏通脑脉,溶栓疗瘫之功效。能从根本上整体调理治疗,更有针对性的治疗脑血栓,达到恢复健康的目的,具有安全可靠,疗效确切,迅速,无痛苦,无毒副作用。同时本发明工艺简单,疗效好,使用方便,有效率高达100%,但疗程长,易受外界环境破坏。
申请号201610481540.5,名称为溶解血栓的纳豆激酶组合物及其制备方法,该组合物包括纳豆激酶、当归和淫羊藿组成;每g组合物中纳豆激酶的纤溶活性为200~400IU。其制备方法包括:1)利用产纳豆激酶的枯草杆菌制备纳豆激酶发酵液,离心,取上清液浓缩,得到纳豆激酶浓缩液;2)将淫羊藿加水提取,将提取液浓缩成淫羊藿浸膏;3)将当归加水提取,将提取液浓缩成当归浸膏;4)将淫羊藿浸膏、当归浸膏与纳豆激酶浓缩液混合,即得。本发明以纳豆激酶、当归和淫羊藿为原料,这三种组分发挥协同增效的作用,具有较好的预防血栓形成以及溶栓效果。
发明内容
针对现有技术的不足,本发明的目的在于提供一种负载纳豆激酶的血栓通络药物及其制备方法,该药物的活性成分来源于纳豆,降低成本的同时,保证了药物的疗效。
一种负载纳豆激酶的血栓通络药物,包括以下按重量份数计的组分:纳豆激酶0.01-0.05份、白果18-25份、杏仁1-9份、丝瓜藤15-24份、当归12-35份、铁皮石斛1-8份、淀粉45-63份、维生素E1-4份、天麻24-39份、皇菇1-4份、磷脂酰丝氨酸12-28份、有机硒1-5份、香茅草秸秆25-37份。
作为改进的是,上述负载纳豆激酶的血栓通络药物,包括以下按重量份数计的组分:纳豆激酶0.02份、白果22份、杏仁6份、丝瓜藤21份、当归30份、铁皮石斛7份、淀粉58份、维生素E 3份、天麻35份、皇菇2份、磷脂酰丝氨酸16份、有机硒1.2份、香茅草秸秆30份。
作为改进的是,纳豆激酶的酶活性为1500FU/g。

作为改进的是,步骤4中去离子水的加入量为黄菇的1-3倍。
作为改进的是,步骤5中喷雾干燥的温度为30-40℃。
本发明药物机理如下:
白果,据《本草纲目》记载:“熟食温肺、益气、定喘嗽、缩小便、止白浊;生食降痰、消毒杀虫”。现代科学证明:银杏种仁有抗大肠杆菌、白喉杆菌、葡萄球菌、结核杆菌、链球菌的作用。
杏仁,含有丰富的黄酮类和多酚类成分,这种成分不但能够降低人体内胆固醇的含量,还能显著降低心脏病和很多慢性病的发病危险。
丝瓜藤,舒筋,活血,健脾,杀虫,治腰膝四肢麻木,月经不调,水肿;齿露,鼻渊,牙宣。
当归,补血活血,调经止痛,润肠通便。用于血虚萎黄、眩晕心悸、月经不调、经闭痛经、虚寒腹痛、肠燥便秘、风湿痹痛、跌扑损伤、痈疽疮疡。
铁皮石斛,养阴清热润燥,是治疗糖尿病的专用药。临床研究表明,铁皮石斛不仅可以增强胰岛素活性,同时能显著降低血糖水平,使血正常;可促进循环、扩张血管、降低血胆固醇和甘油三脂。
维生素E,改善脂质代谢,预防冠心病、动脉粥样硬化。
天麻,有镇静、镇痛、抗惊厥作用;能增加脑血流量,降低脑血管阻力,轻度收缩脑血管,增加冠状血管流量;能降低血压,减慢心率,对心肌缺血有保护作用;天麻多糖有免疫活性。
香茅草,含有柠檬醛,有消毒、杀菌与治疗神经痛、肌肉痛的效果,因而香茅草被誉为“消痛剑客”。用它泡茶喝,可提高消化机能,达健胃消脂之功,可减缓筋骨酸痛、腹部绞痛或痉挛,并能处理消化问题;也可治疗腹泻、感冒及发烧、头痛等不适。此外,对于女性也有利尿、防止贫血及润湿皮肤等功效。
与现有技术相比,本发明负载纳豆激酶的血栓通络药物具有活血化瘀、滋补肝肾、增强抵抗力的功效,主治血栓,且成分多为食物,副作用小,成本低,减轻患者的经济负担。

下面通过具体实施例对本发明作进一步详细介绍。
实施例1
一种负载纳豆激酶的血栓通络药物,包括以下按重量份数计的组分:纳豆激酶0.01-0.05份、白果18-25份、杏仁1-9份、丝瓜藤15-24份、当归12-35份、铁皮石斛1-8份、淀粉45-63份、维生素E1-4份、天麻24-39份、皇菇1-4份、磷脂酰丝氨酸12-28份、有机硒1-5份、香茅草秸秆25-37份。
其中,纳豆激酶的酶活性为1500FU/g。
上述负载纳豆激酶的血栓通络药物的制备方法,包括以下步骤:步骤1,称取各组分;步骤2,将白果、杏仁、丝瓜藤、当归、铁皮石斛、天麻、香茅草秸秆投入球磨机中磨碎得粒径为23μm的细粉;步骤3,向细粉内加入其重量3倍的去离子水,文火煎煮3次,每次2h,过滤得第一混合液;步骤4,将皇菇粉碎,再加入等重量的去离子水45℃煎煮5h过滤第二混合液;步骤5,将第一混合液、第二混合液、维生素E、磷脂酰丝氨酸、有机硒、淀粉混合,150rpm的速度下磁力搅拌10min后,30℃下喷雾干燥,即可。
实施例2
一种负载纳豆激酶的血栓通络药物,包括以下按重量份数计的组分:纳豆激酶0.02份、白果22份、杏仁6份、丝瓜藤21份、当归30份、铁皮石斛7份、淀粉58份、维生素E 3份、天麻35份、皇菇2份、磷脂酰丝氨酸16份、有机硒1.2份、香茅草秸秆30份。
其中,纳豆激酶的酶活性为1500FU/g。
上述负载纳豆激酶的血栓通络药物的制备方法,包括以下步骤:步骤1,称取各组分;步骤2,将白果、杏仁、丝瓜藤、当归、铁皮石斛、天麻、香茅草秸秆投入球磨机中磨碎得粒径为35μm的细粉;步骤3,向细粉内加入其重量的4倍的去离子水,文火煎煮4次,每次3h,过滤得第一混合液;步骤4,将皇菇粉碎,再加入其重量的2倍的去离子水54℃煎煮7h过滤第二混合液;步骤5,将第一混合液、第二混合液、维生素E、磷脂酰丝氨酸、有机硒、淀粉混合,380rpm的速度下磁力搅拌14min后,38℃下喷雾干燥,即可。
实施例3
一种负载纳豆激酶的血栓通络药物,包括以下按重量份数计的组分:纳豆激酶0.01份、白果18份、杏仁1份、丝瓜藤15份、当归12份、铁皮石斛1份、淀粉45份、维生素E1份、天麻24份、皇菇1份、磷脂酰丝氨酸12份、有机硒1份、香茅草秸秆25份。
其中,纳豆激酶的酶活性为1500FU/g。
上述负载纳豆激酶的血栓通络药物的制备方法,包括以下步骤:步骤1,称取各组分;步骤2,将白果、杏仁、丝瓜藤、当归、铁皮石斛、天麻、香茅草秸秆投入球磨机中磨碎得粒径为40μm的细粉;步骤3,向细粉内加入其重量的5倍的去离子水,文火煎煮5次,每次4h,过滤得第一混合液;步骤4,将皇菇粉碎,再加入其重量的3倍的去离子水68℃煎煮8h过滤第二混合液;步骤5,将第一混合液、第二混合液、维生素E、磷脂酰丝氨酸、有机硒、淀粉混合,120rpm的速度磁力搅拌15min后,40℃喷雾干燥,即可。
实施例1-3制备的血栓通络药物可以为片剂、胶囊或冲剂。
以片剂为例,用法用量:日服三次、饭后服用,每次1-2粒。
选200名脑血栓患者分为实验组和对照组,实验组用实施例2的药物治疗,对照组用西药常规治疗,结果如下表所示。

典型病例:
张某某,女,80岁,于2017年无诱因的情况下出现下半身行动不便,医院诊断为脑血栓,医院治疗3个月,毫无起色,随着服用实施例2的药物,50天出现症状减轻,90天后症状消失,之后间断性服药加以巩固,至今未复发。
刘某,男,55岁,于2017年因洗澡出现左侧上肢乏力,被诊断为中风,服用实施例2的药物后,40天后手部已可发力,50天后症状小时,停药后,无复发。
另外,本发明不限于上述实施方式,只要在不超出本发明的范围内,可以采取各种方式实施本发明。
Claims (8)
1.一种负载纳豆激酶的血栓通络药物,其特征在于,包括以下按重量份数计的组分:纳豆激酶0.01-0.05份、白果18-25份、杏仁1-9份、丝瓜藤15-24份、当归12-35份、铁皮石斛1-8份、淀粉45-63份、维生素E1-4份、天麻24-39份、皇菇1-4份、磷脂酰丝氨酸12-28份、有机硒1-5份、香茅草秸秆25-37份。
2.根据权利要求1所述的一种负载纳豆激酶的血栓通络药物,其特征在于,包括以下按重量份数计的组分:纳豆激酶0.02份、白果22份、杏仁6份、丝瓜藤21份、当归30份、铁皮石斛7份、淀粉58份、维生素E 3份、天麻35份、皇菇2份、磷脂酰丝氨酸16份、有机硒1.2份、香茅草秸秆30份。
3.根据权利要求1所述的一种负载纳豆激酶的血栓通络药物,其特征在于,纳豆激酶的酶活性为1500FU/g。
4.基于权利要求1所述的一种负载纳豆激酶的血栓通络药物的制备方法,其特征在于,步骤1,称取各组分;步骤2,将白果、杏仁、丝瓜藤、当归、铁皮石斛、天麻、香茅草秸秆投入球磨机中磨碎得细粉;步骤3,向细粉内加入去离子水,文火煎煮3-5次,每次2-4h,过滤得第一混合液;步骤4,将皇菇粉碎,再加入去离子水45-68℃煎煮5-8h过滤第二混合液;步骤5,将第一混合液、第二混合液、维生素E、磷脂酰丝氨酸、有机硒、淀粉混合,磁力搅拌10-15min后,喷雾干燥,即可。
5.根据权利要求4所述的一种负载纳豆激酶的血栓通络药物的制备方法,其特征在于,步骤2中细粉的粒径为23-40μm。
6.根据权利要求4所述的一种负载纳豆激酶的血栓通络药物的制备方法,其特征在于,步骤3中去离子水的加入量为细粉的3-5倍。
7.根据权利要求4所述的一种负载纳豆激酶的血栓通络药物的制备方法,其特征在于,步骤4中去离子水的加入量为黄菇的1-3倍。
8.根据权利要求4所述的一种负载纳豆激酶的血栓通络药物的制备方法,其特征在于,步骤5中喷雾干燥的温度为30-40℃。
