Abstract
本发明公开了一种氨甲环酸致大鼠高凝血状态动物模型及其构建方法。本发明采用氨甲环酸对大鼠进行两次灌胃处理,在2~12小时期间,即可形成高凝血状态动物模型,操作简单,模型成功率高,高凝血状态持续时间长。本发明构建方法操作简单,模型成功率高,高凝血状态持续时间长。本发明成功建立了基于氨甲环酸药物的大鼠高凝血状态动物模型,可以为相关于大鼠高凝血状态研究奠定基础,是一类新型的大鼠高凝血状态动物模型方法。

技术领域
本发明涉及一种氨甲环酸致大鼠高凝血状态动物模型及其构建方法,属于实验动物学领域。
背景技术
高凝血状态又被称作血栓前状态,是指机体呈现出的血液凝固倾向增高的状态。通常认为,高凝血状态的出现与凝血系统凝血机能严重异常有关,原因复杂多样。也有观点认为,高凝血状态可以分为遗传性高凝血状态和获得性高凝血状态。遗传性高凝血症是由于特定的遗传基因突变而导致血栓,发病年龄较低,并且血栓形成的部位比较特殊。就一般病理性分布而言,获得性高凝血状态的发病范围更为广泛。获得性高凝血状态是基于后天因素诱发形成的血液凝固倾向增高的状态。也有观点认为,高凝血状态是很多因素引起的止血、凝血和抗凝系统失调的一种病理过程,具有易导致血栓形成的多种血液学变化。其变化反映在:血管内皮细胞受损或受刺激;血小板和白细胞被激活或功能亢进;凝血因子含量增高或被活化;抗凝因子含量减少或结构异常;纤溶因子含量减少或功能减弱;血液粘度增高或血流减慢(王鸿利,宋善俊.分子标志物在诊断血栓前状态的意义.国外医学输血及血液学分册.1995,18(2):68-72;Piatek C,O’Connell CL,Liebman HA.Treating venousthromboembolism in patients with cancer.Expert Rev Hematol.2012,5(2):201-209;Lyman GH,Khorana AA,Falanga A,et al.American Society of ClinicalOncology.American Society of Clinical Oncology guideline:recommendations forvenous thromboembolism prophylaxis and treatment in patients with cancer.JClin Oncol.2007,25:5490-505;Khorana AA.Venous thromboembolism and prognosisin cancer.Thromb Res.2010,125(6):490-493;Barbosa M.What is the best treatmentfor a cancer patient with thrombosis Clin Med Insights Oncol.2014,8:49–55.)。
多种疾病,如动脉粥样硬化、肿瘤、糖尿病、肾脏综合症等诸多疾病过程也会伴随血脂代谢异常、血管内皮损伤、组织因子和促凝血蛋白水平增高而催生高凝血状态的形成。一些正常的生理过程,如妊娠、口服避孕药等,则可通过影响促凝因子的浓度、活力、半衰期而改变凝血平衡,通过加剧凝血或促进凝血蔓延,诱发高凝血状态的形成(Herzog E,Kaspereit F,Krege W,et al.Correlation of coagulation markers and 4F-PCC-mediated reversal of rivaroxaban in a rabbit model of acutebleeding.Thrombosis Research,2015,135(3):554-560;Geddings JE,Mackman N.Tumor-derived tissue factor-positive microparticles and venous thrombosis in cancerpatients.Blood.2013,122:1873-1880.)。
探究高凝血状态及所对应的疾病标志物,对于人类疾病的研究、预防和检测也有着重要的意义。当前,血纤维蛋白原(Fib)和纤维蛋白二聚体(D-dimer)已经被用于对深静脉血栓和肺栓塞进行排查,但直接针对动脉血栓形成的实验室诊断缺乏共识(BattistoniA,Rubattu S,Volpe M.Circulating biomarkers with preventive,diagnostic andprognostic implications in cardiovascular diseases.International Journal ofCardiology.2012,157,160-168)。但以上参考标准,严格意义上而言,应该算作是血栓形成后的生物标志物,并不能视作严格意义上的高凝血状态的生物标志物。血栓出现的原因、产生的部位及影响因素,是极为复杂的。目前来看,还没有真正意义上的高凝血状态生物标志物。有观点认为,有可能多种生物标志物分子可以单独或一起出现在高凝血状态。对于高凝血状态生物标志物的研究,目前尚处于探索研究阶段。高凝血状态的相关研究,其基础是首先建立高凝血状态动物模型。迄今为止,尚没有真正意义上的高凝血动物模型。对于血栓病理方面的相关研究,目前学术界基本上应用的是形成血栓的动物模型,即血栓形成是目的,且明确动物模型机体内已经形成血栓。目前,已知的比较成熟的血栓动物模型主要有以下几类,它们均是血栓构成模型,即动物体内已经形成血栓。
1、机械损伤法静脉血栓动物模型。其原理是用刮匙搔刮损伤血管内膜,使内皮下细胞外基质(ECM)裸露,促使血小板与ECM(主要是胶原纤维)接触而被激活和粘附。同时裸露的胶原纤维激活XII因子,损伤的内皮细胞释放出组织因子,启动了内源性和外源性凝血过程,导致血栓形成。此法依据血栓形成基本原理,在动物体内形成血栓,方法简便、易行,成功率100%,术后24h即开始有血栓形成。此方法主要适用于:1)动态观察血栓形成过程中血栓形态及血栓变化规律的研究。2)评价药物溶栓作用的实验研究。3)与形成和溶解血栓有关的其他实验研究。此法在24h内即可成栓,属于比较快的一种成栓动物模型。但确定的成栓时刻是无法准确预计或者估算的(陈铁军,陈瑞芳,杨果杰.一种家兔股动脉血管内制作血栓模型的简便方法.实验动物科学与管理.1997,14(2):33-34)。
2、股动脉异物法血栓动物模型。其原理是于股动脉内放置螺旋钢丝线圈,造成动脉内膜损伤,局部血流动力学改变,激活凝血系统,促进血小板凝集并释放一系列活性物质,致血栓形成。此法可靠易行,重复性好,血栓由血小板、白细胞及纤维蛋白构成。目前多用于血栓放射免疫显像方面的研究(万卫星.血栓动物模型的99Tcm-重组水蛭素显像研究.中华核医学与分子影像杂志.1999,19(4):201-203)。
3、电流损伤法血栓动物模型。原理:1.5mA直流电刺激颈动脉壁可使血管损伤,血管内膜变得粗糙不平,引起血小板粘附聚集及释放一系列活性物质,激活内源性凝血系统,同时受损内膜也可激活外源性凝血系统,于是动脉管腔内逐渐形成肉眼可见的混合血栓。此实验中形成的血栓成分在电刺激区以血小板及纤维蛋白为主,有少量红细胞及白细胞,而在电刺激远端以纤维蛋白及红细胞为主,所以1.5mA直流电刺激动脉壁形成的血栓在形态结构上与人类动脉血栓相似,有较好的可比性,适用于抗血栓形成药物的筛选。此模型操作简单,但受动物年龄、环境温度等的影响较大,标准差较大,因而应用时有一定局限性(张丽萍,牟善初,余霞君.蚓激酶对实验性血栓的预防作用.中国循环杂志.1995,10(1):679-680)。
4、结扎法血栓动物模型。原理:粗丝线结扎下腔静脉,引起局部血流淤滞、缺氧,导致血管内皮细胞损伤,启动内源性凝血系统。此外,粘聚的血小板与局部形成的凝血因子也因血流受阻停留在局部,致使血栓形成。一般在结扎后2h血栓形成率约为60%~80%,此时主要观察血栓形成百分率。6h后血栓形成率为100%,此时应观察血栓质量。此方法多用于判定溶栓药物的体内抗血栓作用(Reyer L.Failure of asprin at different doses tomodify experimental thrombosis in rat.Thromb Res.1980,18:669;陈奇.中药药理研究方法学.北京:人民卫生出版社,1993.511)。
5、胰蛋白酶血栓形成法动物血栓模型。原理:胰蛋白酶具有蛋白水解作用,使血管内膜和部分肌层脱落,血小板粘附、聚集,致血栓形成。所形成的血栓富含血小板和纤维蛋白,类似于临床上动脉血栓的形成。方法可靠,重复性好。用于抗血栓药物的筛选和研究(郭丹.中国黑眼镜蛇毒蛋白酶natrahagin抑制血小板凝集和动脉血栓形成的作用.中国药理学与毒理学杂志.2001,15(1):27-30;Imura Y,Terashita Z,Nishikawa K.The roleofthromboxane(TX)A2in rabbit arterial thrombosis induced by endothelialdamage.Thromb Res.1990,59(1):195-205)。
6、过氧化氢(H2O2)血栓形成法动物血栓模型。原理:过氧化氢损伤血管内膜,引起血小板粘附聚集,致血栓形成。所形成的血栓以白色血栓为主,符合动脉血栓的特征,可以用来探索防止血栓形成的方法(谢协驹.降低红细胞压积对家兔颈总动脉血栓形成的影响.中国病理生理学杂志.1995,5(2):198-200)。
7、角叉菜胶血栓形成法动物血栓模型。原理:角叉菜胶是从海藻中提取的含硫酸多糖物质,是一种较强的致炎物质,其诱发的尾动脉局部血栓与血管内炎症密切相关,炎症时释放的大量炎症介质,如白介素-1、肿瘤坏死因子等可破坏正常内皮细胞维持凝血与纤溶之间的平衡作用,从而促发血栓形成。此外,内皮细胞损伤时舒血管物质如乙酰胆碱等分泌减少,缩血管物质如内皮素等增多,使局部血管痉挛,加重局部血管缺血缺氧状况,进一步促进血栓形成及血管内皮损伤。
24h实验动物尾部小静脉和毛细血管内出现大量纤维素和少量白细胞及血小板,血管内皮细胞核浓缩呈骰子状,血管内壁中性粒细胞靠边。72h可见较大血管呈炎症改变,伴有混合性血栓形成。由于血栓发生在尾部,界限明显,可从体表观察血栓形成出现的时间与发展过程,测量其波及的范围或长度,可为研究体内血栓形成全过程提供一个简便、直观的动物模型(胡三觉.一种新的体内血栓形成动物模型.中华血液杂志.1993,14(10):541-542;Rosa M D.Biological properties of carrageenan.J Pharm Pharmacol,1972,24:89-102)。
8、三氯化铁(FeCl3)血栓形成法动物血栓模型。原理:铁离子侵入血管壁造成血管内膜损伤,血小板粘附聚集,血栓形成。方法简单、可靠,重复性好,形成的动脉血栓成分为血小板、红细胞及纤维蛋白等,为混合血栓。而且此方法仍保留有大脑中动脉,可进行血管观察,较接近于临床,且此模型既能反映抗栓药又能反映溶栓药的药效,用已知的抗栓药物阿司匹林和溶栓药物尿激酶均验证了这一点。该模型还可以利用TTC染色技术直观地观察脑梗死程度。其不足之处是动物的颧弓被去除,影响动物进食,不利于作长期观察等,此法还有待改进(刘小光,徐理纳.一种能评价溶栓和抗栓药的大鼠大脑中动脉血栓模型.药学学报.1995,30;662-667;张玉珍.银杏叶提取物(EGB)对大鼠局部脑缺血及颈静脉血栓形成的保护作用.药学学.1998,33(12):901-905)。
9、光化学法动物血栓动物模型。原理:利用虎红B(四碘四氯荧光素钠)在单色绿光下产生并释放单态氧,使血管内皮细胞膜上的多不饱和脂肪酸发生脂质过氧化反应,不仅使细胞膜的通透性改变,Ca2+内流增加,而且脂质过氧化物增加可破坏前列环素(PGI2)与血栓素B2(TXB2)的平衡,促进血小板聚集与粘附,激活凝血过程,形成血栓并进一步阻塞血管导致缺血;同时凝血过程中产生大量血管活性物质及神经毒性物质,破坏脑细胞尤其是血-脑脊液屏障,与人类脑梗死发生极为相似。
此模型成功率100%,模型稳定、可靠,造模时采用分离颞肌、拉开下颌关节,保留术后鼠的咀嚼功能,提高动物存活率。采用分步照射法,在2次注射光敏剂时剂量减半,既可避免动物损伤程度过重,又可制备出较实用、可靠的大脑中动脉血栓模型。由于该模型的形成过程与人类脑血栓形成发病机制、血栓形成过程、损害程度较为相近,加之光照或光敏剂对血管本身无机械性的损害,因而可广泛用于脑血管病的基础与临床研究,以及康复医学的实验研究(杨文清.绿光光照诱导大鼠脑血栓模型后脑组织病理与行为学改变的研究.现代康复.2001,5(4):38-39)。
血栓风险推升了心血管事件、肿瘤等多种疾病的致死率和致残率。如何准确筛查、及早发现和预防血栓风险,具有非常重要的医疗和科学意义。对于血栓形成的预测、筛查目前尚不能有效满足临床需要。大量有关血栓形成的研究,迫切需要合适的高凝血状态动物模型。目前的血栓动物模型,不能满足此类研究的需要,不具备此列研究所需的各项条件。目前对于高凝血状态动物模型,没有既定的现成模型系统可资使用。
发明内容
本发明的目的是提供一种高凝血状态动物模型及其构建方法;本发明采用氨甲环酸进行干预,经氨甲环酸作用后的大鼠的活化部分凝血活酶时间(APTT)和凝血时间(CT)均显著缩短(p<0.01),成功建立了基于氨甲环酸药物的大鼠高凝血状态动物模型,为相关于高凝血状态研究奠定了重要的基础。
本发明所涉及的“高凝血状态”指的是通过抑制纤溶酶活性所导致的高凝态。
本发明所涉及的氨甲环酸,化学名称为4-(氨甲基)环己甲酸,又名为凝血酸、传明酸、止血环酸,分子式为C8H15NO2,分子量为157.21,其分子结构示意图如图1所示;氨甲环酸能与纤溶酶和纤溶酶原上的纤维蛋白亲和部位的赖氨酸结合部位(LBS)强烈吸附,阻抑了纤溶酶、纤溶酶原与纤维蛋白结合,从而强烈地抑制了由纤溶酶所致纤维蛋白分解,从而达到血液高凝态。
本发明所提供的高凝血状态动物模型是以氨甲环酸作为诱导剂。
本发明提供的所述高凝血状态动物模型的构建方法,包括如下步骤:
采用氨甲环酸对动物进行灌胃处理;
所述动物优选大鼠。
上述的构建方法中,所述灌胃处理的剂量可为250~315mg/kg,优选315mg/kg(按照如下方式得到:人类一次口服3g,换算到大鼠的用量:3000mg×6.3/60kg=315mg/kg)。
上述的构建方法中,进行两次所述灌胃处理。
上述的构建方法中,两次所述灌胃处理之间的时间间隔可为0~6h,但不为零,优选为6。
上述的构建方法中,按照优选灌胃时间间隔(6h)和剂量(315mg/kg),首次所述灌胃处理后2~12h期间能够维持高凝血状态,即形成了高凝血状态动物模型。
上述的构建方法中,所述大鼠可为SPF级大鼠,如SPF级雄性Wistar大鼠;
所述大鼠的质量可为180~220g;
所述灌胃处理之前,适应性饲养所述大鼠,可饲养一周的时间。
氨甲环酸在构建高凝血状态动物模型或制备用于构建高凝血状态动物模型的产品中的应用也属于本发明的保护范围。
本发明具体实施方式采用活化部分凝血活酶时间(APTT)和凝血时间(CT)两个指标评估大鼠的凝血状态,以验证大鼠高凝血模型是否成功构建。
本发明基于APTT和CT指标检测结果,明确地表明了高凝血状态的形成过程,明确了大鼠高凝血状态的高峰期。通过对给药次数、剂量、给药时间设定等条件的摸索,明确了氨基己酸条件下大鼠动物模型高凝血状态维持特征。
本发明具体实施方式采用大鼠内眦静脉采血,用于凝血分析。
本发明提供了一种新型的高凝血状态动物模型,采用氨甲环酸进行干预,经两次灌胃处理,2~12h期间造模成功,操作简单,模型成功率高,高凝血状态持续时间较长(第12小时检测结果如附图4所示)。本发明成功建立了基于氨甲环酸药物的大鼠急性高凝血状态动物模型,可以为相关于大鼠高凝血状态研究奠定基础,是一类新型的大鼠高凝血状态动物模型方法。

具体实施方式
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
下述实施例中采用活化部分凝血活酶时间(Activated Partial ThromboplastinTime,APTT)和凝血时间(Clotting Time,CT)评估大鼠的凝血状态,以确定大鼠高凝血模型是否成功建立,具体测定方法如下:
1、活化部分凝血活酶时间的测定
大鼠内眦静脉采血后,按照血液:柠檬酸钠抗凝剂=9:1(体积比)的比例加入柠檬酸钠抗凝剂,上下轻柔颠倒混匀5~6次,离心:4℃,2500g,15min,离心所得上清液即为血浆。取100μL血浆用于部分凝血活酶时间的测定。
测定方法遵循试剂盒的产品说明书:37℃温浴100μL鞣花酸试剂(不超过15min),然后加入100μL待测血浆,37℃温浴5min(不少于5min),之后加入37℃预热的0.5mol/LCaCl2溶液100μL,混匀并开始计时。当出现絮状沉淀时立即停止计时,即为活化部分凝血活酶时间。
2、凝血时间测定
采用毛细玻璃管法测定凝血时间。大鼠内眦静脉采血后,立即将25μL未加抗凝剂的静脉血吸入毛细玻璃管中,开始计时。计时过程中,与水平面呈±60°角度来回颠倒毛细玻璃管,直到玻璃管内的血液不再来回移动,停止计时,即为凝血时间。
实施例1、构建大鼠高凝血状态动物模型
1、实验材料与试剂
(1)实验动物
SPF级雄性Wistar大鼠,180~220g,购自北京维通利华实验动物技术有限公司,标准环境下适应性饲养一周后开始实验。
(2)试剂
所用试剂如表1中所示。

2、实验步骤
雄性wistar大鼠20只,随机等分成两组,每组10只,称重。实验组大鼠灌胃氨甲环酸(315mg/kg),对照组灌胃相应体积生理盐水。6h一次,共2次。将第一次灌胃时间记为0h,第二次灌胃时间则为6h。第一次灌胃后12h,立即内眦静脉采血,并检测大鼠的凝血功能,确定高凝血状态大鼠动物模型形成规格(流程图如图2所示)。
通过APTT和CT两个重要指标来评估大鼠凝血功能的改变。APTT,即活化部分凝血活酶时间,是通过对内源性凝血的全部条件进行体外模拟,测定血浆凝固的时间,从而反映出内源凝血因子是否正常。CT,即凝血时间,是指血液在体外发生凝固所需的时间,用于检测内源性凝血途径中,各种凝血因子的数量和功能正常与否,同时还可检测是否有多余的抗凝物质。血液处于高凝状态时,APTT和CT均表现明显缩短(p<0.01),如图4所示。
本实验室基于此高凝血状态大鼠模型,通过收集上述过程中大鼠尿液,用于蛋白质组学的相关研究。
3、实验结果
通过上述实验步骤,经氨甲环酸作用之后,如图4所示的大鼠的活化部分凝血活酶时间(APTT)和凝血时间(CT)检测结果显示,Wistar大鼠的APTT和CT均显著缩短(p<0.01),表明灌胃氨甲环酸使大鼠凝血功能改变,血液处于高凝血状态,高凝态模型建模成功。
本发明构建的高凝血状态动物模型的大鼠高凝血状态时间能够持续至少12个小时。
本发明还摸索了单次给药的时间对APTT平均值和CT平均值的影响,结果分别如图3(A)和图3(B)所示,图3(A)显示,单次氨甲环酸灌胃给药后,2~6小时期间APTT明显下降,且维持一个比较稳定的平台期,随后开始逐渐恢复正常;图3(B)结果显示,单次氨甲环酸灌胃给药后,2~6小时期间,CT明显呈下降趋势,但6~8小时,基本一致,无明显变化。通过以上数据,2~6小时期间,表现出高凝血状态平台期。
按照上述实验步骤,本发明还摸索了其它剂量,采用250mg/kg体重灌胃剂量,由结果可知,采用250mg/kg体重灌胃时,wistar大鼠也可在灌胃处理后2~6h达到高凝血状态,但是该高凝血状态相关指标不如315mg/kg体重灌胃剂量获得的高凝血状态大鼠模型有效。所以采用的优选剂量为315mg/kg。此时,高凝血状态明显,同时,大鼠生命体征外观无明显异常。
本发明在有效的氨甲环酸给药的情况下,能够保证在2小时即开始形成高凝血状态,在2~12小时内,APTT值和CT值都保持在一个较高的高峰平台期,12小时后,相关APTT和CT检测指标开始恢复正常状态。这个有效的高峰平台期,为开展针对高凝血状态相关基础性科学研究,提供了可能。比如,基于此动物模型,可以开展相关尿液蛋白质组学及相关生物标志物的研究工作。
本实施例表明,本发明方法成功建立了基于氨甲环酸药物的大鼠高凝血状态动物模型,可以为相关于大鼠高凝血状态研究奠定重要的基础,是一类新型的大鼠高凝血状态动物模型方法。
Claims (1)
1.一种高凝血状态动物模型的构建方法,包括如下步骤:
采用氨甲环酸药物对SPF级大鼠进行两次灌胃给药;
所述大鼠质量一般为180~220 g;
所述灌胃给药的剂量为315mg/kg;
两次所述灌胃给药之间的时间间隔为6小时;
大鼠高凝血状态在首次所述灌胃给药处理后2~12小时期间维持。
