技术领域
本发明涉及鱼类产品深加工技术领域,尤其是涉及基于海鲈鱼鳍活性多肽的复配物。
背景技术
功能性食品是指具有营养功能、感觉功能和调节生理活动功能的食品。其研究与开发在我国尚属新兴的研究学科和开发领域,涉及营养学、药学、生理学、预防医学、生物工程、食品科学等多个学科,是多领域、多学科不断交叉融合的产物。另外,功能性食品在人们的社会生活中发挥了越来越重要的作用。海洋生物构成了全球约一半的多样性生物,是不同结构的生物功能成分的良好来源。因此海洋生物是被认为是提供新的具有生物活性的胶原蛋白及其水解物的一个潜在的来源。近年来,海洋胶原蛋白越来越受人们关注,它广泛应用于加工功能性食品、化妆品、生物医学等领域。每年有大量的海洋加工副产物,包括不合格的肌肉、内脏、头、皮、鳍、骨头等废弃物产生。些副产物通常或者丢弃,或者用于动物饲料或肥料。海洋加工副产物往往含有价值高的功能性成分包括胶原蛋白,丢弃处理不但污染了环境,而且提高了成本。因此,需要考虑综合利用这些废料,为其提供解决方案。
海鲈鱼(学名)又称七星鲈、花鲈、青鲈、日本花鲈、日本真鲈,为辐鳍鱼纲鲈形目真鲈科花鲈属,分布于西太平洋,我国沿海均产。海鲈鱼是广东省产量最高的海洋经济鱼,在其加工过程中会产生大量鱼鳍、鱼皮、鱼头、鱼骨等副产物,此类副产物中含有丰富的胶原蛋白,提取的量大,可作为制备胶原蛋白肽的理想原料,为食品、保健品的加工提供优良原辅料。
现有技术如授权公告号为CN 103239705 B的中国发明专利,公开了一种鳄鱼胶原蛋白抗菌肽的组合物及其制备方法,提供一种对关节炎等骨关节疾病治疗方面具有良好的疗效的组合物,其技术要点是:该组合包括下述质量百分比的原料:鳄鱼胶原蛋白抗菌肽1-90%,D-氨基葡萄糖1-50%,硫酸软骨素1-50%,各组分之和为100%。该组合物在改善关节炎等骨关节疾病治疗方面具有良好的疗效,为骨关节疾病的治疗提供了新途径,但制备方法酶解不彻底,得率低,口感较差。
发明内容
本发明的目的在于提供一种营养丰富全面,口感爽滑,易于吸收,无化学防腐剂,无毒副作用,能抑制或者杀伤肿瘤细胞,增强人体免疫力的基于海鲈鱼鳍活性多肽的复配物。
本发明针对上述技术中提到的问题,采取的技术方案为:基于海鲈鱼鳍活性多肽的复配物,复配物的成分及其重量份为:海鲈鱼鳍活性多肽3~6份、胶原蛋白18~24份、肉苁蓉多糖5~11份、枸杞多糖1~4份、羟甲基纤维素5~11份、微量元素0.1~0.3份、维生素E 0.8~1.4份、果粉4~8份、水38~45份。上述复配物的复配物的营养丰富全面、易于吸收,无腥苦味、口感爽滑,无化学防腐剂,无毒副作用,任何消费者均可饮用;能清除机体内自由基,减少自由基对机体的攻击,能激活超氧化物歧化酶的活性,加强分解过多的过氧化物,从而对机体起到保护和防止老化的作用。
基于海鲈鱼鳍活性多肽的复配物,复配物的制备方法包括以下步骤:
脱钙:将鱼鳍粉加入浓度为8~10%的柠檬酸溶液里,超声波震荡脱钙50~60min,料液的重量体积比为1:14~16,用水冲洗,过滤,重复2~4次,烘干,备用,由于鱼鳍中的胶原纤维与羟基磷灰石紧密粘结,因此在提取鱼鳍活性物前应进行脱钙处理,以利于胶原蛋白的溶出,该步骤利用超声波震荡,可大大减少脱钙时间,提高脱钙效率;
脱杂蛋白:将脱钙后的鱼鳍粉放入0.1~0.2mol/L的NaOH溶液浸泡5~8h,料液的重量体积比为1:7~10,用水冲洗,过滤,重复2~4次,烘干,备用,该步骤能去除鱼鳍粉中的非胶原蛋白,提高胶原蛋白的含量,增加活性提取物的纯度;
超声波酶解:将脱杂蛋白鱼鳍粉加水调至底物浓度为5~7%,调节pH至8~9,加入700~900U/g的胰蛋白酶,放入超声波中,在超声功率为200~220W,温度为35~40℃下酶解2~3h,灭酶,取出后经6000~8000r/min、3~5℃的高速冷冻离心机离心15~25min,上清液即为酶解液,结合超声波和蛋白酶酶解,能有效的从鱼鳍粉中提取出高活性的活性多肽,能明显增加蛋白质的酶解速度,不仅不会破坏活性多肽的成分和活性,而且能改善活性多肽的品质,优化酶解条件,操作简单;
纯化:用截留分子量为3~5KDa的超滤膜在0.7~1MPa压力下超滤分离,真空浓缩至固含为23~27%,喷雾干燥,即得海鲈鱼鳍活性多肽,该活性多肽的氨基酸序列为HSHACKLCVCVNCFCAKCYLCRVLHPGKLCVCNCSK,该活性多肽一方面能清除机体内自由基,减少自由基对机体的攻击,减轻组织或细胞的过氧化损伤,减少脂类过氧化物的形成,减少体内脂褐质的堆积,提高胶原纤维含量,延缓衰老,另一方面能激活超氧化物歧化酶的活性,加强分解过多的过氧化物,从而对机体起到保护和防止老化的作用;
复配物配制:按配方称取海鲈鱼鳍活性多肽、胶原蛋白、肉苁蓉多糖、枸杞多糖、羟甲基纤维素、微量元素、维生素E、果粉、水,混合均匀,灌装,灭菌即得海鲈鱼鳍活性多肽的复配物,该复配物的营养丰富全面、易于吸收,无腥苦味、口感爽滑,无化学防腐剂,无毒副作用,任何消费者均可饮用,能清除机体内自由基和激活超氧化物歧化酶的活性,对机体起到保护和防止老化的作用。
与现有技术相比,本发明的优点在于:1)本发明基于海鲈鱼鱼鳍活性多肽的复配物的营养丰富全面、易于吸收,无腥苦味、口感爽滑,无化学防腐剂,无毒副作用,任何消费者均可饮用;2)该复配物的能清除机体内自由基,减少自由基对机体的攻击,能激活超氧化物歧化酶的活性,加强分解过多的过氧化物,从而对机体起到保护和防止老化的作用;3)制备方法结合超声波和蛋白酶进行酶解,增加蛋白质的酶解速度,改善活性多肽的品质,优化酶解条件,操作简单,提高海鲈鱼鱼鳍的资源利用率,具有较大的经济效益和社会效益。
具体实施方式
下面通过实施例对本发明方案作进一步说明:
实施例1:
基于海鲈鱼鳍活性多肽的复配物,复配物的成分及其重量份为:海鲈鱼鳍活性多肽5份、胶原蛋白21份、肉苁蓉多糖7份、枸杞多糖3份、羟甲基纤维素7份、微量元素0.15份、维生素E 0.9份、果粉5份、水39份。
基于海鲈鱼鳍活性多肽的复配物,复配物的制备方法包括以下步骤:
1)脱钙:将鱼鳍粉加入浓度为9%的柠檬酸溶液里,超声波震荡脱钙55min,料液的重量体积比为1:15,用水冲洗,过滤,重复3次,在78℃的鼓风烘箱内烘干至恒重,备用,由于鱼鳍中的胶原纤维与羟基磷灰石紧密粘结,因此在提取鱼鳍活性物前应进行脱钙处理,以利于胶原蛋白的溶出,该步骤利用超声波震荡,可大大减少脱钙时间,提高脱钙效率;
2)脱杂蛋白:将脱钙后的鱼鳍粉放入0.13mol/L的NaOH溶液浸泡7h,料液的重量体积比为1:7,用水冲洗,过滤,重复3次,在78℃的鼓风烘箱内烘干至恒重,备用,该步骤能去除鱼鳍粉中的非胶原蛋白,提高胶原蛋白的含量,增加活性提取物的纯度;
3)超声波酶解:将脱杂蛋白鱼鳍粉加水调至底物浓度为6%,调节pH至8.5,加入900U/g的胰蛋白酶,放入超声波中,在超声功率为210W,温度为37℃下酶解2.5h,100℃下灭酶20min,取出后经7000r/min、3℃的高速冷冻离心机离心17min,上清液即为酶解液,结合超声波和蛋白酶酶解,能有效的从鱼鳍粉中提取出高活性的活性多肽,能明显增加蛋白质的酶解速度,不仅不会破坏活性多肽的成分和活性,而且能改善活性多肽的品质,优化酶解条件,操作简单;
4)纯化:用截留分子量为3KDa的超滤膜在0.9MPa压力下超滤分离,真空浓缩至固含为23~27%,喷雾干燥,即得海鲈鱼鳍活性多肽,该活性多肽的氨基酸序列为HSHACKLCVCVNCFCAKCYLCRVLHPGKLCVCNCSK,该活性多肽一方面能清除机体内自由基,减少自由基对机体的攻击,减轻组织或细胞的过氧化损伤,减少脂类过氧化物的形成,减少体内脂褐质的堆积,提高胶原纤维含量,延缓衰老,另一方面能激活超氧化物歧化酶的活性,加强分解过多的过氧化物,从而对机体起到保护和防止老化的作用;
5)复配物配制:按配方称取海鲈鱼鳍活性多肽、胶原蛋白、肉苁蓉多糖、枸杞多糖、羟甲基纤维素、微量元素、维生素E、果粉、水,在500rpm的均质机下搅拌混合均匀,灌装,密封,100℃下灭菌18min,即得海鲈鱼鳍活性多肽的复配物,该复配物的营养丰富全面、易于吸收,无腥苦味、口感爽滑,无化学防腐剂,无毒副作用,任何消费者均可饮用,能清除机体内自由基和激活超氧化物歧化酶的活性,对机体起到保护和防止老化的作用。
实施例2:
基于海鲈鱼鳍活性多肽的复配物,复配物的成分及其重量份为:海鲈鱼鳍活性多肽4份、胶原蛋白20份、肉苁蓉多糖8份、枸杞多糖2份、羟甲基纤维素6份、微量元素0.2份、维生素E 0.8份、果粉6份、水40份。
基于海鲈鱼鳍活性多肽的复配物,复配物的制备方法包括以下步骤:
1)脱钙:将鱼鳍粉加入浓度为8%的柠檬酸溶液里,超声波震荡脱钙50min,料液的重量体积比为1:14,用水冲洗,过滤,烘干,备用;
2)脱杂蛋白:将脱钙后的鱼鳍粉放入0.2mol/L的NaOH溶液浸泡6h,料液的重量体积比为1: 10,用水冲洗,过滤,烘干,备用;
3)超声波酶解:将脱杂蛋白鱼鳍粉加水调至底物浓度为7%,调节pH至9,加入900U/g的胰蛋白酶,放入超声波中,在超声功率为220W,温度为40℃下酶解2h,灭酶,取出后经8000r/min、5℃的高速冷冻离心机离心20min,上清液即为酶解液,结合超声波和蛋白酶酶解,能有效的从鱼鳍粉中提取出高活性的活性多肽,能明显增加蛋白质的酶解速度,不仅不会破坏活性多肽的成分和活性,而且能改善活性多肽的品质,优化酶解条件,操作简单;
4)纯化:用截留分子量为4KDa的超滤膜在0.8MPa压力下超滤分离,真空浓缩至固含为24%,喷雾干燥,即得海鲈鱼鳍活性多肽,该活性多肽的氨基酸序列为HSHACKLCVCVNCFCAKCYLCRVLHPGKLCVCNCSK,该活性多肽一方面能清除机体内自由基,减少自由基对机体的攻击,减轻组织或细胞的过氧化损伤,减少脂类过氧化物的形成,减少体内脂褐质的堆积,提高胶原纤维含量,延缓衰老,另一方面能激活超氧化物歧化酶的活性,加强分解过多的过氧化物,从而对机体起到保护和防止老化的作用;
5)复配物配制:按配方称取海鲈鱼鳍活性多肽、胶原蛋白、肉苁蓉多糖、枸杞多糖、羟甲基纤维素、微量元素、维生素E、果粉、水,混合均匀,灌装,灭菌即得海鲈鱼鳍活性多肽的复配物,该复配物的营养丰富全面、易于吸收,无腥苦味、口感爽滑,无化学防腐剂,无毒副作用,任何消费者均可饮用,能清除机体内自由基和激活超氧化物歧化酶的活性,对机体起到保护和防止老化的作用。
实施例3:
基于海鲈鱼鳍活性多肽的复配物,复配物的成分及其重量份为:海鲈鱼鳍活性多肽6份、胶原蛋白19份、肉苁蓉多糖9份、枸杞多糖3份、羟甲基纤维素8份、微量元素0.2份、维生素E 1份、果粉7份、水41份。
基于海鲈鱼鳍活性多肽的复配物,复配物的制备方法包括以下步骤:
将鱼鳍粉以料液的重量体积比为1:15的比例加入浓度为9%的柠檬酸溶液里,超声波震荡脱钙55min,用水冲洗,过滤,烘干,然后将脱钙后的鱼鳍粉以料液的重量体积比为1:9的比例放入0.2mol/L的NaOH溶液浸泡5h,用水冲洗,过滤,烘干,随后将脱杂蛋白鱼鳍粉加水调至底物浓度为6%,调节pH至8.5,加入880U/g的胰蛋白酶,在超声功率为200W,温度为38℃下酶解2.5h,灭酶,离心,将上清液用截留分子量为5KDa的超滤膜在1MPa压力下超滤分离,真空浓缩至固含为23~27%,喷雾干燥得海鲈鱼鳍活性多肽,最后按配方称取海鲈鱼鳍活性多肽、胶原蛋白、肉苁蓉多糖、枸杞多糖、羟甲基纤维素、微量元素、维生素E、果粉、水,混合均匀,灌装,灭菌即得海鲈鱼鳍活性多肽的复配物,该复配物的营养丰富全面、易于吸收,无腥苦味、口感爽滑,无化学防腐剂,无毒副作用,任何消费者均可饮用,能清除机体内自由基和激活超氧化物歧化酶的活性,对机体起到保护和防止老化的作用。

1.基于海鲈鱼鳍活性多肽的复配物,其特征在于:所述的复配物的成分及其重量份为:海鲈鱼鳍活性多肽3~6份、胶原蛋白18~24份、肉苁蓉多糖5~11份、枸杞多糖1~4份、羟甲基纤维素5~11份、微量元素0.1~0.3份、维生素E 0.8~1.4份、果粉4~8份、水38~45份。
2.根据权利要求1所述的基于海鲈鱼鳍活性多肽的复配物,其特征在于:所述的海鲈鱼鳍活性多肽的氨基酸序列为HSHACKLCVCVNCFCAKCYLCRVLHPGKLCVCNCSK。
3.根据权利要求1所述的基于海鲈鱼鳍活性多肽的复配物,其特征在于:所述的复配物的制备方法包括以下步骤:
1)脱钙:将鱼鳍粉加入柠檬酸溶液里,超声波震荡脱钙,冲洗,过滤,烘干;
2)脱杂蛋白:将脱钙后的鱼鳍粉放入NaOH溶液浸泡,冲洗,过滤,烘干;
3)超声波酶解:将脱杂蛋白的鱼鳍粉加水混合,加入胰蛋白酶,放入超声波中酶解,灭酶,离心,上清液即为酶解液;
4)纯化:超滤分离,真空浓缩,喷雾干燥,即得海鲈鱼鳍活性多肽;
5)复配物配制:按配方称取海鲈鱼鳍活性多肽、胶原蛋白、肉苁蓉多糖、枸杞多糖、羟甲基纤维素、微量元素、维生素E、果粉、水,混合均匀,灌装,灭菌即得海鲈鱼鳍活性多肽的复配物。
4.根据权利要求3所述的基于海鲈鱼鳍活性多肽的复配物的制备方法,其特征在于:所述的脱钙步骤中柠檬酸溶液的浓度为8~10%,料液的重量体积比为1:14~16,超声波震荡脱钙50~60min。
5.根据权利要求3所述的基于海鲈鱼鳍活性多肽的复配物的制备方法,其特征在于:所述的脱杂蛋白步骤中NaOH溶液的浓度为0.1~0.2mol/L,料液的重量体积比为1:7~10,浸泡时间为5~8h。
6.根据权利要求3所述的基于海鲈鱼鳍活性多肽的复配物的制备方法,其特征在于:所述的超声波酶解步骤中底物浓度为5~7%,胰蛋白酶的加入量为700~900U/g,酶解pH为8~9。
