Background
Rhizoma Polygonati is dried rhizome of Polygonatum kingianum, Polygonatum sibiricum Red or Polygonatum cyrtonema of Liliaceae. According to different shapes, it is called "rheum officinale essence", "polygonatum kingianum" and "polygonatum zingiberense". Mainly produced in Guizhou, Hunan, Hubei, Sichuan and Anhui. Collected in spring and autumn, and cleaned after removing fibrous roots. The sealwort is sweet in nature and tasty and refreshing to eat. The fleshy rootstock is fat and thick, contains a large amount of starch, sugar, fat, protein, carotene, vitamins and various other nutritional ingredients, can allay hunger when eaten raw or stewed, has the function of body building, can multiply the vitality of people, has abundant muscles and strong bone marrow, and is very beneficial to the body. Rhizoma polygonati rhizome is shaped like sweet potato, and is often eaten as vegetable by people in mountainous areas. The property of the medicine is sweet and neutral. It enters spleen, lung and kidney meridians. Has the functions of invigorating vital energy, nourishing Yin, invigorating spleen, moistening lung and benefiting kidney. The method is mainly applied to: deficiency of spleen-stomach qi, lassitude, hypodynamia, deficiency of stomach yin, dry mouth and poor appetite; cough due to lung deficiency, cough with blood; deficiency of essence and blood, soreness and weakness of the waist and knees, early white beard and hair, internal heat and diabetes. It has a variety of pharmacological effects: resisting pathogenic microorganism, lowering blood sugar, relieving fatigue, resisting oxidation, delaying aging, stopping bleeding, treating cardiovascular diseases, enhancing anoxia tolerance, and resisting virus.
Steaming rhizoma polygonati: cleaning rhizoma Polygonati, steaming repeatedly until the inside and outside are moist black, slicing into thick pieces, and drying. After steaming, the spicy flavor can be removed, and the stimulation to the throat can be avoided, so that the effects of tonifying the spleen, moistening the lung and tonifying the kidney are mainly achieved, but the effect of nourishing and greasing the spleen is achieved. Can be used for: cough due to lung deficiency, weakness of spleen and stomach, deficiency of kidney essence, and diabetes due to yin deficiency.
The fresh rhizoma Polygonati is directly harvested, removed of fibrous root, cleaned and used. The fresh rhizoma polygonati has the main function of nourishing yin and moistening lung. In the past, steamed sealwort is mainly used, while fresh sealwort is rarely used, and the reason is that the throat of a human body can be stimulated by directly taking the fresh sealwort. The fresh fructus Mori is dry fruit ear of Mori fructus of Moraceae. Mainly produced in Jiangsu, Zhejiang, Hunan and Sichuan provinces. Collected when the fruit turns red in 4-6 months. Its properties are sweet, sour and cold. It enters heart, liver and kidney meridians. Has the functions of nourishing yin, enriching blood, promoting the production of body fluid and moistening dryness. Is mainly applied to liver and kidney yin deficiency, dizziness, tinnitus, palpitation, insomnia and early white beard and hair; thirst due to fluid consumption, internal heat, diabetes, constipation due to intestinal dryness.
The mulberry mainly contains flavonoid components: cyanidin-glucoside, cyanidin-rutinoside; fatty acid component: linoleic acid, oleic acid, stearic acid, and the like; volatile oil: eucalyptol, geraniol, and the like; also contains organic acids, carotene, saccharides, vitamins, etc.
The mulberry can delay aging, improve the activities of peroxidase and catalase in whole blood and glycomacropeptide, enhance the activity of superoxide dismutase, reduce myocardial lipofuscin and lipid peroxide, and improve hydroxyproline in skin; the product has moderate lymphocyte transformation promoting effect, and can promote T cell maturation, recover function of aged T cell, and enhance immunity; the product also has effects in reducing cholesterol, low density lipoprotein, triacylglycerol, and arteriosclerosis-activating index, and increasing high density lipoprotein and arteriosclerosis-resisting index. The mulberry juice also has the functions of preventing leucopenia caused by cyclophosphamide and reducing the bone marrow micronucleus rate and the chromosome aberration rate induced by the leucocyte reduction juice.
The Bulbus Lilii is fleshy dry scale leaf of Liliaceae plant herba Centellae, Bulbus Lilii or Lilii tenuifolia. Mainly produced in Hunan, Hubei, Jiangsu, Zhejiang and Anhui. Collected in autumn, cleaned, peeled off scaly leaves, slightly scalded in boiling water, and dried. The property of the medicine is sweet, cold in nature and entering heart and lung meridians. The efficacy is as follows: nourish yin and moisten lung, clear heart and induce tranquilization. The application mainly comprises the following steps: cough due to yin deficiency, cough with blood; restlessness due to deficiency with pavor, insomnia, dreaminess, absentmindedness. Mainly contains steroidal saponins components, sugar and a small amount of colchicine. The pharmacological action mainly comprises: both the raw product and honey-fried lily water extract have the functions of relieving cough and eliminating phlegm; the lily water extract has the functions of calming, resisting anoxia and resisting fatigue; the lily polysaccharide can also resist oxidation, improve the immune function and reduce the blood sugar of a hyperglycemic model mouse caused by alloxan; the lily ethanol extract and the ethyl acetate extract can inhibit micrococcus luteus, staphylococcus aureus, escherichia coli, yellow mold, enterococcus faecalis and pseudomonas aeruginosa; the lily bulb extract has higher gram-positive bacteria inhibition activity than gram-negative bacteria.
Rhizoma Polygonati Odorati is dried rhizome of Polygonatum odoratum of Liliaceae. Mainly produced in Hunan, Hubei, Jiangsu and Zhejiang. Picking in autumn, removing fibrous root, cleaning, sun drying to soft, repeatedly kneading, sun drying until no hard core exists, and sun drying; or steaming, kneading to be translucent, and drying in the sun. Cut into thick pieces or segments, unprocessed. The property of the medicine is sweet and slightly cold. It enters lung and stomach meridians. Has the effects of nourishing yin, moistening dryness, promoting the production of body fluid and quenching thirst. The application comprises the following steps: lung yin deficiency, cough due to dryness-heat; stomach yin deficiency, dry throat, thirst, internal heat and diabetes. Mainly contains polysaccharide and steroid components.
Meanwhile, the sealwort, the mulberry, the lily and the polygonatum are all medicine-food homologous medicines, and the concept of pursuing health of modern people is met. The additive in the beverage is also selected by utilizing the natural health concept, and healthy and natural substances such as stevioside, erythritol, citric acid, sodium citrate, guar gum, vitamin C, soybean peptide, vitamin B6, vitamin B12, vitamin B3 and the like are selected.
Disclosure of Invention
The invention mainly aims to provide a yin-nourishing and lung-moistening fresh rhizoma polygonati beverage, which takes fresh rhizoma polygonati, fresh mulberry, lily and radix polygonati officinalis as main raw materials, and stevioside, erythritol, citric acid, sodium citrate, guar gum, vitamin C, soybean peptide, vitamin B6, vitamin B12 and vitamin B3 are added, so that the beverage has the effects of reducing blood sugar, resisting fatigue, resisting oxidation, tonifying spleen, moistening lung, delaying senescence and the like.
The invention also aims to provide another usage of the sealwort, which is not the common steamed sealwort but the fresh sealwort and can better reserve the nutrient substances of the raw materials. In addition, the invention squeezes the fresh rhizoma polygonati through a special process, overcomes the stimulation to the throat caused by directly taking the fresh rhizoma polygonati, and solves the problem that the rhizoma polygonati cannot be used fresh all the time.
The technical scheme of the invention is as follows:
the polygonatum health-care beverage comprises active components and water, wherein the active components comprise the following components in parts by weight: 100-300 parts of fresh rhizoma polygonati, 10-20 parts of fresh mulberry, 2-10 parts of lily and 2-10 parts of polygonatum.
The polygonatum health-care beverage also comprises stevioside, erythritol, citric acid, sodium citrate and guar gum in 0.001-0.1 part respectively.
The active components of the polygonatum health-care beverage also comprise 0.001-0.5 parts of vitamin C, vitamin B6, vitamin B12 and vitamin B3 respectively according to the parts by weight.
The active components of the polygonatum health-care beverage also comprise 0.1-3 parts of soybean peptide by weight.
The invention relates to a preparation method of a polygonatum health-care beverage, which is characterized by comprising the following steps: collecting and selecting raw materials, extracting juice, blending, sterilizing, canning and sterilizing;
(1) collecting and selecting raw materials: digging roots and stems of the fresh Huangjing in spring and autumn every year; collecting fresh Mori fructus when fruit turns red in 4-6 months; yuzhu is collected in autumn, and the ground roots and fibrous roots are removed; collecting Bulbus Lilii in autumn, cleaning, removing scale leaves, slightly scalding in boiling water, and drying.

The specific process for preparing the lily juice comprises the following steps: adding water 5-15 times the weight of the cleaned and crushed lily into the lily, adding Vc into the lily, heating the mixture to 60-80 ℃, leaching the mixture for 2-3 hours, filtering the mixture to obtain filtrate, adding water 3-5 times the weight of the mixture into filter residue, heating the mixture to 60-80 ℃, leaching the mixture for 1-2 hours, filtering the mixture to obtain filtrate, and combining the two filtrates to obtain the lily juice.
The preparation method of the rhizoma Polygonati Odorati juice comprises extracting rhizoma Polygonati Odorati with water 5-15 times, leaching at 100 deg.C for 1-2 hr, leaching with 5-10 times of water at 100 deg.C for 0.5-1 hr, and mixing the extractive solutions to obtain rhizoma Polygonati Odorati juice.
(4) Blending: mixing rhizoma Polygonati juice, Mori fructus juice, Bulbus Lilii juice, and rhizoma Polygonati Odorati juice, stirring, adding stevioside, erythritol, citric acid, sodium citrate, guar gum, vitamin C, soybean peptide, vitamin B6, vitamin B12, and vitamin B3 one by one, stirring, and mixing. Fine filtering the beverage with 1.2 μm filter membrane to obtain filtrate;
(5) and (3) sterilization: performing ultrahigh temperature instantaneous sterilization on the filtrate at 130 ℃ to obtain a sterilized solution;
(6) canning: degassing and filling to obtain bottled beverage;
(7) and (3) sterilization: pasteurizing bottled beverage for 30 min.
Thus obtaining the polygonatum health-care beverage.
The polygonatum health-care beverage and the preparation method thereof have the beneficial effects that:
the polygonatum health-care beverage prepared by the invention adopts fresh polygonatum juice, fresh mulberry juice and various nutritional ingredients as active components, and adds natural stevioside, erythritol, citric acid and sodium citrate as flavoring agents, so that the polygonatum health-care beverage has no side effect, is soft in taste and is sour, sweet and delicious.
The polygonatum health-care beverage can obviously prolong the exhaustive swimming time of mice, reduce the BUN content, increase the glycogen storage, reduce the accumulation of MDA (multidiameter multiple access), enhance the activity of SOD (superoxide dismutase), and further improve the anti-fatigue and anti-oxidation capability of the mice. Compared with the beverage only containing rhizoma polygonati, mulberry and polygonatum odoratum, the anti-fatigue and anti-oxidation capability of the beverage is remarkably improved after lily is added for compatibility. Compared with the common polygonatum health care product beverage, the beverage has better tonifying effect.
The invention juices the fresh rhizoma polygonati by a special process combining high-speed juicing and ultrasonic technology, overcomes the stimulation to the throat caused by directly taking the fresh rhizoma polygonati and solves the problem that the rhizoma polygonati cannot be used fresh all the time. The preparation method disclosed by the invention is simple in process, and can be used for extracting the active ingredients in the polygonatum sibiricum as much as possible, so that the utilization rate of the polygonatum sibiricum is improved, and the resource waste is reduced.
Detailed Description
Example 1:
preparation of a polygonatum health drink 1.
Collecting and selecting raw materials: digging roots and stems of the fresh Huangjing in spring and autumn every year; collecting fresh Mori fructus when fruit turns red in 4-6 months; yuzhu is collected in autumn, and the ground roots and fibrous roots are removed; collecting Bulbus Lilii in autumn, cleaning, removing scale leaves, slightly scalding in boiling water, and drying.
Extracting juice: 200 parts of fresh rhizoma polygonati: collecting, cleaning, selecting juice extractor with power of 850w, rotation speed of 38000 rpm, and juicing once for 25 min; and after juicing, ultrasonically oscillating the filtered juice at the frequency of 28/40khz for one time for 30min to obtain the sealwort juice. 15 parts of fresh mulberry: collecting, cleaning and juicing to obtain the fresh mulberry juice. ③ 5 parts of lily: cleaning, crushing, adding 10 times of water by weight, adding Vc, heating to 60 ℃, leaching for 2 hours, filtering to obtain filtrate, adding 4 times of water by weight into filter residue, heating to 60 ℃, leaching for 1 hour, filtering to obtain filtrate, and combining the two filtrates to obtain the lily juice. Fourthly, 5 parts of polygonatum odoratum: cleaning, adding 10 times of water, extracting at 100 deg.C for 1 hr, adding 5 times of water, extracting at 100 deg.C for 0.5 hr, and mixing the extractive solutions to obtain rhizoma Polygonati Odorati juice.
Blending: mixing the sealwort juice, the mulberry juice, the lily juice and the polygonatum juice prepared in the steps, uniformly stirring, then adding 0.05 part of stevioside, 0.03 part of erythritol, 0.01 part of citric acid, 0.01 part of sodium citrate, 0.05 part of guar gum, 0.005 part of vitamin C, 0.008 part of soybean peptide, 60.002 parts of vitamin B, 120.002 parts of vitamin B and 30.001 parts one by one, and uniformly stirring and mixing. Fine filtering the beverage with 1.2 μm filter membrane to obtain filtrate;
and (3) sterilization: performing ultrahigh temperature instantaneous sterilization on the filtrate at 130 ℃ to obtain a sterilized solution;
canning: degassing and filling to obtain bottled beverage;

Extracting juice: 190 parts of fresh rhizoma polygonati: collecting, cleaning, selecting a juicer with power of 900w, rotating speed of 375000 rpm, juicing once, and juicing time of 18 min; and after juicing, ultrasonically oscillating the filtered juice at the frequency of 28/40khz for one time for 20min to obtain the sealwort juice. 20 parts of fresh mulberry: collecting, cleaning and juicing to obtain the fresh mulberry juice. 7 parts of lily: cleaning, crushing, adding 10 times of water by weight, adding Vc, heating to 60 ℃, leaching for 2 hours, filtering to obtain filtrate, adding 4 times of water by weight into filter residue, heating to 60 ℃, leaching for 1 hour, filtering to obtain filtrate, and combining the two filtrates to obtain the lily juice. Fourthly, 8 parts of polygonatum odoratum: cleaning, adding 10 times of water, extracting at 100 deg.C for 1 hr, adding 5 times of water, extracting at 100 deg.C for 0.5 hr, and mixing the extractive solutions to obtain rhizoma Polygonati Odorati juice.
Blending: mixing the sealwort juice, the mulberry juice, the lily juice and the polygonatum juice, stirring uniformly, then adding 0.03 part of stevioside, 0.05 part of erythritol, 0.008 part of citric acid, 0.012 part of sodium citrate, 0.05 part of guar gum, 0.005 part of vitamin C, 0.008 part of soybean peptide, 60.001 parts of vitamin B, 120.002 parts of vitamin B and 30.002 parts of vitamin B one by one, stirring and mixing uniformly. Fine filtering the beverage with 1.2 μm filter membrane to obtain filtrate;
and (3) sterilization: performing ultrahigh temperature instantaneous sterilization on the filtrate at 130 ℃ to obtain a sterilized solution;
canning: degassing and filling to obtain bottled beverage;
and (3) sterilization: pasteurizing bottled beverage for 30 min. Thus obtaining the polygonatum health-care beverage.
Example 3:
the anti-fatigue and anti-oxidation effects of the polygonatum health drink are compared.
Experimental animals: the clean grade ICR mice were 40, half male and half female, and weighed 18-22 g.
Experimental reagent: the liver/myoglycogen kit, the Blood Urea Nitrogen (BUN) kit, the SOD kit and the Malondialdehyde (MDA) kit are purchased from Nanjing institute of bioengineering.
The experimental method comprises the following steps:
1) experimental grouping (i) the polygonatum health drink 1 obtained in example 1 is a group 1; ② according to the method of the embodiment 1, the beverage prepared without adding lily juice (the other components except lily and the process are completely the same) is group 2; ③ Lily juice (prepared according to the preparation method and the dosage of the Lily juice in the example 1) is set 3; and fourthly, blank control group. Each group had 10 animals with no water control (free water, 0.3 g/animal/day). Mice in groups 1 and 2 were gavaged with 5.0g/kg of the drink per day; mice in group 3 were gavaged with 0.12g/kg of the lily juice per day (equivalent to the gavage dose of the lily juice in group 1); the mice in the blank control group are perfused with normal saline 0.2ml/10g every day. Weighing the weight of the mouse every two days, adjusting the gavage amount according to the weight of the mouse, performing gavage once a day, and continuously performing gavage for 14 days.
2) Measurement of exhaustive swimming time 10 mice per group were used to measure exhaustive swimming time, and 30min after the last administration, the mice were placed in a water tank at water temperature (25 ± 2) ° c, and the edges of the water tank were tapped with a wooden stick to stimulate the mice to swim rapidly. And recording the time from the beginning of swimming to the time that the head of the mouse is deep into the water and does not float by using a stopwatch, namely the time of the exhaustion of the mouse for swimming.
3) Measurement of Blood Urea Nitrogen (BUN) 30min after last gastric lavage, placing in water tank with water temperature of 25 deg.C for swimming for 60min, immediately taking blood from eyeball, centrifuging to separate serum, and detecting blood urea nitrogen content with kit.
4) Determination of glycogen and myoglycogen the muscle and liver of hind limb of mouse in the blood urea nitrogen determination experiment are taken, and 1% of glycogen detection solution and 5% of myoglycogen detection solution are prepared according to the specification requirement of kit, and the content of glycogen and myoglycogen is detected.
5) Measuring superoxide dismutase (SOD) and Malondialdehyde (MDA), taking mouse liver and kidney in a blood urea nitrogen measurement experiment, preparing 10% liver homogenate and 10% kidney homogenate according to the specification of a kit, wherein the 10% tissue homogenate is prepared by weighing the weight of a tissue, adding 9 times of volume of a homogenate medium according to the weight (g) and the volume (ml) of the homogenate medium to 1:9, and mechanically homogenizing the mixture under the condition of ice-water bath to prepare 10% tissue homogenate. SOD activity and MDA content in liver and kidney are detected.
The experimental results are as follows:
1) the effect of the anti-fatigue index after exercise in each group of mice is shown in table 1.

The results in tables 1 and 2 show that the drinks of groups 1 and 2 can obviously prolong the exhaustive swimming time of mice, reduce the BUN content, increase the glycogen storage, reduce the accumulation of MDA and enhance the activity of SOD. However, as can be seen from comparative experiments, although the anti-fatigue and anti-oxidation capability of the mice cannot be remarkably improved by the same dosage of lily juice, the anti-fatigue and anti-oxidation capability of the mice can be remarkably enhanced by adding the lily juice into polygonatum sibiricum, mulberry and polygonatum odoratum in a compatible manner.
Example 4:
contrast test of the throat irritation of rats by the health beverage prepared by different juicing processes of fresh rhizoma polygonati.
60 male SD rats of 110-130g were selected and divided into 6 groups of 10 rats. The groups are respectively as follows: group of example 1; example 2 group; group D1: a low-speed common sealwort juicer unit; group D2: a sealwort low-speed common juicer and an ultrasonic group; group D3: a high-speed polygonatum sibiricum juice extractor set; group D4: and (4) a purified water group. Feeding the rats respectively, stopping water and not fasting for 24h before the test, feeding corresponding beverage to the rats respectively, recording the times of scratching throat of the rats within 10min of drinking water, and performing statistical analysis, wherein the results are shown in Table 3

As can be seen from the results in Table 3, the polygonatum health drink prepared by taking the polygonatum juice extracting process of the invention has the effect of remarkably reducing the irritation of the polygonatum juice extracted by the common extracting process. While by group D1: a low-speed common sealwort juicer unit; group D2: a sealwort low-speed common juicer and an ultrasonic group; group D3: the comparison of the sealwort high-speed juice extractor shows that the combination of high-speed juice extraction and ultrasound is necessary for reducing the irritation effect.
Finally, it should be noted that the above embodiments are only used for illustrating the technical solutions of the present invention and not for limiting the protection scope of the present invention, and although the present invention has been described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions can be made on the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention.
