Example 4 inhibition of 5 alpha-reductase assay
The oil content of the skin is one of the most important physiological indexes of the facial skin, and the normal and healthy skin is smooth, fine and elastic. And the skin grease is excessively secreted or excessively secreted, which not only affects the beauty, but also can cause various diseases of the skin.
The effect of sebum regulation is currently reflected by the detection of the inhibitory effect of 5 alpha-reductase.
The 5 alpha-reductase catalyzes and converts testosterone T into dihydrotestosterone under the participation of reduced coenzyme II, and if a 5 alpha-reductase inhibitor exists, the reaction process can be inhibited to a certain extent. Because the reduced coenzyme II has characteristic absorption at the wavelength of 340nm, the characteristic absorption of the oxidized coenzyme II generated after the reaction at the wavelength of 340nm disappears. The ultraviolet spectrophotometer is used for determination, and whether a certain substance has an inhibition effect on the 5 alpha-reductase or not and the size of the inhibition effect can be reflected.
Repeated experiments prove that the 5 alpha-reductase catalytic reaction system with the total volume of 100 mu L is established. Components of the reaction system the amounts (μ L) of the reagents used are shown in Table 1.
TABLE 1
Figure BDA0001297631780000081
Determination of blank control: adding testosterone T, 5 alpha-reductase, phosphate buffer (pH 7.4) and reduced coenzyme II into a reaction tube, mixing, measuring absorbance, incubating at 37 deg.C for 60min, measuring absorbance, deducting blank reduced background value of reduced coenzyme II, and measuring blank reduced value delta A0=0.066。
Determination of the inhibitory Effect: adding testosterone T, 5 alpha-reductase, inhibitor, phosphate buffer (pH 7.4) and reduced coenzyme II into a reaction tube, mixing, measuring absorbance, incubating at 37 ℃ for 60min, measuring absorbance, subtracting blank reduced background value of reduced coenzyme II, and measuring inhibitor reduced value (delta An). The results are shown in Table 2.
And judging whether the activity of the enzyme is inhibited or not by taking finasteride as a positive drug, and preliminarily calculating the inhibition rate. The formula is I (%) ═(ΔA0-ΔAn)/ΔA0X 100%. The calculation results are shown in Table 2.
TABLE 2
Figure BDA0001297631780000091
Remarking: the extract solutions of collard, basil leaf and peanut seed coat in tables 1 and 2 were each the extract solutions prepared in example 1; composition a is the botanical composition provided in example 2; composition B was the botanical composition provided in example 3 (with a test dose of 10 g).
As can be seen from the results in table 2, the individual plant extracts of lupulus, basil leaves and peanut seed coat, composition a and composition B, which were prepared in example 1 of the present invention, all had different degrees of inhibition of 5 α -reductase, but the inhibition rate of composition a was close to that of the positive control finasteride, indicating that the individual plant extracts of lupulus, basil leaves and peanut seed coat, which were prepared by a specific process, were as follows: 3: 1, has good synergistic effect on the inhibition of 5 alpha-reductase.
Example 5 orthogonal assay
The compounding proportion of three functional components of the collard, the basil leaf and the peanut seed coat is determined by adopting a three-factor three-level orthogonal test.
Table 3 orthogonal test protocol table

Figure BDA0001297631780000101
TABLE 4 results of orthogonal experiments
Figure BDA0001297631780000102
The above test combinations were prepared by combining the extracts of the factors in table 3 at the levels indicated.
From the results in table 4, it can be seen that the extracts of lupulus, basil leaf and peanut seed coat all had an effect on the inhibition rate of 5 α -reductase, with the extract of lupulus having the greatest effect on the inhibition rate, followed by the extract of basil leaf; the optimal compound ratio determined by the invention is 6g of the extract of the collard, 3g of the extract of the basil leaf and 1g of the extract of the peanut seed coat, namely A3B3C2Is the best combination.
That is, the extract of the lupulus, the extract of the basil leaf and the extract of the peanut seed coat which are prepared by the specific process according to the invention are mixed according to the ratio of 6: 3: 1, the plant composition prepared by uniformly mixing the components in the weight ratio has the best effect of inhibiting 5 alpha-reductase.
