本发明提出了一种蛹虫草颗粒冲剂及其制备方法,由以下原料制成:蛹虫草素提取物、蛹虫草多糖提取物、配料多糖提取物、填料、甜味剂与酸味剂,所述配料多糖提取物为黄芪多糖提取物、枸杞多糖提取物或者灵芝多糖提取物。制备方法:1)混料:按照配比称取蛹虫草素提取物、蛹虫草多糖提取物、配料多糖提取物、填料、甜味剂与酸味剂进行混合,加入适量去离子水搅拌均匀,使其成为浆液状,得到混合浆液;2)均质化:调配好的混合浆液进入胶体磨,再用放入均质机中进行均质;3)制粒:将均质后浆液通过喷雾干燥机制粒即可。该颗粒冲剂采用配料多糖作为虫草营养素的补充,在增强免疫调节功效的同时,满足不同体质消费群体的需求。

蛹虫草是一种名贵的中药材,性平味甘,具有滋肺补肾、止血化痰的功效。近年来的研究结果表明,蛹虫草是冬虫夏草的重要人工替代品之一,其在降血脂,防治动脉硬化,保护心、脑组织,镇静催眠,增强巨噬细胞活性,抗癌、抗炎、抗菌、抗缺氧等方面具有良好功效。蛹虫草的功能性成分、药理药效作用与天然冬虫夏草基本一致,其子实体富含虫草素和虫草多糖等重要的生理活性成分。据现代科学论证:虫草素是一种具有抗菌活性的核苷类物质,对多聚腺苷酸聚合酶有很强的抑制作用,能抑制癌细胞的生长,并有抗菌抗病毒、免疫调节、清除自由基等多种药理作用;虫草多糖是一种高度分枝的半乳甘露聚糖,它能促进淋巴细胞转化,提高血清IgG的抗体含量和机体的免疫功能,增强机体自身抗癌抑癌的能力。
目前的蛹虫草冲剂类产品通常采用超微粉碎等技术将蛹虫草子实体进行粉碎后直接作为终端产品销售,其产品针对性不强,附加值较低,且产品结构单一,很难满足消费群体的多样化需求。
发明内容
本发明提出一种蛹虫草颗粒冲剂,该颗粒冲剂采用配料多糖作为虫草营养素的补充,在增强免疫调节功效的同时,满足不同体质消费群体的需求。
本发明的技术方案是这样实现的:
一种蛹虫草颗粒冲剂,由以下原料制成:蛹虫草素提取物、蛹虫草多糖提取物、配料多糖提取物、填料、甜味剂与酸味剂,所述配料多糖提取物为黄芪多糖提取物、枸杞多糖提取物或者灵芝多糖提取物。
优选地,本发明一些实施例中,所述填料为麦芽糊精与β-环糊精的混合物,所述甜味剂为果葡糖浆,所述酸味剂为苹果酸。
优选地,本发明一些实施例中,按照重量百分数计算,所述蛹虫草素提取物的含量为30~40%、所述蛹虫草多糖提取物的含量为30~40%、所述配料多糖提取物的含量为10~20%、所述麦芽糊精的含量为10~20%、所述果葡糖浆的含量为5~10%、所述β-环糊精的含量为2~4%以及所述苹果酸的含量为0.05~0.1%。
优选地,本发明一些实施例中,所述蛹虫草多糖提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇或无水乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)多糖提取:将烘干后的滤渣与乙醇溶液混合进行超声波辅助提取,超声波频率为20~40KHz,超声处理时间为30~90分钟,重复处理2~3次,提取结束后,过滤,合并多次滤液,在35~45℃,300~700mmHg下减压浓缩,添加体积浓度为95%乙醇或无水乙醇,静置8~24小时,得到析出的水溶性蛹虫草粗多糖。
优选地,本发明一些实施例中,所述蛹虫草素提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇或无水乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)蛹虫草素提取:将烘干后的滤渣与乙醇溶液混合进行超声波辅助提取,超声波频率为20~40KHz,超声处理时间为30~90分钟,重复处理2~3次,提取结束后,过滤,合并多次滤液,在35~45℃,300~700mmHg下减压浓缩,添加体积浓度为95%乙醇或无水乙醇,静置8~24小时,取醇析后的上清液,在35~45℃,300~700mmHg下减压浓缩至干,向其中加蒸馏水溶解,调pH值至3.0~4.0,上离子交换柱用0.1~0.3mol/L氨水洗脱除掉杂质,收集洗脱液进行再浓缩,冷藏析出晶体,用正丁醇重结晶,得到蛹虫草素提取物。
优选地,本发明一些实施例中,所述配料多糖提取物通过以下方法制备得到的:
将配料粉碎后加入10~15倍的去离子水进行超声波辅助提取,超声波频率为20~40KHz,超声处理时间为30~90分钟,重复处理2~3次,提取结束后,过滤,合并多次滤液,在55~65℃,300~700mmHg下减压浓缩,添加95%乙醇或无水乙醇,静置8~24小时,即可获得配料多糖提取物。
优选地,本发明一些实施例中,所述步骤2)中与滤渣混合的乙醇溶液体积浓度为10%~30%,滤渣:乙醇溶液的质量比为1:10~20。
本发明的另一个目的是提供一种蛹虫草颗粒冲剂的制备方法,包括以下步骤:
1)混料:按照配比称取蛹虫草素提取物、蛹虫草多糖提取物、配料多糖提取物、填料、甜味剂与酸味剂进行混合,加入适量去离子水搅拌均匀,使其成为浆液状,得到混合浆液;
2)均质化:调配好的混合浆液进入胶体磨,再用放入均质机中进行均质;
3)制粒:将均质后浆液通过喷雾干燥机制粒即可。
优选地,所述步骤2)均质时的压力为40MPa。
优选地,所述喷雾干燥机的制粒条件为进风温度160℃,排风温度90℃,转速12000r/min。
本发明的有益效果:
1)本发明的蛹虫草颗粒冲剂将蛹虫草素和蛹虫草多糖进行粗提取后与其他中药提取物进行配伍,大幅度提高了产品中的有效物质含量,具有极好的提高免疫力功效,且颗粒冲剂的形式更易于儿童等特殊群体服用,满足更多消费者的需求。
2)配料多糖中黄芪多糖是黄芪发挥作用的主要成分,具有抗病毒、抗肿瘤、抗衰老、抗辐射、抗应激、抗氧化等作用,枸杞多糖是枸杞子的主要活性成分,具有免疫调节、延缓衰老等多种功效,灵芝多糖是灵芝的重要活性成分,具有提高机体免疫力、加快血液微循环、提高血液供氧能力、消除体内自由基等多种功能。这些配料多糖与蛹虫草素和蛹虫草多糖相配合,在增强免疫调节功效的同时,用于作为蛹虫草营养素的补充,满足不同体质消费群体的需求。在颗粒冲剂的加工过程中,采用蛹虫草素和蛹虫草多糖共同提取的方法进行提取,保证了蛹虫草素和蛹虫草多糖的活性,最大限度的增加了原料利用率,降低了生产成本,便于工业化生产。
具体实施方式
实施例1
一种蛹虫草颗粒冲剂,按照重量百分数计算,由以下原料制成:
含量为35%的蛹虫草素提取物、含量为35%的蛹虫草多糖提取物、含量为10%的黄芪多糖提取物、含量为10%的麦芽糊精、含量为7.7%的果葡糖浆、含量为2.22%的β-环糊精以及含量为0.08%的苹果酸。
蛹虫草多糖提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)多糖提取:将烘干后的滤渣与20%乙醇溶液(滤渣:乙醇溶液的质量比为1:10)混合进行超声波辅助提取,超声波频率为20KHz,超声处理时间为90分钟,重复处理2次,提取结束后,过滤,合并多次滤液,在35℃,700mmHg下减压浓缩,添加体积浓度为95%乙醇,静置24小时,得到析出的水溶性蛹虫草粗多糖。
蛹虫草素提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)蛹虫草素提取:将烘干后的滤渣与10%乙醇溶液(滤渣:乙醇溶液的质量比为1:20)混合进行超声波辅助提取,超声波频率为20KHz,超声处理时间为90分钟,重复处理2次,提取结束后,过滤,合并多次滤液,在35℃,700mmHg下减压浓缩,添加体积浓度为95%乙醇,静置24小时,取醇析后的上清液,在45℃,300mmHg下减压浓缩至干,向其中加蒸馏水溶解,调pH值至3.0,上离子交换柱用0.1mol/L氨水洗脱除掉杂质,收集洗脱液进行再浓缩,冷藏析出晶体,用正丁醇重结晶,得到蛹虫草素提取物。
黄芪多糖提取物通过以下方法制备得到的:
将黄芪粉碎后加入10倍的去离子水进行超声波辅助提取,超声波频率为40KHz,超声处理时间为30分钟,重复处理3次,提取结束后,过滤,合并多次滤液,在55℃,700mmHg下减压浓缩,添加无水乙醇,静置18小时,即可获得黄芪多糖提取物。
蛹虫草颗粒冲剂的制备方法:
1)混料:按照配比称取蛹虫草素提取物、蛹虫草多糖提取物、黄芪多糖提取物、麦芽糊精、β-环糊精、果葡糖浆与苹果酸进行混合,加入适量去离子水搅拌均匀,使其成为浆液状,得到混合浆液;
2)均质化:调配好的混合浆液进入胶体磨,再用40MPa的压力在均质机中进行高压均质;
3)制粒:将均质后浆液通过喷雾干燥机制粒即可,喷雾干燥机的制粒条件为进风温度160℃,排风温度90℃,转速12000r/min。
实施例2
一种蛹虫草颗粒冲剂,按照重量百分数计算,由以下原料制成:
含量为30%的蛹虫草素提取物、含量为30%的蛹虫草多糖提取物、含量为15%的灵芝多糖提取物、含量为15%的麦芽糊精、含量为6.5%的果葡糖浆、含量为3.4%的β-环糊精以及含量为0.1%的苹果酸。
蛹虫草多糖提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)多糖提取:将烘干后的滤渣与10%乙醇溶液(滤渣:乙醇溶液的质量比为1:20)混合进行超声波辅助提取,超声波频率为40KHz,超声处理时间为30分钟,重复处理3次,提取结束后,过滤,合并多次滤液,在45℃,300mmHg下减压浓缩,添加无水乙醇,静置8小时,得到析出的水溶性蛹虫草粗多糖。
蛹虫草素提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)蛹虫草素提取:将烘干后的滤渣与10%乙醇溶液(滤渣:乙醇溶液的质量比为1:20)混合进行超声波辅助提取,超声波频率为40KHz,超声处理时间为30分钟,重复处理2次,提取结束后,过滤,合并多次滤液,在35℃,700mmHg下减压浓缩,添加无水乙醇,静置8小时,取醇析后的上清液,在45℃,700mmHg下减压浓缩至干,向其中加蒸馏水溶解,调pH值至4.0,上离子交换柱用0.3mol/L氨水洗脱除掉杂质,收集洗脱液进行再浓缩,冷藏析出晶体,用正丁醇重结晶,得到蛹虫草素提取物。
灵芝多糖提取物通过以下方法制备得到的:
将灵芝粉碎后加入15倍的去离子水进行超声波辅助提取,超声波频率为20KHz,超声处理时间为90分钟,重复处理3次,提取结束后,过滤,合并多次滤液,在65℃,300mmHg下减压浓缩,添加无水乙醇,静置8小时,即可获得灵芝多糖提取物。
蛹虫草颗粒冲剂的制备方法:
1)混料:按照配比称取蛹虫草素提取物、蛹虫草多糖提取物、灵芝多糖提取物、麦芽糊精、β-环糊精、果葡糖浆与苹果酸进行混合,加入适量去离子水搅拌均匀,使其成为浆液状,得到混合浆液;
2)均质化:调配好的混合浆液进入胶体磨,再用40MPa的压力在均质机中进行高压均质;
3)制粒:将均质后浆液通过喷雾干燥机制粒即可,喷雾干燥机的制粒条件为进风温度160℃,排风温度90℃,转速12000r/min。
实施例3
一种蛹虫草颗粒冲剂,按照重量百分数计算,由以下原料制成:
含量为30%的蛹虫草素提取物、含量为35%的蛹虫草多糖提取物、含量为15%的枸杞多糖提取物、含量为10%的麦芽糊精、含量为6%的果葡糖浆、含量为3.95%的β-环糊精以及含量为0.05%的苹果酸。
蛹虫草颗粒冲剂的制备方法:
1)混料:按照配比称取蛹虫草素提取物、蛹虫草多糖提取物、枸杞多糖提取物、麦芽糊精、β-环糊精、果葡糖浆与苹果酸进行混合,加入适量去离子水搅拌均匀,使其成为浆液状,得到混合浆液;
2)均质化:调配好的混合浆液进入胶体磨,再用40MPa的压力在均质机中进行高压均质;
3)制粒:将均质后浆液通过喷雾干燥机制粒即可,喷雾干燥机的制粒条件为进风温度160℃,排风温度90℃,转速12000r/min。
试验例1
增强免疫实验
为了验证上述三种实施例的蛹虫草颗粒冲剂的免疫增强效果,进行小鼠足跖部厚度或肿胀度的实验,测试过程及结果如下:
受试对象:体重相近且性别相同的BALB/C小鼠4组。
检测方法:4组小鼠分别为对照组和3组受试组,将蛹虫草颗粒冲剂用水溶解,3组受试组分别给予实施例1-3的颗粒冲剂的水溶液,给药量为人体推荐量的10倍,对照组给予等量溶媒,在实验条件下适应喂养3天后开始灌胃,每天一次,连续28天。末次灌胃3小时后小鼠用2%(v/v)SRBC腹腔或静脉免疫,每只鼠注射0.2mL(约1×108个SRBC),免疫后4天,测量左后足跖部厚度或肿胀度,然后在测量部位皮下注射20%(v/v)SRBC,每只鼠20μl(约1×108个SRBC),注射后于24h测量左后足跖部厚度或肿胀度,同一部位测量三次,取平均值,以足跖部厚度或肿胀度来判断小鼠免疫力的高低。
测验结论:4组小鼠的足跖部厚度或肿胀度分别为:对照组:2.158mm,实施例1:1.341mm,实施例2:1.116mm,实施例3:1.628mm,足跖部厚度或肿胀度越小,说明其免疫力越强,故3种实施例冲剂均具有良好的增强免疫力效果,其中,以实施例2效果最好。
试验例2
增强免疫实验
为了验证上述三种实施例的蛹虫草颗粒冲剂的免疫增强效果,进行小鼠吞噬指数的实验,测试过程及结果如下:
受试对象:体重相近且性别相同的BALB/C小鼠4组。
检测方法:4组小鼠分别为对照组和3组受试组,将蛹虫草颗粒冲剂用水溶解,3组受试组分别给予3种实施例水溶液,给药量为人体推荐量的10倍,对照组给予等量溶媒,在实验条件下适应喂养3天后开始灌胃,每天一次,连续28天。末次灌胃3小时后,称体重,从小鼠尾静脉注入稀释的印度墨汁,按每10g体重0.1mL计算,待墨汁注入,立即计时。注入墨汁后10min,分别从内眦静脉丛取血20μl,并立即将其加到2mL 0.1%Na2CO3溶液中。用分光光度计或全自动酶标仪在600nm波长处测光密度值(OD),以Na2CO3溶液作空白对照。并将小鼠处死,取肝脏和脾脏,用滤纸吸干脏器表面血污,称重。以吞噬指数表示小鼠碳廓清的能力,即免疫力的强弱,吞噬指数的计算方法为:
测验结论:4组小鼠吞噬指数分别为:对照组:0.670.06,实施例1:0.920.04,实施例2:1.170.07,实施例3:0.890.06,故3种实施例冲剂均具有较好的免疫增强效果,其中,以实施例3效果最好。

Claims (10)
1.一种蛹虫草颗粒冲剂,其特征在于,由以下原料制成:蛹虫草素提取物、蛹虫草多糖提取物、配料多糖提取物、填料、甜味剂与酸味剂,所述配料多糖提取物为黄芪多糖提取物、枸杞多糖提取物或者灵芝多糖提取物。
2.根据权利要求1所述的蛹虫草颗粒冲剂,其特征在于,所述填料为麦芽糊精与β-环糊精的混合物,所述甜味剂为果葡糖浆,所述酸味剂为苹果酸。
3.根据权利要求2所述的蛹虫草颗粒冲剂,其特征在于,按照重量百分数计算,所述蛹虫草素提取物的含量为30~40%、所述蛹虫草多糖提取物的含量为30~40%、所述配料多糖提取物的含量为10~20%、所述麦芽糊精的含量为10~20%、所述果葡糖浆的含量为5~10%、所述β-环糊精的含量为2~4%以及所述苹果酸的含量为0.05~0.1%。
4.根据权利要求1所述的蛹虫草颗粒冲剂,其特征在于,所述蛹虫草多糖提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇或无水乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)多糖提取:将烘干后的滤渣与乙醇溶液混合进行超声波辅助提取,超声波频率为20~40KHz,超声处理时间为30~90分钟,重复处理2~3次,提取结束后,过滤,合并多次滤液,在35~45℃,300~700mmHg下减压浓缩,添加体积浓度为95%乙醇或无水乙醇,静置8~24小时,得到析出的水溶性蛹虫草粗多糖。
5.根据权利要求1所述的蛹虫草颗粒冲剂,其特征在于,所述蛹虫草素提取物通过以下方法制备得到的:
1)原料处理:将蛹虫草子实体粉碎,向粉碎的原料中加入体积浓度为95%乙醇或无水乙醇以冷渗法进行提取,将醇溶性色素的杂质以滤液的形式初步分离出来,弃滤液,将滤渣回收并真空烘干;
2)蛹虫草素提取:将烘干后的滤渣与乙醇溶液混合进行超声波辅助提取,超声波频率为20~40KHz,超声处理时间为30~90分钟,重复处理2~3次,提取结束后,过滤,合并多次滤液,在35~45℃,300~700mmHg下减压浓缩,添加体积浓度为95%乙醇或无水乙醇,静置8~24小时,取醇析后的上清液,在35~45℃,300~700mmHg下减压浓缩至干,向其中加蒸馏水溶解,调pH值至3.0~4.0,上离子交换柱用0.1~0.3mol/L氨水洗脱除掉杂质,收集洗脱液进行再浓缩,冷藏析出晶体,用正丁醇重结晶,得到蛹虫草素提取物。
6.根据权利要求1所述的蛹虫草颗粒冲剂,其特征在于,所述配料多糖提取物通过以下方法制备得到的:

8.如权利要求1所述的蛹虫草颗粒冲剂的制备方法,其特征在于,包括以下步骤:
1)混料:按照配比称取蛹虫草素提取物、蛹虫草多糖提取物、配料多糖提取物、填料、甜味剂与酸味剂进行混合,加入适量去离子水搅拌均匀,使其成为浆液状,得到混合浆液;
2)均质化:调配好的混合浆液进入胶体磨,再用放入均质机中进行均质;
3)制粒:将均质后浆液通过喷雾干燥机制粒即可。
9.根据权利要求8所述的蛹虫草颗粒冲剂的制备方法,其特征在于,所述步骤2)均质时的压力为40MPa。
10.根据权利要求8所述的蛹虫草颗粒冲剂的制备方法,其特征在于,所述喷雾干燥机的制粒条件为进风温度160℃,排风温度90℃,转速12000r/min。
