Background
Malignant tumor is a disease caused by the over proliferation of cells in the body due to the loss of normal regulation and control. Hyperproliferative cells are called cancer cells, which often invade surrounding tissues (infiltration, i nvas ion) and may even be transferred to other parts of the body via the systemic circulation and/or the lymphatic system (cancer metastasis). There are many types of cancer, and the severity of the condition depends on the location of the cancer cells, the degree of malignancy, and whether metastasis has occurred. Once diagnosed, treatment is often performed in combination with surgery, chemotherapy, and radiation therapy. If the cancer is untreated, the end result will often lead to death.
At present, the traditional therapy for malignant tumor mainly adopts radiotherapy and chemotherapy, and the traditional chemotherapy has a plurality of side effects, including immunosuppression, mucosal injury and the like, which directly affect the life quality of patients, so that the substitute of the chemotherapy is urgently needed to be searched.
The anti-tumor traditional Chinese medicine is mainly summarized into two methods of eliminating pathogenic factors and strengthening body resistance, namely a method of eliminating pathogenic factors, clearing away heat and toxic materials, activating blood and removing stasis, eliminating phlegm and resolving masses and a method of combating poison with poison; the methods of strengthening the body resistance, which are based on the orientation of the excess and deficiency of yin, yang, qi, blood, viscera and meridians, are classified into four methods of invigorating spleen, nourishing blood, tonifying kidney and nourishing yin. Mainly takes syndrome differentiation treatment as a main treatment, and can effectively control tumor development, improve survival time and improve survival quality. Many herbal medicines contain anti-tumor components and inhibit or kill tumor cells through multiple pathways and multiple targets. Research suggests that the anticancer mechanism of traditional Chinese medicine mainly has the following aspects: the traditional Chinese medicine has the advantages of relatively small side effect, can prevent cancer cells from relapse and transferring and improve the immunity of patients, so the traditional Chinese medicine is more and more widely applied to clinical application.

The main active ingredients are as follows:
wolfberry polysaccharide: the lycium barbarum polysaccharide is a water-soluble polysaccharide, is the main active ingredient in lycium barbarum, has a relative molecular mass of 68-200, and becomes a hot point of research at home and abroad. The study on the immunoregulation and anti-tumor effects of lycium barbarum polysaccharides is the most. Many studies show that the lycium barbarum polysaccharide has the effects of promoting immunity, resisting aging, resisting tumors, eliminating free radicals, resisting fatigue, resisting radiation, protecting the liver, protecting and improving reproductive function and the like.
Betaine: the chemical name is 1-carboxyl-N, N, N-trimethyl glycine lactone, which is similar to amino acid in chemical structure and belongs to quaternary ammonium base material. Betaine is one of the main alkaloids in the fruits, leaves and stalks of Chinese wolfberry. The action of lycium barbarum on lipid metabolism or lipotropism is mainly due to the betaine content, which acts as a methyl donor in the body. The research on lycium barbarum betaine is limited to domestic aspects of content determination, extraction process and physiological action (salt tolerance enhancement) research on lycium barbarum plants, and few pharmacological and pharmacodynamic researches on lycium barbarum betaine are carried out.
Medlar pigment: the Chinese wolfberry pigment is various color-producing substances existing in Chinese wolfberry berries and is an important physiological active ingredient of Chinese wolfberry seeds. Mainly including carotene, lutein and other colored substances. The carotenoid contained in the medlar has very important medicinal value. Many studies have proved that the lycium barbarum seed pigment has the effects of improving the human body immunity, preventing and inhibiting tumors, preventing atherosclerosis and the like. Carotene is the main active component of Lycium barbarum pigment, and has important physiological functions of resisting oxidation and serving as the synthesis precursor of vitamin A.
Honeysuckle is a good medicine for clearing heat and removing toxicity. It is sweet and cold in nature and fragrant in nature, and sweet and cold in nature and clearing heat without damaging the stomach. It is fragrant and can eliminate pathogens. The chemical components mainly comprise organic acid, flavonoid, triterpenoid saponin, volatile oil and the like. The honeysuckle can disperse wind-heat and also can clear away heat and toxic material, and has obvious effect on various heat diseases such as heat generation, eruption, macula, sore and carbuncle due to heat toxin, sore throat and the like. The honeysuckle comprises the following main active components:
organic acids: chlorogenic acid compounds are main effective components of flos Lonicerae, and comprise chlorogenic acid (chlorochlorogenic acid) and isochlorogenic acid (isochlorogenic acid), wherein isochlorogenic acid is a mixture of 7 isomers, which are respectively 4, 5-dicaffeoylquinic acid, 3, 4-dicaffeoylquinic acid, 3, 5-dicaffeoylquinic acid, 1, 3-dicaffeoylquinic acid, 3-feruloylquinic acid, 4-feruloylquinic acid, 5-feruloylquinic acid, and other organic acids including caffeic acid and palmitic acid.
Luteolin (luteolin) is firstly separated from honeysuckle in flavonoid Zhanqitailang of 1949, loniceraside is separated from him in 1961, and 4 flavonoids are firstly separated from honeysuckle in Gaoyamine of 1995.
Triterpene saponin, wherein a novel triterpene saponin containing six glycosyl groups is separated from honeysuckle flower in 1990. In 1993, Maoqing and the like are also separated to obtain 3 triterpene saponins. In 1994, Chenmin et al isolated 2 new dicaffeoylquinic acid vinegar compounds. Three triterpenoid saponins are separated from Mongolian snakegourd fruit and the like.
Volatile oil is the effective component of honeysuckle. Separating out linalool, bisabolol, palmitic acid, dihydrogeraniol, methyl tetracosanate, ethyl octadecadienoate, ethyl palmitate, 1-dicyclohexyl and other components by gas chromatography-mass spectrometry. The honeysuckle flower and dried flower have large component difference. The fresh flower volatile oil component is mainly linalool and has a content of more than 14%, and the dry flower volatile oil component is mainly palmitic acid and generally accounts for more than 26%. 5-hydroxy 7, 4-dimethoxy flavone was first discovered from this plant by the enzyme chennge et al, 2002, and quercetin, loniceraside, oleanolic acid and daucosterol were first discovered from the flower buds of this plant. The pharmacological actions of honeysuckle flower are mainly as follows: antibacterial, antiviral, antiinflammatory, antipyretic, cholagogue, hepatoprotective, blood lipid reducing, and immunity improving effects.
In vivo experiments show that the antitumor capability of the aqueous extracts of the medlar and the honeysuckle is obviously improved after the aqueous extracts of the medlar and the honeysuckle are fermented by lactic acid bacteria, saccharomycetes and acetic acid bacteria.

Using inguinal inoculation: mice were exposed to the groin, sterilized with 75% alcohol, and the needle was inserted from 1cm below the groin at an angle of 30 degrees to the subcutaneous level, and each mouse was injected with 0.2mL of an ascites diluent.
The groups of mice and the treatment regimen are shown in table 1.
TABLE 1
Group of | Mode of treatment |
A: blank group (not)Make mould) | Normal saline for gastric perfusion |
B: negative control group | Normal saline for gastric perfusion |
C: positive control group | Fluorouracil injection 0.2mL (20mg/kg bw) |
D: high dose group | Gavage fermented beverage stock solution 0.4mL |
E: middle dose group | Gavage fermented beverage 2 times diluent 0.4mL |
F: low dose group | Gavage fermented beverage 4 times diluent 0.4mL |
G: aqueous extract group | Gavage unfermented aqueous extract 0.4mL |
Each group of 15 mice
The mice are treated every day from the next day after model building, the treatment is carried out for 7d totally, the food intake and the body weight of the mice are recorded every day, the mice are killed at the 9 th day, the complete tumor tissues are separated, weighed and recorded.
The experimental results are as follows:
effect of fermented beverages on food intake of mice:
as shown in fig. 4, it can be seen from the experimental results that: the food intake of the chemotherapy groups is the lowest and is obviously different from that of the normal group, so that the food intake of the mice is greatly influenced by chemotherapy, the food intake of the blank group is the highest, the food intake of the high-dose group is the highest, and the food intake of the medium-dose group and the low-dose group are sequentially reduced, but are obviously different from those of the chemotherapy groups and are higher than those of the negative control group. The water extract group had higher food intake than the chemotherapy group, but the difference was not significant. The experiment shows that the beverage can effectively improve the food intake of tumor-bearing mice.
Effect of fermented beverages on mouse body weight:
as shown in fig. 5, it can be seen from the experimental results that: the body weight of the mice in the chemotherapy group is obviously reduced, the body weight and the nutrition condition of the mice are greatly influenced by chemotherapy, the body weight of the blank group is the highest, the body weight of the high-dose group is next to that of the blank group, and the body weights of the medium-dose group and the low-dose group are sequentially reduced, but are obviously different from those of the chemotherapy group and higher than that of the negative control group. The weight of the water extract group is higher than that of the chemotherapy group, and the difference is obvious. The experiment shows that the beverage can effectively increase the body weight of tumor-bearing mice.
The effect of fermented beverages on tumor inhibition in mice is shown in table 2:
TABLE 2
Group of | Number of | Tumor weight (g) | Inhibition ratio (%) |
A: blank group | 15 | 0 | -- |
Negative control group (No treatment) | 15 | 0.74±0.38 | 0 |
C: positive control group (Fluorouracil) | 15 | 0.31±0.16 | 57.8 |
D: high dose group | 15 | 0.40±0.22 | 46.1 |
E: middle dose group | 15 | 0.54±0.29 | 27.1 |
F: low dose group | 15 | 0.57±0.28 | 22.3 |
G: aqueous extract group | 15 | 0.71±0.46 | 4.5 |
The experimental results show that: fluorouracil has the strongest inhibition to mouse tumor, the inhibition rate reaches 58.7%, the inhibition effect of high-dose group to mouse tumor is the second to 46.1%, the unfermented water extract group also has certain inhibition to mouse tumor, but only 4.5%.
The effect of fermented beverages on intestinal flora in mice is shown in table 3:
TABLE 3
The experimental results show that the intestinal flora of the mice is greatly changed after chemotherapy: the number of enterobacteria, enterococcus, lactobacillus and bifidobacterium is reduced obviously. The unfermented aqueous extract has certain recovery and protection effects on the mouse intestinal flora, and has obvious difference compared with the chemotherapy group. The high-dose group and the medium-dose group can effectively improve the number of various intestinal flora of mice, and are obviously different from the water extract group.
In vitro tests show that the inhibition rates of the traditional Chinese medicine fermented beverage on HT29 (human colon cancer cell line) S180 (mouse ascites type sarcoma cell line) HpG2 (human liver cancer cell line) for 72h in vitro are 78.56%, 82.13% and 68.38% respectively, and the inhibition rates of aqueous extracts are only as follows: 28.7%, 38.03%, 29.34%. Through S180 cell tumor-bearing mouse experiments, the product can improve the micro-ecological environment in a mouse body, enhance the immunity of the mouse and effectively inhibit the growth of cancer cells in the animal body, and the inhibition rate of the product reaches 46.1 percent. And can improve the food intake, weight and mental state of the mice.
In conclusion, the anti-tumor capability of the Chinese wolfberry and honeysuckle co-extract can be effectively improved after the mixed fermentation of acetic acid bacteria, lactic acid bacteria and saccharomycetes. Compared with the traditional chemotherapy method, the fermented beverage has the advantages that the tumor inhibition rate is far higher than that of the traditional Chinese medicine extract although being lower than that of fluorouracil chemotherapy, the oral acceptability is high, the animal nutrition condition and the ingestion condition are obviously improved, and the intestinal flora can be protected and repaired.
The above detailed description of the fermented beverage of traditional Chinese medicine with anti-tumor effect and the preparation method thereof with reference to the embodiments is illustrative and not restrictive, and several embodiments can be enumerated according to the limited scope, so that changes and modifications without departing from the general concept of the present invention shall fall within the protection scope of the present invention.
