Description

Chitin-containing virus insecticidal suspending agent and preparation method thereof
Technical Field
The invention belongs to the technical field of biological insecticides, and particularly relates to a chitin-containing virus insecticidal suspending agent and a preparation method thereof.
Background
China is a big agricultural country, has various crop varieties and is a country with multiple and frequent biological disasters such as plant diseases and insect pests, and the plant diseases and insect pests have various varieties, high degrees, high frequency and wide regions, thereby causing great threat to the growth of crops in China. At present, agricultural and forestry pests are mainly prevented by chemical pesticides in China, the pests have drug resistance due to long-term and single use of the chemical pesticides, and in order to achieve the effect of pest control, the toxicity and the use amount of the chemical pesticides have to be improved, so that not only is great damage to food safety and ecological environment caused, but also a large number of natural enemies of the pests are killed, and the pests are rampant. Despite the above-mentioned negative effects, no more ideal alternative agents have been developed, and the continued use of chemical pesticides cannot be effectively prevented. Therefore, it is urgently required to develop an insecticide which is highly effective, free from contamination and free from side effects.
The main host of baculovirus is agriculture and forestry pests, and when the virus enters the insect body, the host insect can be infected by baculovirus and die. Polyhedrin is dissolved in the alkaline environment of insect midgut to release virus particles, quickly invade midgut cells and amplify in the order of magnitude in the cell nucleus. The amplified virus particles infect pests systemically along with the hemolymph circulation, and a large amount of virus particles are generated after all tissues of the pests are infected by the virus, and at the moment, as long as the outside lightly touches the pests, the virus particles can be instantly changed into liquid containing countless polyhedrosis viruses, so that the pests die. The liquid can be spread on the upper generation and the lower generation of pests, and has good lasting effect. Meanwhile, insect viruses have the advantages of no environmental pollution, no harm to natural enemies, difficult pest resistance generation and the like in production and use, so that the control of pests in agriculture and forestry by using baculovirus is an important development direction.
Chitin, also called chitin amine, chitosan and chitin, is the only homogeneous polysaccharide containing amino groups in nature and is named [ (1,4) -2-acetamido)2-deoxy- β -D-glucose]The molecular formula is (C)8H13NO5)nThe natural high molecular polymer is only positively charged in nature at present, belongs to straight-chain aminopolysaccharide and is also called glucosamine. Chitin is widely present in shells of lower plant fungi and marine invertebrates, the annual biosynthesis amount can reach 2000 hundred million tons, the yield in nature is only second to plant fibers, and the chitin is an inexhaustible resource for human beings. But this material has limited its widespread use because of its insoluble nature. Chitosan, a natural cationic polysaccharide, can be obtained after deacetylation of chitin, and the molecular formula is (C)6H11NO4)nThe molecular weight is about 10 ten thousand, and the polysaccharide is a polymer of glucose molecules, and the structure of the polysaccharide is similar to that of cellulose. Chitin and its derivatives have the characteristics of solubility, film-forming property, degradability, nontoxicity, adsorptivity, flocculation property, induced disease resistance, moisture retention, biocompatibility and the like, and can be widely applied to the industries of agriculture, industry, medicine, environmental protection and the like. In agricultural production, chitin has an insecticidal effect. In addition, it is widely recognized as a bio-regulator, promoting plant growth, increasing yield, improving quality, inducing broad-spectrum disease resistance of plants, and thus is known as "biomaterial of the 21 st century".

(2) and (2) adding chitin into the virus insecticidal suspending agent obtained in the step (1), and uniformly mixing to obtain the virus insecticidal suspending agent containing chitin.
Compared with the prior art, the virus insecticidal suspending agent provided by the invention has the following main advantages:
1. improves the insecticidal effect of the virus insecticide, is environment-friendly and has low cost
Chitin has insecticidal effect, and can be compounded with baculovirus insecticide to improve insecticidal effect of the insecticide. Researches find that chitin can be used as an exciton to induce the expression of a protein gene related to the disease course and induce plants to generate chitinase, and the chitinase can be used as an insecticide to directly control pests. Chitin is a major constituent of insect exoskeletons and peritrophic membranes, and typically accounts for 25-40% of the dry weight of insect body walls. The insect's peritrophic membrane and exoskeleton are important physiological barriers for insects to defend against pathogens, and chitin is an attractive potential target for insecticidal action in insects. Chitin can induce plants to generate chitinase, destroy the peritrophic membrane of insects, and promote the infection of insect viruses, thereby providing the insecticidal speed. In addition, chitin has certain light protection to insect viruses, so that the viruses are more stable in the field. Therefore, the compounding of the chitin and the virus insecticidal suspending agent obviously improves the insecticidal capacity of the insecticide. Chitin is a natural straight-chain amide polysaccharide, widely exists in the hard shell of crustaceans and the cell wall of fungi, and is one of the most abundant natural organic matters on the earth.
2. Can improve disease resistance of plant
Chitosan can inhibit the growth of various bacteria and has antifungal activity. The mechanism of the antibacterial is as follows: firstly, anions formed by amino groups of chitin and anions such as phospholipid and the like forming microbial cell walls attract each other to stimulate the freedom degree of microbes and cause fertility disorders; secondly, chitin is further degraded to enter the inside of microbial cells, so that the transcription of genetic factors DNA and RNA is blocked, and the birth inhibition is caused. The chitin can prevent the growth and development of plant pathogenic bacteria cells, induce host plants to protect pathogenic bacteria and improve the disease resistance of the plants.
3. Can improve soil property and culture soil fertility
Chitin contains a large amount of N, C elements, can be decomposed into plant nutrients by microorganisms in soil, can form a polymer film with good permeability, and improves the permeability, water retention, erosion resistance and tiltability of soil, thereby changing soil microorganism flora and inhibiting the growth and reproduction of soil pathogenic bacteria. Meanwhile, the chitin can also effectively improve the granular structure of soil and reduce the adverse effect of continuous cropping on the growth and development of crops, and is an ideal soil conditioner.
4. Increase the yield and improve the quality
Chitin can activate and enhance physiological and biochemical mechanisms of plants, promote root system cell meristem, promote root hair to occur in large quantities, make root systems more developed, and make absorption and antibiotic stronger, and chitin can induce root to secrete antibacterial substances, and prevent and treat diseases such as root rot. Researches show that when the chitin is sprayed on the leaf surfaces of plants, the photosynthesis can be improved, the accumulation of organic matters is promoted, and the chitin is particularly remarkably expressed on fruit and vegetable plants.

a virus insecticide suspending agent containing chitin comprises virus raw powder, chitin, a stabilizer, a surfactant, a synergist, a light protective agent, a dispersant and water, wherein:
the concentration of the virus raw powder in the virus insecticidal suspending agent is 200 hundred million PIB/ml;
the content of the stabilizer in the virus insecticidal suspending agent is 3 wt%;
the content of the surfactant in the virus insecticidal suspending agent is 5 wt%;
the content of the synergist in the virus insecticidal suspending agent is 0.2 wt%;
the content of the light protective agent in the virus insecticidal suspending agent is 0.1 wt%;
the content of the dispersing agent in the virus insecticidal suspending agent is 2 wt%;
the balance being sterile water.
The virus raw powder is diamondback moth granules;
the stabilizer is glycerol;
the surfactant is calcium lignosulfonate;
the synergist is chlorfluazuron;
the photo-protecting agent is tyrosine;
the dispersant is polyvinyl alcohol;
the components are added in sequence during preparation, the chitin is added after being stirred uniformly to enable the final concentration to be 30mg/L, and the cabbage looper nuclear polyhedrosis virus insecticidal suspending agent containing the chitin is obtained after being stirred uniformly and subpackaged. The prepared diamond back moth granulosis virus insecticidal suspending agent containing the chitin is poured into a hand-held sprayer, and the spraying is carried out until the plant leaves are completely wet. During the growth of the vegetables, treatment was performed once a week while using a cabbage looper nuclear polyhedrosis virus insecticidal suspension containing no chitin as a control (except for containing no chitin, the other formulation ingredients were the same as above).
TABLE 11 Effect of chitin-containing Heliothis virescens nuclear polyhedrosis virus insecticidal suspension on the quality and yield of cherry tomatoes
Figure BDA0001292620760000191
As can be seen from table 11, the chitin treatment significantly increased the yield of cherry tomatoes by 16.2% compared to the control group in the case of containing and not containing chitin. Meanwhile, the virus insecticide containing chitin can promote the increase of Vc content in tomato fruits, and is improved by 20.7 percent compared with a control group. The total acidity of tomatoes treated with the chitin-containing viral insecticide was slightly increased and the sugar-acid ratio was also reduced.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various changes and modifications can be made without departing from the overall concept of the present invention, and these should also be considered as the protection scope of the present invention.
