本发明目的在于提供一种具有增强皮肤细胞活力的中药提取物,该中药提取物能够增强皮肤细胞尤其是表皮细胞线粒体活性和/或提高表皮细胞ATP含量,可以作为功效添加剂加入到化妆品中使用,用于皮肤护理。
背景技术
皮肤由表皮、真皮和皮下组织构成,是人体最大的器官。皮肤覆盖全身,是保护人体的天然屏障,既可以保护人体内各组织和器官免受病原微生物等外界物质以及物理等机械损伤的侵袭,还可以保持人体的水分、电解质和营养物质等的流失。在有效保持人体内环境稳定的同时,皮肤还参与人体的新陈代谢过程。
新陈代谢是指机体与外界环境之间的物质和能量交换,以及生物体内物质和能量自我更新的过程,是生命现象最基本的特征之一,包括物质代谢和能量代谢两个方面。机体通过物质代谢,从外界摄取营养物质,经过体内分解吸收,将其中蕴藏的化学能释放出来,转化为组织和细胞可以利用的能量,维持机体正常的生命活动。
细胞的活力主要反映在其新陈代谢水平上。皮肤的新陈代谢旺盛,细胞活力充沛,皮肤就呈现富有弹性、自然、容光焕发的健康状态;皮肤的新陈代谢缓慢,细胞活力减退,就会出现皮肤老化、暗沉、敏感等肌肤问题。表皮是皮肤细胞新陈代谢的主要功能细胞,因此,促进皮肤新陈代谢,提高皮肤细胞的活力对于保持健康、年轻的皮肤是必须的。
线粒体是真核细胞的重要细胞器,是细胞氧化代谢的关键部位,是细胞产生ATP的重要场所,被称为“细胞的能量工厂”,其活性可以直接反映细胞的新陈代谢能力。线粒体可以促进细胞的更新,激励皮肤细胞功能性增强,增加细胞能量,从而增加皮肤的亮度和皮肤抗衰老的作用。表皮细胞中的线粒体,通过参与细胞的有氧代谢过程,为皮肤细胞的其他各项生理功能和化学反应提供能量,保持细胞的正常运行,从而保持皮肤细胞充沛的活力。若线粒体收到损伤或者失去活性,皮肤细胞能量不足,功能性降低,就会导致皮肤老化,产生皱纹,失去光泽。因此,表皮细胞中的线粒体活性大小是衡量细胞活力的重要指标。
ATP,即三磷酸腺苷,是各种活细胞内普遍存在的一种高能磷酸化合物,是细胞内的直接能源物质,常被称为“细胞中的能量通货”。细胞内的ATP在ATP酶的作用下,水解时可以释放出大量的能量,为细胞新陈代谢等化学反应的发生和进行提供动力。通过提高ATP水平,可以为肌肤细胞提供新生能量,使皮肤达到良好的新陈代谢平衡状态,全面改善肌肤状态,重新恢复自然、健康的肌肤状态。由于ATP主要由线粒体等细胞器产生,因此,通过检测细胞内ATP含量可以反映出细胞内线粒体活性的强弱。
随着生物细胞学的研究和发展,近年来研究细胞活力的方法有很多种,主要采用特殊试剂来测定细胞的代谢活力,包括Alamar blue、MTT及其他的四唑盐类等。Alamar blue是一种氧化还原指示剂,根据细胞代谢活性产生亮度变化和荧光信号,其操作简单、无毒性作用,近年来常用于细胞增殖和细胞活力的检测;MTT检测法主要反映细胞的能量代谢,它利用活细胞生长和增殖过程中,线粒体内的脱氢酶可将黄色的MTT分解成蓝紫色的甲臢的原理,检测细胞数量和活力的变化,是检测细胞增殖和活力的一种简便、准确的常用方法。但由于MTT法中生成的甲臢只溶于DMSO,不能溶于水,操作较为复杂,近年来,人们致力于可溶于水的甲臢盐的研究,如CCK-8法、XTT法等。此外,Calbiochem的快速细胞活力试剂盒利用线粒体剪切四唑盐试剂WST-1产生水溶性的formazan盐进行测定;目前还有一种荧光素酶检测细胞内ATP水平来评价细胞或细胞内的线粒体活力。
目前,针对可增强表皮细胞活力或提高表皮细胞ATP含量的中药研究较少。迄今为止,关于能够促进细胞活力和/或提高表皮细胞ATP活力的中药提取物的研究尚未见报道。
雪莲花为菊科凤毛菊属植物大苞雪莲花Saussurea involucrata(Kar.etKir.)Sch.-Bip.、绵头雪莲花Saussurea laniceps Hand.-Mazz.、水母雪莲花Saussurea medusa Maxim.等的带花全株。6~7月间开花时采收,拔起全株,除去泥沙,晾干。别名:雪莲(《西北域记》),雪荷花(《纲目拾遗》),大拇花(《修订增补天宝本草》),大木花(《四川中药志》)。大苞雪莲花也称天山雪莲、新疆雪莲分布于新疆、青海、甘肃等地。雪莲花民间以全草入药,具有活血通经,散寒除湿,强筋助阳等功效。
雪莲花主要含有黄酮类、生物碱类、木脂素类等成分。药典对雪莲花Saussurea involucrata(Kar.et Kir.)Sch.-Bip.药材的评价指标成分为芦丁和绿原酸。有文献报道,天山雪莲中总黄酮平均含量22.30mg/g,芦丁含量3.62mg/g。现代研究表明,雪莲花具有抗光老化、抗氧化、促进胶原蛋白合成、抗辐射、抗炎等多种药理作用。
中医认为雪莲花味苦、辛,性温热,入肝、脾、肾三经,因其性热能温补助阳,御寒除湿,推测对精气流失所致的虚寒性衰老肌肤有着良好的温通补益作用,可以提升肌肤内在能量,促进肌肤细胞的代谢活力;另外其独特的耐寒耐缺氧耐辐射的生存特征,也赋予了它在防晒修护、抗氧化损伤等方面的应用潜能。
因此,本申请首次以雪莲花为研究对象,考察其提取物对皮肤细胞活力的促进作用。
发明内容
本发明首次发现雪莲花提取物对于皮肤细胞活力具有促进作用,表现在能够增强表皮角质细胞线粒体活性和/或提高表皮细胞ATP含量。
一方面,本发明提供了一种雪莲花提取物,以所述雪莲花提取物的总重量计,所述雪莲花提取物中的固含量大于10重量%。在优选的实施方式中,本发明的雪莲花提取物中的固含量为10.87-12.02%重量%。
本发明所述的固含量为雪莲花提取后获得的浓缩液中的干物质的含量。
在一个具体的实施方式中,本发明雪莲花提取物的使用浓度为1mg/mL。
另一方面,本发明提供了雪莲花提取物在促进皮肤新陈代谢和/或提高细胞的活力中的应用。在一个具体的实施方式中,所述促进皮肤新陈代谢和/或提高细胞的活力为通过增强细胞线粒体活性达到。在一个具体的实施方式中,所述促进皮肤新陈代谢和/或提高细胞的活力为通过提高细胞ATP含量达到。
另一方面,本发明提供了雪莲花提取物的提取方法:将雪莲花粉碎成粗粉,乙醇浸提,过滤,滤液减压浓缩,过滤,得到雪莲花提取物。
另一方面,本发明还涉及雪莲花提取物作为具有增强皮肤细胞活力作用的功效添加剂,雪莲花提取物可应用于皮肤外用剂、保健食品和医药,尤其是可应用于化妆品中,根据制剂的不同类型添加不同的用量。
在一个实施方式中,所述化妆品选自:洁面乳、化妆水、乳液、膏霜、啫喱、面膜。
在一个实施方式中,化妆品中雪莲花提取物的用量在0.0001-20重量%。在另一个实施方式中,化妆品中雪莲花提取物的用量在0.001-20重量%。在优选的实施方式中,化妆品中雪莲花提取物的用量为0.001%-10重量%。
所述皮肤外用剂组合物是通常用于皮肤外部的所有成分的统称概念,例如可以是化妆料组合物或药学组合物。所述化妆料组合物中可以是基础化妆料、面部妆容化妆料、身体用化妆料、头发护理用化妆料等,对其剂型无特殊限制,根据不同目的可合理选择。
所述化妆料组合物中根据剂型和目的的不同还含有不同的化妆品学层面允许的介质或基质赋形剂。
可以用于本发明皮肤外用剂组合物的化妆品、皮肤病学或药学上可接受的赋形剂为水相、油相、凝胶、水包蜡型乳液、水包油型乳液或油包水型乳液的形式。水相为一种或多种水溶性或分散性组分的混合物,其在室温(25℃)下可以为液体、半固体或固体。赋形剂包括或可以为在水或水-醇赋形剂中的混悬液、分散液或溶液的形式,其可以含有增稠剂或凝胶剂。本领域技术人员可以基于本领域技术人员掌握的知识选择合适的产品形式,其中包含的组分。
所述的组合物可以包括水相,该水相可以含有水或水与至少一种亲水性有机溶剂的混合物,所述的亲水性有机溶剂诸如醇,尤其是含有2-5个碳原子的直链或支链低级一元醇,如乙醇或丙醇;多元醇,如丙二醇、山梨醇、甘油、泛醇或聚乙二醇及其混合物。
当发明的组合物为乳液形式时,该组合物还可以任选包含表面活性剂。
所述的组合物还可以包含成膜聚合物,如聚氨基甲酸酯、聚丙烯酸均聚物或共聚物、聚酯、基于烃的树脂和/或硅氧烷树脂。可以将聚合物溶于或分散于化妆品可接受的赋形剂中并且任选与增塑剂合并。
本发明的组合物还可以包含油相,所述的油相含有在室温(25℃)下为液体的油溶性或油分散性组分和/或在室温下为油状或蜡状的物质,如蜡、半固体、树胶及其混合物。该油相还可以含有有机溶剂。
通常在室温下为液体,合适的油性物质包括:来源于动物的基于烃的油,如全氢化角鲨烯;基于烃的植物油,如液体的C4-10脂肪酸的甘油三酯类,例如庚酸或辛酸甘油三酯类,或油,例如向日葵油、玉米油、大豆油、葡萄籽油、蓖麻油、鳄梨油、辛酸/癸酸甘油三酯类、霍霍巴油;矿物或合成来源的直链或支链烃类,例如液体石蜡及其衍生物、凡士林;合成酯类和醚类,特别是脂肪醇的酯类,例如肉豆蔻酸异丙酯、棕榈酸2-乙基己酯、硬脂酸2-辛基十二烷基酯、异硬脂酸异硬脂醇酯;羟基化酯类,例如乳酸异硬脂醇酯、羟基硬脂酸辛酯、羟基硬脂酸辛酯、羟基硬脂酸辛基十二烷基酯、脂肪醇的庚酸酯类、辛酸酯类和癸酸脂类;多元醇酯类,例如丙二醇二辛酸酯、新戊二醇二庚酸酯、二甘醇二异壬酸酯和季戊四醇酯类;含有C12-26的脂肪醇类,例如辛基十二烷醇、2-丁基辛醇、2-己基癸醇、2-十一烷基十五烷醇、油醇;基于部分烃的氟油和/或氟硅油,硅油,在室温下为液体或半固体的挥发性或非挥发性的直链或环状聚甲基硅氧烷,例如环状聚二甲基硅氧烷和聚二甲基硅氧烷,其任选包含苯基,例如苯基三甲基硅氧烷、硅氧烷及其混合物。
本发明的组合物可以进一步包含常用于化妆品领域中的任何组分。这些组分包括防腐剂、水相增稠剂(提取物生物聚合物、合成聚合物)和脂肪相增稠剂、芳香剂、亲水性和亲脂性活性剂及其混合物。
本发明的组合物还可以包含另外的颗粒相,所述的颗粒相可以为化妆品组合物中使用的颜料和/或珠光剂和/或填充剂。
颜料可以存在于组合物中,合适的无机颜料包括氧化钛、氧化锆和氧化铈以及氧化锌、氧化铁和铁蓝;合适的有机颜料包括钡、锶、钙和铝色淀和碳黑。
珠光剂可以存在于组合物中,合适的珠光剂包括涂覆了氧化钛、氧化铁或天然颜料的云母。
填充剂可以存在于组合物中,合适的填充剂包括滑石粉、二氧化硅、硬脂酸锌、云母、高岭土、尼龙粉末、聚乙烯粉末、特氟龙、淀粉、一氮化硼、共聚物微球,例如硅氧烷树脂微珠。
本发明组合物的油相可以包含一种或多种蜡、树胶或其混合物。蜡包括基于烃的蜡、氟蜡和/或硅氧烷蜡,并且可以来源于植物、矿物、动物和/或合成来源。合适的蜡包括蜂蜡、巴西棕榈蜡、小烛树蜡、石蜡、微晶蜡、地蜡;合成蜡包括聚乙烯蜡、含有C16-45的硅氧烷蜡。树胶一般为聚二甲基硅氧烷或羧甲基纤维素钠或提取物类,并且半固体物质一般为基于烃的化合物,如羊毛脂及其衍生物。
可以将本发明的组合物配制成任何合适的产品形式。这类产品形式包括,但不限于气溶胶型喷雾剂、霜剂、乳液、固体、液体、分散体、泡沫、凝胶、化妆水、摩丝、软膏、粉剂、贴剂、润发油、溶液、手按泵型喷雾剂、棒状物、面膜和湿纸巾。可以将本发明的组合物通过本领域众所周知的各种方法便利地用于制备或作为化妆品、皮肤病学或药物局部施用产品。
本发明的皮肤外用剂组合物可以包括一种或多种下列成分:抗过敏剂、抗微生物剂、抗氧化剂、螯合剂、着色剂去色素剂、润肤剂、乳化剂、表皮脱落剂、成膜剂、香料、保湿剂、昆虫驱避剂、润滑剂、药物活性剂、增湿剂、耐光剂、防腐剂、护肤剂、皮肤渗透增强剂、防晒剂、稳定剂、表面活性剂、增稠剂、粘度调节剂、维生素或其任意组合。
具体实施方式
本发明的发明人在经过了广泛而深入的研究之后发现:雪莲花提取物具有增强皮肤细胞,尤其表皮角质细胞线粒体活性和/或提高表皮细胞ATP含量的作用。因此,雪莲花提取物可作为功效添加剂加入到皮肤外用剂,尤其化妆品中,增强细胞活力,用于皮肤护理。
文献中均未涉及雪莲花提取物具有增强皮肤细胞,尤其表皮角质细胞线粒体活性和/或提高表皮细胞ATP含量的作用,也未提及将其作为功能添加剂加入化妆品中。因此,本发明首次提出雪莲花提取物具有增强细胞活力的作用,同时该提取物也可以作为功能添加剂加入皮肤外用剂,尤其化妆品中,用于皮肤护理。
下面结合具体的实施例进一步阐述本发明。但是,应该明白,这些实施例仅用于说明本发明而不构成对本发明范围的限制。下列实施例中未注明具体条件的试验方法,通常按照常规条件,或按照制造厂商所建议的条件。除非另有说明,所有的百分比和份数按重量计。
以下实施例中采用的雪莲花均购自中国亳州市京皖中药饮片厂。
实施例1:雪莲花提取物的制备
雪莲花粗粉→70%乙醇(体积浓度)常温浸提→过滤→滤液减压浓缩→过滤→定量至1倍生药质量,即得雪莲花提取物。生药浓度1g/g,固含量12.02%。
实施例2:雪莲花提取物的制备
雪莲花粗粉→50%乙醇(体积浓度)常温浸提→过滤→滤液减压浓缩→过滤→定量至1倍生药质量,即得雪莲花提取物。生药浓度1g/g,固含量10.87%。
实施例3:雪莲花提取物的制备
雪莲花粗粉→80%乙醇(体积浓度)常温浸提→过滤→滤液减压浓缩→过滤→定量至1倍生药质量,即得雪莲花提取物。生药浓度1g/g,固含量11.25%。
实施例4:对表皮角质细胞(HaCaT)线粒体活性的影响
取实施例1-3所得雪莲花浓缩液作为体外功效测试样品,小体积分装后于-20℃保存,每次使用时用细胞培养液稀释,过滤除菌后进行细胞实验。
XTT[2,3-双(2-甲氧基-4-硝基-5-磺苯基)-5-[(苯基氨基)羰基]-2H-四唑鎓盐氢氧化物],是一种四唑氮衍生物,能被活细胞中的线粒体脱氢酶降解还原成水溶性的棕黄色甲臜产物,甲臜的生成量与细胞的线粒体活性呈正相关,故通过XTT法可以用于检测线粒体活性,从而反映细胞的新陈代谢能力。
将HaCaT细胞接种于含DK—SFM培养液的培养瓶中,置于37℃,CO:(体积分数为5%)恒温培养箱中培养5天至细胞融合,隔天换培养液。实验时,将细胞培养瓶取出,用胰蛋白酶将HaCaT细胞消化4min,然后将细胞接种于96孔板孵育24h,加药组每孔加入200μl用培养液稀释的0.1mg/mL固含量的实施例样品,对照组每孔直接加入200μl培养液,于培养箱中培养48h后进行测定。以进行加药孵育,用60℃预热的培养液将XTT溶解,配制成0.2mg/mL的溶液,将96孔板中的细胞培养液移除,每孔中加入XTT/PMS(200:1)后,于37℃条件下培养2h,在酶标仪上450nm波长处测定吸光度值OD。结果显示(表1)。

注:OD/OD0为样品与阴性对照在450nm处吸光度比值。
由实验数据可知,相比阴性对照细胞OD值为1,不同实施例样品在不同浓度下对处理过的细胞线粒体活力均有不同程度的作用,其中实施例1样品在浓度为0.1mg/mL时,OD值为1.1530,提升比例最高,达到17.1%,,能够显著增强表皮角质细胞的线粒体活性(*P<0.05)。
实施例5:对表皮角质细胞(HaCaT)ATP含量的影响
ATP,主要由线粒体等细胞器产生,是细胞内的直接能源物质,其含量与细胞内线粒体活性呈正相关。
将HaCaT细胞接种于含DK—SFM培养液的培养瓶中,置于37℃,CO:(体积分数为5%)恒温培养箱中培养5天至细胞融合,隔天换培养液。实验时,将细胞培养瓶取出,用胰蛋白酶将HaCaT细胞消化4min,然后将细胞接种于96孔板孵育24h,加药组每孔加入200μl用培养液稀释的0.1mg/mL固含量的实施例样品,对照组每孔直接加入200μl培养液,于培养箱中培养48h后进行测定。ATP含量:采用细胞能量检测包检测细胞内ATP含量,操作方法按照试剂盒说明书进行。三磷酸腺苷(ATP)细胞能量检测包,LuminUltra公司。

ATP含量检测结果显示(表2),相比阴性对照细胞,不同实施例在浓度为0.1mg/mL时,对处理过的细胞ATP含量均有不同程度提升,其中实施例1样品提升比例达到35.2%,能够显著增强表皮角质细胞的ATP含量(*P<0.05)。
通过实施例4和5的实验结果表明,实施例雪莲花样品浓度为0.1mg/mL时,可以显著增强表皮角质细胞的线粒体活性,并增强表皮角质细胞的ATP含量。即可以有效提高表皮细胞的活力和能量代谢水平。
取实施例1-3中制备的雪莲花提取物用于皮肤外用剂的制备。所述皮肤外用剂优选为化妆品组合物,例如化妆水、精华液、乳霜等。所述雪莲花提取物在皮肤外用剂中的重量百分比为0.0001%-20%(w/w)。优选的重量百分比为0.001%-20%(w/w)。更优选的重量百分比为0.001%-10%(w/w)。最优选的重量百分比为0.01%-10%(w/w)。
以下是雪莲花提取物在皮肤外用剂中的具体应用的实施例,及其这些剂型的配方和制备方法。以下各表中“-”表示无添加。

Claims (9)
1.一种雪莲花提取物,其特征在于,以所述雪莲花提取物中的总重量计,所述雪莲花提取物的固含量为10.87-12.02重量%。
2.如权利要求1任所述的雪莲花提取物,其特征在于,所述雪莲花提取物通过以下方法提取得到:将雪莲花粉碎成粗粉,乙醇浸提,过滤,滤液减压浓缩,过滤,得到雪莲花提取物。
3.如权利要求2所述的雪莲花提取物,其特征在于,所述乙醇的体积浓度为50-80%。
4.如权利要求1-3任一项所述的雪莲花提取物在促进皮肤新陈代谢和/或提高细胞的活力中的应用。
5.如权利要求4所述的应用,其特征在于,所述促进皮肤新陈代谢和/或提高细胞的活力为通过增强细胞线粒体活性达到。
6.如权利要求5所述的应用,其特征在于,所述促进皮肤新陈代谢和/或提高细胞的活力为通过提高细胞ATP含量达到。
7.如权利要求4-6任一项所述的应用,其特征在于,所述细胞为表皮细胞。
8.一种含有如权利要求1-3任一项所述的雪莲花提取物的皮肤外用剂,其特征在于,所述雪莲花提取物在皮肤外用剂中的重量百分比为0.0001%-20%(w/w),优选的重量百分比为0.001%-20%(w/w),更优选的重量百分比为0.001%-10%(w/w),最优选的重量百分比为0.01%-10%(w/w)。
9.如权利要求1-3任一项所述雪莲花提取物作为功效添加剂在皮肤外用剂、保健食品、药品中的应用。
