Background
Alzheimer's Disease (AD) is a progressive degenerative disease of the nervous system with occult onset. Clinically, it is characterized by generalized dementia such as dysmnesia, aphasia, disuse, agnosia, impairment of visual spatial skills, dysfunction in execution, and personality and behavioral changes. Patients who are older than 65 years are called presenile dementia; the patient after 65 years old is called senile dementia.
The disease is slow in onset or hidden, and when the disease is discovered, symptoms such as decline of cognitive function, mental symptoms, behavior disorder and decline of daily life capacity appear, serious patients completely depend on nursing persons, memory is lost, daily life cannot be self-managed, incontinence of urine and feces appears as mutism, limb stiffness, coma is finally caused, and the disease generally dies from complications such as infection.
From the current research, the possible factors and hypothesis of the disease are as many as 30, such as family history, female, head trauma, low education level, thyroid disease, maternal age, viral infection, neurotransmitter disturbance, gene mutation, free radical damage, neuronal apoptosis, deposition of amyloid β protein and abnormal phosphorylation of tau protein.
Alzheimer's Disease (AD) is clinically manifested by age-related decline in memory and cognitive function, pathologically characterized by intracellular neurofibrillary tangles (NET), formation of extracellular amyloid (a β) plaques and loss of cholinergic nerves, etc.

Vacuum concentrating the filtrate to relative density of above 1.20, vacuum drying for 12 hr to obtain 0.21kg extract, mixing the extract with microcrystalline cellulose 25 wt%, starch 15 wt% and crospovidone 2 wt%, making soft mass with povidone ethanol solution 3 wt% as binder, granulating, drying at 60 deg.C, grading, and encapsulating to obtain Chinese medicinal extract capsule.
Example 4:
new Object discrimination (NOR) experiment examines the influence of Chinese herbal medicine extract on the discrimination ability of new objects in mice with learning and memory impairment induced by LPS.
Materials animals: ICR male mouse, 18-22 g;
medicine preparation: lipopolysaccharide (LPS); chinese herbal medicine extract; donepezil hydrochloride;
the method comprises the following steps: herbal extracts were prepared as described in example 1. ICR mice were taken and randomly divided into 6 groups of 12 mice each: a blank control group, a model control (LPS) group, a Chinese herbal medicine extract group and a positive control group (donepezil hydrochloride). Animals in each group were lightly anesthetized with chloral hydrate and then fixed in their heads for lateral ventriculo-ventricular injection. Model group, Chinese herbal medicine extract low, medium and high dose group (100,200,400mg/kg) and positive control group (donepezil 5mg/kg) were injected with LPS solution (5mg/ml, 3. mu.l/mouse), and blank control group mice were injected with an equal volume of physiological saline. From the next day after LPS injection, the mice of each Chinese herbal medicine extract administration group are intragastrically administered with the solution prepared by the Chinese herbal medicine extract every day according to the weight of the mice, the mice of the positive control group are intragastrically administered with the donepezil hydrochloride solution every day, and the mice of the blank group and the model group are intragastrically administered with the physiological saline with the same volume every day. A novelty discrimination experiment was started on day 8 and the preference index for each mouse was determined. The calculation formula is as follows:
Figure BDA0001164220670000071
as a result: in the NOR experiment, the preference index of mice for new things was significantly reduced by injecting LPS into the lateral ventricle. After the Chinese herbal medicine extract and the positive drug donepezil hydrochloride of 5mg/kg are treated, the preference of mice for new things can be obviously increased. Experimental results show that the Chinese herbal medicine extract can obviously improve the discrimination capability of a new object of a mouse with learning and memory impairment induced by LPS.
Example 5:
the Morris water maze experiment inspects the influence of the Chinese herbal medicine extract on the escape latency of the mouse with learning and memory impairment induced by LPS;
materials animals: ICR male mouse, 18-22 g;
medicine preparation: LPS; chinese herbal medicine extract; donepezil hydrochloride;
the method comprises the following steps: ICR mice were taken and randomly divided into 6 groups of 12 mice each: a blank control group, a model control (LPS) group, a Chinese herbal medicine extract group and a positive control group (donepezil hydrochloride). The Morris water maze experiment was started on day 10 for 5 days. The escape latency of the mice in the experiment of the positioning voyage experiment is determined.
As a result: in the Morris water maze experiment, the escape latency of LPS group mice on the 5 th day of the localized voyage experiment was significantly increased compared to the blank group. The escape latency of the herbal extract group and the positive drug donepezil hydrochloride group was significantly reduced on day 5 compared to the LPS group. There was no significant difference in swimming speed between the groups of animals during the experiment. The experimental result shows that the Chinese herbal medicine extract can obviously improve the space learning and memory ability of mice with learning and memory impairment induced by LPS.
The above description is only for the purpose of illustrating the preferred embodiments of the present invention and is not to be construed as limiting the invention, and any modifications, equivalents and improvements made within the spirit and principle of the present invention are intended to be included within the scope of the present invention.
