下面将结合实施例对本发明的实施方案进行详细描述,但是本领域技术人员将会理解,下列实施例仅用于说明本发明,而不应视为限制本发明的范围。实施例中未注明具体条件者,按照常规条件或制造商建议的条件进行。所用试剂或仪器未注明生产厂商者,均为可以通过市售购买获得的常规产品。
本实施方式提供了马尼地平在制备治疗黄病毒科病毒感染的药物中的应用。黄病毒科病毒包括三个病毒属,即黄病毒属、瘟病毒属和丙型肝炎属。由于黄病毒在病毒体形态结构、传播媒介以及所致疾病等方面与披膜病毒科中的甲组病毒相类似,曾经被归入披膜病毒科的黄病毒属。根据黄病毒的基因组结构、复制方式及形态发生学等方面的特点,1986年被国际病毒命名委员会独立命名为黄病毒科。黄病毒科病毒呈小球形,直径多数为40-70nm,该病毒表面有脂质包膜,其上镶有糖蛋白组成的刺突,包膜内为20面体对称的核衣壳蛋白,黄病毒基因组核酸为单股正链RNA,黄病毒均在细胞质中增殖;黄病毒对热、脂溶剂和去氧胆酸钠敏感,在PH为3-5的条件下不稳定。
马尼地平的化学名为1,4-二氢-2,6-二甲基-4-间硝基苯基-3,5-吡啶二羧酸甲酯2-(4-二苯甲基-1-哌嗪基)乙酯,其化学结构式如式I所示为:
发明人研究发现,马尼地平在无毒性范围内能够有效预防和治疗由黄病毒科病毒引起的病毒感染;且马尼地平已经在临床上有数十年的应用,安全性良好。因此,可进一步开发马尼地平为治疗或预防黄病毒感染的药物,具有广泛的应用前景。
具体地,马尼地平在制备治疗黄病毒属病毒感染的药物中的应用。黄病毒属病毒是黄病毒科病毒的一种,是具有包膜的单正链RNA病毒。这类病毒通常通过吸血的节肢动物(蚊、蜱、白蛉等)传播。研究发现,马尼地平对黄病毒属病毒的抗感染能力较强。
更为具体地,马尼地平在制备治疗流行性乙型脑炎病毒感染的药物中的应用。流行性乙型脑炎病毒属于黄病毒科,黄病毒属,单股正链RNA病毒。流行性乙型脑炎病毒简称乙脑病毒,国外亦称为日本脑炎病毒,是通过蚊叮咬传播,引起乙型脑炎的病原体。乙脑是严重威胁人体健康的一种急性传染病,临床上急起发热,出现不同程度的中枢神经系统症状,重症者病后常留有后遗症。发明人在研究中发现,马尼地平能够有效的抑制流行性乙型脑炎病毒在宿主细胞内的复制,从而能够预防和治疗该病毒感染引起的流行性乙型脑炎。
更为具体地,马尼地平在制备治疗寨卡病毒感染的药物中的应用。寨卡病毒属于黄病毒科,黄病毒属,单股正链RNA病毒,是一种通过蚊虫进行传播的虫媒病毒。流行病学和血清学研究表明,大约20%的ZIKV感染者表现出临床症状,临床症状比较轻微并且通常自愈,表现为体温稍升高、关节疼、皮疹、结膜炎、头疼、肌肉疼。目前尚无有效的疫苗和药物用于ZIKV病毒感染的预防和治疗。发明人在研究中发现,马尼地平能够有效的抑制寨卡病毒在宿主细胞内的RNA复制,从而能够预防和治疗该病毒感染引起的相关症状。
更为具体地,马尼地平在制备治疗西尼罗病毒感染的药物中的应用。西尼罗病毒属于黄病毒科,黄病毒属,单股正链RNA病毒,是一种脑炎病毒,以鸟类为主要的贮存宿主,马、蚊子和人都可以是它的传染宿主。绝大多数为隐性感染,不出现任何症状,少数人表现为西尼罗河热,病人出现发热、头疼、肌肉疼痛、恶心、呕吐、皮疹、淋巴结肿大等类似感冒症状,持续3-6天。极少数人感染后表现为西尼罗河病毒性脑炎、脑膜脑炎和脑膜炎。目前还没有针对西尼罗河病毒的特效药,保护人类的相关疫苗也尚未问世。发明人在研究中发现,马尼地平能够有效的抑制西尼罗病毒在宿主细胞内的RNA复制,从而能够预防和治疗该病毒感染引起的相关症状。
更为具体地,马尼地平在制备治疗登革病毒感染的药物中的应用。登革病毒属于黄病毒科,黄病毒属,单股正链RNA病毒。登革病毒根据其抗原性的差异分别为DENV-1、DENV-2、DENV-3、DENV-4,四种血清型均能引起登革热,表现出发热、皮疹、头疼、肌肉和关节疼痛、恶心等症状。登革病毒感染后引起不同严重程度的症状,有些病毒株感染后为无症状的隐性感染,有些病毒株可造成严重的登革出血热以及可能以死亡转归的登革休克综合征,造成血管渗漏、出血、以及休克等危及生命的病症。在过去的十年中的,登革热的暴发频率逐年增加,受威胁的人口数量也不断增加。目前尚无安全、有效的登革病毒疫苗,也没有针对登革病毒的特效药。发明人在研究中发现,马尼地平能够有效的抑制登革病毒在宿主细胞内的RNA复制,从而能够预防和治疗该病毒感染引起的相关症状。
进一步地,上述的药物均是以(1,4-二氢-2,6-二甲基-4-间硝基苯基-3,5-吡啶二羧酸甲酯2-(4-二苯甲基-1-哌嗪基)乙酯作为药物活性成分制成的剂型。上述药物的剂型为片剂、胶囊、颗粒剂、口服液、缓释制剂、控释制剂、纳米制剂或注射剂。其中,药物中还可以包括辅料,辅料选自增溶剂、助溶剂、乳化剂、粘合剂、崩解剂、填充剂、润滑剂、渗透压调节剂、稳定剂、矫味剂、助悬剂、渗透促进剂、增塑剂、表面活性剂、发泡剂、包合剂、包衣材料中的任意一种或几种。上述药物的剂型为剂、胶囊、颗粒剂、口服液、缓释制剂、控释制剂、纳米制剂或注射剂。
为了更好的了解本发明的实质,下面将用马尼地平在治疗黄病毒科病毒感染的药物的药理实验及结果来说明其在制药领域中的新用途。
实施例1
本实验利用MTT法测定马尼地平对Vero细胞(非洲绿猴肾细胞)的细胞活性的影响。
(1)实验过程
取制备好的Vero细胞悬液(1.0×105个/mL),按照100μL/孔加入到96孔细胞培养板;将马尼地平溶解并在含有质量体积比为2%FBS的DEMN培养基中梯度稀释,分别将终浓度为100μM、50μM、25μM、6.25μM的含有马尼地平的培养基溶液加入到96孔细胞培养板中;同时以含有质量体积比为2%FBS的DEMN培养基溶液作为空白对照。每组重复3孔,于37℃、5%CO2的细胞培养箱中培养24h。
吸弃细胞上清,每孔加入25μL,5mg/mL的MTT溶液,继续在37℃、5%CO2下的细胞培养箱中培养4h;Vero细胞孵育完毕后,吸弃残余MTT溶液,每孔加入DMSO 50μL;在室温下充分溶解15min后,通过酶联免疫检测仪在492nm波长下检测每孔的吸光度值,并根据以下公式计算各孔吸光度值与对照组吸光度值的比值:
细胞存活率(%)=(实验组OD492/对照组OD492)×100%
从而计算各个浓度的马尼地平对细胞活性的影响。
(2)实验结果
如图1所示,根据细胞活性的曲线图可知,马尼地平对于Vero细胞,其CC50在100μM以上,CC50是使得50%的细胞发生病变时的药物浓度;另外,根据实验测定马尼地平的EC50浓度为2.6μM,EC50是指能够有效抑制50%细胞感染病毒的药物浓度。有以上数据可得,马尼地平的安全指数为38.46,因此可见马尼地平的毒性低、疗效高、用药比较安全,为马尼地平在药物应用的安全性方面提供了参考。
实施例2
本实验利用噬斑实验检测马尼地平对JEV(流行性乙型脑炎病毒)的抑制效果。
(1)实验过程
用胰酶消化Vero细胞(1.0×105个/mL),并用细胞计数仪技术,按照100μL/孔加入到96孔细胞培养板;于37℃、5%CO2的细胞培养箱中培养16h后,吸弃上清,分别将终浓度为0μM、2.5μM、5μM、10μM的马尼地平溶液通过12通道排枪加入到对应的细胞孔中,每组重复三孔,并在37℃、5%CO2的细胞培养箱中培养1h;每孔加入40μL JEV(MOI=0.2),每隔15min摇匀一次,感染时间为1h;吸弃上清,用PBS洗涤3次,再分别将含有以上不同浓度的马尼地平的维持培养基加入到相应的细胞孔中,继续培养24h;培养24h后,收取细胞上清作为样品,冻存于-80℃的冰箱中。
将BHK-21细胞(乳仓鼠肾细胞)悬液(1.0×105个/mL)按照2000μL/孔加入到12孔细胞培养板中,于37℃、5%CO2的细胞培养箱中培养16h;将上述的各组样品分别做10倍梯度稀释,然后在12板孔的各个孔中分别加入各组样品进行感染,感染1h,在此期间每隔15min摇匀一次;感染完成后,吸弃上清,在12孔细胞培养板的各个孔中均加入质量体积比为1.5%的甲基纤维素和质量体积比为2%的FBS培养基溶液,于37℃、5%CO2的细胞培养箱中培养72h;取出细胞板,吸弃细胞覆盖物,加入体积分数为3.7%的甲醛溶液,固定细胞12h;吸弃甲醛溶液,加入质量体积比为0.6%的结晶紫染色液,处理细胞30min;回收结晶紫染色液,用自来水冲洗细胞板,并在烘箱中烘干,随后拍照并统计噬斑个数,并根据以下公式计算JEV的滴度:
滴度(PFU/mL)=(X1+X2+…Xn)·d/n·v
X1、X2、Xn:表示同一稀释度在不同的96孔细胞培养板中得到的噬斑数;n表示计数的培养板孔数;v表示病毒量(mL);d表示稀释倍数。
(2)实验结果
如图2所示,马尼地平在0μM、2.5μM、5μM以及10μM的JEV滴度分别为2.465×107PFU/mL、3.747×106PFU/mL、7.067×104PFU/mL、3.467×103PFU/mL。根据图2和各个浓度下JEV滴度的数据分析可知,马尼地平在2.5μmol/L-10μmol/L的浓度范围内可明显降低JEV病毒的滴度,并能够剂量依赖性地抑制JEV对宿主细胞的感染,由此可知马尼地平对JEV感染有抑制效果。
实施例3
本实验利用荧光素酶活性实验检测马尼地平对WNV(西尼罗病毒)的感染情况。
(1)实验过程
用胰酶消化Vero细胞(1.0×105个/mL),并用细胞计数仪技术,按照100μL/孔加入到96孔细胞培养板;于37℃、5%CO2的细胞培养箱中培养16h后,吸弃上清,分别将终浓度为2.5μmol/L、5μmol/L的马尼地平溶液通过12通道排枪加入到对应的细胞孔中,每组重复三孔,并在37℃、5%CO2的细胞培养箱中培养1h;每孔加入40μL WNV病毒(MOI=0.2),每隔15min摇匀一次,感染时间为1h;吸弃上清,用PBS洗涤3次,再分别将含有以上不同浓度的马尼地平的维持培养基加入到相应的细胞孔中,继续培养24h。
培养24h后,吸弃细胞上清,并用PBS洗涤细胞一次;吸弃PBS,并在96孔细胞培养板每孔加入50μL的1×荧光素酶裂解缓冲液;将96孔细胞培养板在摇床上振荡10min,再于-80℃的冰箱中冷冻30min,充分裂解细胞;将100×荧光酶底物加入到1×测定缓冲液中,并避光保存;将20μL细胞裂解液加入到50μL测定缓冲液中,并快速混匀两次,立即测定细胞荧光素酶活性。
(2)实验结果
如图3所示,荧光素酶活性反应了WNV的感染水平,即荧光素酶活性越高,WNV的感染能力越强。本实施例通过荧光素酶活性检测实验得到马尼地平溶液浓度为2.5μM、5μM时的荧光素酶活性分别为0.810和0.459,由此可知,马尼地平可有效对抑制WNV对宿主细胞抑制的感染。
实施例4
本实验利用QPCR(实时定量基因扩增荧光检测系统)来检测马尼地平对ZIKV(寨卡病毒)的基因组复制水平。
(1)实验过程
用胰酶消化Vero细胞(1.0×105个/mL),并用细胞计数仪技术,按照100μL/孔加入到96孔细胞培养板;于37℃、5%CO2的细胞培养箱中培养16h后,吸弃上清,分别将终浓度为5μM、10μM的马尼地平溶液通过12通道排枪加入到对应的细胞孔中,每组重复三孔,并在37℃、5%CO2的细胞培养箱中培养1h;每孔加入40μL ZIKV病毒(MOI=0.2),每隔15min摇匀一次,感染时间为1h;吸弃上清,用PBS洗涤3次,再分别将含有以上不同浓度的马尼地平的维持培养基加入到相应的细胞孔中,继续培养24h;培养24h后,收取细胞上清,并冻存在-80℃冰箱中。
在各组的样品中均加入1000μL Trizol裂解液来裂解待检测的细胞,反复用移液枪吹打或者剧烈震荡以充分裂解Vero细胞;加入200μL氯仿,并剧烈振荡15s,使得两个溶液相充分接触;于13000g(4℃)离心15min,取上清(不能接触到中间相或下层溶液)至新的EP管中;在EP管中加入等体积的异丙醇,并充分混匀,在室温放置10分钟;将EP管在13000g(4℃)离心15min;轻轻倾倒或吸出上清,并加入1mL预冷的用DEPC水配制含75%乙醇的溶液,并将EP管在7500g(4℃)离心5min,轻轻倾倒或吸出上清;用10μL枪头吸出残余溶液,待溶液完全挥发后,加入20μL无核酸酶水,并充分溶解RNA沉淀;再通过逆转录和QPCR来检测ZIKV的基因复制水平。
(2)实验结果
如图4所示,一般情况下ZIKV在相同的培养环境下,其RNA的复制水平相近,本实施例通过检测ZIKV在含有马尼地平溶液浓度不同的培养环境下的基因复制水平,来探索马尼地平对ZIKV的抑制效果。通过上述实验测得马尼地平浓度为5μM、10μM时,ZIKV RNA相对于DENV-2的表达量分别为0.0347和0.006,由此可知,马尼地平可有效对抑制ZIKV RNA在宿主细胞内的复制,并且在无毒性范围内能够有效抑制ZIKV感染宿主细胞。
实施例5
本实验利用荧光素酶活性实验检测马尼地平对DENV(登革病毒)抑制效果。
(1)实验过程
用胰酶消化Vero细胞(1.0×105个/mL),并用细胞计数仪技术,按照100μL/孔加入到96孔细胞培养板;于37℃、5%CO2的细胞培养箱中培养16h后,吸弃上清,分别将终浓度为2.5μM、5μM的马尼地平溶液通过12通道排枪加入到对应的细胞孔中,每组重复三孔,并在37℃、5%CO2的细胞培养箱中培养1h;每孔加入40μL DENV-2病毒(MOI=0.2),每隔15min摇匀一次,感染时间为1h;吸弃上清,用PBS洗涤3次,再分别将含有以上不同浓度的马尼地平的维持培养基加入到相应的细胞孔中,继续培养24h。
培养24h后,吸弃细胞上清,并用PBS洗涤细胞一次;吸弃PBS,并在96孔细胞培养板每孔加入50μL的1×荧光素酶裂解缓冲液;将96孔细胞培养板在摇床上振荡10min,再于-80℃的冰箱中冷冻30min,充分裂解细胞;将100×荧光酶底物加入到1×测定缓冲液中,并避光保存;将20μL细胞裂解液加入到50μL测定缓冲液中,并快速混匀两次,立即测定细胞荧光素酶活性。
(2)实验结果
如图5所示,荧光素酶活性反应了DENV-2的感染水平,即荧光素酶活性越高,DENV-2的感染能力越强。本实施例通过荧光素酶活性检测实验得到马尼地平溶液浓度为2.5μM、5μM时的荧光素酶活性分别为1.346和0.329,由此可知,马尼地平可有效对抑制DENV-2对宿主细胞抑制的感染。另外,根据DENV并不仅限于DENV-2一种病毒,根据抗原性的差异还包括DENV-1、DENV-3和DENV-4。通过上述实施例的实验结果表明,马尼地平是通过抑制病毒RNA在宿主细胞内的复制来达到抑制病毒感染宿主细胞的效果。另外,通过实验也证实了马尼地平对DENV-1、DENV-3和DENV-4有抑制效果,即马尼地平对DENV有抑制效果。
本发明对已知化合物马尼地平发掘了新的医疗用途,开拓了马尼地平在制备治疗黄病毒科病毒感染的药物中的新的应用领域;具体地,马尼地平在制备治疗黄病毒属病毒感染的药物中的应用;更为具体地,马尼地平在制备治疗流行性乙型脑炎病毒感染药物、寨卡病毒感染药物、西尼罗病毒感染药物以及登革病毒感染药物中的应用。马尼地平抑制黄病毒科病毒的RNA在宿主细胞内的复制,从而抑制黄病毒科病毒对宿主细胞的感染,可见马尼地平在制备抗黄病毒科病毒药物的应用中具有实用价值。此外,马尼地平的制备工艺简单,原料来源丰富,且可采用现代制药工艺做成口服剂型、注射剂型、片剂、胶囊剂、颗粒剂等,使用方便,疗效好,应用价值广泛。
尽管已用具体实施例来说明和描述了本发明,然而应意识到,在不背离本发明的精神和范围的情况下可以作出许多其它的更改和修改。因此,这意味着在所附权利要求中包括属于本发明范围内的所有这些变化和修改。

1.马尼地平在制备治疗黄病毒属病毒感染的药物中的应用。
2.马尼地平在制备治疗流行性乙型脑炎病毒感染的药物中的应用。
3.马尼地平在制备治疗寨卡病毒感染的药物中的应用。
4.马尼地平在制备治疗西尼罗病毒感染的药物中的应用。
5.马尼地平在制备治疗登革病毒感染的药物中的应用。
6.根据权利要求5所述的应用,其特征在于,所述登革病毒包括DENV-1、DENV-2、DENV-3以及DENV-4。
7.马尼地平在制备治疗登革热的药物中的应用。
8.根据权利要求1-7任一项所述的应用,其特征在于,所述药物是以(1,4-二氢-2,6-二甲基-4-间硝基苯基-3,5-吡啶二羧酸甲酯2-(4-二苯甲基-1-哌嗪基)乙酯作为药物活性成分制成的剂型。
9.根据权利要求8所述的应用,其特征在于:所述剂型为片剂、胶囊、颗粒剂、口服液、缓释制剂、控释制剂、纳米制剂或注射剂。
