Animal experiments:
the following experiment was performed using the beverage for relieving body fatigue prepared in example 1.
And establishing and grouping exhaustive exercise mouse models. Balb/c mice were fed adaptively for 1 week and randomly divided into 3 groups by body weight, 8 mice per group, i.e., normal group (Control), exhaustive exercise group (Motion), and drink group (NAT). Free-feeding mice were fed with AIN-93M as a standard maintenance feed. Indoor temperature is 21-24 ℃, relative humidity is 40-55%, air circulation is realized, and illumination time is l2 h. Normal group was perfused with normal saline without exhaustive motor intervention; model group and beverage group, using mouse wheel type fatigue instrument to give 2.5h exhaustion exercise, rotating speed of 20 r/min, continuous exercise for 2 days, and interval for 5 days. The same exhaustive exercise program was executed for 16 days. The beverage group perfuses stomach 2.5g/kg of beverage while doing exhaustive exercise, and the model group perfuses stomach equivalent normal saline.
The blood sugar, blood lactic acid, liver glycogen and muscle glycogen storage, superoxide dismutase activity and creatine kinase activity of the mice in each experimental group are measured. As a result, as shown in fig. 1, the exercise of exhaustion resulted in a decrease in blood glucose level, an increase in blood lactate level, and a decrease in the storage of hepatic and muscle glycogen, causing oxidative damage to the intestinal tract, significantly decreasing the SOD level; the above results all indicate that exhaustive exercise results in a significant fatigue stress state in mice. After the beverage is infused into the stomach, the fatigue stress state is obviously recovered.
And (3) measuring indexes such as intestinal villus length, crypt depth, tight junction protein expression, endotoxin level and the like of the mice of each experimental group. The results are shown in fig. 2, and the exhaustive movement causes the structural destruction of the small intestine, which is shown in the following specific points: the small intestine villus length is reduced, and the ratio of villus length to crypt depth is significantly reduced. The beverage can obviously protect the structural integrity of the small intestine under exhaustive exercise. Meanwhile, researches show that the beverage can obviously increase the expression of intestinal tight junction protein, reduce the intestinal permeability and inhibit endotoxin from entering blood.
The content of short chain fatty acid in the feces of the mice in each experimental group is measured. The results are shown in figure 3, where the beverages of the present invention are capable of significantly increasing fecal short chain fatty acid levels, particularly acetate, propionate, and butyrate. Meanwhile, the detection shows that the abundance of the short chain fatty acid producing bacteria (SCFAs) is also increased to a certain extent. Further explanation is as follows: the beverage can increase the content of the short-chain fatty acid by proliferating the short-chain fatty acid producing bacteria. The short chain fatty acid is a product of polysaccharide metabolism by bacteria, can promote the proliferation of intestinal epithelial cells, particularly the generation of butyrate, supplies energy to the intestinal epithelial cells, repairs intestinal barriers and relieves intestinal injury caused by exhaustive exercise.
The results are shown in figure 4, α diversity and β diversity are respectively counted, the results show that the intervention of the beverage has no significant influence on α diversity index, Shannon and Simpson indexes, the Beta diversity is further counted, and the intragroup comparison and the intergroup comparison of all experimental samples are respectively carried out by adopting an Unweighted UniFrac Distance algorithm and a Weighted UniFrac Distance algorithm.
20 mice were randomly divided into 2 groups of 10 mice each, a placebo group (2.5g/kg physiological saline) and a drink group (2.5g/kg drink), and were gavaged 1 time every 1 day for 20 consecutive days. After the last administration for 1h, a lead block with the mass of 5% of the tail root of the mouse is wound on the tail root of the mouse, the mouse is placed in a constant-temperature water bath box with the water depth of 25cm and the water temperature of 25 +/-0.5 ℃, and the time from the water entry to the exhaustion of the mouse immersed in the water for 9s without floating out of the water surface is recorded as the weight-bearing movement time of the mouse. The average swimming time under load of the control group mice is 5.63 +/-0.78 min, and the average swimming time under load of the beverage group mice is 8.76 +/-0.65 min. The result shows that the beverage can effectively relieve the sports fatigue of the mice and obviously prolong the average weight bearing swimming time of the mice.
