Technical Field
The invention relates to the technical field of medicines, and particularly relates to an anti-aging composition, and a preparation method and application thereof.
Background
Aging refers to the degenerative change of tissue morphology, the reduction of physiological functions and the deterioration of psychological functions of the body over time during the life process. At present, the global population is more than 60 hundred million, and by the time of 2025, the global population is predicted to reach 80 million, and then the world will step into an aging society, and people will face two major health problems, namely aging and diseases. Anti-aging research tasks are becoming more urgent.

"delaying aging is a desire of almost everyone, and exploring aging mechanism and prolonging human life is a current problem faced by researchers; especially for female population, the physiological and psychological symptoms of female climacteric syndrome seriously affect the life and health of patients due to the hypofunction of ovaries, the reduction of sex hormone secretion and the increase of gonadotropin, which cause the dysfunction of vegetative nerve and the reduction of immune function. With the accelerated aging of China, the health of the middle-aged and old people and the improvement of the life quality of the middle-aged and old people are concerned, and the middle-aged and old people have aroused wide interest in the medical field; the search for active substances for delaying aging from nature is one of the research hotspots in recent years.
The sea cucumber is recorded by the herbal medicine after the Ming dynasty, is listed as a tonic and is a health food with extremely high nutritive value. With the increasing demand of sea cucumbers, the scale of cultivation and sale of the sea cucumbers is gradually enlarged, so that a large amount of leftovers such as internal organs (sea cucumber flowers) are produced. In the actual production process, the viscera of the sea cucumber are often discarded as the processing waste, which causes great resource waste and environmental pollution. At present, research and development of sea cucumber health products in China are mainly focused on body walls, research reports on sea cucumber visceral function products are few, and the sea cucumber health products are especially rarely applied to the fields of anti-aging medicines, health products and the like.
Disclosure of Invention
The first purpose of the invention is to provide a composition for resisting aging, and the formula rule follows the concept of 'whole body' in traditional Chinese medicine, and mainly aims at tonifying qi and kidney, and strengthening spleen and stomach. On the other hand, the composition not only can resist aging, but also can relieve female climacteric symptoms.
The second objective of the invention is to provide a preparation method of the anti-aging composition, which is simple and feasible, has controllable parameters and strong operability, and is easy for industrial production on the premise of keeping the drug effect.
The third purpose of the invention is to provide the application of the anti-aging composition in preparing anti-aging medicines, health products and foods.

The anti-aging composition and the preparation method thereof provided by the invention have the beneficial effects that:
1. on one hand, the materials of the composition are simple and easy to obtain, and from the theory of traditional Chinese medicine, the formula rule follows the concept of 'whole' of traditional Chinese medicine, and mainly aims at tonifying qi and kidney, and strengthening spleen and stomach, so that yin and yang of internal organs are balanced, and the anti-aging effect is achieved; on the other hand, modern pharmacological research proves that the active ingredients in the composition not only have the functions of removing free radicals, resisting oxidation and improving the immunity of the organism, but also have the effects of simulating estrogens.
2. The composition has reasonable formula, scientific dosage, remarkable and stable curative effect, and no toxic or side effect compared with western medicines.
3. The composition can be used for delaying aging; but also can regulate the female ovarian function and relieve female climacteric symptoms.

In order to make the objects, technical solutions and advantages of the embodiments of the present invention clearer, the technical solutions in the embodiments of the present invention will be clearly and completely described below. The examples, in which specific conditions are not specified, were conducted under conventional conditions or conditions recommended by the manufacturer. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products available commercially.
The following is a detailed description of a composition for anti-aging according to an embodiment of the present invention, and a preparation method and application thereof.
The composition for resisting aging provided by the embodiment of the invention is mainly prepared from the following raw materials in parts by weight: 400-1000 parts of sea cucumber viscera; 5-50 parts of rhodiola rosea; 5-50 parts of acanthopanax; 10-50 parts of polygonatum; 1-30 parts of astragalus; 1-20 parts of codonopsis pilosula; and 1-15 parts of dendrobium.
Further, in the preferred embodiment of the present invention, the raw materials include, by weight: 500-800 parts of sea cucumber viscera; 10-35 parts of rhodiola rosea; 10-35 parts of acanthopanax; 15-30 parts of polygonatum; 5-20 parts of astragalus; 5-12 parts of codonopsis pilosula; and 5-12 parts of dendrobium.
Further, in a preferred embodiment of the present invention, the viscera of the sea cucumber is 600 parts, the rhodiola rosea is 30 parts, the acanthopanax is 30 parts, the polygonatum is 15 parts, the astragalus is 15 parts, the codonopsis pilosula is 10 parts, and the dendrobium is 5 parts.
In the formula, the rule of the formula follows the concept of 'whole body' in traditional Chinese medicine, and the formula mainly aims at tonifying qi and kidney and strengthening spleen and stomach. Wherein the viscera of the sea cucumber is the viscera of the sea cucumber which is a tonic drug. The viscera of Stichopus japonicus comprise digestive tract, gonad, respiratory tree, Vickers tube, hydropneumoniae, Borrelia sac, water ring tube, tentacle, and sieve plate. Medical research proves that the viscera of the sea cucumber not only contains abundant proteins and active substances as the same as body walls, but also contains more precious nutrient components, such as polysaccharide, vanadium, nucleic acid, gonadal pigment and the like, and the natural active components have the effects of delaying senility, preventing tissue aging, promoting sex hormone secretion and the like, almost have no toxic or side effect, and are important components for preventing and treating climacteric syndrome and premature ovarian failure of women.
It should be noted that, in order to optimize the anti-aging effect, the whole visceral tissues of fresh sea cucumbers are selected in this embodiment. In addition, in the embodiment of the invention, the fresh sea cucumber viscera with water can be selected, and the sea cucumber viscera which is processed into a dry product can also be selected.
Kidneys are the innate root and are related to the growth, development, aging and death of the body. Secondly, kidney governs water, and is hidden by the essence of internal organs, so sufficient kidney qi can activate qi and vessels, and the human body is not easy to be aged. Rhodiola rosea has the effects of strengthening body resistance, consolidating constitution, benefiting qi, nourishing blood, clearing heat and moistening lung. Acanthopanax senticosus has the effects of tonifying kidney and spleen. Yu Zhu can nourish yin, moisten dryness, promote the production of body fluid to quench thirst, tonify five internal organs and nourish qi and blood. The three components tonify kidney and qi, smooth the five internal organs and six internal organs, and delay aging of human body. Modern pharmacological research shows that the effective components of rhodiola rosea and polygonatum odoratum can remove high-activity free radicals in organisms, and simultaneously improve the activities of key enzymes such as superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) of the organisms so as to reduce the peroxidation degree of cell lipids, reduce the DNA oxidative damage of eukaryotic cells and achieve the anti-aging effect. The extractive solution of radix Acanthopanacis Senticosi can reduce the content of erythrocyte lipid peroxide (LOP) and myocardial lipofuscin, and increase the content of hydroxyproline in skin.
The spleen and stomach are the acquired root and the source of qi and blood. The spleen and the stomach play a role in lifting and descending the hinge during the activity of the human body, and the innate essence and qi in the kidney also depend on the nourishment of the acquired food essence generated by the biochemical process of the spleen and the stomach, so that the formula is combined with the astragalus and the codonopsis pilosula for tonifying qi and strengthening the spleen, the dendrobium for tonifying the stomach and promoting the production of body fluid, so that the functions of the spleen and the stomach are enhanced, the qi of the spleen and the stomach is sufficient, and the spleen and. Further, in a preferred embodiment of the present invention, the composition is in the form of powder, granules, capsules, tablets, pills, oral liquid or syrup. The preparation formulation selected by the invention is a conventional preparation formulation, and is easy to produce and store; for users, the sea cucumber feed can be taken at any time and in a quantitative manner, and the condition limitation of directly eating sea cucumber resources is overcome. The dosage form of the composition is not limited to the above dosage forms, and other dosage forms can be implemented. For example, the viscera of sea cucumber and the decoction of five traditional Chinese medicines are directly used.
The invention also provides a preparation method of the anti-aging composition, which comprises the following steps:
drying the viscera of the sea cucumber, preferably at 40-60 ℃ in order to prevent the loss of effective components in the viscera of the sea cucumber, and then crushing. Secondly, the crushed sea cucumber viscera are extracted by using an ethanol solution to obtain a first extracting solution. Preferably, the extraction temperature is controlled to be 50-80 ℃ when the viscera of the sea cucumber are extracted. The sea cucumber viscera are dried, crushed and mixed with an ethanol solution, and extraction is carried out at 50-80 ℃. The first extracting solution is the first extracting solution with filter residue filtered. Preferably, ethanol with a volume fraction of 30-70% is used, and the ethanol with the concentration can fully precipitate the active ingredients in the viscera of the sea cucumber. Further, in the preferred embodiment of the present invention, the viscera of the sea cucumber can be extracted 1-3 times by using ethanol solution, so as to improve the extraction rate.
Extracting radix Rhodiolae, radix Acanthopanacis Senticosi, rhizoma Polygonati Odorati, radix astragali, radix Codonopsis and herba Dendrobii with water to obtain a second extractive solution. The second extractive solution is medicinal liquid without residue. Preferably, the five traditional Chinese medicines are soaked for 15-30 min before the raw materials of the five traditional Chinese medicines are extracted. Furthermore, the five traditional Chinese medicines are firstly crushed and then soaked and extracted.
In this embodiment, the method for extracting the Chinese medicine may be: pulverizing rhodiola rosea, acanthopanax, polygonatum, astragalus, codonopsis pilosula and dendrobium, uniformly mixing, soaking in water for 15-30 min, and decocting with slow fire for 1-3 times, wherein each time lasts for 0.25-1 hour. Further, in order to obtain the active ingredient with relatively high purity, the active ingredient can be purified by a water extraction and alcohol precipitation method.
Further, in a preferred embodiment of the present invention, before concentrating the second extract, the method further comprises the step of performing ultrafiltration on the second extract with a 5-15 KDa filter membrane. Preferably, the ultrafiltration is carried out through a 10kDa filter. The ultrafiltration method can separate medicinal residue from medicinal liquid. Meanwhile, other solid-liquid separation methods are not excluded to obtain the second extracting solution without dregs, such as suction filtration and the like.
The order of the steps for obtaining the first extract and the second extract is not limited.
After the first extract and the second extract are obtained, in order to prevent the components in the first extract and the components in the second extract from reacting, the first extract and the second extract are respectively concentrated under reduced pressure and dried into extractum, preferably, vacuum drying is selected, and specific drying methods are not limited, such as spray drying, forced air drying and the like. Drying, recovering solvent, pulverizing the extract into powder, and mixing. Preferably, the particle size of the powder is 80-120 meshes.
The composition for anti-aging provided by the present invention, the preparation method thereof and the application thereof to the preparation of a specific formulation product are further described in detail with reference to the following examples.

Crushing dried sea cucumber viscera, adding 30% ethanol solution by volume fraction, extracting for 1 time, and taking filtrate to obtain a first extract; extracting radix Rhodiolae, radix Acanthopanacis Senticosi, rhizoma Polygonati Odorati, radix astragali, radix Codonopsis and herba Dendrobii with water, and filtering to obtain a second extractive solution; concentrating the first extractive solution and the second extractive solution under reduced pressure, drying, pulverizing into 80 mesh powder, and mixing to obtain the composition.

Drying viscera of Stichopus japonicus at 55 deg.C, pulverizing, adding 40% ethanol solution, extracting at 60 deg.C for 1 hr for 3 times, and mixing filtrates to obtain first extractive solution; soaking radix Rhodiolae, radix Acanthopanacis Senticosi, rhizoma Polygonati Odorati, radix astragali, radix Codonopsis and herba Dendrobii in water for 25min, extracting for 3 times, 40min for the first time, 30min for the second time, and 20min for the third time, mixing filtrates, ultrafiltering with 10KDa filter membrane, and mixing filtrates to obtain second extractive solution; concentrating the first extractive solution and the second extractive solution under reduced pressure, drying, pulverizing into 90 mesh powder, and mixing to obtain the composition.

Test example 1 Effect on body weight of aging mice
1. Preparation of experimental mouse aging model
50 healthy male mice were injected subcutaneously with D-galactose 120mg/Kg per day on the back and neck 1 time per day for 8 weeks to create a subacute aging model; another 10 mice served as blank controls and all mice were routinely fed with normal diet.
2. Method and steps of operation
The aging model mice are randomly divided into 5 groups, each group comprises 10 mice, the model control group (normal saline), the positive control group (lentinan 200mg/Kg) and the composition for resisting aging provided by the embodiment 4 are prepared into low, medium and high dose groups (100mg/Kg, 200mg/Kg and 400mg/Kg), a blank control group (normal saline) is additionally arranged, and the administration is carried out by stomach irrigation for 1 time every day for 8 weeks. Body weight was measured every 1 week from the start to the end of the experiment and the change in body weight was recorded and the results are shown in table 1.

At the end of the experiment, the weight of the mice in the aging model group increased by 4.1 percent, which is only 50 percent of that in the blank control group, and compared with the aging model group, the weight of the mice in the administration group showed obvious difference.
Test example 2 Effect on serum SOD and MDA of aging mice
1. Method of operation
After the procedure of step 1 and step 2 in test example 1, after the last administration for 1 hour, the eyeball was removed under aseptic conditions, blood was collected, left to stand at 4 ℃ for 2 hours, centrifuged at 4000r/min for 10 minutes, and serum was collected, and the procedure was performed according to the MDA and SOD detection kit, and the results are shown in Table 2.

Compared with the model group, the SOD activities of the low, medium and high dose groups of the composition are obviously improved, and the MDA content is obviously reduced.
Test example 3 Effect on thymus and spleen index of aging mice

After the procedures of step 1 and step 2 in test example 1, the thymus and spleen of the bled mice were dissected, washed with physiological saline, the surface blood was drained on a filter paper, and the Thymus Index (TI) and the Spleen Index (SI) were calculated. The calculation method comprises the following steps: the organ index is organ weight (mg)/body weight (g), and the results are shown in table 3.

The thymus of mice in the aging model group is in an atrophied state, the spleen is obviously increased, and compared with the mice in the aging model group, the TI and SI of the sea cucumber internal organ essence composition in the high-dose group are obviously changed.
According to tables 1 to 4, the traditional Chinese medicine considers that aging is mainly related to weakness of the spleen and the stomach, kidney deficiency, phlegm turbidity, blood stasis and the like, the aging of a human body starts from the aging of organs, thymus and spleen are important immune organs, and the organ indexes can reflect the immune state of the body to a certain extent.
Under the experimental conditions, the composition has obvious improvement effect on body weight and organ senescence symptoms generated by the D-galactose-induced mice, so that the immunity of the mice is improved, and the effect of delaying senescence is shown.
Secondly, by influencing the SOD and MDA levels in the blood serum of the aging mouse, the scavenging of free radicals is accelerated, the cell membrane structure is stabilized, and the lipid peroxidation aging caused by the free radicals and metabolites thereof is resisted.
In conclusion, the composition for resisting aging provided by the invention has a remarkable curative effect.
Test example 4 premature ovarian failure test
1. Preparation of experimental mouse aging model
50 healthy female mice are subjected to intraperitoneal injection of 20mg/Kg of cyclophosphamide every day for 5 days continuously to prepare premature ovarian failure models; another 10 were used as blank controls, and daily intraperitoneal injections of physiological saline were given; all mice were routinely fed with normal diet.
2. Method of operation
Aging model mice were randomly divided into 5 groups of 10 mice each, and the model control group (normal saline), the positive control group (estradiol valerate 0.1mg/Kg), and the composition for anti-aging provided in example 4 were prepared into low, medium, and high dose groups (200mg/Kg, 400mg/Kg, 600mg/Kg), and a blank control group (normal saline) was added, and the administration was performed by gavage for 1 time a day for 4 weeks.
3. Influence on sexual organ index
After 24h of the last administration, the patient is killed by cervical dislocation, and the ovary and uterus are taken out after laparotomy, and the sexual organ indexes (ovary index, uterus index, mg/g) are respectively calculated.
4. Results and analysis

Compared with the blank group, the ovary index and the uterus index of the mice in the model group are obviously lower, and the sex organ index of the mice in each group is obviously improved compared with the mice in the model group.
The research shows that the premature ovarian failure of women is premature ovarian failure or premature ovarian failure, the secretion of sex hormones is reduced, gonadotropin is increased, vegetative nerve functional disturbance is caused, and the immune function is reduced at the same time.
In conclusion, the composition for resisting aging, the preparation method and the application thereof provided by the invention have the advantages of reasonable formula, scientific dosage, obvious and stable curative effect and capability of delaying aging; but also can regulate the female ovarian function and relieve female climacteric symptoms. The preparation method is simple to operate, has controllable parameters, is beneficial to large-scale production, and has great social significance and economic value.

Claims (10)
1. The composition for resisting aging is characterized by being capable of regulating female ovarian function and relieving female climacteric symptoms, and comprises the following raw materials in parts by weight: 400-1000 parts of sea cucumber viscera; 5-50 parts of rhodiola rosea; 5-50 parts of acanthopanax; 10-50 parts of polygonatum; 1-30 parts of astragalus; 1-20 parts of codonopsis pilosula; and 1-15 parts of dendrobium;
the composition is prepared by respectively concentrating a first extracting solution and a second extracting solution, recovering a solvent, preparing into powder and mixing;
wherein the first extracting solution is obtained by mixing dried and crushed sea cucumber viscera with an ethanol solution for extraction;
the second extract is obtained by extracting the rhodiola rosea, the acanthopanax senticosus, the polygonatum odoratum, the astragalus membranaceus, the codonopsis pilosula and the dendrobium with water.
2. The anti-aging composition according to claim 1, wherein the composition comprises the following raw materials in parts by weight: 500-800 parts of sea cucumber viscera; 10-35 parts of rhodiola rosea; 10-35 parts of acanthopanax; 15-30 parts of polygonatum; 5-20 parts of astragalus membranaceus; 5-12 parts of codonopsis pilosula; and 5-12 parts of dendrobium.
3. The anti-aging composition according to claim 1, wherein the sea cucumber viscera is 600 parts, the rhodiola rosea is 30 parts, the acanthopanax senticosus is 30 parts, the polygonatum odoratum is 15 parts, the astragalus membranaceus is 15 parts, the codonopsis pilosula is 10 parts, and the dendrobium nobile is 5 parts.
4. The anti-aging composition according to claim 3, wherein the composition is in the form of powder, granules, capsules, tablets, pills, oral liquid or syrup.

The method for preparing the anti-aging composition according to claim 1, wherein the sea cucumber viscera are dried, pulverized, mixed with an ethanol solution, and extracted to obtain a first extract; extracting the rhodiola rosea, the acanthopanax senticosus, the polygonatum odoratum, the astragalus mongholicus, the codonopsis pilosula and the dendrobe with water to obtain a second extracting solution; concentrating the first extractive solution and the second extractive solution respectively, recovering solvent, making into powder, and mixing.
6. The method for preparing the anti-aging composition according to claim 5, wherein the extraction temperature is 50 to 80 ℃ when the viscera of the sea cucumber are extracted.
7. The method for preparing the anti-aging composition according to claim 5, wherein the volume fraction of ethanol in the ethanol solution is 30 to 70 percent.
8. The method for preparing the antiaging composition according to claim 5, characterized in that before the concentration of the second extract, the method further comprises a step of subjecting the second extract to ultrafiltration with a 5-15 kDa filter membrane.
9. The method for preparing the anti-aging composition according to claim 5, wherein the powder has a particle size of 80 to 120 mesh.
10. Use of the composition for anti-aging according to any one of claims 1 to 4 for preparing an anti-aging drug.
