Abstract
本发明涉及干细胞技术领域,尤其涉及鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的方法与试剂盒。该方法包括:将视网膜祖细胞培养至对数生长期后进行流式细胞鉴定,鉴定结果符合鉴定标准的为具有治疗视网膜退行性病变功能的视网膜祖细胞。采用本发明提供的方法鉴定的视网膜祖细胞能够保持良好的增殖能力和干细胞特性,将其用于治疗视网膜退行性病变能够起到良好的疗效。实验表明,将采用本发明提供方法鉴定的视网膜祖细胞注射入RCS大鼠眼睛玻璃体腔后,对视网膜外核层细胞有保护作用、多焦电生理显示细胞注射组视网膜感光细胞传导的电位变化比仅注射HBSS缓冲液组有显著变化。水迷宫实验也表明,注射后大鼠视觉功能得到改善。

技术领域
本发明涉及干细胞技术领域,尤其涉及鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的方法与试剂盒。
背景技术
视网膜退行性病变是一种以视网膜感光细胞和色素上皮细胞凋亡和坏死为主要特点的一类视网膜疾病,主要包括萎缩性年龄相关性黄斑变性 (Age-related maculardegeneration,AMD)、视网膜色素变性(Retinitis Pigmentosa,RP)及视网膜萎缩等。视网膜退行性病变是目前主要的致盲疾病之一,多数具有遗传倾向性,发病初期视网膜细胞出现功能紊乱,随着疾病的进展逐渐出现视网膜神经上皮和色素上皮的凋亡萎缩,视力出现不可逆性下降,严重者失明。因其发病机制主要为细胞凋亡,目前该类疾病尚无有效的预防和治疗方法,多年来各国科学家努力探索尝试各种方法治疗视网膜退行性病变,到目前为止方法有以下三种:
1、基因治疗方式:主要指利用病毒或非病毒载体将目的基因或功能基因转运到眼内,使载体遗传信息与受体遗传信息进行重新整合,从而从基因水平修复错配、突变等基因,达到治疗目的。目前虽然取得一些成绩,不过外源载体的问题一直没有解决,离临床应用还有很长的路程。
2、神经营养因子注射法:试验证明神经营养因子可以推迟或控制感光细胞的凋亡,从而达到治疗视网膜退行性病变的目的。但是,绝大多数的营养因子只有通过眼内注射起到治疗作用,而由于该方法不能长效供应营养,需要多次注射,易造成感染和视网膜脱离等并发症,其风险远远大于所获得的收益,所以该类方法仍需要进一步探索和研究。
3、细胞治疗方式:将细胞通过各种途径运输到眼内,通过细胞持续释放神经保护因子或细胞替代等方式治疗视网膜退行性病变,是目前公认的最有可能实现临床治疗的方法。常用于治疗的细胞主要有:(1)胚胎干细胞(ES):它具有分化成多种细胞的能力,但是这种细胞存在伦理、免疫排斥和畸胎瘤等问题,不适合临床应用。(2)多能诱导干细胞(iPS):指将正常的成熟细胞在体外重编程为多能诱导干细胞,从理论上具有多向分化能力,但是它存在病毒整合和表达异常等问题。(3)间充质干细胞(MSC):这种细胞具有有限的分化能力,取材方便,可以自体治疗。但是由于它有限的分化能力,因此在视网膜疾病治疗中的成功率并不高。(4)视网膜祖细胞(RPC):这种细胞能够专一性的分化为视网膜各层细胞,体外可以增殖传代。使用这种细胞治疗的主要问题是存在个体免疫排斥现象,但是由于眼球具有免疫豁免性,可以有效避免全身免疫排斥问题,因此RPC将成为理想的细胞治疗产品。
视网膜祖细胞(RPC)属于一种神经干细胞,能够表达多种神经营养和抗凋亡因子。根据目前的研究进展,在玻璃体腔内注射视网膜祖细胞可能是最长效、安全治疗视网膜退行性疾病的方法。但是,注射所采用的视网膜祖细胞应当具有优良的品质,未经鉴定的视网膜祖细胞注射入玻璃体腔治疗失败的风险较高,严重时会导致并发症的产生。然而,目前尚无任何研究表明应当对哪些方面进行鉴定才能够保证得到的视网膜祖细胞能够具有良好的治疗视网膜退行性病变的效果。
因此,建立一种鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的方法具有十分重要的意义。
发明内容
有鉴于此,本发明要解决的技术问题在于提供鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的方法及试剂盒。采用本发明提供的方法鉴定的视网膜祖细胞能够有效的治疗视网膜退行性病变。
本发明提供的鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的方法包括:将视网膜祖细胞培养至对数生长期后进行流式细胞鉴定满足以下条件的细胞为具有治疗视网膜退行性病变功能的视网膜祖细胞:Nestin的表达量大于90%;Pax6的表达量大于70%;Ki-67的表达量大于60%;Chx10的表达量大于85%;Sox2的表达量大于50%;CD38的表达量小于5%;HLA-DR 的表达量小于10%。
本发明采用的视网膜祖细胞培养方法采用申请号为201210268933.X的方法进行,其中视网膜祖细胞的来源可以为自行制备,也可由购买获得,本发明对此不做限定,其实施皆在本发明的保护范围之内。
Nestin(巢蛋白),是一种中间丝类型的蛋白,能够特异性的表达在神经上皮干细胞中,是神经干细胞的特征性标志物。由于视网膜祖细胞本质上是一种组织干细胞,而Nestin在干细胞分化过程中表达下降,因此,为了鉴定到具有良好干性的视网膜祖细胞以用于治疗视网膜退行性病变,Nestin需保持较高的表达量。
人类配对盒基因(paired-box gene 6,Pax6)编码一个转录因子,Pax6正常表达在眼部的各种组织中,在眼部器官发育中起到决定性作用。Pax6的缺失或突变能够阻碍胚胎期细胞向神经外胚层细胞的发育,导致形成眼内发育异常,因此,具有治疗视网膜退行性病变功能的视网膜祖细胞中Pax6的表达量不应过低。
细胞增殖抗原标记物Ki-67,是一种细胞增殖相关基因,仅存在于细胞周期分裂期的一种结构与功能都和染色体密切关联的大分子核抗原。现有研究结果表明,细胞中Ki-67表达量升高与多种恶性肿瘤密切相关。但细胞中Ki-67 表达过低则意味着细胞增殖不旺盛,活性较低。
视网膜前体细胞标志物Chx10,是视网膜胚胎发育阶段原始细胞表达的特异性标记,是视网膜早期发育阶段的调控基因,表达于尚未分化的视网膜前体细胞中,因此,合格的用于治疗视网膜退行性病变的视网膜祖细胞中 Chx10的表达量应维持在较高的表达量。
Sox2是一种干细胞相关的标志物,其对早期胚胎发育至关重要,且对晶状体和视网膜的发育中发挥重要作用。其表达对视网膜祖细胞多能状态的维持是不可或缺的。
CD38是一个定位于膜上的糖蛋白,催化环腺苷二磷酸核糖的合成和降解,现有技术中,常作为肿瘤相关标志物,研究表明,CD38的高表达与恶性肿瘤密切相关。因此,合格的用于治疗视网膜退行性病变的视网膜祖细胞中 CD38的表达量不应过高。
HLA-DR是一种组织相容性抗原,目前常用于外周血B淋巴细胞及单核细胞的计数或鉴定淋巴瘤及白血病,研究表明,原发性视网膜色素变性患者的视网膜色素上皮细胞中HLA-DR抗原表达呈阳性,而健康人则无此种表现。因此,鉴定用于治疗视网膜退行性病变的视网膜细胞中HLA-DR的表达不应过高。
视网膜祖细胞(RPC)属于一种神经干细胞,将其用于治疗视网膜退行性病变应具有稳定的干细胞特性和旺盛的增殖力。本发明通过实验证实,多种细胞因子中,本发明提供的鉴定方法中采用的因子Nestin、Pax6、Ki-67、Chx10、 Sox2呈高表达,而CD38、HLA-DR呈低表达,如此鉴定的视网膜祖细胞能够保持良好的增殖能力和干细胞特性,将其用于治疗视网膜退行性病变能够起到良好的疗效。采用本发明提供的方法鉴定视网膜祖细胞,各因子的表达量检测分别进行,即7种鉴定抗体分别与视网膜祖细胞混合后,进行流式细胞检测。
将采用本发明提供的方法鉴定得到的视网膜祖细胞保存于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC NO.10419。
在本发明的实施例中,流式细胞鉴定采用鉴定抗体和鉴定抗体的同型对照抗体。
同型对照抗体是指使用与鉴定抗体相同种属来源、相同剂量及同种免疫球蛋白的相同亚型的抗体作为对照。设置同型对照抗体的主要目的是确定鉴定抗体的结合是特异性的,而不是与非特异性的受体或与其他蛋白的相互作用。
鉴定抗体Nestin、Pax6、Ki-67、Chx10、Sox2、CD38、HLA-DR及它们的同型对照抗体可为自制也可于市场购得,本发明对此不作限定,其实施皆在本发明的保护范围之内。
在本发明的实施例中,流式细胞鉴定包括:将视网膜祖细胞培养至对数生长期,经消化、稀释后与鉴定抗体混合,孵育、洗涤、重悬后以流式细胞仪检测。
在一些实施例中,消化的酶为TrypLETM-Express、胰酶或胶原酶。
作为优选,消化的酶为TrypLETM-Express。

在本发明的实施例中,流式细胞鉴定的具体步骤为:将视网膜祖细胞培养至对数生长期后,用TrypLETM-Express、胰酶或胶原酶消化视网膜祖细胞,以洗涤液A(生理盐水、DPBS缓冲液或Hank’s液)稀释按照0.5×105个/mL 的密度将细胞分别与鉴定抗体及同型对照抗体混合,室温避光孵育15min,然后加入1ml洗涤液(DMEM培养基、MEM培养基或DMEM/F12培养基),然后离心500rpm~800rpm,离心3min。用30μl洗涤液A(生理盐水、DPBS 缓冲液或Hank’s液)重悬上机检测。
采用本发明提供的方法鉴定得到的视网膜祖细胞能够有效治疗视网膜退行性病变。实验表明:将该视网膜细胞注射入RCS大鼠眼睛玻璃体腔内,8 周后,视网膜祖细胞注射组RCS大鼠视网膜外核层细胞有所留存,而同时仅注射HBSS缓冲液的对照大鼠的视网膜外核层则完全萎缩。ERG实验表明,注射本发明提供方法鉴定得到的视网膜祖细胞8W后,RCS大鼠的视网膜神经感光细胞传导的电位变化显著高于仅注射HBSS缓冲液的对照组。水迷宫实验也表明,注射本发明提供方法鉴定得到的视网膜祖细胞后,大鼠视觉功能得到改善,但其运动能力并未发生改变。另外,经注射本发明提供方法鉴定得到的视网膜祖细胞后,玻璃体液中有25种因子表达升高,主要是BDNF(脑源性神经营养因子)、GDF-15(生长分化因子-15)、FGF-4(成纤维细胞生长因子 4)、FGF-7(成纤维细胞生长因子7)、NGF(神经生长因子)、GDNF(胶质细胞源性神经营养因子)、IGF-Ⅰ(胰岛素样生长因子-Ⅰ)、IGF-Ⅱ(胰岛素样生长因子-Ⅱ)、NT-3(神经营养因子-3)、NT-4(神经营养因子-4)、IGFBP-1(胰岛素生长因子结合蛋白1)、IGFBP-2(胰岛素生长因子结合蛋白2)、 IGFBP-3(胰岛素生长因子结合蛋白3)、IGFBP-4(胰岛素生长因子结合蛋白 4)、IGFBP-6(胰岛素生长因子结合蛋白6)、OPG(骨保护素)、TGF-β1(转移生长因子-β1)、TGF-β3(转移生长因子-β3)、PDGF-AA(血小板衍生生长因子-AA)、VEGF(血管内皮生长因子)、CNTF(睫状神经营养因子)、TNF(肿瘤坏死因子)、HSP27(热休克蛋白27)、HSP60(热休克蛋白60)和HSP70(热休克蛋白70)。而体外培养本明提供方法鉴定得到的视网膜祖细胞经鉴定结果表明,其上清液中持续释放的细胞保护因子与玻璃体也中高表达的因子具有一致性。而这些因子在现有技术的研究中,多具有治疗视网膜退行性病变的功能,说明本发明提供方法鉴定得到的细胞注射入玻璃体腔内,能够持续释放这些具有治疗视网膜退行性病变功能的细胞因子的表达,从而起到治疗视网膜退行性病变的作用。而与注射细胞因子相比,注射本发明提供方法鉴定得到的视网膜祖细胞的释放具有持续性。
因此,本发明还提供了一种具有治疗视网膜退行性病变功能的视网膜祖细胞,其中Nestin的表达量大于90%;Pax6的表达量大于70%;Ki-67的表达量大于60%;Chx10的表达量大于85%;Sox2的表达量大于50%;CD38 的表达量小于5%;HLA-DR的表达量小于10%。
在本发明的实施例中,注射入RCS大鼠玻璃体腔的细胞制剂中包括:保藏编号为CGMCC NO.10419的视网膜祖细胞及平衡盐溶液。
在一些实施例中,制剂中细胞的密度为1×107个/mL~10×107个/mL。
作为优选,每只眼睛用量为106个保藏编号为CGMCC NO.10419的视网膜祖细胞。
作为优选,平衡盐溶液为HBSS。
本发明还提供了鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的试剂盒,其中,包括鉴定抗体和鉴定抗体的同型对照抗体;

在本发明的实施例中,本发明提供的试剂盒还包括洗涤液A、洗涤液B 和消化液;
洗涤液A为生理盐水、DPBS缓冲液或Hank’s液;
洗涤液B为DMEM培养基、MEM培养基或DMEM/F12培养基;
消化液为TrypLETM-Express、胰酶溶液或胶原酶溶液。

本发明提供了鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞方法及其试剂盒,该方法包括:将视网膜祖细胞培养至对数生长期后进行流式细胞鉴定,鉴定结果符合鉴定标准的为具有治疗视网膜退行性病变功能的视网膜祖细胞。采用本发明提供的方法鉴定得到的视网膜祖细胞能够保持良好的增殖能力和干细胞特性,将其用于治疗视网膜退行性病变能够起到良好的疗效。实验表明,将采用本发明提供方法鉴定得到的视网膜祖细胞注射入大鼠眼睛玻璃体腔后,对外核层细胞有保护作用、而视网膜神经感光细胞传导的电位变化显著高于仅注射HBSS缓冲液的对照组、水迷宫实验也表明,注射后大鼠视觉功能得到改善。并且,所有注射了采用本发明提供方法鉴定得到的视网膜祖细胞的大鼠皆未发现异常。

本发明采用的仪器皆为普通市售品,皆可于市场购得。
下面结合实施例,进一步阐述本发明:
实施例1视网膜祖细胞的培养
视网膜祖细胞的成球培养:组织消毒后,取视网膜组织,用酶消化后, 1000r离心3分钟,取沉淀,用DMEM/F12基础培养基(加入N-2添加物,浓度为1:100、表皮生长因子EGF、浓度为10ng/ml、碱性生长因子bFGF、浓度为 10ng/ml、L-谷氨酰胺、浓度1:100)混匀沉淀计数、按照1-2×105/cm2接种于低吸附的T25培养瓶中,置于37℃,5%CO2恒温培养箱内培养,次日细胞成球。
海藻酸钠凝胶的制备:以去离子水作为海藻酸钠的溶剂,先称取海藻酸钠粉末,将粉末缓缓加入装有少量溶剂(20ml)的离心管中,同时边加粉末边加剩余溶剂,最后用旋涡混合器震荡5min后55℃水浴加热过夜,次日高压蒸汽灭菌,然后在超净工作台内灌装于细胞培养瓶中,恢复到室温既可以使用。
视网膜祖细胞的三维空间培养:细胞成球后,收集细胞液离心,收集沉淀,用视网膜专用培养基混匀细胞,种植铺有海藻酸钠凝胶的细胞培养瓶中,细胞即能在海藻酸钠凝胶的三维空间中快速生长和繁殖。
视网膜专用培养基由DMEM/F12基础培养基、N-2添加物(浓度为1: 100)、表皮生长因子EGF(浓度为10ng/ml)、碱性生长因子bFGF(浓度为 10ng/ml)、L-谷氨酰胺(浓度1:100)、三碘甲状腺氨酸T3(浓度为20ng/ml)、神经营养素NT4(浓度为20ng/ml)、神经生长因子NGF(浓度为20ng/ml)、神经营养物质-3NT-3(浓度为20ng/ml)组成。
实施例2视网膜祖细胞的鉴定
取实施例1培养的增殖旺盛的第五代视网膜祖细胞进行流式细胞仪鉴定:用TrypLETM-Express消化视网膜祖细胞,用DPBS重悬细胞后计数,按照0.5 ×105个细胞量将细胞平均分配到流式检测管中,然后向其中加入鉴定抗体 (Nestin/Pax6//Ki67/Chx10/Sox2/CD38/HLA-DR)及同型对照抗体室温避光孵育15min,然后加入1ml DMEM/F12离心500-800rpm,离心3min。用30μL DPBS重悬上机检测。
鉴定抗体和同型对照抗体购自BD和Abcam。
鉴定标准为:Nestin的表达量大于90%;Pax6的表达量大于70%;Ki-67 的表达量大于60%;Chx10的表达量大于85%;Sox2的表达量大于50%;CD38 的表达量小于5%;HLA-DR的表达量小于10%。
符合鉴定标准的视网膜祖细胞即为具有治疗视网膜退行性病变功能的视网膜祖细胞。
保存于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为 CGMCCNO.10419。
实施例3视网膜祖细胞制剂的制备
将保藏编号为CGMCC NO.10419的视网膜祖细胞悬浮于HBSS平衡盐溶液,细胞浓度为4×107个/mL。
实施例4保藏编号为CGMCC NO.10419的视网膜祖细胞对大鼠的治疗作用
以实施例3制得的视网膜祖细胞制剂验证采用本发明提供方法鉴定的视网膜祖细胞对RCS大鼠视网膜退行性疾病的治疗效果。实验方法如下:
实验动物选用皇家外科学院(royal college surgeons,RCS)大鼠,它是一种常染色体隐性遗传的经典动物模型鼠,3周龄,体重250~300g,雌雄不限,共计30只。分组如下:
A组:正常对照组,同种野生型含色素RCS大鼠(rdy+)6只,未进行任何处置。
B组:实验对照组,只进行HBSS(平衡盐溶液)治疗,左眼玻璃体腔内单次注射,10μl/只/次,12只。
C组:实验治疗组,移植具有治疗作用的视网膜祖细胞,左眼玻璃体腔内单次注射,10μl/只/次,12只。
形态学检测:B组和C组实验动物分别于注射后8周取材,制备石蜡切片,HE染色观察视网膜各层细胞变化。图1a示B组大鼠注射8周后视网膜组织石蜡切片,放大400倍;图1b示C组大鼠注射8周后视网膜组织石蜡切片,放大400倍。HE染色显示B组视网膜外核层完全萎缩,胶质化;而C 组大鼠眼球同一部位经细胞治疗后有留存的外核层细胞。
多焦视觉电生理(multifocal visual electroretinogram,ERG):分别于注射后和注射后2W,4W,8W进行暗适应ERG观察视网膜神经感光细胞传导的电位变化。如图2所示,通过统计分析后,注射4W和8w后细胞注射组和HBSS 组有显著性差异。
动物行为学-水迷宫检测:注射后8w对三组动物进行水迷宫检测,结果如表1,行为学实验说明玻璃体腔内移植RPC后,只改变了大鼠的视觉功能,未改变其运动能力。

表1显示,C组细胞移植组RCS鼠的运动路程和时间与B组对照组相比有显著提高,这说明具有治疗潜能的RPC移植后使RCS鼠的视功能改善。以上结果说明,本发明提供方法鉴定得到的视网膜祖细胞能够具有治疗视网膜退行性病变的功能。且C组大鼠在实验过程中皆表现出良好的治疗效果,未见产生任何不良反应,未见恶性肿瘤的产生,也未见产生任何的并发症。采用本发明其他实施例制得的视网膜祖细胞制剂进行实验的结果与此相似。
实施例5保藏编号为CGMCC NO.10419的视网膜祖细胞体内外的表达情况
1、体外培养:以实施例1提供的方法培养保藏编号为CGMCC NO.10419 的视网膜祖细胞,分别于培养15天、30天、60天后检测细胞上清液中因子的表达情况。检测采用RAYBIOTEC芯片进行检测,主要检测包括炎性因子芯片、生长因子芯片、细胞因子芯片和凋亡因子芯片。结果如图3所示。
2、体内培养:取实施例4中B组和C组的注射后8W的RCS鼠,将玻璃体腔液裂解后取其裂解液,用RAYBIOTEC芯片进行检测,主要检测包括炎性因子芯片、生长因子芯片、细胞因子芯片和凋亡因子芯片。结果如图4 所示。
结果表明,经过玻璃体腔移植RPC治疗后,有25种因子高表达。主要是: BDNF(脑源性神经营养因子)、GDF-15(生长分化因子-15)、FGF-4(成纤维细胞生长因子4)、FGF-7(成纤维细胞生长因子7)、NGF(神经生长因子)、GDNF (胶质细胞源性神经营养因子)、IGF-Ⅰ(胰岛素样生长因子-Ⅰ)、IGF-Ⅱ(胰岛素样生长因子-Ⅱ)、NT-3(神经营养因子-3)、NT-4(神经营养因子-4)、IGFBP-1(胰岛素生长因子结合蛋白1)、IGFBP-2(胰岛素生长因子结合蛋白 2)、IGFBP-3(胰岛素生长因子结合蛋白3)、IGFBP-4(胰岛素生长因子结合蛋白4)、IGFBP-6(胰岛素生长因子结合蛋白6)、OPG(骨保护素)、TGF-β1(转移生长因子-β1)、TGF-β3(转移生长因子-β3)、PDGF-AA(血小板衍生生长因子-AA)、VEGF(血管内皮生长因子)、CNTF(睫状神经营养因子)、TNF (肿瘤坏死因子)、HSP27(热休克蛋白27)、HSP60(热休克蛋白60)和HSP70(热休克蛋白70)。而在体外培养的视网膜祖细胞中,这些因子也得到了高度的表达。可见,本发明提供方法鉴定的视网膜祖细胞能够高度表达上述生长因子,因此,能够具有治疗视网膜退行性病变的功能。
以上仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (5)
1.鉴定具有治疗视网膜退行性病变功能的视网膜祖细胞的方法,其特征在于,包括:将视网膜祖细胞培养至对数生长期后进行流式细胞鉴定,满足以下条件的细胞为具有治疗视网膜退行性病变功能的视网膜祖细胞:Nestin的表达量大于90%;Pax6的表达量大于70%;Ki-67的表达量大于60%;Chx10的表达量大于85%;Sox2的表达量大于50%;CD38的表达量小于5%;HLA-DR的表达量小于10%。
2.根据权利要求1所述的方法,其特征在于,所述流式细胞鉴定采用鉴定抗体和鉴定抗体的同型对照抗体。
3.根据权利要求2所述的方法,其特征在于,所述流式细胞鉴定包括:将视网膜祖细胞培养至对数生长期,经消化、稀释后与鉴定抗体、鉴定抗体的同型对照抗体混合,孵育、洗涤、重悬后以流式细胞仪检测。
4.根据权利要求3所述的方法,其特征在于,所述消化的酶为TrypLETM-Express、胰酶或胶原酶;所述稀释采用生理盐水、DPBS缓冲液或Hank’s液;所述洗涤采用DMEM培养基、MEM培养基或DMEM/F12培养基;所述重悬采用生理盐水、DPBS缓冲液或Hank’s液。
5.根据权利要求3所述的方法,其特征在于,所述消化的酶为TrypLETM-Express;所述稀释采用DPBS缓冲液;所述洗涤采用DMEM/F12培养基;所述重悬采用DPBS缓冲液。
