The method comprises the steps of taking chitosan and magnetic powder as raw materials, forming core-shell structure particles of the magnetic powder coated by the crosslinked chitosan under the action of a crosslinking agent, and forming magnetic beads for protein purification through reduction and carboxymethylation. The magnetic beads for protein purification are used for separating and purifying proteins with his-tag, so that the separation efficiency is greatly improved.
