step one, extracting coarse powder of Guangxi big-fruit hawthorn leaves with ethanol, taking a first extracting solution, filtering to obtain a first filtrate and a first filter residue, and removing the ethanol in the first filtrate until no alcohol smell exists to obtain a first concentrated solution; wherein the weight of the ethanol used for extraction is 6 times of the weight of the coarse powder of the Guangxi big fruit hawthorn leaves;
step two, adding water into the first filter residue for extraction to obtain a second extracting solution, and filtering to obtain a second filtrate and a second filter residue, wherein the using weight of the water for extraction is 6 times of the weight of coarse powder of the Guangxi big fruit hawthorn leaves;
and step three, evaporating the second filtrate to 1/2% of the weight of the original Guangxi big fruit hawthorn leaf coarse powder, adding ethanol into the mixture until the volume fraction of the ethanol in the mixed solution reaches 60%, standing for 12 hours to obtain an upper layer clear solution and a lower layer suspension, respectively filtering the upper layer clear solution and the lower layer suspension to obtain a third filtrate and a fourth filtrate, merging the third filtrate and the fourth filtrate, removing the ethanol in the merged filtrate until no alcohol smell exists to obtain a second concentrated solution, mixing the second concentrated solution with the first concentrated solution obtained in the step one to obtain a mixed concentrated solution, and evaporating and concentrating until the relative density of the mixture reaches 1.30/80 ℃ to obtain the Guangxi big fruit hawthorn leaf extract.

Pulverizing 1[ Taiwan Chinese pear-leaved crabapple Malus doumeri (Bois) Chev ] leaf of Guangxi large-fruit hawthorn, sieving with a 20-mesh sieve, performing reflux extraction with 80% ethanol for 2 times, adding 10 times of alcohol for the first time, performing reflux extraction for 2 hours, adding 6 times of alcohol for the second time, performing reflux extraction for 1.5 hours, filtering an alcohol extract, recovering ethanol from a filtrate until no alcohol smell exists, collecting and refrigerating for later use; adding 8 times of water into filter residues, boiling for two times, adding 6 times of water into the filter residues for two times, boiling for 1 hour, filtering a water extracting solution, reducing the pressure of filtrate, evaporating and concentrating until the total amount is 1/2 of the weight of the medicinal materials, adding 90% ethanol into a concentrated solution to ensure that the alcohol content of the solution reaches 70%, uniformly stirring, standing for 12 hours, filtering and collecting an ethanol precipitation solution, recovering ethanol until no alcohol smell exists, mixing the remaining concentrated solution with the ethanol extraction concentrated solution while hot, uniformly stirring and mixing, and continuously evaporating and concentrating until the relative density is 1.35/80 ℃, thereby obtaining the extract 1.

Pulverizing leaves of fructus crataegi 2 (Malus leiocaulica S.Z.Huang) of Guangxi fructus, sieving with 20 mesh sieve, extracting with 80% ethanol by reflux for 2 times, adding 10 times of alcohol for the first time, extracting for 2 hr by reflux, adding 6 times of alcohol for the second time, extracting for 1.5 hr by reflux, filtering the extractive solution, recovering ethanol from the filtrate until no alcohol smell exists, collecting, and refrigerating; adding 8 times of water into filter residues, boiling for two times, adding 6 times of water into the filter residues for two times, boiling for 1 hour, filtering a water extracting solution, reducing the pressure of filtrate, evaporating and concentrating until the total amount is 1/2 of the weight of the medicinal materials, adding 90% ethanol into a concentrated solution to ensure that the alcohol content of the solution reaches 70%, uniformly stirring, standing for 12 hours, filtering and collecting an ethanol precipitation solution, recovering ethanol until no alcohol smell exists, mixing the remaining concentrated solution with the ethanol extraction concentrated solution while hot, uniformly stirring and mixing, and continuously evaporating and concentrating until the relative density is 1.35/80 ℃, thereby obtaining the extract 2.
Pharmacodynamic experiment method for treating rat hyperlipidemia by Guangxi big fruit hawthorn leaf extract
Experimental methods
2.1 establishment of rat model for hyperlipemia
2.1.1 animal grouping and preliminary testing
96 male rats are randomly divided into 8 groups (normal control, model, positive 1 (simvastatin), positive 2 (simvastatin + Xuezhikang), extract 1 high dose, extract 1 low dose, extract 2 high dose and extract 2 low dose) and 12 rats in each group (the grouping conditions are shown in table 1), after each group is quantitatively fed with basal feed for 7 days, the weight is weighed, the fasting is not forbidden for 16 hours, after 10% chloral hydrate local anesthesia, cardiac puncture is carried out to collect blood, serum is separated, and four indexes of total cholesterol, triglyceride, high density lipoprotein and low density lipoprotein blood fat are detected, the result is shown in table 2 below

2.1.2 rat model establishment of hyperlipidemia
From the next day after the completion of the test, the rats in the normal group were gavaged with 10ml/kg of distilled water containing 10% tween-80 every morning, and the rest groups were gavaged with 10ml/kg of the formula emulsion every morning; the basal diet was given to the rats simultaneously in each group for free feeding. Each group is continuously perfused for 2 weeks, after 2 weeks, rats are fasted and are not forbidden to be watered for 16 hours, after 10% chloral hydrate is locally anesthetized, the heart is punctured to take blood, serum is separated, and four indexes of total cholesterol, triglyceride, high density lipoprotein and low density lipoprotein are detected; the results are given in Table 3/4 below

The preparation method of the high fat milk comprises the following steps:
1) taking 10 (50 mg/tablet) propylthiouracil tablets, adding 50ml purified water to fully dissolve the propylthiouracil tablets so as to obtain 10mg/ml propylthiouracil liquid medicine;
2) weighing lard oil in a beaker, heating and stirring the lard oil by using a magnetic stirrer to melt the lard oil, weighing cholesterol and the salt of the lard oil for 6-8 times, adding the cholesterol and the salt of the lard oil into the beaker, weighing a proper amount of tween-80, fully stirring and emulsifying the mixture evenly, finally adding 20ml of thiouracil propionate liquid medicine, adding purified water to the scale, fully stirring and emulsifying the mixture evenly, and cooling the mixture to below about 40 ℃ so as to be used for gastric lavage. After the stomach is filled, the mixture is refrigerated and stored in a refrigerator, when in use, the mixture is dissolved in water at 50 ℃ and heated fully, stirred uniformly and emulsified, and then used after the temperature is moderate.
2.2 grouping and administration
Feeding rats in cages, quantitatively feeding basic feed and water to a normal control group, and feeding purified water with the same amount to the rats through gastric lavage; except for the stomach-filling administration of the corresponding drugs, the other groups were quantitatively fed with high-fat diet and water (grouping and administration schedule are shown in table 5), the weights were measured every 3 days, the animals of each group were fasted and not prohibited after 4 weeks, and the animals of each group were treated after 12 hours. Adopting cardiac puncture to take blood, then centrifugally separating serum, and detecting four indexes of total cholesterol, triglyceride, high density lipoprotein and low density lipoprotein blood fat; the results are given in Table 6/7 below:

From the above results, it can be seen that:
compared with the rats in the normal control group, the cholesterol of each group is increased compared with that of the normal group; after drug treatment, cholesterol of the hyperlipidemic rats of each administration group is reduced to different degrees. Compared with the model group, the cholesterol in the high-dose and low-dose groups of the extract 1 and the extract 2 has statistical significance, and the reduction degree of the high-dose group and the low-dose group of the extract 1 and the extract 2 is more obvious.
Compared with normal control rats, the triglyceride level of each group is increased, and the triglyceride of the hyperlipemia rats of each administration group is reduced to different degrees after the drug treatment. Compared with the model group, the data of the positive 2 groups, the extractum 1 and 2 high dose groups and the extractum 1 and 2 low dose groups for reducing triglyceride have statistical significance; wherein, the reduction degree of the high-dose groups of the extractum 1 and the extractum 2 is more obvious.
Compared with the normal control group rats, the serum LDL concentration level of each group is increased, and after the drug treatment, the serum LDL concentration level of each administration group is reduced to different degrees. Compared with the model group, the data of the reduction of the concentration of the low-density lipoprotein in the serum of each administration group has statistical significance, wherein the reduction degree of the positive 2 group, the extract 1 group and the extract 2 high-dose group is more obvious.
Compared with a normal control group, the effect of the medicines of each group in the experiment on the content of the high-density lipoprotein in the serum of the rat is small.
From the above experiments, it was confirmed that the extracts of the leaves of the wide-west crabapple (Malus pumila) thus prepared in example 7 and the leaves of the wide-west crabapple (Malus leiocalyca s.z. huang) thus prepared in example 8 all have the effect of reducing the concentrations of total cholesterol, triglyceride and low-density lipoprotein in the serum of hyperlipidemic rats.
