Abstract

技术领域
本公开涉及用于执行靶细胞的体外光分离置换的方法且更具体地涉及作为光分离置换处理的一部分的利用辐照接收器检测红细胞压积的系统和方法。
背景技术
全血由诸如悬浮在其液体成分中的红细胞、白细胞和血小板的各种细胞和非细胞成分、血浆构成。全血可以分离成其构成成分(细胞、液体或其它)并且分离的成分可以在需要该特定成分时施用到患者。
血液和/或血液成分的施用在处理有疾病的患者时是常见的。不是注入全血,单独的成分可以在患者的需要要求时被施用到患者。例如,血小板的施用(注入)可能通常指定用于其制造血小板的能力已经被化学治疗损害的癌症患者。在细胞已经经历某些另外的处理或治疗之后注入白细胞(即,单核细胞)也由于治疗原因被要求,包括具体地涉及白细胞的疾病治疗。因此,可能希望从全血分离和收集期望的血液成分且然后利用具体的血液成分治疗患者。剩余的成分可以返回到供体或者保留用于其它用途。
存在被认为主要地涉及单核细胞的疾病或病症,诸如皮肤T细胞淋巴瘤、在移植术之后的器官同种异体移植排斥和自身免疫疾病,诸如类风湿性关节炎和系统性硬化病等。
皮肤T细胞淋巴瘤(CTCL)是用于描述宽范围的病症的术语。通常,CTCL是免疫系统的癌症的类型,其中T细胞(一种类型的单核细胞)以不受控制的方式变异或生长,迁移到皮肤并形成痒的、鳞状的斑块或斑片。疾病的更晚期也影响淋巴结。CTCL的治疗处理选择之前已经是有限的。尽管已经使用化学选择,但是该特定形式的处理也具有许多相关的不希望的副作用,诸如降低的对感染的抵抗力、出血、青肿、恶心、不育症和脱发,仅举几个例子。
器官同种异体移植排斥可能以排斥非宿主原有的组织,包括移植的心脏组织以及肺、肝和肾脏移植物为特征。在移植之后的免疫抑制药物治疗是常见的。然而,存在包括由于由该类型的治疗引起的免疫系统的受损能力再出现的感染的可能缺陷。
类似地,移植物抗宿主病(GVHD)是在干细胞或骨髓移植之后可能发生的并发症,其中新移植的材料攻击移植物受体的身体。供体细胞和受体组织之间的差异常常使来自供体的T细胞将受体的身体组织作为外来物质,从而引起新移植的细胞攻击受体。GVHD可使干细胞或骨髓移植更麻烦,从而可能限制这些挽救生命的治疗。因此,在移植之后,受体通常被施用抑制免疫系统的药物,这帮助减少GVHD的机会或严重性。
包括类风湿性关节炎(RA)和系统性硬化病(PSS)的自身免疫疾病可以将身体自己的组织错误认为是外来物质的过度活跃的免疫系统为特征。结果是,身体产生攻击正常细胞和组织的自身抗体。同时,通常用于调节免疫系统和抑制过量反应或自身免疫的调节T细胞失去该能力。这除此之外还可导致在RA中的关节破坏和在PSS中的结缔组织的发炎。
发明内容
根据示例性实施方式,本公开涉及一种用于光分离置换的辐照设备,包括:曝光室,其配置为接收容纳靶细胞悬液的照射容器;辐照发射器,其配置为辐照所述照射容器和靶细胞悬液;辐照接收器,其配置为检测来自所述辐照发射器的辐射的吸收;以及处理电路,其联接到所述辐照接收器,并且配置为确定所述靶细胞悬液的红细胞压积是否超过预定的阈值红细胞压积,并且如果预定的阈值被超过,则利用处理剂量的辐射处理所述靶细胞悬液。
根据示例性实施方式,本公开涉及一种在体外光分离置换程序期间检测红细胞压积的方法,包括以下步骤:提供曝光室,其配置为接收容纳靶细胞悬液的照射容器,该靶细胞悬液包含选定量的光活化剂;提供具有辐照发射器的辐照设备,所述辐照发射器配置为辐照在所述照射容器内的内含物,其中所述辐照设备包含至少一个辐照接收器;利用所述辐照设备辐照所述靶细胞悬液,同时利用所述辐照接收器检测红细胞压积;以及当所述辐照接收器检测到红细胞压积低于最小可接受的红细胞压积时提供响应行为。

具体实施方式
存在可以在下面描述和要求保护的设备和系统中单独地或一起实施的本发明主题的多个方面。这些方面可以单独地或与本文描述的主题的其它方面组合地使用,并且这些方面共同的描述预期不排除单独地使用这些方面或者这样的方面的单独保护或者以所附权利要求中阐述的不同组合。
在用于治疗一种或多种疾病的现有疗法可能导致某些无意的副作用的情况下,可能希望或需要另外的治疗。已经表明在疾病的治疗和/或涉及单核细胞的现有疗法的副作用中是有效的一种程序是体外光分离置换或“ECP”。体外光分离置换(有时也称为体外光化学疗法)是包括以下步骤的过程:(1)从患者收集单核细胞(MNC),(2)收集的MNC细胞的光活化处理;以及(3)处理的细胞(MNC)再输注到患者。更具体地,ECP涉及与诸如8-甲氧补骨脂素或“8-MOP”的光活性化合物组合的周围血液单核细胞的体外曝光,光活性化合物然后被紫外线光活化,随后是处理的单核细胞的再输注。8-MOP和UV辐射的组合可引起ECP处理的T细胞的细胞凋亡或编程性细胞死亡。
在ECP处理期间,已知光活化引起8-MOP不可逆转地共价结合到T细胞核中包含的DNA链。当光化学破坏的T细胞被再输注时,引起细胞毒效应。例如,细胞毒T细胞或“CD8+细胞”在暴露于感染的或破坏的细胞时释放细胞毒素或者以其它方式攻击在表面上携带某些外来或异常分子的细胞。细胞毒素瞄准破坏的细胞膜并且进入靶细胞,其最终地导致靶向细胞的细胞凋亡或编程性细胞死亡。换句话说,在处理的单核细胞返回到主体之后,免疫系统认识到染色的异常细胞并且开始产生健康的淋巴细胞(T细胞)以战胜那些细胞。
体外光分离置换还可以引起单核白细胞(单核细胞的一种)变异成能够吞噬和处理细胞调亡的T细胞的树突细胞。当这些活化的树突细胞被再输注到系统循环中时,它们可以引起类似上面描述的对处理的细胞凋亡的T细胞抗原的以系统细胞毒素CD8+T淋巴细胞为媒介的免疫响应。
ECP可导致患者中的免疫耐受响应。例如,在移植物抗宿主疾病的情形中,凋亡细胞的输注可以刺激调整的T细胞生成,阻止炎性细胞因子产生,造成有效T细胞的消除且导致其它响应。参见Peritt的“PotentialMechanismsofPhotopheresisinHematopoieticStemCellTransplantation,”BiologyofBloodandMarrowTransplantation12:7-12(2006)。尽管目前免疫耐受响应的理论出现在主流的解释中,但是仍存在关于ECP相对于移植物抗宿主疾病以及其它疾病状态的行为的机理的其它理论。
在执行用于MNC的ECP程序中,希望向在MNC悬浮在其中的悬液中的可光活化材料输送正确剂量的光能量,特别是如果悬液包括基本上不透光的材料(诸如红细胞)使得其衰减光活化所预期的光能量。正确的剂量可以通过利用红细胞压积传感器和算法来确定和管理,该算法利用关于悬液的厚度、红细胞压积和透光率值的信息。
在缺乏确定红细胞压积的红细胞压积传感器时,UV剂量也可以通过UV辐照接收器(例如,光传感器)来监测,UV辐照接收器成角度以检测从UV发射器(例如,光源,诸如灯泡)发射的UV光和从在每一个发射器(例如,一组灯泡)后面的镜表面反射的UV光(且假定较少的光借助于吸收光的处理的细胞产物反射回来)。如果产物红细胞压积太高,则通过UV辐照接收器(例如,光传感器)单独监测可能没有充分地考虑被红细胞吸收的UV光且因此可能需要操作者或光分离置换系统来经由稀释将产物红细胞压积调节到足够低的水平以允许通过可光活化的基板进行能量的足够吸收。
某些实施方式可能使得能够确定MNC产物红细胞压积是否高于最小红细胞压积值以优化输送到靶细胞的辐照水平。
某些实施方式可以使得能够在不存在专用的红细胞压积传感器或细胞计数器的情况下确定红细胞压积。
某些实施方式可以以离线光分离置换方法检测红细胞压积的下限以避免产物的过度辐照。
在某些实施方式中,在ECP程序期间MNC的过度辐照可以被避免,使得细胞不会在再进入患者的血流之前过早地经历细胞凋亡或坏死,从而最小化对指定的免疫响应和ECP程序的治疗效果的损害。
图1总体上示出了构成ECP系统5且在本文描述的方法中的一个或多个中使用的机械部件。系统5可包括分离部件10和处理(即辐照)部件20。辐照部件20可以是独立的且与分离部件10分开地容纳,或者部件20和10可以集成到单个设备中。在其中部件20和10分开地容纳的实施方式中,分离设备10和辐照设备20可以邻近彼此定位,允许操作者或临床医生在特定的处理程序期间接入两个设备。患者可以连接到图1、2、4示出的流体回路200,流体回路200提供在分离部件10和辐照部件20之间的无菌封闭路径并且可以共同地安装在分离设备10的硬件上。分离设备10可以具有分离性输血设备的一个或多个特征,诸如由伊利斯诺州的苏黎世湖的Fenwal公司作为分离器出售的系统,如美国专利5,868,696中更详细描述的,其通过引用整体并入本文,但是可以使用任何合适的分离设备。尽管本文公开的实施方式结合分离设备10描述,但是本实施方式可以单独地适用于辐照设备20,在此情形中,靶细胞群可以在被收集到其它地方之后被提供到辐照设备20。
参考图1,全血可以从患者抽出并被引入分离部件10,在分离部件10处,全血被分离以提供靶细胞群。在一个实施方式中,靶细胞群可以是单核细胞(MNC)或特定类型的MNC(淋巴细胞、单核白细胞和/或树突细胞等)。从全血分离的其它成分,诸如红细胞(RBC)、血浆和/或血小板可以返回到患者或者被收集在血液处理组的预附接的容器中。
例如单核细胞的分离的靶细胞群可以然后被在处理部件20中处理和辐照。如上面论述的,单核细胞的处理可以涉及已经与单核细胞组合的光活化剂的光活化。使用诸如的设备的单核细胞收集在美国专利6,027,657中更详细地描述,该专利的内容通过引用整体并入本文。优选地,收获、收集和再输注单核细胞使用的装置可以是“多功能”自动化分离性输血设备,如分离器的情形。换句话说,分离部件10可以是能够执行各种收集实验方案和/或用于如可能被特定医院或设施需要的多用途的多功能自动化装置,使得其可以不仅在用于执行本文描述的MNC的光分离置换处理的系统和方法中使用,而且也可以用于其它目的,包括血液和包括血小板、血浆、红细胞、粒细胞的血液成分的收集和/或执行血浆/RBC交换,以及医院或医疗设施需要的其它功能。
图2-4描述了具有安装在其上的流体回路200的分离器10(图2),流体回路(图4)具有限定适合于从全血收获单核细胞(MNC)的分离室12的血液处理容器14(图3)。如图2所示,一次性处理组或流体回路200(其包括容器14)可以安装在分离器10的前面板上。流体回路200可以包括带有用于与分离器10上的蠕动泵关联的管道环路的多个处理盒23L、23M和23R。流体回路200还可以包括管道和预连接的容器的网络,用于建立与患者的流动连通且用于处理和收集流体以及血液和血液成分,如图4中示出的。如图2和图4中看到的,一次性处理组200可包括用于供应抗凝剂的容器60、用于从在处理和洗涤单核细胞的过程中的一个或多个步骤收集废弃物的废弃物容器62、用于容纳盐或其它洗涤或再悬浮介质的容器64、用于收集血浆的容器66、用于收集单核细胞的容器68,和可选地,用于容纳光活化剂的容器69。
容器68也可以用作照射容器,且照射容器68可以被预附接到一次性组20且与一次性组20成一体。可替代地,容器68可以通过诸如无菌对接等的已知的无菌连接技术附接到组200。在图2中,容器68被示出为从设备10悬挂。然而,容器68可以容纳在相邻的分离容纳的辐照设备20内(如图4中的虚线示出的),从而省略操作者将容器68放入辐照设备20内的步骤。引导到流体回路200中的容器68和/或从流体容器200中的容器68引导的管道可以是足够长度的以达到邻近分离设备但与分离设备分离地容纳的辐照设备20。
参考图4,流体回路200可包括入口管路72、用于从容器60输送抗凝剂(AC)的AC管路74、用于将红细胞从容器14的室12输送到容器67的RBC管路76、用于将贫血小板血浆(PPP)输送到容器68的PPP管路78和用于将单核细胞在血液处理容器14和收集/照射容器68之间输送的管路80。血液处理组可以包括用于进入患者的循环系统的一个或多个静脉穿刺针。如图4中示出的,流体回路200可包括入口针70和返回针82。在可替代的实施方式中,单个针可以用作入口针和出口针。
穿过流体回路200的流体流动可以与设备10和流体回路200的阀、泵、重量计和传感器配合地通过基于微处理器的控制器来驱动、控制和调节,它们的细节在前述的美国专利6,027,657中描述,但是可以使用任何合适的控制器。
根据本公开,流体回路可以进一步适合于与辐照设备20相关联。合适的辐照设备的一个示例在美国专利7,433,030中描述,该专利通过引用整体并入本文,但是可以使用任何辐照设备。辐照设备20可以包括用于在处理期间接收一个或多个容器的托盘或其它保持器。
参考图3,分离室12通过柔性处理容器14的壁限定,柔性处理容器14被承载在由旋转卷盘元件18和外部碗元件(未示出)限定的环形间隙内。血液处理容器14可以采取在使用之前围绕卷盘元件18缠绕的细长管的形式。碗和卷盘元件18可以在轭状物上在直立位置和悬挂位置之间枢转。在操作中,离心机10可以使悬挂的碗和卷盘元件18围绕轴线28旋转,在处理容器14内产生离心场。用于引起所述的卷盘18和碗元件的相对运动的机构的细节在题为“CentrifugewithSeparableBowlandSpoolElementsProvidingAccesstotheSeparationChamber”的美国专利5,360,542中公开,该专利也通过引用整体并入本文,但是可以使用任何合适的分离机构。
图5描述了处理单核细胞的在线方法的一个实施方式。“在线”光分离置换系统包括在集成的系统中的血液分离设备和辐照设备两者。在线系统提供将处理的靶细胞再输注到患者。首先可以穿过入口针70从患者抽出全血(步骤30)且将全血引入处理组200的容器14的分离室12内,在此处,全血经受离心场。离心场可以从红细胞、血小板和血浆分离靶细胞群,即单核细胞(步骤32)。诸如红细胞和血小板的成分可以返回到患者或者可以输送到容器(例如,容器67)以进一步处理。单核细胞的收集可以在一个或多个循环中处理,在给定的治疗程序中进行的处理循环的数量取决于待收集的MNC的总量。尽管图5描述了处理MNC的在线方法,但是也可以利用离线方法。在离线方法中,分离性输血设备可以用于收集靶细胞。通常地容纳在一个或多个收集容器中的收集的靶细胞从在收集期间使用的管道切断或以其它方式分离,在此处,它们稍后在单独的辐照或UVA光设备中处理,后面是随后将处理的细胞再输注到患者。在这种离线方法期间,当细胞从分离性输血设备传递到辐照设备(该设备可以位于另一个房间或实验室)时,切断与患者的连通且细胞从患者分离。
利用光对MNC的有效处理可以通过在具有合适的红细胞压积的悬液中收集单核细胞来促进。待处理的MNC悬液的红细胞压积的水平影响由MNC吸收的UV光的量,假定在MNC悬液中的红细胞阻挡至少一部分UV光到达靶向MNC。可以不存在红细胞压积的控制,特别是对于其中高精度细胞计数或专用红细胞压积传感器没有被集成的系统,视情况而定。假如辐照设备的光源配置为辐照一组光强度或有限设定的光强度值,红细胞压积的控制可能是希望的,但是假如强度和曝光设定可以容易根据红细胞压积进行调节,则红细胞压积控制可能也是希望的。辐照设备的发射器(例如,一组灯泡)就发射强度而言是不可调节的且因此可以发射恒定强度的光是常见的。如果悬浮的MNC的红细胞压积太高(使得红细胞通过MNC防止光的吸收),可能希望用稀释溶液(诸如血浆或盐水)稀释单核细胞,如步骤33中示出的(图5),以控制红细胞压积,使得希望量的UV光将达到靶向MNC。稀释的单核细胞(在容器68中)可以然后在步骤34中与合适的光活化剂组合。另一方面,如果悬浮的MNC的红细胞压积太低,则RBC可能不提供足够的辐射阻挡,导致MNC在ECP程序期间被过渡辐照,导致细胞在再进入患者的血流之前过早地经历细胞凋亡或者甚至坏死。在这种情形中,指定的免疫系统响应可能受损且可逐渐损坏ECP程序的治疗效果。
为了确定悬挂的MNC的红细胞压积不太低,由位于辐照设备内的UV-A辐照接收器(例如,传感器)观察的UV-A光强度可以用于确定是否已经达到最小红细胞压积以便防止MNC产物的过渡辐照。前面提到的美国专利7,433,030公开了UV-A传感器,但是可以使用任何合适的辐照接收器。由UV-A辐照接收器观察的光强度取决于由UV-A光源发射的UV-A光的强度、曝光室的内表面反射的UV-A光和由MNC悬液吸收的UV-A的量或百分比,MNC悬液可以包括靶细胞和非靶细胞两者以及悬浮介质。红细胞可以吸收由UV-A光源发射的UV-A光的大部分,其中由RBC进行的较大吸收随着MNC悬液的厚度增加而发生。因此,为了补偿由非靶材料吸收的光,UV-A光源可以配置为发射比由在悬液内的靶MNC实际吸收的多超过10倍的UV-A光。
通过使用UV-A辐照接收器确定适合于辐射的最佳红细胞压积可以通过绘制标准吸收率与红细胞压积曲线来实现,标准吸收率与红细胞压积曲线涉及通过MNC悬液实现的UV-A光的百分比吸收率和MNC悬液的实际红细胞压积,如图6中示出的。用于产生标准曲线的UV-A辐照接收器(例如,传感器)100可以安装在辐照设备中的多个UV-A灯泡(即,UV-A发射器)的上部集合体102a上方,如图7所示。反射板104可以布置在灯泡的上部集合体102a上方以反射由灯泡发射的光。由UV透明材料构成的曝光平面106可以布置在灯泡的上部集合体102下方以支撑照射容器68。多个UV-A灯泡的下部集合体102b可以布置在曝光平面106下方。第二反射板104可以布置在灯泡的下部集合体102b下方以反射由灯泡发射的光。第二UV-A辐照接收器100可以可选地安装在灯泡的下部集合体102b下方。辐照设备可以包括任何数量的UV-A辐照接收器100,取决于期望的精度水平,且UV-A辐照接收器100可以经由滤波器调谐以排除不同于从辐照发射器发射的UV频率光的光频率以最小化环境光干扰。
基线吸收率可以通过在不存在MNC悬液或容器的情况下在辐照期间测量UV吸收率来确定。图6的点A示出了在该示例中没有容器被装载到辐照设备内且UV-A辐照接收器100感测发射的光的最大量的点。因此,点A已经设计成其中UV-A光吸收率为0%的点,因为在该示例中在辐射设备中没有容器。点B示出了在该示例中放置在曝光平面106上的空辐照容器吸收近似5%的发射光,如由UV-A辐照接收器测量的,UV-A辐照接收器接收到比接收器在点A已经接收的光少5%的光。在可替代的示例中的其它材料的使用可产生其它结果。
在有RBC或没有RBC的情况下变化厚度T的MNC产物可以被测试,如在图6中作为正方形(T=2mm)和菱形(T=4mm)描绘的。图6的点C示出了当没有包含RBC的MNC悬液被辐照时,UV-A光吸收率近似为15%。不包含RBC的MNC悬液可以通过对从健康受试者获得的血液执行密度梯度分离(Ficoll)来获得。净化的MNC可以悬浮在近似40%的血浆和60%的盐水中以产生包含0%红细胞压积(Hct)的MNC悬液。已知量的RBC可以添加到0%Hct的MNC悬液以获得变化红细胞压积的悬液,同时维持预期的产品厚度T,且产生的吸收率百分比可以被绘制。图6示出了当MNC悬液达到近似0.5%的Hct水平时,接收器信号饱和,且标准曲线在近似40%的吸收率渐进。
测试已经揭示当MNC悬液包含给定厚度T的RBC阈值水平时靶MNC实际吸收的光量的变化最小。这种测试的示例在国际申请公布WO/2014/123521中公开。在图6的数据被获得的参数和条件下,该阈值在该实施方式中凭经验示出为对于在2和4mm之间的产物厚度T来说为近似0.5%Hct。然而,在可替代的实施方式中,阈值可以小至0.1%Hct或大至10%Hct。通过利用由图6的标准曲线提供的信息,是否已经实现最小红细胞压积因此可以通过UV-A辐照接收器确定,而没有红细胞压积检测器。图6的标准曲线示出在40%吸收率的渐近线水平下的吸收率水平表示红细胞压积低于0.5%。ECP系统5或辐照部件20可以经由处理电路来配置使得当如由接收器观察的在辐照期间的吸收率水平下降到低于40%,则可以执行响应行为。响应行为可以包括终止程序、通知操作者低于阈值的产物和/或处理另外的全血以便增加MNC产物红细胞压积的处理电路。处理电路可包括配置为或编程为执行本文描述的任何功能的模拟和/或数字电部件。处理电路可以包括一个或多个微处理器、微控制器、专用集成电路、可编程逻辑设备等,其可以进一步通过在有形存储器设备上存储的操作系统、应用程序和/或其它计算机程序来程序化。存储器可以包括用于在执行其功能时支持处理电路的各种类型的RAM、闪存、易失性存储器和/或非易失性存储器。
阈值红细胞压积可以设定为操作者期望的任何数。例如,阈值红细胞压积可以设定为0.25%Hct,在此情形中,根据图6的标准曲线,阈值吸收率水平可以是设定为近似35%。阈值吸收率水平也可以设定为从设定数偏离一定范围。在阈值吸收率水平设定为35%的示例中,偏离范围可以例如设定为5%使得在UV-A接收器检测到吸收率水平低于30%或大于40%时执行响应行为。
吸收率与红细胞压积曲线可以对于每一种照射容器设计或配置以及辐照设备设计或配置是不同的。在对于ECP程序使用来自相同或不同制造商的不同容器设计或辐照设备设计的情形中,用于特定容器设计和辐照设备设计组合的标准曲线可以利用本文公开的方法建立。

Claims (16)
1.一种用于光分离置换的辐照设备,包括:
曝光室,所述曝光室被配置为接收容纳靶细胞悬液的照射容器;
辐照发射器,所述辐照发射器被配置为辐照所述照射容器和靶细胞悬液;
辐照接收器,所述辐照接收器被配置为检测对来自所述辐照发射器的辐射的吸收;以及
处理电路,所述处理电路被联接到所述辐照接收器,并且被配置为确定所述靶细胞悬液的红细胞压积是否超过预定的阈值红细胞压积,并且如果预定的阈值被超过,则利用处理剂量的辐射处理所述靶细胞悬液。
2.根据权利要求1所述的辐照设备,其中,所述辐照设备被配置为,如果所述靶细胞悬液的红细胞压积不超过所述预定的阈值红细胞压积,则引起响应行为,所述响应行为包括终止光分离置换程序、产生错误通知和处理另外的细胞悬液以增加红细胞压积中的至少一种。
3.根据权利要求1所述的辐照设备,其中,所述处理电路被配置为使用所述辐照发射器和辐照接收器检测所述靶细胞悬液的红细胞压积,并且控制所述辐照发射器以利用所述处理剂量的辐射处理所述靶细胞悬液。
4.根据权利要求3所述的辐照设备,其中,所述处理电路被配置为使用来自所述辐照接收器的信号来控制所述处理剂量的辐射的大小。
5.根据权利要求1所述的辐照设备,其中,所述辐照设备是在线或离线系统的一部分,并且其中,所述辐照发射器发射UV-A光并且所述辐照接收器接收UV-A光。
6.根据权利要求1所述的辐照设备,其中,所述靶细胞包括单核细胞,并且所述靶细胞悬液包括光活化剂8-甲氧补骨脂素。
7.根据权利要求1所述的辐照设备,其中,所述辐照发射器被配置为发射比由在所述悬液内的靶细胞实际吸收的辐射多超过10倍的辐射。
8.根据权利要求1所述的辐照设备,其中,所述辐照接收器被调谐以排除不同于从所述辐照设备的所述辐照发射器发射的UV频率的辐照频率并且减少环境光干扰。
9.根据权利要求1所述的辐照设备,其中,所述辐照接收器被布置在所述曝光室内。
10.一种在体外光分离置换程序期间检测红细胞压积的方法,包括以下步骤:
提供曝光室,所述曝光室被配置为接收容纳靶细胞悬液的照射容器,所述靶细胞悬液包含选定量的光活化剂;
提供具有辐照发射器的辐照设备,所述辐照发射器被配置为辐照在所述照射容器内的内含物,其中所述辐照设备包含至少一个辐照接收器;
利用所述辐照设备辐照所述靶细胞悬液,同时利用所述辐照接收器检测红细胞压积;以及
当所述辐照接收器检测到红细胞压积低于最小可接受的红细胞压积时,提供响应行为。
11.根据权利要求10所述的方法,其中,所述靶细胞悬液包括单核细胞,并且所述光活化剂包括8-甲氧补骨脂素。
12.根据权利要求10所述的方法,其中,所述辐照发射器被配置为发射比由在所述悬液内的靶细胞实际吸收的辐射多超过10倍的辐射。
13.根据权利要求10所述的方法,其中,所述辐照设备被配置为,如果所述靶细胞悬液的红细胞压积不超过预定的阈值红细胞压积,则引起响应行为,所述响应行为包括终止光分离置换程序、产生错误通知和处理另外的细胞悬液以增加红细胞压积中的至少一种。
14.根据权利要求10所述的方法,其中,所述辐照设备是在线系统的一部分,并且其中,所述辐照发射器发射UV-A光且所述辐照接收器接收UV-A光。
15.根据权利要求10所述的方法,其中,所述辐照接收器被调谐以排除不同于从所述辐照设备的所述辐照发射器发射的UV频率的辐照频率并且减少环境光干扰。
16.根据权利要求10所述的方法,其中,所述辐照接收器被布置在所述曝光室内。
