Abstract

本发明公开了一种SIS组织制备食管修复材料的方法,首先取小肠粘膜制备SIS内层,将SIS裁剪成方形SIS单元,取一个SIS单元沿着圆柱体模具外壁的圆周铺设制成SIS内层,同时将羟基丁酸酯?羟基己酸酯共聚物与聚乳酸?聚乙醇酸共聚物混合后制备成混合溶液,然后在SIS内层的外表面上刷一层粘合剂,再将混合溶液均匀的涂抹在小肠粘膜下层的外表面上作为外层,得到食管修复材料;本发明采用小肠粘膜下层为内层,外层使用羟基丁酸酯?羟基己酸酯共聚物与聚乳酸?聚乙醇酸共聚物的混合物,食管修复材料生物力学强度和降解速率适宜,释放生长因子的能力与小肠粘膜下层一样,细胞相容性好,免疫原性低,溶血性低,可用于制备人工食管。

一种 SIS 组织制备食管修复材料的方法
技术领域
本发明属于生物技术领域,特别涉及一种SIS组织制备食管修复材料的方法。
背景技术
小肠粘膜下层(Small Intestinal Submucosa,简称SIS)是一种可降解的天然生物学材料,其在组织修复与再生方面具有突出的生物学优势和广阔的应用前景。
SIS是以猪小肠粘膜下层为原料,原料来源广泛,通过生物衍生化处理技术,研制开发的新型的组织再生修复材料。SIS是当前广泛用于组织工程研究的天然细胞外基质类生物材料,具有良好的细胞相容性、含有多种生长因子、无免疫原性、具有抗微生物活性、各向异性、组织特异性诱导再生等特点,是较为理想的组织工程食管支架材料。
羟基丁酸酯-羟基己酸酯共聚物(PHBHHx)是微生物细胞内的一种高分子聚合物,具有良好的力学性能,其降解产物3-羟基丁酸是人体的常见代谢物,无毒副作用、易于塑形、但降解时间较长;聚乳酸-聚乙醇酸共聚物(polylacide-co-glycolide acid,PLGA)可通过改变聚乳酸与羟基乙酸的比例来调节降解时间,但材料力学强度不足。这类材料难以诱导组织再生,体内修复效果不好。
发明内容
发明目的:本发明提供一种SIS组织制备食管修复材料的方法,以解决现有技术中的问题。
技术方案:为实现上述目的,本发明采用的技术方案为:
一种SIS组织制备食管修复材料的方法,首先取小肠粘膜下层,经消毒、机械剥除、脱脂、脱细胞和去垢后制备SIS内层,将SIS裁剪成方形SIS单元,取一个SIS单元沿着圆柱体模具外壁的圆周铺设,刮除气泡后将此SIS单元互相覆盖的部分采用粘合剂粘合,再将一个新的SIS单元覆盖于上一个SIS单元,且低于上一个SIS单元1cm,依次向下设置若干个SIS单元制成SIS内层,同时将羟基丁酸酯-羟基己酸酯共聚物与聚乳酸-聚乙醇酸共聚物混合后溶解在二氯甲烷中,制备成混合溶液,然后在SIS内层的外表面上刷一层粘合剂,再将混合溶液均匀的涂抹在小肠粘膜下层的外表面上作为外层,待凝固后冻干,取出圆柱体模具,得到食管修复材料。
进一步的,所述SIS单元的边长比圆柱体模具横截面的外圆周长长0.3mm。
进一步的,所述内层的每层SIS单元之间均通过粘合剂粘合。
进一步的,所述外层的复合量为0.5g /cm2,所述外层的羟基丁酸酯-羟基己酸酯共聚物与聚乳酸-聚乙醇酸共聚物的摩尔比为1:1。
进一步的,所述羟基丁酸酯-羟基己酸酯共聚物中,羟基丁酸酯与羟基己酸酯的摩尔比为15:3。
进一步的,所述聚乳酸-聚乙醇酸共聚物中,聚乳酸与聚乙醇酸的摩尔比为1:1。
进一步的,所述粘合剂为浓度为10%的明胶溶液。
有益效果:本发明采用小肠粘膜下层为内层,外层使用羟基丁酸酯-羟基己酸酯共聚物与聚乳酸-聚乙醇酸共聚物的混合物,食管修复材料生物力学强度和降解速率适宜,释放生长因子的能力与小肠粘膜下层一样,细胞相容性好,免疫原性低,溶血性低,可用于制备人工食管。

下面结合实施例对本发明作更进一步的说明。

1.一种SIS组织制备食管修复材料的方法,其特征在于:首先取小肠粘膜下层,经消毒、机械剥除、脱脂、脱细胞和去垢后制备SIS内层,将SIS裁剪成方形SIS单元,取一个SIS单元沿着圆柱体模具外壁的圆周铺设,刮除气泡后将此SIS单元互相覆盖的部分采用粘合剂粘合,再将一个新的SIS单元覆盖于上一个SIS单元,且低于上一个SIS单元1cm,依次向下设置若干个SIS单元制成SIS内层,同时将羟基丁酸酯-羟基己酸酯共聚物与聚乳酸-聚乙醇酸共聚物混合后溶解在二氯甲烷中,制备成混合溶液,然后在SIS内层的外表面上刷一层粘合剂,再将混合溶液均匀的涂抹在小肠粘膜下层的外表面上作为外层,待凝固后冻干,取出圆柱体模具,得到食管修复材料。
2.根据权利要求1所述的SIS组织制备食管修复材料的方法,其特征在于:所述SIS单元的边长比圆柱体模具横截面的外圆周长长0.3mm。
3.根据权利要求2所述的SIS组织制备食管修复材料的方法,其特征在于:所述内层的每层SIS单元之间均通过粘合剂粘合。
4.根据权利要求1所述的SIS组织制备食管修复材料的方法,其特征在于:所述外层的复合量为0.5g /cm2,所述外层的羟基丁酸酯-羟基己酸酯共聚物与聚乳酸-聚乙醇酸共聚物的摩尔比为1:1。
5.根据权利要求4所述的SIS组织制备食管修复材料的方法,其特征在于:所述羟基丁酸酯-羟基己酸酯共聚物中,羟基丁酸酯与羟基己酸酯的摩尔比为15:3。
6.根据权利要求4所述的SIS组织制备食管修复材料的方法,其特征在于:所述聚乳酸-聚乙醇酸共聚物中,聚乳酸与聚乙醇酸的摩尔比为1:1。
7.根据权利要求1所述的SIS组织制备食管修复材料的方法,其特征在于:所述粘合剂为浓度为10%的明胶溶液。
