Abstract

<b>本发明是提供一种芽胞恢复生长培养基,其由液体或固体基质和发芽因子组成。发芽因子诱导芽胞发芽,液体或固体基质为芽胞提供营养物质或生长环境,芽胞发芽后即可在后续实验中对其检测。本发明通过在液体或固体基质中,添加特异的嗜热脂肪芽胞杆菌芽胞发芽因子,诱导芽胞发芽,而特异的嗜热脂肪芽胞杆菌芽胞发芽因子快速诱导芽胞发芽,同时液体或固体基质为芽胞提供营养物质或生长环境,在二者共同作用下,促进芽胞的发芽与恢复生长。</b>

芽胞恢复生长培养基
技术领域
本发明涉及微生物培养基领域,尤其涉及一种嗜热脂肪芽胞杆菌的芽胞恢复生长培养基。
背景技术
在生存条件恶劣下,如营养物质不充分、代谢废物累积等,产芽胞菌种生长停滞,产生芽胞。芽胞是营养细胞的休眠体,在较长时间内保持休眠状态,能抵抗多种外界压力,如高温,高压,高寒,营养缺乏等。在合适条件下,通常是在营养物质的激活下,同芽胞营养性发芽受体结合,引发细胞内级联信号转导,诱导芽胞发芽并恢复营养菌体生长。芽胞发芽是微生物芽胞在暴露于能支持微生物生长的条件之后发生的一系列不可逆、复杂的生化事件,使芽胞进入营养菌体繁殖阶段。芽胞的高度抗逆性提高了细菌的存活,也可利用其高度的抗性进行灭菌监测。
生物指示剂是灭菌监测的金标准,通过标准化的菌株及符合要求的抗力来对特定灭菌过程进行监测。生物指示剂中所用菌株比器械或敷料上真实的微生物负荷的抗力更大,因此通过生物指示剂可对灭菌过程进行检测,保证灭菌有效性。
嗜热脂肪芽胞杆菌对热具有较高抗性,嗜热脂肪芽胞杆菌芽胞比其营养菌体的热抗性更强,常用作压蒸汽灭菌生物指示剂的指示菌种。包含有芽胞的生物指示剂在进行活菌计数,抗力测试或灭菌结束进行培养时,需诱导芽胞发芽生长。在芽胞恢复生长培养基的诱导下芽胞发芽并生长,但普通培养基诱导芽胞发芽率极低。
发明内容
本发明的目的针对嗜热脂肪芽胞杆菌芽胞,提供一种芽胞恢复生长培养基,以制备出芽胞恢复生长培养基,促进嗜热脂肪芽胞杆菌芽胞发芽,并恢复至营养菌体繁殖阶段,由此以用于芽胞活菌计数,抗力测试或灭菌结束培养验证。
本发明通过在液体或固体基质中,添加特异的嗜热脂肪芽胞杆菌芽胞发芽因子,诱导芽胞发芽。其中液体或固体基质可为培养基如营养肉汤培养基,营养琼脂培养基,胰蛋白胨大豆肉汤培养基,胰蛋白胨大豆琼脂培养基,液体或固体基质也可为缓冲液如PBS,生理盐水。发芽因子诱导芽胞发芽,液体或固体基质为芽胞提供营养物质或生长环境,芽胞发芽后即可在后续实验中对其检测。
为达成上述发明目的,本发明的技术方案如下:一种芽胞恢复生长培养基,其用于嗜热脂肪芽胞杆菌芽胞恢复生长。
在上述技术方案的基础上,进一步包括如下附属技术方案:
其原料由以下成分组成:液体或固体基质、和发芽因子。
所述液体或固体基质为营养肉汤培养基、或营养琼脂培养基、或胰蛋白胨大豆肉汤培养基、或胰蛋白胨大豆琼脂培养基、或缓冲液、或上述成分的任意组合。
所述营养肉汤培养基由以下质量比的原料组成:蛋白胨5g/L~20g/L,牛肉粉1g/L~10g/L,氯化钠3g/L~10g/L,pH6.5~7.8。
所述营养肉汤培养基由以下质量比的原料组成:蛋白胨7g/L~10g/L,牛肉粉3g/L~7g/L,氯化钠4g/L~6g/L,pH7.0~7.5。
所述营养琼脂培养基由以下质量比的原料组成:蛋白胨5g/L~20g/L,牛肉粉1g/L~10g/L,氯化钠3g/L~10g/L,琼脂10g/L~30g/L,pH6.5~7.8。
所述营养琼脂培养基由以下质量比的原料组成:蛋白胨7g/L~10g/L,牛肉粉3g/L~7g/L,氯化钠4g/L~6g/L,琼脂15g/L~25g/L,pH7.0~7.5。
所述胰蛋白胨大豆肉汤培养基由以下质量比的原料组成:胰蛋白胨10g/L~30g/L,大豆蛋白胨1g/L~8g/L,葡萄糖1g/L~5g/L,磷酸氢二钾1g/L~5g/L,氯化钠3g/L~10g/L,pH6.5~7.8。
所述胰蛋白胨大豆肉汤培养基由以下质量比的原料组成:胰蛋白胨15g/L~20g/L,大豆蛋白胨1.5g/L~5g/L,葡萄糖1.5g/L~4g/L,磷酸氢二钾1.5g/L~4g/L,氯化钠3.5g/L~7g/L,pH7.0~7.5。
所述胰蛋白胨大豆琼脂培养基由以下质量比的原料组成:胰蛋白胨10g/L~30g/L,大豆蛋白胨1g/L~8g/L,葡萄糖1g/L~5g/L,磷酸氢二钾1g/L~5g/L,氯化钠3g/L~10g/L,琼脂15g/L~25g/L,pH6.5~7.8。
所述胰蛋白胨大豆琼脂培养基由以下质量比的原料组成:胰蛋白胨15g/L~20g/L,大豆蛋白胨1.5g/L~5g/L,葡萄糖1.5g/L~4g/L,磷酸氢二钾1.5g/L~4g/L,氯化钠3.5g/L~7g/L,琼脂15g/L~25g/L,pH7.0~7.5。
所述发芽因子为酵母浸膏、或磷酸二氢钾、或磷酸氢二钾、或组氨酸、或脯氨酸、或L-缬氨酸、或L-亮氨酸、或淀粉、或葡萄糖、或肌酐、或吡啶二羧酸DPA、或吡啶二羧酸钙盐CaDPA中的一种或多种。
所述磷酸二氢钾、或磷酸氢二钾、或组氨酸、或脯氨酸、或L-缬氨酸、或L-亮氨酸、或葡萄糖、或肌酐、或吡啶二羧酸DPA、或吡啶二羧酸钙盐CaDPA的含量为10mmol/L~200mmol/L。
所述酵母浸膏、或淀粉的含量为1g/L~5g/L。
所述芽胞恢复生长培养基同时含有上述液体或固体基质和发芽因子。
本发明的有益效果如下:
本发明通过在液体或固体基质中,添加特异的嗜热脂肪芽胞杆菌芽胞发芽因子,诱导芽胞发芽,而特异的嗜热脂肪芽胞杆菌芽胞发芽因子快速诱导芽胞发芽,同时液体或固体基质为芽胞提供营养物质或生长环境,在二者共同作用下,促进芽胞的发芽与恢复生长。
具体实施方式
下面结合实施例对本发明做进一步说明。
实施例一
一种嗜热脂肪芽胞杆菌的芽胞恢复生长培养基由以下质量比的原料组成:
蛋白胨7g/L
牛肉粉4g/L
氯化钠5g/L
酵母浸膏2g/L
去离子水1L
室温搅拌至完全溶解后,使用NaOH调节pH7.2~7.4。
121°30min灭菌待用。
实施例二
一种嗜热脂肪芽胞杆菌的芽胞恢复生长培养基由以下质量比的原料组成:
蛋白胨7g/L
牛肉粉4g/L
氯化钠5g/L
磷酸二氢钾20mmol/L
葡萄糖20mmol/L
琼脂20g/L
去离子水1L
加热至完全溶解后,使用NaOH调pH7.2~7.4,
121°30min灭菌待用。
实施例三
一种嗜热脂肪芽胞杆菌的芽胞恢复生长培养基由以下质量比的原料组成:
胰蛋白胨15g/L,
大豆蛋白胨3g/L,
葡萄糖1.5g/L,
磷酸氢二钾2g/L,
肌酐30mmol/L,
氯化钠3.5g/L,
琼脂15g/L,
加热至完全溶解后,使用NaOH调pH7.2~7.4,
121°20min灭菌待用。
实施例四
一种嗜热脂肪芽胞杆菌的芽胞恢复生长培养基由以下质量比的原料组成:
氯化钠9g/L,
葡萄糖20mmol/L
吡啶二羧酸钙盐CaDPA60mmol/L,
室温搅拌至完全溶解后,使用NaOH调pH7.2~7.4,
121°20min灭菌待用。
当然上述实施例只为说明本发明的技术构思及特点,其目的在于让熟悉此项技术的人能够了解本发明的内容并据以实施,并不能以此限制本发明的保护范围。凡根据本发明主要技术方案的精神实质所做的等效变换或修饰,都应涵盖在本发明的保护范围之内。

1.一种芽胞恢复生长培养基,其特征在于:用于嗜热脂肪芽胞杆菌芽胞恢复生长。
2.根据权利要求1所述的芽胞恢复生长培养基,其特征在于,其原料由以下成分组成:液体或固体基质、和发芽因子。
3.根据权利要求2所述的芽胞恢复生长培养基,其特征在于:所述液体或固体基质为营养肉汤培养基、或营养琼脂培养基、或胰蛋白胨大豆肉汤培养基、或胰蛋白胨大豆琼脂培养基、或缓冲液、或上述成分的任意组合。
4.根据权利要求3所述的芽胞恢复生长培养基,其特征在于:所述营养肉汤培养基由以下质量比的原料组成:蛋白胨5g/L~20g/L,牛肉粉1g/L~10g/L,氯化钠3g/L~10g/L,pH6.5~7.8。
5.根据权利要求4所述的芽胞恢复生长培养基,其特征在于:所述营养肉汤培养基由以下质量比的原料组成:蛋白胨7g/L~10g/L,牛肉粉3g/L~7g/L,氯化钠4g/L~6g/L,pH7.0~7.5。
6.根据权利要求3所述的芽胞恢复生长培养基,其特征在于:所述营养琼脂培养基由以下质量比的原料组成:蛋白胨5g/L~20g/L,牛肉粉1g/L~10g/L,氯化钠3g/L~10g/L,琼脂10g/L~30g/L,pH6.5~7.8。
7.根据权利要求6所述的芽胞恢复生长培养基,其特征在于:所述营养琼脂培养基由以下质量比的原料组成:蛋白胨7g/L~10g/L,牛肉粉3g/L~7g/L,氯化钠4g/L~6g/L,琼脂15g/L~25g/L,pH7.0~7.5。
8.根据权利要求3所述的芽胞恢复生长培养基,其特征在于:所述胰蛋白胨大豆肉汤培养基由以下质量比的原料组成:胰蛋白胨10g/L~30g/L,大豆蛋白胨1g/L~8g/L,葡萄糖1g/L~5g/L,磷酸氢二钾1g/L~5g/L,氯化钠3g/L~10g/L,pH6.5~7.8。
9.根据权利要求8所述的芽胞恢复生长培养基,其特征在于:所述胰蛋白胨大豆肉汤培养基由以下质量比的原料组成:胰蛋白胨15g/L~20g/L,大豆蛋白胨1.5g/L~5g/L,葡萄糖1.5g/L~4g/L,磷酸氢二钾1.5g/L~4g/L,氯化钠3.5g/L~7g/L,pH7.0~7.5。
10.根据权利要求3所述的芽胞恢复生长培养基,其特征在于:所述胰蛋白胨大豆琼脂培养基由以下质量比的原料组成:胰蛋白胨10g/L~30g/L,大豆蛋白胨1g/L~8g/L,葡萄糖1g/L~5g/L,磷酸氢二钾1g/L~5g/L,氯化钠3g/L~10g/L,琼脂15g/L~25g/L,pH6.5~7.8。
11.根据权利要求10所述的芽胞恢复生长培养基,其特征在于:所述胰蛋白胨大豆琼脂培养基由以下质量比的原料组成:胰蛋白胨15g/L~20g/L,大豆蛋白胨1.5g/L~5g/L,葡萄糖1.5g/L~4g/L,磷酸氢二钾1.5g/L~4g/L,氯化钠3.5g/L~7g/L,琼脂15g/L~25g/L,pH7.0~7.5。
12.根据权利要求2所述的芽胞恢复生长培养基,其特征在于:所述缓冲液为磷酸盐缓冲液,生理盐水中的一种或多种。
13.根据权利要求2所述的芽胞恢复生长培养基,其特征在于:所述发芽因子为酵母浸膏,磷酸二氢钾,磷酸氢二钾,组氨酸,脯氨酸,L-缬氨酸,L-亮氨酸,淀粉,葡萄糖,肌酐,吡啶二羧酸DPA,吡啶二羧酸钙盐CaDPA中的一种或多种。
14.根据权利要求13所述的芽胞恢复生长培养基,其特征在于:所述磷酸二氢钾、或磷酸氢二钾、或组氨酸、或脯氨酸、或L-缬氨酸、或L-亮氨酸、或葡萄糖、或肌酐、或吡啶二羧酸、或吡啶二羧酸钙盐的含量为10mmol/L~200mmol/L。
15.根据权利要求13所述的芽胞恢复生长培养基,其特征在于:所述酵母浸膏、或淀粉的含量为1g/L~5g/L。
