Description
技术领域
本发明属于生物医药领域,具体地说,本发明涉及一种治疗肠道病毒感染的组分和联合用药方法。
背景技术
肠道病毒是带有单股正链RNA的微小核糖核酸病毒科病毒,目前共发现上百种血清型。多数肠道病毒感染并不引起严重的症状或仅仅引起较轻微的疾病,但在儿童和免疫缺陷人群中通常造成严重后果[1]。肠道病毒属中的肠道病毒71型(EV71)和柯萨奇病毒A16(CVA16)是引起亚太地区婴幼儿手足口病的主要病原体[2]。手足口病的症状通常较轻微如发热、咽痛、腹泻、局部出疹等,但也有些病人会发展成中枢神经系统(CNS)疾病,如无菌性脑膜炎、致死性脑炎甚至死亡[3-5]。而EV71是引起重症手足口病的主要病原体[6]。迄今为止尚无有效针对肠道病毒感染的药物治疗手段,现有的治疗手段仅限于支持性护理、静脉注射免疫球蛋白或利巴韦林[5,7],因此寻找抗病毒药物迫在眉睫。肠道病毒使用病毒自身的RNA依赖性RNA聚合酶合成基因组,所以在RNA复制过程中,子代RNA很容易发生突变。
因此,为了有效地预防或治疗肠道病毒感染,本领域迫切需要开发新型的预防和/或治疗肠道病毒感染的技术。
发明内容
本发明的目的在于提供一种治疗肠道病毒感染的组分和联合用药方法。
本发明的第一方面,提供了一种抑制肠道病毒组合物,所述组合物包括第一活性成分和第二活性成分,
其中,所述第一活性成分为肠道病毒(如,EV71)的3C蛋白抑制剂;
所述第二活性成分选自下组:
肠道病毒(如,EV71)的3A蛋白抑制剂;和
肠道病毒(如,EV71)的3D蛋白抑制剂。
在另一优选例中,所述第一活性成分包括芦平曲韦、其类似物、或其药学上可接受盐;
所述第二活性成分选自下组:伊曲康唑、其类似物、或其药学上可接受盐;和法匹拉韦、其类似物、或其药学上可接受盐。
在另一优选例中,所述芦平曲韦的类似物包括对肠道病毒的作用靶点与芦平曲韦相同的物质(如:AG7404)。
在另一优选例中,所述伊曲康唑的类似物包括对肠道病毒的作用靶点与伊曲康唑相同的物质。
在另一优选例中,所述肠道病毒选自下组:肠道病毒71型(EV71)、柯萨奇病毒A16(CVA16)、CVB3型、PV1型或EV68型,以及鼻病毒。
在另一优选例中,所述第一活性成分和第二活性成分的摩尔比约为1~20:10~100,优选地为1~10:10~100,更更优选地为1~5:10~100。
在另一优选例中,所述第一活性成分具有肠道病毒(如,EV71)的3C蛋白抑制活性。
在另一优选例中,所述第二活性成分具有肠道病毒(如,EV71)的3A蛋白抑制活性;和/或3D蛋白抑制活性。在本发明中,伊曲康唑具有3A蛋白抑制活性,法匹拉韦具有3D蛋白抑制活性。
在另一优选例中,所述组合物中还包括药学上可接受的载体或赋形剂。
在另一优选例中,所述药物组合物的剂型包括片剂、颗粒剂、胶囊、丸剂、注射剂、或口服液。
在另一优选例中,所述的组合物为单元剂型,每个单元剂型中所述第一活性成分和所述第二活性成分的含量约为日剂量的0.1至1(或0.25-1,或0.5-1),其中所述日剂量为20-100mg。
在另一优选例中,所述的日剂量为25-70mg,如25mg、40mg、50mg。
本发明的第二方面,提供了如本发明第一方面所述的抑制肠道病毒组合物的用途,用于制备预防和/或治疗肠道病毒感染的药物。
本发明的第三方面,提供了一种预防和/或治疗肠道病毒感染的方法,所述方法包括步骤:
给需要的对象施用本发明第一方面所述的抑制肠道病毒组合物,从而抑制所述对象体内的肠道病毒。
在另一优选例中,所述的对象包括人和非人哺乳动物(如啮齿动物)。
在另一优选例中,所述的施用的用量为10-100mg/kg体重,较佳地15-70mg/kg体重,更佳地10-50mg/kg体重,以所述第一活性成分的重量计。
应理解,在本发明范围内中,本发明的上述各技术特征和在下文(如实施例)中具体描述的各技术特征之间都可以互相组合,从而构成新的或优选的技术方案。限于篇幅,在此不再一一累述。

具体实施方式
本发明人通过广泛而深入的研究,首次意外地发现了一种抑制肠道病毒组合物,所述组合物中以芦平曲韦为第一活性成分,以伊曲康唑和/或法匹拉韦为第二活性成分,实验结果表明,此药物组合具有显著的协同作用,而且在被测浓度组合下均未发现增强的细胞毒性。
本发明公开了能够有效抑制肠道病毒感染的联合用药方法。针对肠道病毒的抑制活性,本发明人经过大量的测试意外的发现芦平曲韦和伊曲康唑、芦平曲韦和法匹拉韦表现出显著地协同作用。而且上述药物组合,在被测浓度组合下均未发现增强的细胞毒性。芦平曲韦和伊曲康唑组合能够阻止耐药病毒的产生。
在描述本发明之前,应当理解本发明不限于所述的具体方法和实验条件,因为这类方法和条件可以变动。还应当理解本文所用的术语其目的仅在于描述具体实施方案,并且不意图是限制性的,本发明的范围将仅由所附的权利要求书限制。
除非另外定义,否则本文中所用的全部技术与科学术语均具有如本发明所属领域的普通技术人员通常理解的相同含义。如本文所用,在提到具体列举的数值中使用时,术语“约”意指该值可以从列举的值变动不多于1%。例如,如本文所用,表述“约100”包括99和101和之间的全部值(例如,99.1、99.2、99.3、99.4等)。

芦平曲韦及其类似物
芦平曲韦(rupintrivir)最初用于治疗鼻病毒感染,研究表明它主要通过抑制EV71的3C蛋白从而抑制病毒复制[8-9]。

伊曲康唑及其类似物
伊曲康唑(itraconazole)是一种口服的三唑类广谱抗真菌剂,能够抑制曲霉菌和白色念珠菌[10-11],也能有效治疗儿童的霉菌感染[12-13],它也是一种最近被报道的肠道病毒广谱抑制剂,通过作用于病毒的3A蛋白和宿主的氧化固醇结合蛋白抑制病毒生命周期[14-15]。
在本发明的一个优选地实施方式中,根据本发明的伊曲康唑的类似物包括对肠道病毒的作用靶点与伊曲康唑相同的物质。
法匹拉韦及其类似物
法匹拉韦(favipiravir)是一种具有广谱抗病毒活性的RdRP抑制剂[20]。经过深入的研究本发明人意外地发现法匹拉韦能够在体外抑制EV71复制,机制研究发现其作用位点位于EV71的3D蛋白。
在本发明的一个优选地实施方式中,根据本发明的法匹拉韦的类似物包括对肠道病毒的作用靶点与法匹拉韦相同的物质。
苏拉明及其类似物
苏拉明(suramin)在临床上用于治疗锥体虫病,它能够与EV71的衣壳蛋白作用从而抑制病毒吸附进入细胞[16-17]。
在本发明的一个优选地实施方式中,根据本发明的苏拉明的类似物选自下组:

在本发明的一个优选地实施方式中,根据本发明的苏拉明的类似物包括对肠道病毒的作用靶点与苏拉明相同的物质(如:NF449、NF110、NM16)。
GW5074及其类似物
GW5074是Raf信号通路激酶抑制剂,对肠道病毒复制具有抑制作用,但并不作用于该细胞通路,它通过作用于脊髓灰质炎病毒的3A蛋白发挥抗病毒活性[18-19]。
本申请所涉及的化合物的结构式如下:1为伊曲康唑,2为芦平曲韦,3法匹拉韦,4为GW5074,5为苏拉明。
在联合用药疗法中,伊曲康唑和芦平曲韦,苏拉明和法匹拉韦表现出显著的强协同作用,且在被测浓度下没有检测到明显增强的细胞毒性;芦平曲韦和法匹拉韦表现出弱协同作用;芦平曲韦和苏拉明表现出叠加作用;而伊曲康唑和苏拉明,伊曲康唑和法匹拉韦,GW5074和伊曲康唑表现出强拮抗作用。
组合物
如本文所用,术语“组合物”包括药物组合物。
发明第一方面所述的组合物包含抑制肠道病毒的活性成分,和药学上可接受的载体。抑制肠道病毒的活性成分包括第一活性成分和第二活性成分,其中,所述第一活性成分包括芦平曲韦、其类似物、或其药学上可接受盐;而第二活性成分选自下组:伊曲康唑、其类似物、或其药学上可接受盐;和法匹拉韦、其类似物、或其药学上可接受盐。
本发明的药物组合物还可以包含各种与所含化合物或组合物相适应的药物辅料,并通过常规方法制备成有利于给药的剂型,如:但不仅限于水溶液注射剂、粉针剂、丸剂、散剂、片剂、贴剂、栓剂、乳剂、霜剂、凝胶剂、颗粒剂、胶囊剂、气雾剂、喷雾剂、粉雾剂、缓释剂和控释剂等。所述药用辅料既可以是各种制剂中常规使用的,如:但不仅限于等渗剂、缓冲液、矫味剂、赋形剂、填充剂、粘合剂、崩解剂和润滑剂等;也可以是为了与所述物质相适应而选择使用的,如:但不仅限于乳化剂、增溶剂、抑菌剂、止痛剂和抗氧剂等,这类辅料能有效提高组合物所含化合物的稳定性和溶解性或改变化合物的释放速率和吸收速率等,从而改善各种化合物在生物体内的代谢,进而增强组合物的给药效果。此外,还可以为实现特定的给药目的或方式,如:缓释给药、控释给药和脉冲给药等,而使用的辅料,如:但不仅限于明胶、白蛋白、壳聚糖、聚醚和聚酯类高分子材料(如:但不仅限于,聚乙二醇、聚氨酯、聚碳酸酯及其共聚物等)。所述有利于给药的主要表现有:但不仅限于提高治疗效果、提高生物利用度、降低毒副作用和提高患者顺应性等。
在水溶液注射剂中,辅料一般包括等渗剂和缓冲液,以及必要的乳化剂(如:Tweeen-80、Pluronic和Poloxamer等)、增溶剂和抑菌剂等。此外,还包括含有药学上可接受的其它药用辅料,如:抗氧剂、pH调节剂和止痛剂等。
用于制取口服液体制剂的辅料一般包括溶剂,以及必要的矫味剂、抑菌剂、乳化剂和着色剂等。
用于制取片剂的辅料一般包括填充剂(如:淀粉、糖粉、糊精、乳糖、可压性淀粉、微晶纤维素、硫酸钙、磷酸氢钙和甘露醇等)、粘合剂(如:乙醇、淀粉浆、羧甲基纤维素钠、羟丙基纤维素、甲基纤维素、乙基纤维素、羟丙基甲基纤维素、明胶溶液、蔗糖溶液和聚乙烯吡咯烷酮的水溶液或醇溶液等)、崩解剂(如:干淀粉、羧甲基淀粉钠、低取代羟丙基纤维素、交联聚乙烯吡咯烷酮和交联羧甲基纤维素钠)和润滑剂(如:硬脂酸镁、微粉硅胶、滑石粉、氢化植物油、聚乙二醇4,000、聚乙二醇6,000和月桂醇硫酸镁等)等。
用于制取乳剂的辅料一般为水、油(如:脂肪酸)、乳化剂,以及必要的防腐剂和矫味剂等。
用于制取颗粒剂的辅料与片剂类似,但造粒过程不同。根据需要,将制得的颗粒剂与助流剂混合后装入胶囊即得胶囊剂。
如本文所用,术语“对象”、“生物体”、“动物”或“患者”包括人、野生动物和家畜(Livestock)。野生动物为自然状态下未经人工驯化的动物。家畜是为了提供食物来源而人工饲养的动物,如:但不仅限于狗、猫、鼠、大鼠、仓鼠、猪、兔、奶牛、水牛、公牛、绵羊、山羊、鹅和鸡等。给予治疗的“患者”或“生物体”优先选择哺乳动物,尤其是人。
如本文所用,术语“预防”是指在未被临床标准认定的疾病前,各种用于防止疾病发生或发展的手段或措施,包括医学、物理或化学的方法,以阻止和降低疾病各种症状的发生或发展。
如本文所用,术语“治疗”是指为了阻止和降低疾病的发生或发展,使疾病病程的发展或加重得以抑制、遏制、减轻、改善、减缓、停止、延迟或反转,所描述的保持和/或用药时的疾病的、紊乱的或病理学状态的各种指标包括减轻或减少症状或并发症,或治愈或消除疾病、紊乱或状况。
如本文所用,术语“药物”是指可以用于预防或治疗某种疾病的单一化合物、多种化合物形成的组合物,或指以单一化合物为主要活性成分的组合物或制剂(formulation),还指由多种化合物为活性成分的组合物或制剂。“药物”应理解为不仅指根据一国之法律规定,通过其设立的行政机构审批并准予生产的产品,还指在为了获得通过审批和准予生产的过程中,所形成的含单一化合物为活性成分的各类物质形态。“形成”应理解为通过化学合成、生物转化或购买等途径获得。
本发明提供的作为药物组合物的给药途径,包括但不仅限于,口服(Oral)、鼻腔(Nasal)、(面)颊(Buccal)、透皮(Transdermal)、肺部(Pulmonal)、阴道(Vaginal)、皮下(Subcutaneous)或静脉(Intravenous)给予生物体。
本发明的主要优点在于:
(1)首次揭示了芦平曲韦、伊曲康唑、和/或法匹拉韦针对肠道病毒的协同抑制作用;
(2)本发明组合物起效量低;
(3)本发明的组合物可大幅降低临床用药量,既可降低生产成本,也能减轻患者负担。
(4)本发明的组合物可以阻止耐药株的产生。
下面结合具体实施例,进一步详陈本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明详细条件的实验方法,通常按照常规条件如美国Sambrook.J等著《分子克隆实验室指南》(黄培堂等译,北京:科学出版社,2002年)中所述的条件,或按照制造厂商所建议的条件。除非另外说明,否则百分比和份数按重量计算。以下实施例中所用的实验材料和试剂如无特别说明均可从市售渠道获得。
材料和方法:
细胞、病毒和化合物
RD(人横纹肌瘤)、Vero(非洲绿猴肾)细胞在含有1%青霉素/链霉素(penicillin/streptomycin,P/S)和10%胎牛血清(fetal bovine serum,FBS)的DMEM培养基中,于37℃,5%CO2的培养箱中培养。EV71FY573株(GenBank登录号HM064456)用于评估化合物抗病毒活性、病毒效价减少实验和联合用药实验。EV71G082用于病毒效价减少实验、突变病毒筛选以及评估实验。化合物伊曲康唑、芦平曲韦、法匹拉韦和GW5074分别购自Sigma-Aldrich、Santa Cruz和Chembest公司,并溶解于DMSO中进行实验。苏拉明购自拜尔公司并溶解于培养基中。
化合物的评估实验
从Sigma-Aldrich购得伊曲康唑,溶于DMSO中,终浓度为10mM,从拜尔购得苏拉明,溶于含有2%FBS的培养基中,终浓度为50mM,从Chembest购得法匹拉韦,溶于DMSO中,终浓度为400mM,从Santa Cruz公司购得芦平曲韦,溶于DMSO中,终浓度为2mM,从Sigma公司购得GW5074,溶于DMSO中,终浓度为10mM。为了评估五种化合物抑制EV71诱导的CPE活性,本发明人进行了剂量依赖实验。向96孔白板(Corning Costar)的每孔中加入50μl含有10000个RD细胞的DMEM,在37℃,5%CO2的培养箱中培养24h后,每孔分别加入梯度稀释的被测化合物(对于溶解于DMSO中的化合物,DMSO终浓度为0.25%),对照组中加入5μl 0.25%的DMSO或培养基。然后加入45μl含150PFU的病毒稀释液,每孔的终体积为100μl,培养96小时后,取出,于室温下平衡30分钟。而后每孔加入50μl CellTiter-Glo(Promega)试剂,在室温下放置10到30分钟,使用Veritas Microplate Luminometer(Turner BioSystem)酶标仪进行检测。为了测定化合物对细胞的作用,本发明人进行了细胞毒性实验,实验方法与剂量依赖实验相同,但是不再加入病毒液,取而代之的是等体积的含2%FBS和1%P/S的DMEM。
联合用药实验
为评估联合用药疗法对EV71感染的抑制效果,本发明人采用了棋盘法进行实验[21-22]。向96孔白板(Corning Costar)的每孔中加入50μl含有10000个RD细胞的DMEM,在37℃,5%CO2的培养箱中培养24h后,向96孔板的中央60个孔中分别加入5μl两种2倍稀释的被测化合物,对照组中加入5μl0.25%的DMSO或培养基。然后加入40μl含150PFU的病毒稀释液,每孔的终体积为100μl,培养96小时后,取出,于室温下平衡30分钟。而后每孔加入50μlCellTiter-Glo(Promega)试剂,在室温下放置10到30分钟,使用Veritas MicroplateLuminometer(Turner BioSystem)酶标仪进行检测。为了测定同时加入两种化合物对细胞的影响,本发明人进行了细胞毒性实验,实验方法与联合用药实验相同,但是不再加入病毒液,取而代之的是等体积的含2%FBS和1%P/S的DMEM。实验结果用MacSynergy II软件分析并得出3D示意图。
病毒效价的测定
测EV71G082株和重组病毒的效价,向12孔板(Corning Costar)的每孔中加入1ml含3×105个Vero细胞的DMEM,培养24h。病毒进行10倍倍比稀释,即将27μl病毒液与243μl的含2%FBS和1%P/S的DMEM混匀。吸出12孔板中的培养基,每孔加入200μl病毒液。放置在37℃,5%CO2的培养箱中感染1h,每隔15分钟轻轻摇动。然后吸出病毒液,加入1ml含0.8%甲基纤维素(AquacideⅡ,Calbiochem)和2%FBS的DMEM,在37℃,5%CO2的培养箱中培养6天,置于3.7%的福尔马林中固定1h后,用1%的结晶紫染色。
EV71FY573株的滴度是通过半数组织培养感染剂量(TCID50)测定的。向96孔透明板每孔中加入20000个RD细胞,培养24小时后加入100μl 10倍倍比稀释的病毒(从10-1到10-8),每个稀释度的病毒加入到10个孔中。感染1小时后,吸出病毒,加入含2%FBS的DMEM。在37℃,5%CO2的培养箱中培养7天后置于3.7%的福尔马林中固定1h后,用1%的结晶紫染色。用Reed-Muench方法测病毒滴度并表示为TCID50/ml。
病毒效价减少实验
在12孔板中接种RD细胞,3×105个/孔,37℃培养过夜,24小时后加入MOI=0.1的EV71病毒液和2倍倍比稀释的伊曲康唑、芦平曲韦、法匹拉韦、苏拉明和GW5074,在37℃培养48h后,收集上清液,放到-80℃冰箱中冻存,然后测定病毒滴度。为测定伊曲康唑和芦平曲韦联合用药对EV71效价的抑制作用,本发明人在12孔板中接种Vero细胞,3×105个/孔,37℃培养过夜,24小时后加入MOI=0.1的EV71病毒液和不同浓度的伊曲康唑和芦平曲韦,在37℃培养48h后,收集上清液,放到-80℃冰箱中冻存,然后测定病毒滴度。
免疫染色实验
为定性检测耐药株筛选实验中的病毒,本发明人进行了免疫染色。在24孔板中预先接种3×105个Vero细胞,在37℃,5%CO2的培养箱中培养24h后,向每孔加入病毒原液和10倍稀释的病毒,放置在37℃,5%CO2的培养箱中感染1h,每隔15分钟轻轻摇动。然后吸出病毒液,加入1ml含0.8%甲基纤维素(AquacideⅡ,Calbiochem)和2%FBS的DMEM,在37℃,5%CO2的培养箱中培养6天。用4%甲醛溶液固定细胞,固定好的细胞用含有0.05%的吐温-20的PBS(PBS-T)洗涤两次,然后与抗肠道病毒71型的一抗(MAB979,Merck Milipore)在室温下孵育1h,之后用PBS-T洗涤三次,与连接有辣根过氧化酶的二抗(羊抗鼠,Bethyl,Montgomery,TX)于室温孵育1h。再经过3次PBS洗涤,加入TrueBlue过氧化物酶底物(50-78-02)显色,至对照组形成明显蓝色斑点,利用蒸馏水停止反应,烘干并记录结果。产生蓝色的细胞为阳性,未产生蓝色的细胞则不含有病毒。
数据分析
把原始数据输入到Excel表格中计算信背比(S/B),信噪比(S/N),Z因子以及待测化合物对病毒的抑制率。计算公式如下:S/B=μc/μv,μc表示细胞对照组信号的平均值,μv表示病毒对照组信号的平均值,S/N=(μc-μv)/(σc-σv),σc表示细胞对照组信号的标准差,σv表示病毒对照组信号的标准差,Z=1-((3σc+3σv)/|μc-μv|),Z因子在0.5与1之间表示实验方法能够有效的区分对照组之间的差异[23]。化合物的抗病毒活性CPE抑制率=(μcpd-μv)/(μc-μv)×100%,μcpd表示待测化合物的平均信号强度,化合物对细胞的作用细胞存活率=μcpd/μc×100%。半数最大效应浓度(EC50)是指能引起50%最大效应的浓度。半数细胞毒性浓度(CC50)是指引起50%细胞毒性反应的药物浓度,在此实验中表示为实验组比对照组荧光强度降低50%。在利用Macsnergy II分析两种化合物的相互作用效果时,在95%置信度,大于零的体积代表两个化合物的相互作用为协同效果,负值代表拮抗作用。位于-25和+25之间的数值代表两种化合物见的作用不显著,位于25和50之间的数值代表显著但较弱的协同作用,位于50和100间的数值意味着中等协同作用,大于100的数值表示强协同作用。利用Chou-Talalay法分析药物相互作用时,CI>1时,药物之间为拮抗作用;CI=1时,药物间为叠加作用;CI<1时,药物间为协同作用。CI<0.1时,药物间为非常强的协同作用;0.1<CI<0.3时,两个药物为强协同作用[24]。
实施例1单组分活性测定
伊曲康唑、芦平曲韦、法匹拉韦、苏拉明和GW5074能够有效抑制EV71对细胞的感染,呈剂量依赖关系。

实施例2组合活性测试
伊曲康唑、芦平曲韦、法匹拉韦、苏拉明和GW5074相互组合在治疗EV71感染中产生协同、叠加或者拮抗三种不同效果。结果如图3所示。

实施例3伊曲康唑和芦平曲韦协同抑制病毒效价
伊曲康唑和芦平曲韦不仅能够抑制病毒感染过程中产生的细胞病变效应,还能够协同抑制病毒产出。结果如图4所示。

实施例4利用Chou-Talalay法分析芦平曲韦和伊曲康唑在联合用药中的相互作用
使用Compusyn软件,根据Chou-Talalay法分析了伊曲康唑和芦平曲韦的相互作用,结果发现两种化合物能产生协同作用。且在摩尔浓度比例为伊曲康唑/芦平曲韦=10/1时,协同效果最明显。结果如表2所示。

表2.在Vero细胞中加入EV71和不同浓度的伊曲康唑和芦平曲韦或DMSO共同培养,48小时后收集上清,用空斑形成实验测定病毒滴度。通过与对照组比较得出不同浓度组合对EV71感染的抑制效果。利用Compusyn软件分析两种药物在达到n%抑制效果时的相互作用,使用CI(combination index)和DRI(dose reduction index)作为评估参数。CI<0.1表示非常强的协同作用,0.1<CI<0.3表示强协同作用。DRI表示相对于使用一种化合物进行抗病毒治疗时,为达到相同的抑制效果,联合用药中两种化合物所需浓度减少的程度(倍数)。图中的数据来自两个独立的平行实验。
实施例5伊曲康唑和芦平曲韦联合用药能够阻止耐药病毒产生并清除所有病毒
将EV71、伊曲康唑和0.5μM或1μM的芦平曲韦同时加入Vero细胞中,传代培养病毒,并收集上清。传代20代后,本发明人通过免疫染色实验分别检测了第10、16和20代上清里的病毒,未检测出病毒。病毒效价减少实验证明,伊曲康唑[15]不能清除细胞培养中的全部病毒。实验结果表明,伊曲康唑和芦平曲韦联合疗法能够阻止耐药病毒的产生,且完全抑制EV71在体外复制。

Claims (10)
1.一种抑制肠道病毒71型(EV71)组合物,其特征在于,所述组合物的活性成分由第一活性成分和第二活性成分组成,
其中,所述第一活性成分为芦平曲韦或其药学上可接受盐;
所述第二活性成分选自下组:
伊曲康唑或其药学上可接受盐;和
法匹拉韦或其药学上可接受盐。
2.如权利要求1所述的组合物,其特征在于,所述第一活性成分和第二活性成分的摩尔比为1~20:10~100。
3.如权利要求2所述的组合物,其特征在于,所述第一活性成分和第二活性成分的摩尔比为1~10:10~100。
4.如权利要求2或3所述的组合物,其特征在于,所述第一活性成分和第二活性成分的摩尔比为1~5:10~100。
5.如权利要求1所述的组合物,其特征在于,所述组合物中还包括药学上可接受的载体或赋形剂。
6.如权利要求1所述的组合物,其特征在于,所述药物组合物的剂型选自下组:片剂、颗粒剂、胶囊、丸剂、注射剂、或口服液。
7.如权利要求1所述的组合物,其特征在于,所述的组合物为单元剂型,每个单元剂型中所述第一活性成分和所述第二活性成分的含量为日剂量的0.1至1倍,其中所述日剂量为20-100mg。
8.如权利要求7所述的组合物,其特征在于,每个单元剂型中所述第一活性成分和所述第二活性成分的含量为日剂量的0.25-1倍。
9.如权利要求7或8所述的组合物,其特征在于,每个单元剂型中所述第一活性成分和所述第二活性成分的含量为日剂量的0.5-1倍。
10.如权利要求1所述的抑制肠道病毒71型(EV71)组合物的用途,其特征在于,用于制备预防和/或治疗肠道病毒71型(EV71)感染的药物。
