Abstract

接种疫苗广泛应用于主要的每年供应的禽类(鸡、火鸡、珍珠鸡、鹌鹑、鹅、鸭……),然而造成了两个难题:
·疫苗的生产成本必须与农业保持匹配,对农民仅产生低的利润空间。尽管在接种疫苗的个体方面市场是相当大的(例如,在法国2011年4.4千万产蛋母鸡),然而疫苗仅能以超过几欧分/剂量的价格销售。
·必须确保生物安全,特别是在减毒活疫苗的使用中潜在的病原菌污染的载体。应召回接种了疫苗的饲养动物,并且在筛选金字塔(pyramidedesélection)的顶部的意外污染,对整个生产链具有巨大的影响。
在许多病毒起源的病理中,马立克氏病(Marek’sDisease-MD)是一种影响鸡的T细胞的感染性淋巴组织增生疾病。MD的临床表现主要影响母鸡(原鸡(Gallusgallus)),但是其它来自鸡形目(Galliformes)的鸟类同样可被感染并显现症状(鹌鹑或火鸡)。在60年代,这种疾病作为对世界家禽养殖的主要限制逐渐出现。马立克氏病被认为是产蛋母鸡的第二感染性疾病,据估算有10亿美元/年的总体影响,这些损失与病理和接种疫苗的成本有关(FAO数据,2002)。
马立克氏病是由一种疱疹病毒所导致,所述的疱疹病毒是马立克氏病毒属的α-疱疹病毒科的亚家族,被称为马立克氏病病毒(levirusdelamaladiedemarek,MDV)或禽疱疹病毒2型(Gallidherpesvirustype2,GaHV-2)。马立克氏病毒属包括其他四种禽病毒:禽疱疹病毒3型(GaHV-3)、更为特异地影响火鸡的吐绶鸡疱疹病毒(Meleagridherpesvirus,MeHV-HVT代表火鸡疱疹病毒)、特别地影响鸽形目(Columbiformes)的鸽疱疹病毒1型(Colombidherpesvirus1,CoHV-1)、以及特别地影响鸭科(anatidae)(鸭、鹅)的鸭疱疹病毒1型(Anatidherpesvirus1,AHV-1)。
MDV通过健康的动物与感染的鸡的直接接触,或者通过暴露于来自感染的动物的皮屑碎片所污染的处所、落物、灰尘,在家禽农场中迅速传播。在呼吸道发生病毒的进入,其中肺B淋巴细胞在疾病的进展中发挥重要的作用。
在20世纪90年代出现与高致病性病毒的出现相关的这种疾病极其急性的形式。
自70年代以来,疫苗提供了在家禽农场中控制马立克氏病的可能性。仅活疫苗有效,并且存在两种类型的疫苗,它们通过减毒GaHV-2(同源的)或者衍生自天然的致病性(apathogènes)株MeHV-1或GaHV-3(异源的)所获得。衍生自MeHV-1的疫苗制剂卵内施用于17–18日龄的胚胎,且同源的疫苗注射至在孵化中的小鸡。GaHV-2病毒的感染度与活的受感染细胞相关:在上清液中和细胞裂解液中,检测不到的游离的病毒颗粒(Churchill,1968)。因而,GaHV-2感染的传播需要在体外使感染的细胞与初始细胞共培养,或者在体内通过注射至动物施用活的感染的细胞。因此,目前的GaHV-2疫苗由培养或冷冻细胞中感染的细胞组成。
现有技术
病毒和细胞之间的关系只能使用所谓的受纳细胞进行研究,且对于MDV,复制局限于禽细胞。当细胞允许病毒增殖并且产生新的感染性病毒颗粒时,则它被称为是受纳的。细胞的基因型和分化状态是决定细胞对病毒感染的受纳性的两个主要因素。
目前,细胞培养中马立克氏病毒的传播限制于原代或二代取出的禽细胞。目前在四种不同细胞类型中获得了令人满意的复制水平:鸡胚成纤维细胞(CEF)、鸭胚成纤维细胞(DEF)(Solomon等人,1968)、鸡肾细胞(CKC)(Churchill,1968)、或者鸡胚皮肤细胞(CESkC)(Dorange等人,2000)。
少量允许禽病毒的有效复制的受纳禽细胞系,为科学家和实业家、疫苗生产者带来了难题。
该问题由对马立克氏病毒的极其敏感性(acuité)引起,迄今没有可用的未转变的禽细胞系以允许所有的马立克氏病毒有效复制,即生产。因此,疫苗仍然完全(exclusivement)通过在培养胚胎细胞、或通过酶解鸟(鸡形目或雁形目)取出的器官所获得的原代禽细胞上生产。这需要承担巨大的成本,生产工序和复杂的控制以允许确保每一生产周期中不存在污染物质。到目前为止,已证明使用从器官或胚胎取出的细胞分离细胞系的试验不是很有成效。
目前,仅有一个衍生自DF-1系(JB-J1)的系似乎能实现病毒疫苗的工业化生产的基本标准(Geerligs等人2008),没有证明该系可以用作基底(substrat)用于复制不“适应”细胞培养的病毒(在感染的动物上恢复“野生”病毒)。对于这些“野生”病毒,唯一报告的结果涉及使用过表达病毒糖蛋白基因gE的鹌鹑转化的细胞系(QM7)(Schumacher等人,2002)。这些称为SOgE的细胞支持病毒MDV-1株生长,生长水平与在胚胎细胞的原代培养上所观察到的水平相当。然而,这最后一个细胞系衍生自转化的细胞系,即在动物内能够无限增殖,与永生化过程相关的潜在的肿瘤生产能力;因此使用这类细胞制备旨在注射至鸡的疫苗制剂具有危险,因为其增殖不被控制。此外,SOgE细胞是经基因修饰的生物体,它的使用是严格规范的,并且可能不允许在疫苗制剂中使用。
应该注意的是,如文件WO98/006824中所述的,DF1细胞(以及从其衍生的JB–J1)通过从孵育10天的胚胎所获得的原代胚胎成纤维细胞的自发性建立所获得。这些细胞因此衍生自已经参与分化途径的体细胞。
以同样的方式,在专利申请EP775746中所述的CHCC-OU2细胞是衍生自孵育数日的胚胎细胞。如Ogura和Fujiwara的公开中所述的(1987),这些细胞通过永生化作用构建成系,所述的永生化作用使用N-甲基-N’-硝基N-亚硝基胍(N-Methyl-N’-nitroN-nitrosoGuanidine,MNNG)对原代细胞进行化学处理。这类处理对于禽细胞的作用是已知的,并且导致永生化系的获得,但其具有致癌特性。因此,例如同样通过化学处理获得的鹌鹑成纤维细胞QT6具有肿瘤形成特性(Moscovici等人,1977)。
此外,没有建立在复制马立克氏病毒中CHCC-OU2细胞的受纳性。在目前的情况下,CHCC-OU2细胞没有形成列为马立克氏病毒受纳的系统。此外,在专利申请EP775743中,CHCC-OU2的衍生系是使用MDV病毒长期感染的系,并且不是对复制马立克氏病毒受纳的细胞系。
以同样的方式,Lee等人(2013)描述了通过已孵育的胚胎的肝细胞的永生化所获得细胞,如获得LMH系,其本身衍生自化学处理之后所建立的胚胎肝细胞(Kawaguchi等人,1987)。如同QT6细胞的情况,这些细胞为强致癌性。它们并不像成纤维细胞所述的,并且不受纳或不是很受纳马立克氏病毒(感染滴度=10空斑形成单位(unitésformantplaques,ufp)对比ESCDL-1的500,000):因此,它未建立能够真正复制病毒的细胞,剩余的10ufp可能由于与CEL.im.细胞共培养中DF-1细胞的存留(persistance)(由感染的细胞组成的病毒接种物)所导致。
最后,专利申请EP2572728中所述的EB66细胞实际起源于胚胎,如同cES细胞作为本发明的ESCDL1细胞的起源。然而,它们保留了ES细胞的特征,尤其是表达标志物cPOUV/OCT4和NANOG。此外,这些EB66细胞不受纳马立克氏病毒。
发明内容
本发明涉及一种允许获得对禽病毒的复制受纳的非转化的细胞系方法,所述的细胞系可用于疫苗制剂。
本发明涉及一种用于诱导禽干细胞分化的方法,其导致获得衍生自胚胎干细胞(ES)的非转化的系,不再表达干细胞的特征标志物(cPOUV和NANOG),建立后在超过至少60代具有稳定的表型,并且允许禽病毒的复制。
所述的方法至少包括以下步骤:
a)存在基质(stroma)时,将禽胚胎干细胞培养至少三天;
b)在低血清浓度介质中将来自步骤a)、或c)、或d)之一的细胞培养至少两天;
c)在含1至10mM的六亚甲基二乙酰胺(hexaméthylènebisacétamide,HMBA)的低血清浓度介质中将来自步骤a)、或b)、或d)之一的细胞培养至少2天;
d)在低血清浓度介质中将来自步骤a)、或b)、或c)之一的细胞培养至少10天;
e)培养或冷冻步骤a)、b)、c)和d)中所得的允许禽病毒复制的禽细胞系。
本发明还涉及按上述的方法所获得的细胞系,以及其作为基底用于禽病毒的体外生产的用途。
本发明还涉及上述细胞系作为基底用于体外生产禽病毒或用于滴定禽病毒的用途。
此外,本发明的另一个目的涉及一种用于体外制备疫苗制剂的方法,所述的方法包括培养上述的系,并且使用至少一种禽病毒感染,还涉及疫苗制剂,严格地说,所述的疫苗制剂包括感染有至少一种禽病毒的上述的细胞系。
发明详述
本发明涉及一种用于获得允许在体外复制禽病毒的未转化的禽细胞系的方法,其至少包括以下步骤:
A)存在基质时,将禽胚胎干细胞培养至少三天;
B)在低血清浓度介质中培养至少两天;
C)在含1至10mM的六亚甲基二乙酰胺(HMBA)的低血清浓度介质中培养至少两天;
D)在低血清浓度介质中培养至少10天;
E)培养或冷冻允许禽病毒复制的禽细胞系。
根据本发明的方法具有禽胚胎干细胞(ES细胞)作为“原产品”、或起始细胞或原始细胞,具体地如Lavial等人2007所述的表达标志物cPOUV和NANOG。
被暴露于降低的血清浓度的禽ES细胞的分化表现为形态学变化(细胞大小的增加以及细胞质/细胞核比例的增加),并且对“适应”在细胞上进行培养的病毒获得低受纳性。
根据基于暴露于至少一种细胞外基质复合物和HMBA的细胞分化作用的诱导逻辑(logiqued’induction),实施从禽ES细胞建立受纳的系。
本发明提供了获得对禽病毒的复制受纳的细胞(尤其包括CHCC-OU2细胞)的可能性,与现有技术的细胞不同,所述的细胞建立成系不依靠致癌剂导致的转化,或者通过例如化学试剂的外部试剂的方式永生化方法。本发明的细胞通过分化建立,尤其通过细胞分化化学处理(HMBA)的方法,且不通过转化或永生化。因此,这些受纳细胞的建立为非自发的,与现有技术中的某些细胞如DF1细胞不同,因为它们衍生自已建立的细胞(ES细胞)并且不来自原代体细胞。
定义
一般地,在本发明的范围内,“禽病毒的复制”的表述应被理解为病毒的有效和生产性的复制的含义。因此,对于“对禽病毒的复制受纳的细胞系”意思是一种细胞系或细胞能够完成病毒周期的全部步骤,导致由感染性病毒颗粒所组成的子代病毒的生产,以及尤其是马立克氏病毒和双RNA病毒(Birnaviruses)(在马立克氏病毒的情况中,感染性病毒颗粒与接种物中的细胞相关)。如上所述,受纳的细胞允许病毒的进入和生产性复制。对禽病毒的复制的受纳性,在不同病毒间是可变的,本领域技术人员根据有关病毒能够理解不同的情况。本领域技术人员将因此能够根据待测试的病毒的类型,对每一种原始颗粒确定寻找复制因素。
熟于细胞培养的本领域技术人员容易理解术语“在合适的介质中培养”。它是指在体外、以及在合适的营养介质通常为液体中、在对细胞的生长优化的条件下(尤其是在温度和CO2浓度控制方面),细胞生长的事实。“合适的介质”是指包含血清的适于禽细胞的常规培养介质。
术语“血清”是指清除了它的细胞和凝血蛋白的血液液体。它是血液离心后获得的上清液,不含有任何凝血抑制因子。这种液体包含大量营养素(氨基酸、维生素……)、可溶性蛋白(抗体、白蛋白、细胞因子、生长因子等)、以及各种离子(钠、氯等)。在本申请中,术语“血清”是指常规用于细胞培养的胎牛血清,和鸡血清。可使用二者的组合以使各血清的优势结合。根据总体积的百分比添加血清至培养介质,即在一升的总介质中约100ml的血清,表述为“10%的血清”浓度。
“低血清浓度或含量”的表述是指低于5%的血清的百分比,具体为0.5%、1%、1.5%、2%、2.5%、3%、或4%的血清。
根据本发明,术语“基质”是指衍生自细胞的细胞外基质(matrice)复合物。例如,根据本发明的基质可以根据Coraux(Coraux等人,2013)所述的程序,从鸡皮肤的原代细胞制备;应注意的是,在这个公开中这种基质用于将鼠胚胎干细胞分化为表皮组织。这种基质还可以衍生自对禽病毒受纳的细胞(基质细胞衍生的诱导活性-SDIA-(Kawasaki等人,2000))或者进一步来自对病毒非受纳的任何类型的细胞。后者中包括以下非穷尽的列举:鼠胚胎原代成纤维细胞(MEF),目前用作通过丝裂霉素或辐射的作用失活后的营养层,鼠成纤维细胞所建立的系,如允许ES细胞的维持的STO细胞(ATCCCRL1503),3T3-J2细胞(Panacchia等人,2010),骨髓基质细胞如PA6细胞(也称为MC3T3G2),其允许神经外胚层(neurectodermique)的分化(Correia等人,2008),骨髓基质细胞如OP9细胞(ATCCCRL2749)(Nakano等人,1994),其允许诱导淋巴细胞增殖分化,或者人类的或动物的角化细胞或者对本领域技术人员有利的任何细胞类型。在本发明的范围内,本领域技术人员将能够使基质的类型与分化的类型相适应。
已知六亚甲基二乙酰胺(HMBA-化学式CH3CONH(CH2)6NHCOCH3)分化因子对鼠细胞的红白血病(érythroleucémique)系的细胞分化作用,其中分化与增殖能力的丧失相关(Marks和Rifkind,1989)。在被疱疹病毒感染的情况中,通过HMBA所进行的诱导,既赋予细胞提高的受纳性,允许某些缺陷病毒的复制(Preston和McFarlane,1998),又提高了转录因子PBX/HOX的表达以促进病毒复制(Storlie等人,2006)。目前,HMBA以1至10mM的剂量作为鸡胚皮肤原代细胞的分化诱导试剂而使用,鸡胚皮肤的原代细胞用作马立克氏病毒的复制的基底(Denesvre等人,2007)。
术语“代”是指细胞的培养到达它们的支持物最高的占用水平。通过酶或蛋白酶的混合物(胰蛋白酶、链霉蛋白酶、中性蛋白酶、胶原蛋白酶)的快速作用,细胞从它们的支持物上分离,使细胞彼此分离,并促进它们从细胞外基质分离(“胰蛋白酶消化(trypsination)”作用),并且,在培养介质中稀释后,接种于新的支持物进行数日的新培养。通常地,从它们的支持物分离的细胞进行计数,随后以一定的细胞/cm2的比例接种于培养皿,培养皿使用含0.1%牛明胶溶液的磷酸盐缓冲液(PBS)预处理至少20分钟,目的在于给转移的细胞提供细胞外基质以促进它们对塑料支持物的粘附。
以需要的时间实施不同的步骤a)、b)、c)和d)。具体地,本领域技术人员将以显微镜的方式观察跟踪细胞的分化、它们的生长和它们的活力,以及可确定步骤是否继续延长超过示出的最低时间。
存在基质时,培养步骤a)将实施至少2天,优选地至少3天,尤其是3天,更优选地3至5天。
在低血清浓度介质中的培养步骤b)将实施至少2天,优选地3天。
在含HMBA的介质中的培养步骤c)将持续至少2天,优选地3天。
在低血清浓度介质中的培养步骤d)将实施至少10天,优选地15天。
优选地,在b)、c)和d)各步骤后,所述的方法进一步包括用于在合适的生长介质中培养细胞的步骤,具体在含高于5%血清的介质中,以及优选地约10%血清。
最后步骤e)相当于获得病毒的复制受纳的细胞系,根据情况,随后其可用于病毒的复制,或者可根据本领域技术人员熟知的技术冷冻。
所述的方法可详细地如下示出:
a)存在基质时,培养禽胚胎干细胞4天;
b)在低血清浓度介质中培养细胞3天;
c)在含10%血清的介质中培养细胞10天;
d)在含1至10mM的HMBA(六亚甲基二乙酰胺)的低血清浓度介质中培养细胞3天;

f)培养或冷冻允许禽病毒的复制的禽细胞系。
以更为详细的方式,该方法可包括以下步骤:
a)存在基质时,培养禽胚胎干细胞4天;
b)在低血清浓度介质中培养细胞3天;
c)在含10%血清的介质中培养细胞10天,并通过胰蛋白酶消化传代;
d)在含1至10mM的HMBA(六亚甲基二乙酰胺)的低血清浓度介质中培养细胞3天;
e)通过胰蛋白酶消化将细胞在含10%血清的介质中传代3天;
f)在低血清浓度介质中培养细胞15天;
g)通过胰蛋白酶消化将细胞在含10%血清的介质中传代12天;
h)培养或冷冻禽细胞系,所述的细胞系在合适的培养介质中在至少60代具有稳定的表型,且允许禽病毒的复制。
如上所述的本发明的方法的全部步骤a)至d)、a)至f)和a)至h)可以任何顺序实施,有利地,它们以从a)至d)、从a)至f)和从a)至h)的顺序实施。例如当步骤a)至d)以这种顺序实施时,本发明的方法至少包括以下步骤:
A)存在基质时,培养禽胚胎干细胞至少3天;
B)在低血清浓度介质中培养来自步骤A)的细胞至少2天;
C)在含1至10mM的六亚甲基二乙酰胺(HMBA)的低血清浓度介质中培养来自步骤B)的细胞至少两天;

本方法的产物包含对禽病毒的复制受纳的细胞,所述的细胞在合适的培养介质中,在至少60代保持相同的表型,并且不再表达ES细胞的特征性标志物cPOUV和NANOG。
这些细胞命名为“ESCDL-1”,代表“胚胎干细胞衍生的系1(EmbryonicStemCellDerivedLine1)”。ESCDL-1为成纤维细胞样(aspectfibroblastique)的贴壁细胞,即与原代成纤维细胞相比,更为分散(étalées)且较不伸展的(étirées)。通过它们的形态学、它们的生长特征、以及ES细胞的标志物的表达消失与ES细胞的系相区分。
这个最后的特征还提供了将它们与现有技术的细胞相区分的可能性,例如EB66细胞(见EP2572728)或者DF1、LMH、CEF和cES细胞(BP25)(见本申请的实施例2),它们保持了ES细胞的标志物的表达。因此,本发明的ESCDL–1细胞不表达以下标志物的至少一种:cPOUV/OCT4、NANOG、SSEA1、EMA1、IXF1、KRT8和KRT19。更优选地,它们不表达这些标志物中的至少两种、至少三种、至少四种、至少五种或者至少六种。甚至更优选地,它们不表达这些标志物中的任一个。
此外,本发明的ESCDL–1细胞强表达OLFM3基因,以及与原代成纤维细胞中所观察到的水平相当的WISP1、THBS2和EDN2基因。
本发明的细胞与ES细胞相比,具有低端粒酶活性。ESCDL1细胞是未转化的细胞,并且建立成系,所述的细胞增殖但不致瘤,与以化学方法或通过外源永生化试剂的作用所建立的系不同。ESCDL-1细胞的增殖能力(20%的细胞在S期),高于鸡胚皮肤原代细胞(CEPP)(3%的细胞在S期)。此外,值得注意的是ESCDL-1细胞对凋亡诱导剂敏感,并且在生长因子耗尽的或者存在自噬诱导剂的介质中,具有强诱导自噬过程的倾向。
根据本发明的方法提供了获得对于多种禽病毒(尤其是疱疹病毒、痘病毒、冠状病毒、双RNA病毒、呼肠弧病毒(Réo)和轮状病毒(Rotavirus)的复制受纳的细胞系的可能性,以及尤其用于感染性黏液囊炎(IBDV)的中介的马立克氏病毒和双RNA病毒。
根据本发明的一个优选的方面,起始或原始禽胚胎干细胞是鸡细胞。优选地,ES细胞来自具有杂合子Cou-nu基因型(Na/na)的鸡。
更优选地,原始禽胚胎干细胞是X期至XIV期的胚盘(blastoderme)分离细胞(根据EyalGiladi和Kovak,1976)。与现有技术中的某些细胞不同,根据本发明的细胞系来源于胚胎,因其衍生于胚胎干细胞而未孵育,它们本身从这些未孵育胚盘细胞的培养获得。
这些原始禽胚胎干细胞表达多种标志物基因,如在专利申请WO2007/110343中所示的那些,以及特别是标志物基因cPOUV和NANOG。
根据本发明一个优选的方面,所获得的禽细胞系从它建立起,在至少60次连续的传代具有稳定的表型,优选地至少70次传代,更优选地至少80次传代。
优选地,根据本发明的禽细胞的表型特征更具体的为以下的:
■成纤维细胞/间叶细胞型的形态;
■表达CD44、整合素β1、和胶原蛋白1;
■不表达ES细胞的标志物,尤其包括cPOUV和NANOG、KRT19和KRT8,但强表达OLFM3基因,以及以与原代成纤维细胞中所观察到的相当的水平持续表达WISP1、THBS2和EDN2基因;以及
■对马立克氏病毒和禽双RNA病毒的复制的受纳性。
间叶细胞成纤维细胞是指这样的一种细胞,其具有与结缔组织的未充分分化(peudifférenciées)细胞相似的形态,尤其具有长伸展(longsprolongements)和在细胞的中央卵形的细胞核。
根据本发明的另一方面,所获得的禽细胞系可被基因修饰以表达感兴趣的蛋白。感兴趣的蛋白可以是“报告”蛋白(荧光蛋白;酶例如不同形式的荧光素酶或磷酸酶……)、用于修饰基因组的酶(重组酶)、可作为某些基因已被删除的病毒的互补干预的自体的或异源的病毒蛋白、或者过表达可对病毒的复制具有影响的其它细胞蛋白。
如实施例中所示,即使如上述的基因修饰,根据本发明的禽细胞系保留了它对禽病毒的受纳性。
本发明还涉及可通过上述的方法获得的细胞系,即在体外允许禽病毒的复制的未转化的细胞系,尤其是马立克氏病毒和双RNA病毒。
具体地,该细胞系表达以下标志物:CD44+、整合素β1以及胶原蛋白1。在另一方面,它不再表达被抗体IXF1、MC-480(SSEA1)和EMA-1所识别的抗原,同时后者被原始胚胎干细胞所表达。它们不再表达基因cPOUV/OCT4、NANOG、KRT19和KRT8,但是强表达基因OLFM3,以及与原代成纤维细胞中所观察到的水平相当的基因WISP1、THBS2和EDN2(在图中等于1)。在成纤维细胞DF1中,这些基因的水平低10至100倍(图5)。以同样的方法,肝细胞LMH完全不具有这些标志物相同的表达谱。本发明还涉及上述的细胞系作为基底用于禽病毒的体外生产的用途。
本发明还涉及上述的细胞系用于禽病毒的滴定以及因此确定病毒感染的严重性的用途。
最后,本发明还涉及一种用于体外制备疫苗制剂的方法,其包括培养上述的系,以及用至少一种禽病毒感染所述的系。也就是说,本发明涉及用于体外制备疫苗制剂的方法,其至少包括以下步骤:
A)存在基质时,培养禽胚胎干细胞至少3天;
B)在低血清浓度介质中培养至少两天;
C)在含1至10mM的六亚甲基二乙酰胺(HMBA)的低血清浓度介质中培养至少2天;
D)在低血清浓度介质中培养至少10天;
E)培养或冷冻允许禽病毒的复制的禽细胞系;
F)使用至少一种禽病毒感染所获得的细胞系。
本申请还涉及一种疫苗制剂,其包含感染有至少一种禽病毒的上述的细胞系。
在上述不同的具体用途的范围内,禽病毒尤其选自马立克氏病毒和双RNA病毒。

材料和方法
1.生长因子、化学品和抗体
用于生长禽胚胎干细胞的生长因子(PeproTech-法国)按以下浓度使用:
■IGF–1(PeproTech-100-11)以5ng/ml
■IL6(PeproTech-200-06)以2ng/ml
■sIL6–Ra(PeproTech-200-06R)以2ng/ml
■SCF(PeproTech-300-07)以1ng/ml
HMBA(N,N′–六亚甲基二乙酰胺-编号224235),维甲酸(编号R2625),DMSO(二甲基亚砜-编号D2650),丙戊酸钠(丙戊酸,钠盐-编号P4543)购自Sigma-Aldrich(SigmaAldrich-法国)并且以文中所示出的浓度使用。
2.细胞
a.BP25干细胞:细胞在所述的条件下维持(Pain等人,1996),在经辐射的STO细胞的饲养层上,在DMEM/F12介质(1:1)(Gibco-Invitrogen–法国)中添加有10%的胎牛血清(SVF)(PAN-D.Dutscher-法国)、丙酮酸钠(1mM-Lonza-法国)、谷氨酰胺(2mM-Lonza-法国)和非必需氨基酸(NEAA100X-Lonza–法国)以及包含前文中所示浓度的生长因子(ES介质)。
b.ESCDL-1细胞:通过BP25细胞的分化获得ESCDL-1细胞(参见下文)。细胞在37℃、含5%的CO2的环境中、在DMEM/F12介质(1:1)中(Invitrogen-法国)培养,其中所述介质中添加有10%的SVF(PAN-D.Dutscher-法国)、丙酮酸钠(1mM-Lonza-法国)、谷氨酰胺(2mM-Lonza-法国)以及非必需氨基酸(NEAA100X-Lonza-法国)。对细胞进行胰蛋白酶消化(trypsinées)一周两次(胰蛋白酶10X-T4549-SigmaAldrich-法国),并且根据1比3的比例分配。
c.鸡胚皮肤原代细胞(CESkC):原代细胞根据所述的程序获得(Dorange等人,2000)。
d.3867K和NWB细胞:在V.Nair的实验室获得转化有RB-1B-UL47EGFP病毒(Jarosinski等人,2012)的系,在RPMI1640介质(Lonza-法国)中培养,其中所述的介质含有10%的SVF、10%的胰蛋白示磷酸肉汤(TPB)(编号T8782-SigmaAldrich-法国)、以及4.5g/l的葡萄糖。两种转化的淋巴细胞系3767K和NWB来自使用RB-1B-UL47EGFP病毒感染的鸡中采样的肾肿瘤(该系由英国伯克郡的康普顿Pirbright研究所的LorraineSmith提供给我们)。
3.病毒
a.GaHV-2(MDV)
i.Bac20和衍生病毒:Bac20病毒(Schumacher等人,2000),Bac20eGFPVP22(Denesvre等人,2007)以及Bac20UL17-mRFP(Chbab等人,2009)通过转染含亲本(Bac20)或突变的病毒基因组的杆状病毒(bacmides)获得。
ii.RB-1B和衍生病毒:在Bac20所述的相似的杆状病毒转染的条件下,从L.Petherbridge(Petherbridge等人,2004)所述的棒状病毒获得病毒RB-1B。根据与Jarosinski(Jarosinski等人,2007)所述的RB-1B棒状病毒的内容中Bac20UL17-mRFP相同的逻辑,构建病毒RB-1B-UL17mRFP。病毒RB-1B-UL47EGFP(Jarosinski等人,2012)通过将试验细胞与3867K或NWB细胞接触所获得。
b.MeHV1:疫苗株FC-126从冻干疫苗制剂(-Mérial-法国)恢复(passée)至培养。所有的马立克氏病毒(GaHV-2和MeHV1)的接种物由感染的、液氮中保存的冻干细胞组成。
c.GaHV-1(ILTV):所使用的病毒与疫苗株(Intervet-法国)一致。该病毒在来自冻干疫苗制剂的LMH细胞上生产。病毒接种物来自以200xg离心后的感染的细胞的上清液,保存在-80℃(Fuchs和Mettenleiter,1999)。
d.IBDV:感染性鸡黏液囊炎的双RNA病毒,疫苗株CT,由B.Delmas以感染的培养上清提供给我们(Lejal等人,2000)。
4.干细胞的分化
a.暴露于细胞分化试剂:胰蛋白酶消化处于对数生长期的BP25细胞,计数并以1·105细胞/cm2接种于通过含有牛明胶的溶液预处理的培养皿中(含0.1%的牛明胶的PBS-编号G9391-Sigma–法国)。在37℃的含有10%的胎牛血清的EdeWilliam介质中24小时后,细胞在含有3%的鸡血清(SP)(Gibco-InvitroGen-法国)和2%的胎牛血清(SVF)的相同的介质中孵育24至48小时。融合层在含1%的SVF和1.5%的SP的EdeWilliam介质(WE1-1.5%)中暴露于不同的细胞分化分子24小时,然后感染(参见下文)。
b.ESCDL–1:36代的细胞BP25以1.5·106个细胞接种在75cm2的皿中,所述的皿包含根据C.Coraux所公开的合适的程序(Coraux等人,2003)由原代细胞(CESkC)所制备的细胞外基质。细胞首先在含有10%SF的WE介质中培养4天(图1),然后更换介质,在含有1%SVF-1.5%SP的WE介质中培养3天。在胰蛋白酶消化后,细胞在75cm2的皿中在含有10%的SVF的WE中再培养10天,胰蛋白酶消化并且暴露于WE1%SVF-1.5%SP中通过HMBA(5mM)诱导72小时(3代ESCDL-1)。细胞经胰蛋白酶消化,接种于明胶培养支持物,并且在含10%的SVF的WE中培养一代(P4)。然后在含1%SVF-1.5%SP的WE中培养细胞15天,胰蛋白酶消化并在含10%的SF的DMEM-F12介质中培养(P5至P7)。从7代,细胞每周胰蛋白酶消化两次,以1比3的比例分配,并且以相同的介质培养至18代。细胞从该代(P18)开始保存于液氮中。
5.转染:通过使用AmaxaTM技术(AmaxaNucleofector-Lonza-法国),使用BasicFibroblast试剂盒(编号VPI–1002)转染ESCDL-1细胞,并且对于4·106个细胞,质粒和棒状病毒ADN的量在4至8μg不等。转染后,将细胞分配至明胶预处理的6-孔培养板的3个孔中,并在含10%的SVF的DMEM/F12介质中培养16小时。细胞置于与实验相匹配的介质中(参见下文)。为筛选在pCDNA3中的pCMV启动子下表达转基因的稳定的系,细胞置于含有0.5mg/ml的G418(编号-SigmaAldrich)的10%SVF的DMEM/F12选择介质中,随后在转染后24至48小时首次胰蛋白酶消化。在编码转基因的质粒缺乏筛选基因的情况下,编码转基因的质粒与提供潮霉素抗性的基因的质粒(pTK-HYG2)共转染。在这种情况下,在转染后24小时立刻将细胞置于选择介质(含80μg/ml潮霉素的10%SVFDMEM/F12)。
6.病毒感染
a.接种物的制备
i.棒状病毒的转染:在ESCDL-1细胞中,通过Amaxa技术的方式转染包含病毒基因组的棒状病毒。在诱导的CESCBP25中,棒状病毒遵循制造商的说明通过试剂FuGENE(Roche)进行转染。转染后,ESCDL-1细胞在10%SVF的DMEM/F12介质中孵育16小时,然后在含1%的SVF、1.5%的SP以及0.3mM的抗坏血酸的DMEM/F12介质中孵育。诱导的BP25细胞放回含所述浓度的诱导剂的WE1-1.5%中。
ii.分选的感染的细胞的使用:已描述制备分选的感染的细胞(Denesvre等人,2007)。在37℃孵育1小时30至2小时后移出接种物(分选的细胞),并且ESCDL-1细胞放回含有1%的SVF、1.5%的SP以及0.3mM的抗坏血酸的DMEM/F12介质中。诱导的BP25细胞放回含有所述浓度的诱导剂的WE1-1.5%中。
iii.转化的淋巴细胞的使用:离心NWB或3867K细胞,置于低胎血清浓度的介质中(DMEM/F12),并且接种于ESCDL-1细胞的融合层。在37℃孵育1小时30分钟后,移出接种物,并将介质替换为含有1%的SVF、1.5%的SP以及0.3mM的抗坏血酸的DMEM/F12介质。
iv.冷冻接种物:在每种情况中,以具有最大细胞病变效应的感染细胞层制备接种物。在胰蛋白酶消化和离心(200xg持续10分钟)后,细胞团(culotscellulaires)被95%的SVF和5%的DMSO所组成的冷冻混合物重悬(reprisdans),并且根据3管/75cm2皿的比例分配至细胞冻存管。在-80℃恒温箱(boiteisothermal)中24小时后,管转移至液氮中。
b.滴定:在接种后48小时的细胞层上,在原代细胞(CESkC)或在ESCDL-1细胞上实施滴定。早期描述的方法(Blondeau等人,2008)适用于ESCDL-1细胞的情况,所述的方法中DMEM/F12介质被替换为WE,并且在接种后,细胞层置于半固体介质中,所述半固体介质是含1%的甲基纤维素(Méthylcellulose,编号25449.182-VWR-Prolabo)、1%的SVF、1.5%的SP和3mM的抗坏血酸的DMEM/F12或WE(ESCDL-1)。3天后(Bac20和突变体)或者4天后(RB-1B和突变体)细胞层通过在含4%的多聚甲醛溶液(编号P6148-SigmaAldrich-法国)的PBS缓冲液中室温孵育20分钟以固定。随后移除固定溶液,并使用PBS冲洗细胞层。为检测病毒抗原,根据所述的程序(Dorange等人,2000)对细胞进行渗透和免疫荧光反应。根据N.Richerioux所述的程序实施(Richerioux等人,2012)不同细胞情况的空斑(plages)大小的测量,平均数量为80个独立的空斑(platesindividualisées)。
c.病毒载量:根据K.Jarosinski所述的程序(Jarosinski等人,2002),通过使用Taqman型定量PCR反应(qPCR),估算每细胞病毒基因组的拷贝数。简言之,通过每个基因的一对引物和特异性探针的方式,定量样本中细胞基因iNOS和病毒基因ICP4(表2)。为了量化拷贝数的测量,使用每个目标基因已知ADN拷贝数的稀释范围。iNOS的标准品为pBS–iNOS质粒(K.Jarosinski赠予)调整至5ng/μL的浓度,存储于-20℃。ICP4标准品是BAC-20棒状病毒,调整至50ng/μL,存储于4℃。标准品的稀释范围是在水中10-3至10-7所生产。使用Absolute-BlueqPCRMix&ROX试剂盒(ABgene)、引物对和探针(Eurogentec)在96-孔板中实施qPCR。在终体积20μL中进行反应(10μL的MixFastBlue2X+0.10μL的100μM每种引物+0.5μL的10μM探针+9.5μL的ADN)。
7.细胞和病毒抗原的检测
a.免疫荧光:将直径为14mm且厚度为0.17mm玻片在0.1%明胶的PBS中孵育30分钟后,在24-孔板中(编号353935BDFalcon-法国)在所述玻片(Marienfeld-VWR-法国)上培养细胞。使用4%多聚甲醛固定细胞。对经固定的和渗透的细胞上完成免疫荧光反应的条件已被描述(Chbab等人,2009)。对于非渗透的细胞,在含2%的牛血清白蛋白(BSA)(fractionV-PAA-法国)的PBS缓冲液中实施整个孵育过程。所使用的一抗在表1中描述。所使用的二抗,鼠抗IgG、兔抗IgG或鸡抗IgG偶联有Alexa488或者594(MolecularProbes-Invitrogen–法国)。聚合肌动蛋白(F–actin)标记有偶联了Alexa594的鬼笔环肽(编号A12381-MolecularProbes-Invitrogen-法国)。使用配备有ColibriAxioCamMRm相机的Zeiss装置的ZeissAxiovert显微镜拍摄照片,并且以Axiovision软件(CarlZeissSA-法国)控制。
b.免疫印迹(WB)。从ESCDL-1细胞提取物中检测病毒抗原的条件,与用于原代细胞的所述的条件(Chbab等人,2009)相同。
8.透射电子显微镜法:在ESCDL-1细胞上进行转染后,用第3代的Bac20病毒感染第25代同样的细胞。这些细胞在6-孔板UpCell(NuncUpCellsurface编号174901-ThermoScientific-法国)上,在所述的感染条件下(参见下文)培养,(或者对于对照细胞进行“模拟”感染),并且根据供应商所述的程序对层进行回收,以便随后在Trump固定剂中固定。根据前述的程序(Denesvre等人,2007),制备在电子显微镜下检测的感染的和未感染的细胞。
结果
实施例1.ESCDL-1系的分离
根据基于依次暴露于衍生自受纳细胞的细胞外基质复合物以及HMBA的细胞分化作用的诱导逻辑,从BP25细胞建立受纳系(基质细胞衍生的诱导活性-SDIA-(Kawasaki等人,2000))。根据Coraux所述的程序实施来自鸡皮肤原代细胞的细胞外基质的制备(Coraux等人,2003)。所获得的培养支持物在4℃储存45天,并且在存储结束时检测无细胞生长。在诱导的0天,以26,500细胞/cm2的量,在含10%的SVF的WE介质中,在含有细胞外基质的有盖(Pétri)皿中接种干细胞(图1)。在暴露于降低的血清浓度和两次胰蛋白酶消化的周期后,在含1%的SVF和1.5%的SP的WE介质中,将3代细胞暴露于HMBA(5mM)的作用3天。建立的程序(图1)持续进行了25天,包括3次胰蛋白酶消化,在建立的培养后得到第6代。随后细胞转移至含10%的SVF的DMEM/F12介质,在该介质中培养细胞。胰蛋白酶消化频率从每周1次(6代和7代)过渡至对8代和以后代次的每周两次,以平均每cm245,000细胞接种。与系的构建的同时,在13代一次性试验对病毒复制的受纳性,揭示Bac20病毒在ESCDL-1上的增殖。系从23代被认为是已被建立,并且从18代开始制备冷冻细胞的库存。
实施例2.ESCDL-1系的特征
ESCDL-1细胞具有与成纤维细胞相似的形态(图2),它们的倍增期约为40小时,并且它们似乎从第100代开始显示逐渐丧失它们的增殖能力。它们的生长曲线在图3中显示。
检测BP25-36b和ESCDL-1间的谱系(filiation),以便确定细胞不是衍生自被用于建立细胞外基质的细胞中的持久存活的细胞。对于从裸颈(cou-nu)鸡胚胎获得的BP25细胞(Pain等人,1996),我们使用PCR以从干细胞(BP25)、从ESCDL-1(P38)细胞和从皮肤胚胎原代细胞(LD1母鸡)所提取的基因组ADN,将LD1动物与裸颈动物区分(Mou等人,2011)。所获得的片段的分析显示,BP25和衍生自裸颈基因型(杂合子-Na/na)胚胎的ESCDL-1相似,然而原代细胞起源于“wt”基因型(纯合子na/na)。
图4示出了通过RTPCR技术在BP25和ESCDL-1细胞中的Oct4和NANOG标志物的信使ARN的检测。仅干细胞BP25表达标志物OCT4(7)和NANOG(12)的ARNm。在ESCDL-1细胞(孔10和15)中,未检测到这些ARNm。在所有的ARN制剂中GAPDH的ARNm的检测(1至5),证实了皮肤ARNm制剂(1-6-11)、干细胞BP25(2-7-12)、肝(3-8-13)细胞、肾(4-9-14)细胞以及ESCDL-1细胞(5-10-15)质量的一致性。这些结果被qRTPCR所证实。在免疫荧光中证明ESCDL-1细胞中不能观察到干细胞的其他标志物的表达(表1)。
表1:BP25细胞和ESCDL-1细胞上多能标志物的表达的比较(使用MDCK细胞作为对照)。通过间接免疫荧光,BP25细胞上检测到膜抗原SSEA-1(ES细胞的特征)和EMA-1膜抗原(被认为是PGC细胞良好的标志物),而ESCDL-1细胞不表现任何标志。
最后在ESCDL-1细胞中检测端粒酶活性,与LMH细胞(阳性对照)和其他细胞系(DF1和CLEC)以及原代细胞(CESkC)相比较;与DF1和CLEC细胞所观察到的活性相比(Esnault等人,2011)可检测出低活性,但是显著低于在LMH中所观察到的活性。
通过RT-PCR(Lavial等人2007)比较分析DF1、LMH、CEF、cES(BP25)以及根据实施本发明所获得的ESCDL-1细胞中多能基因(5a)和成纤维细胞的标志基因(5b)的表达,以及某些标志物的存在。该分析所用引物在下表2中列出。该分析的结果归纳于图5a和5b中。

实施例3.ESCDL-1细胞对禽病毒的复制的受纳性
研究以下病毒:
1.适应细胞培养的减毒病毒:
·Bac20病毒和衍生物:使用病毒Bac20UL17mRFP(Chbab等人,2009)作为研究受纳性的主要工具,因为它的操作灵活性(红色荧光蛋白融合至蛋白UL17)以及其相对于亲本病毒(未标记的Bac20)它的复制特性的保留。编码Bac20和Bac20EGFPUL49病毒的棒状病毒(Blondeau等人,2008)也转染至ESCDL-1细胞。还使用以病毒Bac20-eGFPUL49感染的细胞(鸡皮肤原代细胞)进行感染(Denesvre等人,2007)

实施例3A.Bac20UL17mRFP
第一阶段对适应细胞培养的病毒实施试验,所述病毒衍生自编码Bac20病毒的基因组的棒状病毒。转染棒状病毒至ESCDL-1细胞提供了与原代细胞不同的研究模型。

实施例3B.Bac20EGFP-UL49和Bac20
转染棒状病毒至ESCDL-1细胞提供了再分离复制性病毒的可能性,并且在转染后第四天立刻通过细胞病变效应(感染细胞的空斑)的发生而表达。
在ESCDL-1中,从转染棒状病毒获得的Bac20病毒是复制的,并且保留与CESkC中所述相似的病毒形态发生的主要步骤。
如下表所显示,使用Bac20病毒感染的ESCDL-1细胞显示A、B和C型的核内衣壳,胞质内衣壳C以及内和外核膜之间的包裹衣壳(capsidesenveloppées)。

在20个ESCDL-1细胞中1084个衣壳的计数,然而提供了检测到与所述的CESkCs分布略有不同的可能性,无疑地与实验设计的差异相关。然而值得注意的是,胞质内包裹衣壳(成熟衣壳)的百分比在两种细胞模型中相同(Denesvre等人,2007)。
实施例3C.BacRB-1B:
在受纳马立克氏病毒复制的系的发育的范围内,检测该系允许衍生自BacRB-1B的完全致病性病毒的复制是重要的(Petherbridge等人,2004)。转染在UL17蛋白上标记有mRFP的编码RB-1B病毒的棒状病毒,其表现为转染后第四天立刻出现细胞病变病毒效应(ECP)。在ESCDL-1中3次连续传代后,在CESkC和ESCDL-1上滴定该病毒,所计算的滴度相似(2.5至3.5x105UFP/ml),并且平均于目前所观察到的在原代细胞上生产该类型病毒(“标准品”细胞基底)。我们还试验了来自淋巴细胞NWB和3867K病毒感染的传播的可能性。将复活病毒RB-1BUL47GFP的转化细胞与ESCDL-1细胞进行共培养,这导致了细胞的感染,以及导致了病毒的传代在ESCDL-1细胞中进入裂解周期。
实施例3D.MeHV1病毒
在示出的条件下接种病毒于ESCDL-1细胞,并且该接种表现为出现细胞病变效应。在CESkC和ESCDL-1上滴定来源于首次传代的病毒。以UFP测量的病毒滴度相同(4.05±0.9x104)。
实施例3E:双RNA病毒
适应细胞培养的IBDV病毒株CT(CT株,访问编号GENBANKEMBLAJ310185)在CEPP或在ESCDL-1中生产,并在两细胞系统中以有限稀释滴定子代病毒。在CESkC上获得的滴度为1x106.79TCID50,在ESCDL-1为1x106.77TCID50。
在原代细胞(IBDV-PP1H+或H–)或ESCDL-1细胞(IBDV-ESCDL-134或69H+或H–)在2代细胞(PP2)或39代的ESCDL-1细胞在上生产的IBDV病毒的滴定结果在下表5中显示:
CESkCP2:通过胰蛋白酶消化获得的二代细胞并将CESkC放回培养
该表提供了ESCDL-1细胞的表型的稳定性的观点,其证明了在34代或69代对IBDV感染的敏感。病毒滴度(根据Reed和Muench方法计算)在CESkC和ESCDL-1细胞间相似。

实施例4B.ESCDL-1UL37:反式互补系的重要病毒基因。
我们生产了数个棒状病毒,其编码包括突变的病毒,所述突变使编码包膜(tégument)蛋白的一个重要基因UL37无效。该突变涉及整个基因(删除)或者编码“伪亮氨酸拉链(pseudoleucinezipper)”单元的2个参与(participates)亮氨酸的两个密码子,其中一个单元在很多UL37直系同源中保留(在全部疱疹病毒中保留的基因)。筛选转染有pCDNA3-UL37(ESCDL-1_37)或者pCDNA3-UL37-mut(ESCDL-1_37mut)的ESCDL–1系,并且试验它们对来自转染的棒状病毒的突变和亲本病毒的复制能力。我们仅能够在ESCDL-1_37细胞上观察到两种突变病毒(删除或一次性突变)空斑的形成以及子代病毒的产生。ESCDL-1和表达突变蛋白的ESCDL-1_37mut都不能够补充突变病毒。
在目前的科学情况下,对于这类研究ESCDL-1系似乎是仅“能使用的”系,早期所述的QM7细胞(Schumacher等人,2002)不具有对全部的马立克氏病毒的相同的受纳性。
实施例4C.ESCDL-1Lc3-GFP和LBR-GFP:表达融合至EGFP(增强型绿色荧光蛋白)的细胞蛋白的细胞克隆
生成不同的ESCDL-1细胞克隆以表达Lc3-EGFP蛋白(自噬标志物)和LBR-EGFP蛋白(核纤层蛋白B受体)。通过转基因的稳定表达,这些系提供了在病理过程中研究细胞反应(自噬和核纤维层的不稳定)的可能性,尤其是在ADN和ARN病毒的感染中(例如,禽流感病毒能够激活自噬程序)。这些稳定的系的主要益处是实时跟踪在细胞感染中Lc3和LBR蛋白的再分布(relocalisation)。
还获得了表达重组酶、翻转酶(Flippase)的ESCDL–1系;建立了转基因的表达以及表征了翻转酶的活性。
由于表达标记细胞蛋白的ESCDL-1系的建立既简单又快捷(约1个月),可预期将该系统延伸至更广泛的蛋白。

Claims (14)
1.一种用于获得允许禽病毒体外复制的非转化的禽细胞系的方法,其至少包括以下步骤:
a)存在基质时,培养禽胚胎干细胞至少三天;
b)在低血清浓度的介质中培养至少两天;
c)在含1至10mM的六亚甲基二乙酰胺(HMBA)的低血清浓度的介质中培养至少2天;
d)在低血清浓度的介质中培养至少10天;
e)培养或冷冻允许禽病毒的复制的禽细胞系。
2.根据权利要求1所述的方法,其特征在于,所述的禽病毒是马立克氏病毒或双RNA病毒。
3.根据权利要求1或2中的任一项所述的方法,其特征在于,所述的禽胚胎干细胞来自鸡。
4.根据权利要求1至3中的任一项所述的方法,其特征在于,所述的禽胚胎干细胞分离自在X期至XIV期的胚盘。
5.根据权利要求1至4中的任一项所述的方法,其特征在于,在步骤b)、c)和d)后,所述的方法进一步包括用于在合适的生长介质中培养细胞的步骤。
6.根据权利要求1至5中的任一项所述的方法,其特征在于,所获得的禽细胞系在合适的培养介质中至少60次传代中具有稳定的表型。
7.根据权利要求1至6中的任一项所述的方法,其特征在于,所获得的禽细胞系经基因修饰以表达感兴趣的蛋白。
8.一种细胞系,其可通过权利要求1至7中的任一项所述的方法获得。
9.根据权利要求8所述的细胞系,其表达以下标志物:CD44+、整合素β1、胶原蛋白1和OLFM3。
10.根据权利要求8或9中的任一项所述的细胞系,所述的系不表达以下标志物:cPOUV/OCT4、nanog、SSEA1、EMA1、IXF1、KRT8和KRT19。
11.权利要求8至10中的任一项所述的细胞系作为基底用于体外生产禽病毒的用途。
12.权利要求8至10中的任一项所述的细胞系用于禽病毒的滴定的用途。
13.一种用于体外制备疫苗制剂的方法,其包括培养权利要求8至10中的任一项所述的细胞系,以及用至少一种禽病毒感染所述的细胞系。
14.一种疫苗制剂,其包含权利要求8至10中的任一项所述的细胞系,所述的细胞系感染有至少一种禽病毒。
