Abstract
本发明涉及一种表皮生长因子在制备治疗慢性萎缩性胃炎药物中的应用,它是从动物颌下腺中提取的,据对这种表皮生长因子ECG的活性试验、毒性试验和临床观察表明,ECG对胃粘膜上皮细胞的生长有促进作用,对慢性萎缩性胃炎的总有效率达94.54%。对肠化和异形增生的逆转也有较好疗效,总有效率分别达90.57%和93.75%,且无副作用,因此,ECG是治疗慢性萎缩性胃炎有特效的新药物。
Description
本发明涉及一种表皮生长因子在制备治疗慢性萎缩性胃炎药物中的应用。
萎缩性胃炎(CAG)是世界上公认的胃癌前期病变,癌变率在5%左右,有关治疗CAG的药物,国外尚无有效的理想的专题报导,国内也仅有用中草药治疗CAG的报告,但仍缺乏前后病理对照,且治疗的总有效率不高,以疗效较好的胃疡安治疗CAG为例,其总有效率仅为56.52%。
本发明的目的是提供一种表皮生长因子在制备治疗慢性萎缩性胃炎药物中的应用。
这种表皮生长因子是从动物颌下腺中提取的。
提取方法:
解剖取出动物颌下腺,去除周围组织,绞碎颌下腺,加水并加盐酸,使其呈微酸性,PH在3-6,于-15℃下冰冻保存,然后快速解冰,80℃加温30-120分钟,再经3000转/分,45分钟的离心,将上清液除菌过滤,得液态ECG,将滤液冰冻干燥,得粉状ECG,反复上述过程数次,将提高ECG纯度。
活性试验:
测定ECG对体外胃粘膜上皮细胞生长的影响,取人胃粘膜上皮细胞在含ECG和不含ECG的RPNI 1640培养液中培养,用I 125UdR掺入法测定细胞DNA合成,结果示含ECG组为21980 9997.8Cpm,不含ECG组为4168 171.5Cpm,表明细胞生长数目前者明显多于后者,ECG对胃粘膜上皮细胞的生长有促进作用。
毒性试验:
一、急性毒性试验:口服:在进行LD50时,给予一次量为治疗量10000倍,十五次时未有动物死亡。静注;静注能受量350μg/kg以上,未有动物死亡。
二、长期毒性试验:用ECG对狗进行半年观察,体重、红细胞、白细胞、血小板、白细胞分类、血清转氨酶、碱性磷酸酶、胆固醇、甘油三脂、尿素氮、肌酐、血清总蛋白及球蛋白等,与对照组比较均无显著差异。
临床观察:
温州医学院采用提取的ECG口服液对55例慢性萎缩性胃炎病员进行治疗,临床观察表明治疗后病员症状改善,食欲增进,体重增加;用药前后胃镜及病理检查资料证实,ECG对慢性萎缩性胃炎的总有效率分别达90.57%和93.75%,且用药半年以上无任何副作用。
因此,基础研究和临床证明,ECG是治疗慢性萎缩性胃炎有特效且无副作用的药物。
