技术领域
本发明涉及监测液体(fluid)中的单糖(simple sugar)(或单糖(monosaccharide))含量。更具体地,本发明与偏振器结合地使用光能源以确定被试液体(subject fluid)(诸如,血液)相对于基线浓度的糖水平(例如,葡萄糖)的改变。
背景技术
单糖根据等式Θ=α×L×C改变通过其的光能的偏振,其中L是通过其中糖聚集的液体的能量的行进长度,C是糖浓度,并且α是依赖于糖的类型、能量的波长和液体的常量。如果已知L和α,则通过测量相对于基线测量的通过含糖液体的能量的偏振的改变,可以推导出液体的糖浓度。
例如,可以使用这个原理非入侵地确定人类血液的葡萄糖浓度。正常血液具有非零葡萄糖浓度C,这引起通过血液的能量的偏振的改变。对于70mg/dL的葡萄糖浓度和α=45.62(x10-6)度/mm/(mg/dL),633nm的波长的能量和3.0mm的路径长度将具有0.00958度的旋转Θ。测量由糖引起的旋转的改变允许推导出当前糖浓度。
发明内容
本发明可以用以监测液体中的糖(例如,葡萄糖),并且相对于依赖标准偏振分析仪(该分析仪需要有源移动部分和到0.01度的角分辨率精度)的传统技术提供很多优点。第一,本发明是非侵入的,这降低感染的风险。第二,本发明可以提供流实时、连续数据的能力。第三,本发明提供低操作成本。
本发明包括:光能源,具有发射器,该发射器具有发射图案;第一偏振器,横切发射图案;第二偏振器,与第一偏振器间隔距离并且横切发射图案,第二偏振器相对于第一偏振器旋转第一旋转量Θ;第一光检测器,横切发射图案;第二光检测器,接近第二偏振器定位,第一偏振器和第二偏振器定位在光能源和第二光检测器之间,第二光检测器横切发射图案;补偿电路,耦合至第二光检测器;以及减法器电路,耦合至补偿电路和第一光检测器。

具体实施方式
图1示出本发明的一个实施例20,其包括:光能源22,第一偏振器24,与第一偏振器24间隔一距离的、具有相对于第一偏振器24的旋转Θ的第二偏振器26,第一光能检测器28,与第一检测器28并列的第二光能检测器30,以及电路46。第一和第二光检测器28、30中的每个被定向为接收通过空间32的光能。在优选实施例中,检测器28、30是硅检测器。如本文使用的,“并列”意味着相互邻近定位,以便在所有其他条件都相同的情况下,来自公共源的光将以近似相等的强度进入每个检测器。此外,虽然实施例公开了使用硅检测器,但是可以使用其他类型的检测器(诸如,光敏电阻器)。
当被激励时,能量源22产生具有发射图案36的初始光能34。能量源22优选是诸如红色发光二极管(LED)或者激光器的红色光源,但替代地可以是近红外的。最终,初始光能34必须有可以受到被试液体中存在糖影响。同时也通过包含该液体的其他容器的波长。
第一偏振器24接近源22定位,以便初始光能34通过第一偏振器24并且变成偏振能量38。偏振能量38横穿第一和第二偏振器24、26之间的空间 32,其中偏振能量38的第一部分40由第一光检测器28检测,并且偏振能量38的第二部分42通过第二偏振器26到第二光能检测器30。值得注意地,第一检测器和第二检测器28、30并列,尽管第二偏振器26接近第二检测器30。因为在图1中空间32是空的,所以通过空间32的偏振能量38不由于例如液体中糖的存在而旋转。
优选地,第一和第二偏振器24、26是线性偏振膜,因为这样的膜相比于其他可用替代物便宜。然而,这样的膜对于可见光谱中的能量波长是最优的。可以使用其他偏振器,只要选取能量源22的选择波长以最优地对应。例如,替代偏振器可以是线栅(wire-grid)或者全息的,其最优地配置用于具有近红外或者红外波长的能量的本发明中。
优选地,偏振器24、26之间的旋转的差是45度(或者四十五度的整数倍)加上由基线引起的旋转。在该最优情况下,相对于基线的浓度的改变至少初始地沿着正弦波的最线性部分移动,这使得与移动进一步远离波的斜率是1的地方并且进一步朝向斜率是0的地方(即,正弦波的波峰和波谷)相比,更容易地检测旋转的改变。例如,当以基线葡萄糖浓度100mg/dL在长度L上使用时,Θ等于0.014度。在该情况下,偏振器之间的旋转应该是45.014度。然而,从基线的浓度的改变越大,旋转与浓度的改变的相关性越非线性。
第一和第二检测器28、30电耦合到电路46。电路46具有补偿电路48、减法器电路50和增益电路52。第一检测器28直接耦合到减法器电路50。第二检测器30耦合到补偿电路48,其将由第二检测器30产生的信号的增益升高足够的量,以补偿可归因于偏振能量38的部分42通过偏振膜、以及由于液体中的基线浓度引起的偏振效应的强度的损失,但是,补偿电路48不补偿由于从某一基线本身的浓度改变引起的偏振的改变所产生的强度的损失。减法器电路50产生作为从第一和第二检测器28、30接收到的信号之间的差异的信号。增益电路52将信号放大至可使用的电平。
值得注意地,在替代实施例中,补偿电路48可以是耦合到第一检测器28的衰减器,均衡接收到的光能的强度,其目的在于由第一检测器28和第二检测器30看到的能量的差异与能量的旋转而不是其幅度有关。类似地,减法器电路50可以由惠斯通(Wheatstone)或者类似电桥取代。
参考图2,第一和第二检测器28、30的输出提供到电路46。电路46包括具有电位计Ro1的补偿电路48、减法器电路50、包括Ro1和C1以及Ro2 和C2的第一和第二30Hz低通滤波器和增益电路52。减法器电路50和增益电路52并入了OPA 211KP运算放大器IC 66。低通滤波器排除在检测器28、30处的任何噪声。偏振输出53和非偏振输出55馈送到包括Ro3、Ro4、Rl3和Rl4的减法器电路50。减法器电路输出54然后提供到包括Ro5和C3的增益电路52。最终信号提供在增益电路输出56。实施例包括用于相位匹配的目的可选单位增益电路57。
图3示出与人耳68一起使用的实施例20,人耳68的至少一部分占据空间32。空间32内的耳朵68的优选定向使得偏振能量38通常平行于横轴地通过耳朵68,其中L是沿着测量液体的轴的距离。对于大多数人耳,L接近富毛细血管和富血管的皮肤的三毫米。
当被激励时,能量源22产生具有发射图案36的初始光能34。初始能量34通过第一偏振器24,并且是耳朵68的非糖成分(即,皮肤、血液、组织、软骨)至少在某种程度上对于其透明的波长。
在通过第一偏振器24之后,初始能量34变成偏振能量38。然而,耳朵68中的血液内的葡萄糖将根据Θ=α×L×C使得能量38的偏振改变,使得退出耳朵的旋转能量70具有第一旋转Θ1。
旋转能量70的第一部分72的强度由第一检测器28检测。旋转能量70的第二部分74的强度通过第二偏振器26并且由第二检测器30检测。第一和第二检测器28、30的每个产生代表接收到的强度的信号。因为由第二检测器30接收到的旋转能量70的强度只是通过第二偏振器26的旋转能量分量的强度,所以通过测量检测器28、30处的强度的差异,可以推导出由耳朵68中的葡萄糖引起的旋转,从该旋转可以确定相对于基线的葡萄糖浓度的改变。
为了确定基线,在使用之前,通过改变电位计60以补偿由第一和第二检测器28、30接收到的能量的强度的差异,将实施例20校准为70mg/dL的基线葡萄糖浓度(对于人类血液的“正常”浓度)。因此,测量到的旋转的任何改变代表从某一基线(例如,70mg/dL)的葡萄糖浓度的改变。
本发明的替代实施例使用650nm的波长校准为100mg/dL的基线葡萄糖浓度,导致第二偏振器相对于第一偏振器的45.028度的旋转。这导致对于30mg/dL和300mg/dL之间的葡萄糖浓度,加上或者减去0.2度的产生的基线的旋转的范围。因此,30mg/dL的葡萄糖浓度将导致0.0096度的检测器之间的旋转差异,而300mg/dL的葡萄糖浓度将导致在30mg/dL浓度的方向的相反方向上的.0273度的旋转差异。
存在至少两个用于校准本发明的方法。首先并且优选地,在每个传感器的制造期间,将旋转偏振状态已知量的样本对照(sample control)血清或者类似成分将被插入空间。该对照将提供仿真的已知葡萄糖浓度,用于调整设备为适当校准设置。可替代地,用户/佩戴者可以利用传感器取得初始读数,并且另外地利用另一葡萄糖传感器(例如,血液棒计(blood stick meter))取得接近同时的读数。来自另一传感器的该值将利用用户输入部件(例如,连接到微控制器的旋钮、按钮等)被输入到传感器。
图4A-4C示出来自用以检测75mg/dL、150mg/dL和300mg/DL的葡萄糖浓度的本发明的实施例的实际数据。每个示例的左侧示出从偏振检测器28和非偏振检测器30接收到的实际信号。每个示例的右侧示出减法器电路的输出。对于75mg/dL的基线校准该实施例。在图4A中,减法器电路平均化为零,以指示没有从基线的改变。在图4B中,减法器电路平均化为接近.00005伏。在图4C中,减法器电路的输出平均化为接近.0001伏或者中间示例的两倍,只要底部示例的浓度是图4B所示的浓度的两倍就预期这一点。

Claims (15)
1.一种用于测量相对于基线浓度的液体中的糖浓度的改变的装置,该装置包括:
光能源,所述源具有发射器,所述发射器具有发射图案;
第一光检测器,其与所述源间隔距离;
第二光检测器,其以向所述第一光检测器和所述第二光检测器提供相同强度的光源的光能的方式与所述第一光检测器并列,;
第一偏振器和第二偏振器,放置在所述光源和所述第一和第二检测器之间,其中,
所述第一偏振器,其横切发射图案并且放置在所述源附近;以及
所述第二偏振器,其相对于所述第一偏振器被旋转第一旋转量Θ,与所述第一偏振器间隔距离,并且接近所述第二光检测器,所述第二光检测器接收所述光能穿过其中;
其中所述第一偏振器和所述第二偏振器之间的距离足够使得液体的体积的光学定位能横切所述第一偏振器和所述第二偏振器之间的所述发射图案并且在所述第一偏振器和所述第一检测器之间;以及
至少一个电路,其耦合至所述第一光检测器和所述第二光检测器,所述至少一个电路包括:
