Abstract
静宁鸡是甘肃唯一特有的地方鸡种,其肉质鲜嫩,风味独特,具有良好的地方口碑。然而目前却处于濒临灭绝的状况,前景堪忧。一种检测静宁鸡肌肉特殊生长发育规律的方法,是以静宁鸡鸡胚为研究对象,选择特色肉质性状调控基因,如生长发育相关蛋白:CECR2和IGF-1;脂肪酸代谢相关蛋白:Ex-FABP和LPL;蛋白质降解相关蛋白:CAST和CAPN1。用免疫组织化学和Western?blotting等方法检测鸡胚发育HH19-31期后肢芽和体节中以上蛋白的表达分布及其表达水平的变化,并与白羽肉鸡、黄羽肉鸡、海兰褐蛋鸡进行比较。本方法探讨了生长发育调控功能基因与生长性能和肉品质间的关系,为转基因鸡构建目的基因提供了依据。此外,有利于大力开发和利用这种资源优势,为当地经济的发展注入新的活力。

技术领域
在研究鸡肌肉品质领域,本发明涉及到了特色肉质性状调控基因领域,尤其涉及一种检测静宁鸡肌肉特殊生长发育规律的方法。
背景技术
随着现代化养鸡业的发展,人们的生活水平不断提高,对鸡肌肉品质研究已成为科技工作者共同关注的课题。然而,现有的研究大都只将肌纤维类型、密度和直径作为重要因素,或者对肌纤维在不同生长发育阶段生长变化规律进行研究,且发育时期的研究主要从刚孵化出的雏鸡开始。科学技术,特别是涉及生命科学领域的实验技术日新月异,现有的研究方法只是从宏观的角度牵强的说明肌肉品质的良莠,远远不能作为肌肉口感优劣的佐证,而且从雏鸡开始研究肌肉发育规律,本身是不系统也是不科学的。本方法是针对目前研究鸡肌肉品质这个特殊背景,提供了包括以甘肃省唯一特有的地方鸡种—静宁鸡作为研究对象,从鸡胚发育HH19期开始,筛选并检测了静宁鸡特色肉质性状调控基因表达水平的变化,是一种较科学的检测肌肉特殊生长发育规律的方法。
一种检测静宁鸡肌肉特殊生长发育规律的方法,是针对目前的研究鸡肌肉品质现状,立足于人们对生活品质的不断追求,基于现有的技术、设备和才智资源,利用新型实验手段进行开拓创新,构建一个操作性强,科学性高,应用性广的检测鸡肌肉品种优劣的系统实验测定方法,该测定方法可被借鉴于其他商业鸡种(特指肉鸡)肌肉优劣的测定及科学评价,从而有效地节约时间和经费,并促进商品鸡产业的发展乃至成为当地经济发展的新引擎。
发明内容
本发明的内容基于现有技术、设备和人力资源,利用新型实验技术进行开拓创新,为实现上述发明目的,本发明采用如下的技术方案:
1 本发明首次以甘肃省唯一特有的地方鸡种-静宁鸡作为研究对象,检测肌肉特殊发育规律,探究其肉质鲜嫩、口感上乘和与其他鸡种肌肉的不同之处。
2 本发明从影响肌肉发育的众多基因中筛选出静宁鸡特色肉质性状调控基因—CECR2、IGF-1、Ex-FABP、LPL、CAST、CAPN1,并检测了其在肌肉发育中的表达水平。
3 本发明从鸡胚发育HH19-31期开始,用免疫组织化学和Western blotting方法系统性的检测了鸡胚HH19、HH21、HH24、HH27和HH31以上特色肉质性状调控基因的时空性表达,并与白羽肉鸡、黄羽肉鸡、海兰褐蛋鸡鸡胚中的表达进行比较。
具体实施方式
下面结合实验测定方法对本发明的具体实施方式作进一步说明:
1 本发明首先是深入甘肃省静宁县各个乡,在当地畜牧局工作人员和村民的热心帮助下,按照静宁鸡特征及mtDNA测序,获取静宁鸡种鸡品系,并与农户建立即时联系方式,以便于获得种蛋。
2 其次,种蛋(静止24h)孵化(温度37±1℃;湿度79±2%)。在鸡胚HH19、HH24、HH27、HH31各期及孵化13天(E13)、孵化18天(E18)进行取材。
3 再次,筛选特色肉质性状调控基因,如生长发育相关蛋白:猫眼综合症候选基因2(CECR2)、胰岛素样生长因子1(IGF-1);脂肪酸代谢相关蛋白:细胞外脂肪酸结合蛋白(Ex-FABP)、脂蛋白脂酶(LPL);蛋白质降解相关蛋白:钙蛋白酶抑制蛋白(CAST)、钙蛋白酶1(CAPN1)。
4 最后用免疫组织化学方法观察鸡胚发育HH19-31期后肢芽和体节中以上蛋白的表达分布,Western blotting方法检测静宁鸡鸡胚在HH19、HH24、HH27、HH31各期及孵化13天(E13)、孵化18天(E18)后肢肌肉中以上蛋白的表达水平变化,并与白羽肉鸡、黄羽肉鸡、海兰褐蛋鸡鸡胚中的表达进行比较。

1.一种检测静宁鸡肌肉特殊生长发育规律的方法,其检测方法:选择特色肉质性状调控基因,如生长发育相关蛋白:CECR2、IGF-1;脂肪酸代谢相关蛋白:Ex-FABP、LPL;蛋白质降解相关蛋白:CAST、CAPN1;用免疫组织化学和Western blotting等方法检测鸡胚发育HH19-31期后肢芽和体节中以上蛋白的表达分布及其表达水平的变化,并与白羽肉鸡、黄羽肉鸡、海兰褐蛋鸡进行比较,系统探究了静宁鸡肌肉特殊生长发育规律。
2.根据权利要求1所述的一种检测静宁鸡肌肉特殊生长发育规律的方法,其特征在于:首次以甘肃省唯一特有的地方鸡种---静宁鸡作为研究对象,检测肌肉特殊发育规律,探究其肉质鲜嫩、口感上乘和与其他鸡种肌肉的不同之处。
3.根据权利要求1所述的一种检测静宁鸡肌肉特殊生长发育规律的方法,其特征在于:现有的方法只限于对肌纤维类型、密度和直径的检测从而对肌肉品质进行表观评价;相反,本方法从影响肌肉发育的众多基因中筛选出静宁鸡特色肉质性状调控基因—CECR2、IGF-1、Ex-FABP、LPL、CAST、CAPN1,并检测了其在肌肉发育中的表达水平。
4.根据权利要求1所述的一种检测静宁鸡肌肉特殊生长发育规律的方法,其特征在于:本方法从鸡胚发育HH19-31期开始,系统性的检测了鸡胚HH19、HH21、HH24、HH27和HH31各期特色肉质性状调控基因的时空性表达。
