本发明涉及用于对唾液样本实施化验的装置和方法,尤其涉及侧向横流装置和方法。
发明背景
侧向横流装置被用于多种化验并且是用来测试样本中目标的存在(或不存在)的简单装置。侧向横流装置可被用于各种各样的应用中,包括妊娠测试、艾滋病病毒测试和疟疾测试。该测试能被应用于,例如,尿、唾液、血液或粪便样本。
EP1277053公开了一种用于口腔流体包括唾液的诊断测试的方法和装置。这个专利涉及唾液测试时遇到的唾液样本不沿侧向横流测试条流动的问题。EP1277053的发明人认为这个问题起因于样本中粘蛋白和其它粘性的蛋白质材料的浓度相对高。该发明人提出在装置中包括垫,该垫上布置有盐或其它公知的已知可分解和降低含粘蛋白流体的粘性的粘蛋白分解物质。所述垫由玻璃纤维基质构成,玻璃纤维基质提供用于从试样过滤被分离的蛋白质材料和其它微粒材料的手段。该装置据称允许方便并快速地测试相对小的唾液样本而无需为了使其沿测试条迁移而预处理唾液。
在EP1277053中所描述的纤维垫包括具有约0.2毫米厚度的市售玻璃纤维材料的冲切部。所述垫位于留置有检测试剂的连结垫上。唾液试样位于分解粘蛋白物质的玻璃纤维盐垫上,粘蛋白物质随后被盐垫中的玻璃纤维和底层玻璃纤维样本垫中的玻璃纤维过滤。然后测试试样穿过连结垫,在此处,如果化验物存在于试样中则发生化学反应。然后测试试样进入测试结果可以被读取的捕获区。
本发明的发明人已发现,对于某些唾液化验,包括胃蛋白酶分析,唾液可沿侧向横流测试条充分流动以能够获得测试结果。然而在或许约10%的测试中,唾液没有充分流动。其原因不明。然而发明人已发现,使用与EP1277053中建议的相比相对厚的过滤件可使唾液充分流动以获得有意义的测试结果。因此,没有必要让唾液穿过承载有能够分离蛋白质材料的材料如盐的垫。
发明内容
根据本发明提供一种用于测试口腔流体如唾液的装置,所述装置包括具有用于接收待测样本的样本部以及用于显现测试结果的测试部的吸收性侧向横流测试条,所述样本部包括过滤垫,在使用中,唾液在到达所述测试部之前穿过所述过滤垫,所述过滤垫具有至少0.5毫米的厚度。
本发明还提供一种用于实施口腔流体试样诊断测试的方法,所述方法包括:提供如前述任一权利要求所述的装置,在所述样本部上留置口腔流体试样,显现在所述测试部上提供的测试结果。
优选地,所述的装置包括容纳诊断条的壳体,所述诊断条提供所述样本部和测试部。
优选地,所述样本部位于所述诊断条的一端。
优选地,吸收部位于所述诊断条的另一端。
优选地,所述过滤垫为无粘合剂型纤维垫。更优选地,所述纤维垫为玻璃纤维垫。最优选地,为微玻璃垫。
优选地,所述纤维垫由硼硅酸盐微玻璃构成。
优选地,所述样本部包括粘附有所述纤维垫的支承基板。更优选地,所述支承基板为粘性聚苯乙烯。
优选地,所述纤维垫的厚度为至少0.7毫米,更优选地,在0.75毫米和1毫米之间。

具体实施方式
参照附图中的图1,侧向横流装置1包括容纳于塑料壳体3中的侧向横流化验机构,塑料壳体3包括用于接收唾液试样的样本井5。容纳在所述壳体内的诊断条7包括样本接收部、用于显现测试结果的测试部以及在远离所述样本接收部的端部处的吸收部。
在下LFD的样本端部,无粘合剂的硼硅酸盐微玻璃纤维垫直接位于样本井5的下方并粘附至粘性聚苯乙烯支承基板。微玻璃纤维垫具有120.4克每平方米(gm2)的基重、0.85毫米的厚度以及75.2升/平方米/秒的弗雷泽渗透率。合适的纤维垫的一个例子是由Ahlstrom制造的142级垫。
纤维垫便于样本缓慢地过滤、输送和迁移至与其连通的相邻玻璃纤维连结垫。所得到的“经处理的”唾液使固定不动的聚苯乙烯标记再水化,随后迁移至硝酸纤维膜测试区。
随着唾液通过毛细管作用流动穿过纤维垫,任何微粒均因尺寸选择而被滤除。其它唾液成分的输送被调节,经处理的样本缓慢并持续地释放至连结垫。一旦输送至测试区并最终至吸收区,唾液通过持续的毛细管作用被抽出。
据信,所述垫可充分地使样本改性,以使那些“有问题的”个体被调节成具有正常的表现并以与“没有问题的”试样相同的方式流过膜的间隙空间,获得相同的化验性能。
图1示出了“有问题的”唾液试样的化验结果。可以看到,在另一个LFD中,窗口7没有出现对照线。在示出相同的但移除了壳体的多个LFD的图2中可以看到,在上LFD中,80微升的唾液样本已凝集且不流动。

Claims (14)

1.一种用于测试口腔流体如唾液的装置,所述装置包括具有用于接收待测样本的样本部以及用于显现测试结果的测试部的吸收性侧向测试条,所述样本部包括过滤垫,在使用中唾液在到达所述测试部之前穿过所述过滤垫,所述过滤垫具有至少0.5毫米的厚度。
2.根据权利要求1所述的装置,包括容纳诊断条的壳体,所述诊断条提供所述样本部和测试部。
3.根据权利要求2所述的装置,其中,所述样本部位于所述诊断条的一端。
4.根据权利要求2或3所述的装置,其中,吸收部被设置在所述诊断条的另一端。
5.根据前述任一权利要求所述的装置,其中,所述过滤垫为无粘合剂型纤维垫。
6.根据权利要求5所述的装置,其中,无粘合剂型纤维垫由微玻璃垫构成。
7.根据权利要求6所述的装置,其中,所述无粘合剂型纤维垫为硼硅酸盐微玻璃。
8.根据前述任一权利要求所述的装置,其中,所述样本部包括粘附有所述纤维垫的支承基板。
9.根据权利要求8所述的装置,其中,所述支承基板为粘性聚苯乙烯。
10.根据前述任一权利要求所述的装置,其中,所述纤维垫的厚度为至少0.7毫米。
11.根据权利要求10所述的装置,其中,所述纤维垫具有在0.75毫米和1毫米之间的厚度。
12.根据权利要求1并大致如本文所述的装置。
13.一种用于测试口腔流体如唾液的装置,所述装置大致如本文参照附图所述。
