本发明的目的在于提供一种全新的化合物1-(4-氟苯基)吡咯及制备方法,本发明首次合成并纯化该化合物,使其纯度达到一定要求,能够对其进行进一步的实验或开发使用。

该化合物的制备方法如下:
将4-氟苯胺溶解在醋酸中,加入2,5-二甲氧基四氢呋喃回流3h,当溶液变黑,生成黑色固体后,用TLC检测至4-氟苯胺反应完毕。停止加热,旋出醋酸,加DCM,有机层用饱和NaHCO3洗涤三次,有机层柱层析分离,得到黄白色固体,用PE重结晶,用油泵拉干PE得到白色结晶性固体1-(4-氟苯基)吡咯。
所述4-氟苯胺与2,5-二甲氧基四氢呋喃的摩尔比为0.9:1.08;所述4-氟苯胺溶于醋酸的浓度为:每0.90mol4-氟苯胺采用500ml醋酸溶解。
本发明的优点和积极效果是:
1、本发明使用4-氟苯胺、2,5-二甲氧基四氢呋喃、醋酸等原料首次合成了具有复杂结构的1-(4-氟苯基)吡咯,制备终产物的产率高,且结晶效果好,纯度高,为后续的化学和生物实验提供高纯度的标样或样品。

本发明的反应路线如下:
原料
4-氟苯胺、2,5-二甲氧基四氢呋喃、醋酸等原料均为市场购买,无特殊要求。
该化合物的合成方法如下:
将4-氟苯胺(100g,0.9mol)溶在500ml醋酸中,然后加入2,5-二甲氧基四氢呋喃(142.72g,1.08mol)回流3h,当溶液变黑,生成黑色固体后,TLC(PE:EA=5:1)检测4-氟苯胺反应完毕。停止加热,旋出醋酸,加DCM300ml,有机层用饱和NaHCO3洗涤三次,有机层柱层析分离(展开剂用纯PE),得到104g黄白色固体,用150mlPE重结晶,固体用油泵拉干PE得94.87g白色结晶性固体1-(4-氟苯基)吡咯。

所述4-氟苯胺与2,5-二甲氧基四氢呋喃的摩尔比为0.9:1.08;所述4-氟苯胺溶于醋酸的浓度为:每0.9mol4-氟苯胺采用500ml醋酸溶解。
