背景技术
酒精性肝损伤是由于长期大量饮酒，酒精与肝脏细胞频繁接触而导致的肝脏损伤，一种或几种受损伤的肝脏细胞产生自由基等系列介质，使肝脏细胞脂质过氧化，造成肝脏细胞坏死、凋亡、炎症、纤维化等病理变化，最终导致肝纤维化、肝硬化乃至肝癌等各种肝脏疾病。据世界卫生组织统计结果，我国长期饮酒者约2亿，一般饮酒者超过5亿，且随着人们生活水平的提高及经济活动的活跃，饮酒者数量迅速增加，酒精性肝损伤发生率逐年增长。大量或长期饮酒者80％以上有不同程度的脂肪肝，其中10％-20％将发展为肝硬化，每年有近11万人因酒精中毒而死亡。可见，酒精性肝损伤已对国民健康造成了极大的危害。目前对肝损伤的治疗主要依赖于皮质激素、S-腺普甲硫氨酸等药物，但它们的疗效有限，更严重的是这些药物的长期服用会产生新的肝损伤。因此，探索新的预防、治疗酒精性肝损伤的保健品、药品已成为食品、医药领域的研究热点之一。
正常情况下肝脏所含脂肪约占肝脏湿重的2%～4%,当肝脏内脂肪积聚超过5%时,或组织学上每单位面积见1/3以上肝细胞脂变时,即形成脂肪肝(Fatty liver,FL)。脂肪肝并非临床上的一个独立性疾病,而是多种疾病和原因引起的肝内脂质(主要为甘油三酯)蓄积过多的一种病理状态,也是多种肝脏疾病发展中的常见病理过程。随着我国人民生活饮食结构的变化以及临床诊断技术的不断提高,各种诱因所致的脂肪肝在我国的诊断率逐年增加。目前,根据饮酒与否,将脂肪肝分为酒精性脂肪肝和非酒精性脂肪肝,其中后者进一步肝纤维化的发生率约高达25%,且约1.5%～8.0%患者可进展为肝硬化,故积极防治脂肪肝对阻止慢性肝病进展和改善愈后十分重要。目前,脂肪肝的发病机制尚未明确,西医缺乏理想的针对性治疗药物。相比而言,中医药治疗脂肪肝具有疗效稳定、持久、无副作用、价廉等优势,因此深入进行中医药治疗脂肪肝的临床及实验研究是有其现实意义的。

本发明上述目的是通过以下技术方案予以实现的：
本发明提供一种预防治疗酒精肝和脂肪肝的中草药保健口服液，其特征在于： 1000份口服液中中药原料及质量配比为：丹参5-10份、石斛10-15份、三七8-12份、当归10-15份、浒苔15-20份、栀子10-13份、水飞蓟8-12份、益母草10-15份、槲皮10-16份、白茅根10-18份、山楂15-20份、郁金10-15份、车前子12-18份、鸡内金10-15份、川芎8-12份、菊花5-10份、鸡血藤5-10份、生鳖甲10-15份、半夏10-15份、白芥子10-15份、余甘子8-12份、巴戟天6-10份、甘草10-15份、绿茶15-20份。
本发明的优选技术方案是：一种预防治疗酒精肝和脂肪肝的中草药保健口服液，其特征在于：其中药原料及质量配比为：丹参8份、石斛13份、三七11.5份、当归12份、浒苔16份、栀子12份、水飞蓟10份、益母草14份、槲皮15份、白茅根15份、山楂18份、郁金12份、车前子15份、鸡内金12份、川芎10份、菊花7份、鸡血藤8份、生鳖甲13份、半夏12份、白芥子13份、余甘子11份、巴戟天8份、甘草13份、绿茶18份。
本发明提供一种预防治疗酒精肝和脂肪肝的中草药保健口服液，其特征在于通过以下步骤制备：
（1）分拣称量：将所用中草药分拣，去除杂质等，按照上述比例称重；
（2）超声波粉碎：将中草药超声波粉碎，过40目筛；
（3）醇提：将中草药混合后加入4-6倍量的50%乙醇，加热提取2次，提取时间1-1.5h，过滤；
（4）水煎：将醇提后的中草药加入12-15倍量的水，煎煮3次，每次1.5-2h，水煎液过滤合并；
（5）减压浓缩：将合并后的水溶液浓缩至1g/ml的清膏，放冷；
（6）醇沉：向清膏中加入95%的乙醇使含醇量达到60%-70%，4℃条件下放置48h以上
（7）合并抽滤：将醇沉后的液体与醇提液合并抽滤；
（8）减压回收：将抽滤液进行减压回收乙醇至无醇味；
（9）添加辅料：加入0.10-0.15%山梨酸钾、1-2%木糖醇、0.01-0.02%硒，加水搅拌定容；
（10）静置：将口服液半成品常温下静置48h以上；
（11）离心：将口服液半成品3000r/min离心5-10min；
（12）定容分装：将离心后的口服液定容、分装即得口服液成品。
本发明中药物组合是在传统中医理论和实践指导下精选药方。本发明中药组合物中：丹参具有活血调经、祛瘀止痛、凉血消痈、清心除烦、养血安神之功效；石斛具有护肝利胆、益胃生津、滋阴清热之功效；三七具有抗肝损伤之功效；当归具有补血活血、调经止痛、润肠通便等功效；浒苔多糖具有降血脂、保肝利胆等功效；栀子具有泻火除烦、清热利湿、凉血解毒等功效；水飞蓟具有清热解毒、保肝、利胆等功效；益母草具有活血、祛瘀、调经、消水等功效；白茅根具有凉血、止血、清热利尿等功效；山楂具有益消积花滞、收敛止痢、活血化淤等功效；郁金具有行气解郁、凉血破瘀等功效；车前子具有清热利尿、渗湿止泻、明目祛痰等功效；鸡内金具有消积滞、健脾胃等功效；川芎具有活血祛瘀、行气开郁、祛风止痛等功效；菊花具有散风热、平肝明目、消咳止痛的功能；鸡血藤具有补血、活血、通络等功效；生鳖甲具有滋阴清热、潜阳熄风、坚散结之功效；半夏具有化湿健脾、消食止泻等功效；白芥子具有温肺豁痰利气、散结通络止痛等功效；余甘子具有清热利咽、润肺止咳等功效；巴戟天具有补肾助阳、强筋壮骨、祛风除湿等功效；甘草具有清热解毒、祛痰止咳等功效；本发明精选中草药配方，其药物组分经辨证配伍，在作用上相互佐助，共行清热利湿、祛浊解毒、疏肝理气、活血化瘀、软坚化痰的功效，从而起到预防、治疗酒精肝和脂肪肝的效果。
    本发明提供一种预防治疗酒精肝和脂肪肝的中草药保健口服液，其优势在于：一是科学配伍多种中草药，共行清热利湿、祛浊解毒、疏肝理气、活血化瘀、软坚化痰功效，酒精肝和脂肪肝预防治疗效果好；二是在口服液的制备过程中采用“醇提+水煎”的工艺，充分糅合中草药中的预防治疗酒精肝和脂肪肝的功能活性成分，提高治疗效果；三是具有扶正培本、益气养血、补肝益肾、预防和治疗酒精肝和脂肪肝的功效，快速有效的降低转氨酶，而且可以益气养血、养心安神，增强机体免疫力。
具体实施方式
    下面结合实施例对本发明做进一步说明。
实施例1：
一种预防治疗酒精肝和脂肪肝的中草药保健口服液，通过以下步骤制备：
（1）分拣称量：将所用中草药分拣，去除杂质等，按照丹参10份、石斛10份、三七12份、当归10份、浒苔20份、栀子10份、水飞蓟12份、益母草10份、槲皮16份、白茅根10份、山楂20份、郁金10份、车前子18份、鸡内金10份、川芎12份、菊花5份、鸡血藤10份、生鳖甲10份、半夏15份、白芥子10份、余甘子12份、巴戟天6份、甘草15份、绿茶15份比例称重；
（2）超声波粉碎：将中草药超声波粉碎，过40目筛；

实施例2：
一种预防治疗酒精肝和脂肪肝的中草药保健口服液，通过以下步骤制备：

（11）离心：将口服液半成品3000r/min离心5-10min；
（12）定容分装：将离心后的口服液定容、分装即得口服液成品。
实施例3：
一种预防治疗酒精肝和脂肪肝的中草药保健口服液，通过以下步骤制备：
（1）分拣称量：将所用中草药分拣，去除杂质等，按照丹参8份、石斛13份、三七11.5份、当归12份、浒苔16份、栀子12份、水飞蓟10份、益母草14份、槲皮15份、白茅根15份、山楂18份、郁金12份、车前子15份、鸡内金12份、川芎10份、菊花7份、鸡血藤8份、生鳖甲13份、半夏12份、白芥子13份、余甘子11份、巴戟天8份、甘草13份、绿茶18份比例称重；
（2）超声波粉碎：将中草药超声波粉碎，过40目筛；
（3）醇提：将中草药混合后加入4-6倍量的50%乙醇，加热提取2次，提取时间1-1.5h，过滤；

（9）添加辅料：加入0.12%山梨酸钾、1.5%木糖醇、0.015%硒，加水搅拌定容；
（10）静置：将口服液半成品常温下静置48h以上；
（11）离心：将口服液半成品3000r/min离心5-10min；
（12）定容分装：将离心后的口服液定容、分装即得口服液成品。

2.根据权利要求1所述的一种预防治疗酒精肝和脂肪肝的中草药保健口服液，其特征在于：其中药原料及质量优化配比为：丹参8份、石斛13份、三七11.5份、当归12份、浒苔16份、栀子12份、水飞蓟10份、益母草14份、槲皮15份、白茅根15份、山楂18份、郁金12份、车前子15份、鸡内金12份、川芎10份、菊花7份、鸡血藤8份、生鳖甲13份、半夏12份、白芥子13份、余甘子11份、巴戟天8份、甘草13份、绿茶18份。
