本发明公开了一种山羊精液玻璃化冷冻和解冻配方与方法,其中山羊精液玻璃化冷冻配方包括:1)基础液:含20mg/ml牛血清白蛋白的MEM培养基;2)平衡液:基础液添加20%乙二醇;3)玻璃化冷冻液:于基础液中添加70%乙二醇、0.8mol/ml海藻糖、10%聚乙烯吡咯烷酮。解冻配方包括解冻液1(TS1):基础液添加0.4mol/L海藻糖解冻;解冻液2(TS2):基础液。本发明还选择了合适的冷冻及解冻方法,提高了解冻后精子活力,具有冷冻过程简单易于操作,冷冻效果好的特点。

一种山羊精液玻璃化冷冻和解冻配方与方法
技术领域
本发明动物生殖生理与繁殖技术领域,具体涉及一种山羊精液玻璃化冷冻和解冻配方与方法。
背景技术
羊冷冻精液的研究国外起始于20世纪30年代,我国起始于1978年,但到目前为止仍未形成成熟稳定的程序化冷冻技术。羊的精子和鸡、牛精子相比更加脆弱,在冷冻、解冻过程极易形成损伤,从而失去受精能力;虽然用于冷冻的精液要求冷冻前活率达到0.8以上,但因不同公羊个体差异所产生的精子活力不同,精子冷冻效果很不稳定。同时,现有羊冷冻精液解冻后活率低于0.35且存在很多“衰老”精子和弱死精子,有效精子少,活力低,质量差。羊冷冻精液在我国辽宁、贵州有少量生产,2008年山西省岢岚县引进部分羊冷冻精液,人工授精后妊娠率不足30%,无法进行批量生产。
授权公告号为CN102657149B,发明名称为“一种超高活力羊细管精液冷冻方法”的中国专利中公开了其制备方法:“吸取羊新鲜精液注入装有清洗液的第一离心管底部,在CO2培养箱中静置30-60min,高活力精子上游至上清液,吸取上清液,注入第二离心管中离心,离心管底部获得高浓度、高活力的精液;吸取该浓缩的高活力精液加入冷冻稀释液中,混合均匀后灌装细管,程序冷冻,液氮保存,输精前解冻”,羊细管冷冻精液冷冻前的活率为0.8以上,解冻后活率为0.45-0.60。但是这种方法冷冻过程繁琐,冷冻效率还有提升空间。
发明内容
针对这一问题,本发明提供一种山羊精液玻璃化冷冻方法,采用了玻璃化冷冻方法并优化了冷冻液的配方,通过于基础液中添加70%乙二醇、0.8mol/ml海藻糖、10%聚乙烯吡咯烷酮的玻璃化冷冻液进行保存,然后通过解冻液解冻,提高其解冻后的活率,具有冷冻过程简单易于操作,冷冻效果好的特点。
为解决上述技术问题,本发明采用的一个技术方案是:
山羊精液的冷冻保存,包括冷冻配方的选择,选择合适的冷冻方法,选择合适的解冻配方及方法。具体如下:
山羊精液玻璃化冷冻配方,包括:1)基础液:含20mg/ml牛血清白蛋白的MEM培养基;2)平衡液:基础液添加20%乙二醇;3)玻璃化冷冻液:于基础液中添加70%乙二醇、0.8mol/ml海藻糖、10%聚乙烯吡咯烷酮。
山羊精液玻璃化冷冻的方法,包括如下步骤:
(1)用上述的山羊精液玻璃化冷冻配方——基础液、平衡液、玻璃化冷冻液在常温下平衡;
(2)按照常规方法获取羊精液,液化后进行CASA分析;
(3)将平衡液与等量的羊精液混合,平衡5~10分钟;
(4)将等量的玻璃化冷冻液加入到混合液中;
(5)立即将混入了玻璃化冷冻液的精液装入500μl冷冻脉管中;
(6)冷冻脉管投入液氮罐中保存;
(7)记录本次冷冻结果,保存位置等信息。
山羊精液玻璃化解冻配方,其特征在于,包括解冻液1(TS1):基础液添加0.4mol/L海藻糖解冻;解冻液2(TS2):基础液。
山羊精液玻璃化解冻方法,包括如下步骤:
(1)将权利要求3中的解冻液1和解冻液2常温下平衡;
(2)冷冻脉管从液氮中取出,于37度水浴60sec;
(3)剪去脉管塞一端,从管中吹出冷冻精液到TS1中,300g混匀离心,去上清;
(4)沉淀用TS2稀释到需要浓度,显微镜分析,备用。
具体步骤见图1。
有益效果:
减少程序冷冻时间,使冷冻过程简单化,便于在畜牧生产中过程中操作。

具体实施方式
选择8月龄波杂山羊(公)作为试验对象,试验前对山羊精液进行常规分析,达到精液质量要求的进入实验,剔除精液质量较差的山羊。对其进行如下处理:
配备好山羊精液玻璃化冷冻配方,包括:1)基础液:含20mg/ml牛血清白蛋白的MEM培养基;2)平衡液:基础液添加20%乙二醇;3)玻璃化冷冻液:于基础液中添加70%乙二醇、0.8mol/ml海藻糖、10%聚乙烯吡咯烷酮。然后将上述的基础液、平衡液、玻璃化冷冻液在常温下平衡;按照常规方法获取羊精液,液化后进行CASA分析;将平衡液与等量的羊精液混合,平衡5分钟;将等量的玻璃化冷冻液加入到混合液中;立即将混入了玻璃化冷冻液的精液装入500μl冷冻脉管中;然后将冷冻脉管投入液氮罐中保存;记录本次冷冻结果,保存位置等信息。
在使用时对其进行解冻处理:
首先配制好山羊精液玻璃化解冻配方,包括解冻液1(TS1):基础液添加0.4mol/L海藻糖解冻;解冻液2(TS2):基础液,然后将解冻液1和解冻液2常温下平衡;再将冷冻脉管从液氮中取出,于37度水浴60sec;剪去脉管塞一端,从管中吹出冷冻精液到TS1中,300g混匀离心,去上清;沉淀用TS2稀释到需要浓度,冻后活力检查,精子图像计算机辅助分析仪进行活力的检查,重复检查两次结果取平均值。
本实验共采用10只山羊进行实验,玻璃化冷冻组(实验组)和常程序冷冻组(对照组)各5只,每只羊进行3次取精、冷冻,取平均值,其比较结果见表1。

试验组与对照组在冷冻前精子活率差异不显著(p<0.5),解冻后在精子活率、顶体完整率、情期受胎率三个衡量指标上,试验组与对照组差异不显著(p<0.5)。但试验组花时间少,使用方便。

1.一种山羊精液玻璃化冷冻液,其特征在于:包括:1)基础液:含20mg/ml牛血清白蛋白的MEM培养基;2)平衡液:基础液添加20%乙二醇;3)玻璃化冷冻液:于基础液中添加70%乙二醇、0.8mol/ml海藻糖、10%聚乙烯吡咯烷酮。
2.一种山羊精液玻璃化解冻液,其特征在于,包括解冻液1(TS1):基础液添加0.4mol/L海藻糖解冻;解冻液2(TS2):基础液。
3.一种山羊精液玻璃化解冻方法,其特征在于,包括如下步骤:
(1)将权利要求2中的解冻液1和解冻液2常温下平衡;
(2)冷冻脉管从液氮中取出,于37度水浴60sec;
(3)剪去脉管塞一端,从管中吹出冷冻精液到TS1中,300g混匀离心,去上清;
(4)沉淀用TS2稀释到需要浓度,显微镜分析,备用。
