背景技术
在自然的动物消化和微生物消化时以及在以蛋白酶或含蛋白酶的微生物对含蛋白质的原料进行工业处理时,在蛋白质酶促水解下产生肽混合物。原料中的蛋白质通常是由20种不同的α-L-氨基酸形成的、具有100到数千个单体单元的聚酰胺,蛋白质与蛋白质间的聚酰胺序列是强烈地变化的。相应地,在以蛋白水解的方式处理蛋白质混合物以及即使是以蛋白水解的方式处理纯粹单一的蛋白质时,都产生非常多样的聚合物碎片。
在酶促技术的水解过程中产生的肽混合物取决于所使用的原料、所使用的酶以及所设定的水解度地在其组成上有所区别。而在同样的水解条件下组成是可同样方式地重现的。
对于非常多种的由蛋白质水解产物形成的肽(部分甚至在原始水解产物中没有富集)已经证明了特殊的功能特性和生物活性:

通过含蛋白质原料的水解,可以借助单一的或者组合的蛋白酶几乎以无限的变化来制造肽混合物,其中一些在文献中被加以描述,一些被商业化地例如作为饲料或者食物提供并且一些拥有以下特性,即,因为这些特性其可以用在食品应用、化妆品应用或者制药应用中。富集功能性肽或者从不期望的副成分中的分离出功能性肽根据现有技术在个别情况下通过沉淀法或者超滤进行。两种方法是相对非特异性地并且每个方法步骤仅允许分离出两种分离组分。

这些固定相的典型代表是带有阴离子或阳离子官能化的二乙烯基苯交联的聚苯乙烯树脂、交联的乙烯醇-苯乙烯共聚物或交联的苯酚-甲醛缩聚物,例如陶氏化学公司(Dow Chemicals)的产品系列Amberlite或者朗盛公司(Lanxess)的产品系列Lewatit。

水解产物通常可以例如通过蒸发而进一步浓缩、调节到期望的pH值、在必要时通过例如过滤而澄清化并且以直至0.3床体积(BV)、优选0.1床体积加入到填充有固定相的层析柱上。这些料液()具有优选5%到50%干物质的浓度。
洗脱优选通过无其他添加物的纯水进行,但是在必要时大多还少量地添加有盐或溶剂。
根据所期望的组分分离,使用羟基官能化的固定相,借此实现了根据疏水性的分离;或者固定相是阴离子或阳离子官能化的,借此实现了根据离子排斥或离子交换的分离。
针对层析可以以多种方式使用分离树脂,特别地,针对层析作为固定相使用的分离树脂是大孔聚合物树脂或凝胶状聚合物树脂,其优选具有允许直至10000Da分子尺寸的局部渗透的交联度,并且具有在300Da和10000Da之间的凝胶过滤效果。
在一些情况下,针对层析还可以有利地使用未官能化的工业吸附剂作为固定相,例如由吸附技术所公知的粒状碳(Kornkohle)或者经热解的分离树脂,其由于热解而可以具有与粒状碳近似的吸收特性。
优选地,在多个层析循环进程中,来自混合物的、被吸附的肽的结合平衡适应于分离树脂,该结合平衡通过相对未结合的肽的不同的亲和性大幅影响分离进程。
其他的过程参数为在-4℃和98℃之间的洗脱温度下,尤其是在70℃和95℃之间的洗脱温度下的在每小时0.2和0.4床体积之间的层析流量。
在层析柱的洗脱液中,在建立新的分离时首先确定所寻求的功能特性。分离树脂和分离条件的选择以以下方式进行,即,具有不期望的特性的组成部分尽可能地不在所期望的功能性范围中洗脱。分离组分范围的确定根据洗脱液的可容易地测量的物理化学参数确定。
层析的结果是,根据可预定的功能特性富集肽和/或从不期望的副产物(例如有色物质、苦味物质或者气味物质)中纯化出肽。
如此获得的肽混合物可以通过进一步的、特别还是层析方式的提纯步骤进一步组分分离直至达到一种或多种肽任意高的富集程度,所述一种或多种肽可以在饲料、食品、化妆制品中作为功能性物质使用,并且可以作为制药学的有效成分使用。
无功能的分离组分被聚集、浓缩并且被供应给其在食物、饲料、发酵培养基或肥料领域中的原始利用。
具体实施方式
借助实施例进一步阐述本发明的实质,这些实施例不具有限定本发明的性质。
示例1
由商业购得的具有约90%的肽份额、具有在250和2500Da之间的分子量的酶解小麦蛋白质水解产物通过分析型分离柱借助于HPLC设备完成了组成图谱。小麦蛋白质水解产物调节为50%(w/w)(质量百分数)的水溶液以及4.5的pH值、经过过滤并且示例性地在以下所示的一些特性方面定性:
关于%干物质的导电性
关于%干物质的颜色成分(E430/cm)
1%溶液的发泡行为(在1分钟后物料充气后的泡沫体积,0.1样品体积/秒)

双罩壳玻璃柱(内直径:35mm、长度:1000mm)填充有DVB交联的、大孔的磺化聚苯乙烯树脂,其是钙型的、单体分散的以及0.4mm的颗粒尺寸的。双罩壳借助于循环温控器加热到85℃。小麦蛋白质水解产物以50%(w/w)的浓度和50ml的体积(相当于0.1床体积(BV))加入到柱中并且以中等硬度的热自来水和1BV/h的体积流量层析。在1.5BV的洗脱体积之后,进行0.1BV小麦蛋白质水解产物的重新加料并且以水重新洗脱。不对分离树脂再生地,该工序重复至少10次,其在原则上是可以任意多次的。每个层析循环的洗脱液以每(à)100ml15个分离组分收集。
示例性地,可以通过在线跟踪pH变化、颜色变化、导电性和在214nm的吸光度在洗脱进程中读取带有相应的特性的肽的富集。
各个分离组分的肽组成图谱通过以上所描述的HPLC方法记录。
各个分离组分的HPLC图谱是明显地彼此不同的并且各个肽明显地被富集到不同的分离组分中。
此外,对于定性小麦蛋白质水解产物所考虑的功能特性也在各个分离组分中加以研究并且与初始溶液相比较。
洗脱分离组分具有不同的E430/TS比例,颜色成分在一些分离组分中富集,在另外的分离组分中贫化。一些分离组分相对于料液是无色的,不再是苦味的并且在相同的浓度下表现出提高30倍的泡沫稳定性。已经由料液已知的且验证的头发修复特性同样可以在一些分离组分中重新发现,而在其他分离组分中则不能重新发现。
示例2
具有1500Da的平均肽尺寸的大豆蛋白水解产物如在示例1中所描述的那样定性并且准备用于层析。层析示例性地在两个不同的试验中在5.0的pH值的情况下和在10.0的pH值的情况下施行。
双罩壳玻璃柱(内直径:10mm、长度:300mm)填充有DVB交联的聚苯乙烯-乙烯醇共聚物,其是未经离子官能化的、单体分散的并且0.2mm的颗粒尺寸的。双罩壳借助于循环温控器冷却到10℃。大豆蛋白水解产物以35%(w/w)的浓度和3ml的体积(0.1BV)加入到柱中并且以蒸馏水和1BV/h的体积流量层析。在1.5BV的洗脱体积之后,进行0.1BV小麦蛋白质水解产物[w1]的重新加料并且以水重新洗脱。
不对分离树脂再生地,该工序重复至少3次。每个层析循环的洗脱液以每(à)3ml15个分离组分收集。由洗脱循环的分离组分,示例性地通过以上所描述的HPLC方法记录肽组成图谱。此外,用于定性大豆蛋白质水解产物所考虑的特性也在各个分离组分中加以研究并且与初始溶液相比较。
各个分离组分的HPLC图谱是明显地彼此不同的并且各个肽明显地被富集到不同的分离组分中。特别是在pH5.0下的试验中获得与在pH10.0下的试验中全然不同的分离组分。
洗脱分离组分区别在于其颜色成分、其泡沫形成行为和其感官特性。
示例3
由具有小于2000Da的肽尺寸的酶解胶原蛋白水解产物通过分析型分离柱借助于HPLC设备完成了组成图谱。胶原蛋白水解产物以40%(w/w)溶解于水中、pH值调节到8.0并且如在示例1中所描述的那样定性。
双罩壳玻璃柱(内直径:35mm、长度:1000mm)填充有DVB交联的、大孔的磺化聚苯乙烯树脂,其是钠型的、单体分散的以及0.4mm的颗粒尺寸的。双罩壳借助于循环温控器加热到85℃。酶胶原蛋白水解产物以40%(w/w)的浓度和50ml的体积(0.1BV)加入到柱中并且以中等硬度的热去离子水和1BV/h的体积流量层析。在1.5BV的洗脱体积之后,进行0.1BV胶原蛋白水解产物的重新加料并且以水重新洗脱。不对分离树脂再生地,该工序重复至少10次。每个循环的洗脱液以每(à)100ml15个分离组分收集。

根据本发明的方法在每种实施方式中都提供肽分离组分,这些肽分离组分在其形式上是新颖的、具有有利的功能特征并且用于比所使用的原料明显更高的应用价值。以上所描述的分离方法也可以以其能想到的组合或应用而有能力,将蛋白水解物的巨大功能性潜力用于广泛的用途。
通过过程参数变化和在不同条件下的多个层析的组合,总是可以生产新的肽混合物乃至纯化肽。根据本发明的方法以及由其产生的产品展示了出自食品和农产品的副产物组分的显著附加值,否则这些副产品组分就仅是劣质的或者是根本无法利用的。对于特定应用拥有不够充足的功能特性的所有子分离组分,至少可回到原料的原始利用中,通常这些子分离组分还可以通过酶促分解而得以提升价值,而在任何情况下它们都不是无价值的。
层析的洗脱在所有情况下通过不带有其他附加物的水实现,而不需要通过化学试剂再生层析柱。对于层析所使用的、通过例如其在水处理中的使用而公知的分离树脂,以及所使用的出自饲料和食品应用的酶都是可商业购得的。
Claims (19)
1.一种用于由含蛋白质的原料制造经富集的肽分离组分的方法,其特征在于,蛋白质水解产物根据所述蛋白质水解产物的物理化学性质通过层析经由固定相以水溶液作为洗脱剂分离。
2.根据权利要求1所述的方法,其特征在于,在所述层析之前,所述原料以通过特异性的蛋白酶水解的方式被调节到1到100个氨基酸的肽尺寸上。
3.根据前述权利要求中一项或多项所述的方法,其特征在于,在层析之前对所述蛋白质水解产物进行pH值调节、浓度调节和/或澄清化。
4.根据前述权利要求中一项或多项所述的方法,其特征在于,对于所述层析,使用未经官能化的或者经过中性亲水性、酸性或碱性官能化的离子交换剂或者工业吸附剂作为固定相。
5.根据前述权利要求中一项或多项所述的方法,其特征在于,对于所述层析,使用分离树脂作为吸附剂作为固定相。
6.根据权利要求5所述的方法,其特征在于,所述分离树脂是大孔聚合物树脂或凝胶状聚合物树脂。
7.根据前述权利要求中一项或多项所述的方法,其特征在于,对于所述层析,使用吸附剂:粒状碳或者经热解的分离树脂作为固定相。
8.根据前述权利要求中一项或多项所述的方法,其特征在于,料液具有5%到50%干物质的浓度。
9.根据前述权利要求中一项或多项所述的方法,其特征在于,所述固定相具有在300Da和10000Da之间的分子筛效果的交联度。
10.根据前述权利要求中一项或多项所述的方法,其特征在于,所述层析的流量在每小时0.2到4床体积之间。
11.根据前述权利要求中一项或多项所述的方法,其特征在于,洗脱温度在-4℃和98℃之间。
12.根据权利要求10所述的方法,其特征在于,洗脱温度在70℃和95℃之间。
13.根据前述权利要求中一项或多项所述的方法,其特征在于,纯水作为洗脱剂。
14.根据前述权利要求1到11中一项或多项所述的方法,其特征在于,带有附加的盐、酸、碱或溶剂的、经稀释的水溶液作为洗脱剂。
15.根据前述权利要求中一项或多项所述的方法,其特征在于,在层析中,肽的富集根据能够预定的功能特性来实现。
16.根据前述权利要求中一项或多项所述的方法,其特征在于,在层析中,从不期望的副成分中纯化出所述肽。
17.根据前述权利要求中一项或多项所述的方法,其特征在于,根据前述权利要求制造的肽混合物通过进一步的提纯步骤、特别是层析提纯步骤进行进一步的组分分离直至达到一种或多种肽的任意高的富集程度。
18.一种经富集的肽分离组分,所述肽分离组分根据前述权利要求中一项或多项所述的方法制造。
19.一种根据前述权利要求获得的肽的用途,用作在饲料、食品、化妆制品中的功能性物质以及用作制药学有效成分。
