本发明公开了一种蚕丝丝素肽粉止血材料的制备方法。将蚕茧脱胶后得精练蚕丝丝素蛋白备用;精炼蚕丝丝素蛋白以浴比1:20浸入配制盐溶液中溶解得到蚕丝丝素蛋白溶液;再经冷冻干燥制得蚕丝丝素肽粉止血材料。本发明的材料是蚕丝丝素蛋白水解的中间产物,与蚕丝丝素的氨基酸组成相同,分子量比蚕丝丝素蛋白小;无菌、无细胞毒性、无致敏和刺激作用。可用缓慢出血和快速、大量出血。本发明操作简单,耗能少,不使用强酸强碱,不破坏氨基酸,基本无污水和废渣,简易可行,具有实用价值。

技术领域
本发明涉及生物医学材料技术,尤其是涉及一种蚕丝丝素肽粉止血材料的制备方法。
技术背景
当人体失血超过800 mL时就会危及生命,而一旦意外伤害导致动脉出血,可能在10分钟内出血超过800mL,因此在急救医学上对于动脉出血抢救有“白金10分钟”的说法。但在实际应用特别是在战争或交通事故中,因缺乏有效的止血药物,很多伤员在送救途中错过治疗最佳阶段,而丧失生命或造成伤残。每年我国有上百万的人死于失血过多,这些人群主要集中在车祸、摔砸伤、地震灾害、洪水灾害、手术中或手术后以及其它突发事件等出血。在战场上,50%的阵亡士兵是由于出血过多所致。所以为了减少失血造成的不必要的死亡,各国医学界对止血材料的止血性能提出了更高的要求:作为止血敷料或止血剂的材料不仅要具备良好的止血性能,而且要具有优良的生物相容性,无毒副作用,无刺激性,易于加工成型等优异性能,开发出止血效果更佳的材料势在必行。
蚕丝丝素肽粉是蚕丝丝素蛋白水解的中间产物,与蚕丝丝素的氨基酸组成相同,但其分子量比蚕丝丝素要小得多,具有良好的生物相容性,对机体无毒性、无致敏和刺激作用,在不影响细胞活性对细胞有良好的细胞附着率和增殖率,而且具有维持细胞正常形态和功能的作用,被认为是目前生物相容性与胶原蛋白效果相当的物质。所以作者认为蚕丝丝素蛋白作为止血材料的应用前景非常广阔。但蚕丝丝素肽粉的生产,一般采用强酸、强碱法水解丝素或丝素用中性盐溶解后再经酶水解,控制一定的水解条件可以获得各种不同分子量的丝素肽。现有制备方法,均存在过程复杂且会破坏部分氨基酸,产生的废液对环境造成污染、生产成本过高等问题,不适合大规模生产。这些缺陷极大地限制了蚕丝丝素肽的应用。因此为解决上述问题,需要开发出方法操作简单,耗能少,更有效地制备蚕丝丝素肽粉的方法。
发明内容
本发明的目的在于提供一种蚕丝丝素肽粉止血材料的制备方法,该方法制备的蚕丝丝素肽粉止血材料价格便宜、原料易得、止血速度快、抗菌抑菌、与人体生物相容性好、可以生物降解、能促使创面快速修复及其它功能性如止痛等的快速止血材料。
本发明采用的技术方案的步骤如下:
(1)将蚕茧切剖去蛹剥成片状后在质量百分比浓度为2%的中性丝光皂溶液中精练,中间用质量百分比浓度为0.05%的碳酸钠溶液和去离子水反复洗净,用质量百分比浓度为0.5%的苦味酸胭脂红溶液检验至丝胶脱净为止,再用质量百分比浓度为0.05%碳酸钠溶液浸泡,用去离子水洗净后离心脱水,置入恒温干燥箱烘干,得精练蚕丝丝素蛋白备用;
(2)将步骤(1)中得到的精炼蚕丝丝素蛋白以浴比1:20浸入配制盐溶液中,在温度为80~180℃和压强为100~500千帕斯卡条件下溶解2~24小时,得到蚕丝丝素蛋白溶液;
(3)将第步骤(2)中得到的蚕丝丝素蛋白溶液经冷冻干燥制得蚕丝丝素肽粉止血材料。
所述盐溶液为氯化钙、乙醇和水配成的有机无机混合溶剂,其中氯化钙、乙醇和水的重量百分比为0.1-1:0.1-1:1。
本发明具有的有益效果是:
本发明的材料是蚕丝丝素蛋白水解的中间产物,与蚕丝丝素的氨基酸组成相同,分子量比蚕丝丝素蛋白小;无菌、无细胞毒性、无致敏和刺激作用。本发明所制备的蚕丝丝素肽粉可以用于不同医用场合的止血,包括缓慢出血和快速、大量出血。该材料止血效果好,止血速度快,经过肝切口的动物止血实验模型,其止血性能可与美国Medafor公司生产Arista止血粉媲美。
本发明操作简单,耗能少,不使用强酸强碱,不破坏氨基酸,基本无污水和废渣,简易可行,具有重大实用价值。
具体实施方式
实施例1:
(1)将蚕茧切剖去蛹剥成片状后称取20g在质量百分比浓度为2%的中性丝光皂溶液中精练2次,每次50min,中间用质量百分比浓度为0.05%的碳酸钠溶液和去离子水反复洗净,用质量百分比浓度为0.5%的苦味酸胭脂红溶液检验至丝胶脱净为止,然后再在50℃下,用质量百分比浓度为0.05%的碳酸钠溶液浸泡30min,用去离子水洗净后离心脱水,置入40℃恒温干燥箱烘干,得精练蚕丝丝素。
(2)取4克脱胶后的精练蚕丝丝素置于氯化钙、乙醇和水的质量百分比为0.1:0.1:1的80mL配制盐溶液中,在温度为180℃和压强为500千帕斯卡下溶解24小时。冷却后将蚕丝丝素粗溶液抽滤去除不溶物,得到纯净的蚕丝丝素蛋白溶液待用。
(3)将待用的蚕丝丝素蛋白溶液在-80℃预冷冻12h,再将冷冻样品放入冷冻干燥机干燥2天,制得分子量为10000的蚕丝丝素肽粉止血材料。
实施例2:
(1)将蚕茧切剖去蛹剥成片状后称取20g在质量百分比浓度为2%的中性丝光皂溶液中精练2次,每次50min,中间用质量百分比浓度为0.05%的碳酸钠溶液和去离子水反复洗净,用质量百分比浓度为0.5%的苦味酸胭脂红溶液检验至丝胶脱净为止,然后再在50℃下,用质量百分比浓度为0.05%的碳酸钠溶液浸泡30min,用去离子水洗净后离心脱水,置入40℃恒温干燥箱烘干,得精练蚕丝丝素。
(2)取5克脱胶后的精练蚕丝丝素置于氯化钙、乙醇和水的质量百分比为1:1:1的100mL配制盐溶液中,在温度为80℃和压强为100千帕斯卡下溶解2小时。冷却后将蚕丝丝素粗溶液抽滤去除不溶物,得到纯净的蚕丝丝素蛋白溶液待用。
(3)将待用的蚕丝丝素蛋白溶液在-80℃预冷冻12小时,再将冷冻样品放入冷冻干燥机干燥2天,制得分子量为2000的蚕丝丝素肽粉止血材料。
实施例3:
(1)将蚕茧切剖去蛹剥成片状后称取20g在质量百分比浓度为2%的中性丝光皂溶液中精练2次,每次50min,中间用质量百分比浓度为0.05%的碳酸钠溶液和去离子水反复洗净,用质量百分比浓度为0.5%的苦味酸胭脂红溶液检验至丝胶脱净为止,然后再在50℃下,用质量百分比浓度为0.05%的碳酸钠溶液浸泡30 min,用去离子水洗净后离心脱水,置入40℃恒温干燥箱烘干,得精练蚕丝丝素。
(2)取6克脱胶后的精练蚕丝丝素置于氯化钙、乙醇和水的质量百分比为0.6:0.4:1的120mL配制盐溶液中,在温度为150℃和压强为300千帕斯卡下溶解15小时。冷却后将蚕丝丝素粗溶液抽滤去除不溶物,得到纯净的蚕丝丝素蛋白溶液待用。
(3)将待用的蚕丝丝素蛋白溶液在-80℃预冷冻12小时,再将冷冻样品放入冷冻干燥机干燥2天,制得分子量为6000的蚕丝丝素肽粉止血材料。
止血性能的测定:
实验动物:健康成年SD大白鼠,体重为180±10g,由上海长征医院实验动物中心提供。
对照组:美国Medafor公司生产的Arista止血粉
实验方法:老鼠肝切口止血预实验,麻醉成功后,将SD大鼠固定于解剖台上,并用碘尔康溶液消毒,在肋弓下沿着腹中线切一纵行切口,长度约2cm,深度先至皮肤全层,然后提起腹肌沿着腹中线剪开腹肌,注意避开腹内重要脏器,轻轻挤压腹部,肝中叶从切口处被挤出,注意避免损伤肝中叶,用无菌纱布吸干肝脏周围腹腔液,用天平称过重量的无菌纱布垫于肝中叶下,快速切除肝中叶,切线约是肝中叶下缘以上2cm。同时立刻上止血材料,并计时,直至出血停止,记下出血时间,称出吸血的无菌纱布重量,与之前称出的重量相减得出出血量。
止血性能的测定结果:
实验结果见附表1,实施例1中蚕丝丝素肽粉止血材料的止血时间为91秒,出血量为0.94g;实施例2中蚕丝丝素肽粉止血材料的止血时间为83秒,出血量为0.85g;实施例3中蚕丝丝素肽粉止血材料的止血时间为87秒,出血量为0.95g;其中作为对照组的美国Medafor公司生产的Arista止血粉的止血时间为86秒,出血量为0.93g。由此可见,蚕丝丝素肽粉止血材料的可溶性非常好,遇到血液能迅速地溶解,与创面的粘附能力强,止血效果好,与Arista止血粉止血效果相当。

Claims (2)
1.一种蚕丝丝素肽粉止血材料的制备方法,其特征在于,包括以下步骤:
(1)将蚕茧切剖去蛹剥成片状后在质量百分比浓度为2%的中性丝光皂溶液中精练,中间用质量百分比浓度为0.05%的碳酸钠溶液和去离子水反复洗净,用质量百分比浓度为0.5%的苦味酸胭脂红溶液检验至丝胶脱净为止,再用质量百分比浓度为0.05%碳酸钠溶液浸泡,用去离子水洗净后离心脱水,置入40℃恒温干燥箱烘干,得精练蚕丝丝素蛋白备用;
(2)将步骤(1)中得到的精炼蚕丝丝素蛋白以浴比1:20浸入配制盐溶液中,在温度为80~180℃和压强为100~500千帕斯卡条件下溶解2~24小时,得到蚕丝丝素蛋白溶液;
(3)将步骤(2)中得到的蚕丝丝素蛋白溶液经冷冻干燥制得蚕丝丝素肽粉止血材料。
2.根据权利要求1所述的一种蚕丝丝素肽粉止血材料的制备方法,其特征在于:所述盐溶液为氯化钙、乙醇和水配成的有机无机混合溶剂,其中氯化钙、乙醇和水的质量百分比为0.1-1:0.1-1:1。
