发明背景
牙斑在牙齿上形成为膜。其为微生物生长的产物,为由大量嵌入基质的微生物组成的在牙齿表面上累积的致密微生物层。微生物主要为球菌样生物,特别是在早期斑块中,所述球菌样生物在至少一些人的口腔内在几天后变为丝状生物。

聚合物可包含水溶性聚合物和水不溶性聚合物的混合物,条件是在施用时释放颗粒。可用的水不溶性聚合物包括可溶于至少一种有机溶剂的聚合物;例如,丙烯酸共聚物(其中羧酸官能团未被中和)、交联的聚(乙烯基吡咯烷酮)例如可得自BASF的KOLLIDON® CL或CL-M、聚(乙酸乙烯酯) (PVAc)、某些纤维素衍生物例如乙酸纤维素、硝酸纤维素、烷基纤维素例如乙基纤维素、丁基纤维素和异丙基纤维素、乙酸邻苯二甲酸纤维素、虫胶、乙烯-乙酸乙烯酯共聚物、乙酸乙烯酯均聚物、硅酮聚合物(例如,二甲基硅酮)、聚甲基丙烯酸甲酯(PMMA)、不溶于有机溶剂的聚合物例如纤维素、聚乙烯、聚丙烯、聚酯、聚氨酯和尼龙、天然或合成的橡胶和它们的混合物。合适的成膜丙烯酸共聚物的一个实例为LUVIMER® 30E,为丙烯酸叔丁酯/丙烯酸乙酯/甲基丙烯酸共聚物的30重量%乙醇溶液,市售可得自BASF (Florham Park,N.J.,美国)。水不溶性聚合物可制备为分散液(例如,通过乳液聚合)并且可使用合适的乳化剂稳定。一种可用的PVAc乳液例如为KOLLICOAT® SR 30D,为PVAc在水的30重量%分散液,使用2.7重量%PVP和0.3%十二烷基硫酸钠稳定。丙烯酸共聚物分散液的实例为KOLLICOAT® EMM 30D,为丙烯酸乙酯:甲基丙烯酸甲酯共聚物(丙烯酸乙酯与甲基丙烯酸甲酯的重量比为约2:1)的30重量%水性分散液,据报道平均分子量为800,000,可得自BASF。
其它可用的聚合物或水溶性填料不受限制地包括天然树胶,例如藻酸钠、角叉菜胶、黄原胶、阿拉伯树胶、阿拉伯胶、瓜尔豆胶、支链淀粉、琼脂、甲壳质、壳聚糖、果胶、刺梧桐树胶、玉米蛋白、大麦醇溶蛋白、oliadin、刺槐豆胶、黄蓍胶和其它多糖;淀粉,例如麦芽糊精、直链淀粉、高直链淀粉、玉米淀粉、马铃薯淀粉、稻米淀粉、木薯淀粉、豌豆淀粉、甜马铃薯淀粉、大麦淀粉、小麦淀粉、蜡质种玉米淀粉、改性淀粉(例如,羟丙基化的高直链淀粉淀粉)、糊精、果聚糖、爱生兰和面筋;和蛋白质,例如胶原蛋白、乳清蛋白质分离物、酪蛋白、牛奶蛋白质、大豆蛋白质、角蛋白和明胶。涂布聚合物还可包括可分散或可溶胀的填料,例如改性淀粉、藻酸酯、藻酸盐的二价或多价离子盐。
水不溶性聚合物的其它非限制性实例包括乙酸纤维素、硝酸纤维素、乙烯-乙酸乙烯酯共聚物、乙酸乙烯酯均聚物、乙基纤维素、丁基纤维素、异丙基纤维素、虫胶、疏水性硅酮聚合物(例如,二甲基硅酮)、PMMA (聚甲基丙烯酸甲酯)、乙酸邻苯二甲酸纤维素和天然或合成的橡胶;不溶于有机溶剂的聚合物,例如纤维素、聚乙烯、聚丙烯、聚酯、聚氨酯和尼龙。
在水性组合物中,在聚合物涂层中水溶性聚合物和水不溶性和/或部分水溶性聚合物的相对量优选使得涂层在水性组合物中储存稳定,但是在使用组合物期间分解。在各种实施方案中,涂层包括一定量的水溶性聚合物,其按重量计为涂层的0.1%-90%、1%-80%、5%-70%、9%-50%或10%-40%。除了水溶性聚合物以外或作为其代替,涂层可包括部分水不溶性或水可溶胀的聚合物,其量按重量计为涂层的0.1%-50%,优选1%-10重量%。在各种实施方案中,在水性载体环境中稳定亲水性涂层的方法是在涂层中使用水溶性材料和水不溶性材料,使其在储存在产品载体中时均衡以得到稳定性,但其在使用时分解以释放其中所含的活性成分。
在制备涂布溶液中,应选择用于所选聚合物的适当溶剂和辅助溶剂,根据本领域的常识,以生产在操作温度下可流动的成膜涂布溶液。
用于稳定微聚集体中的颗粒的表面活性剂优选为非极性表面活性剂或在形成微聚集体之前或在施用之后能抑制颗粒聚集的任何表面活性剂。因此,合适的聚合物包括,例如,聚氧乙烯-脱水山梨糖醇-脂肪酰基酯、聚氧乙烯-脱水山梨糖醇-脂肪醚、聚羟基乙烯-脂肪单酰基酯、聚羟基乙烯-脂肪二酰基酯或聚羟基乙烯-脂肪醚和它们的混合物,例如聚氧乙烯脱水山梨糖醇单油酸酯(例如,聚山梨酸酯20、聚山梨酸酯40、聚山梨酸酯60、聚山梨酸酯65或聚山梨酸酯80,例如,聚山梨酸酯80,例如,聚氧乙烯(20)脱水山梨糖醇单油酸酯,例如吐温80)以及生育酚聚乙二醇琥珀酸酯(TPGS,例如,D-α-生育酚PEG 1000琥珀酸酯)和前述任何物质的混合物。
口腔护理制剂:根据已知的方法,本发明的微聚集体可配制成为口腔护理组合物。特别优选含有所述微聚集体的牙膏。
氟化物离子源:口腔护理组合物还可包括一种或多种氟化物离子源,例如,可溶性氟化物盐。各种各样产生氟化物离子的材料可用作本发明组合物中的可溶性氟化物的来源。合适的产生氟化物离子的材料的实例可见于Briner等人的美国专利号3,535,421;Parran,Jr.等人的美国专利号4,885,155;和Widder等人的美国专利号3,678,154,其通过引用结合到本文中。
代表性氟化物离子源包括但不限于氟化亚锡、氟化钠、氟化钾、单氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺、氟化铵和它们的组合。在某些实施方案中,氟化物离子源包括氟化亚锡、氟化钠、单氟磷酸钠以及它们的混合物。
在某些实施方案中,本发明的口腔护理组合物也可含有氟化物离子源或提供氟的成分,其量足以供应25 ppm-25,000
ppm的氟化物离子,通常至少500 ppm,例如,500-2000 ppm,例如,1000-1600 ppm,例如,1450 ppm。氟化物的适当水平取决于特定应用。供一般消费者使用的牙膏通常具有1000-1500 ppm,儿科牙膏稍少。用于专业应用的洁齿剂或涂层可具有高达5,000乃至25,000
ppm氟化物。
在一种实施方案中,氟化物离子源可以0.01重量%-10重量%或0.03重量%-5重量%的水平加入到本发明的组合物中,而在另一实施方案中,按重量计为另一实施方案中组合物的0.1重量%-1重量%。提供适当水平的氟化物离子的氟化物盐的重量明显基于盐中的反离子的重量而变。
当组合物包含碳酸氢钙时,出于稳定性原因,比起氟化钠,优选单氟磷酸钠。
研磨剂:本发明的微聚集体可与研磨材料一起加入到洁齿剂中,所述研磨材料例如天然碳酸钙、沉淀碳酸钙、磷酸钙研磨剂例如磷酸三钙(Ca3(PO4)2)、羟基磷灰石(Ca10(PO4)6(OH)2)或二水合磷酸二钙(CaHPO4• 2H2O,在本文中有时也称为DiCal)或焦磷酸钙、和/或二氧化硅研磨剂例如平均颗粒尺寸最多为20 μm的沉淀二氧化硅(例如由J. M.
Huber销售的Zeodent 115®)、偏磷酸钠、偏磷酸钾、硅酸铝、煅烧的氧化铝、膨润土或其它硅质材料或它们的组合。
发泡剂:本发明的口腔护理组合物也可包含当刷拭口腔时提高产生的泡沫量的试剂。提高泡沫量的试剂的说明性实例包括但不限于聚氧乙烯和某些聚合物,包括但不限于藻酸酯聚合物。聚氧乙烯可提高泡沫的量和通过本发明的口腔护理载体组分产生的泡沫的厚度。聚氧乙烯还常称为聚乙二醇(“PEG”)或聚氧化乙烯。适用于本发明的聚氧乙烯的分子量为200,000-7,000,000。在一种实施方案中,分子量为600,000-2,000,000,在另一实施方案中,为800,000-1,000,000。Polyox®为由Union
Carbide生产的高分子量聚氧乙烯的商品名。按重量计,聚氧乙烯可以本发明的口腔护理组合物的口腔护理载体组分的1%-90%的量存在,在一种实施方案中,为5%-50%,在另一实施方案中,为10%-20%。口腔护理组合物中发泡剂的剂量(即,单一剂量)为0.01-0.9重量%,0.05-0.5重量%,在另一实施方案中,为0.1-0.2重量%。
表面活性剂:任选包括在本发明的口腔护理组合物中的另一种试剂为表面活性剂或相容的表面活性剂的混合物。合适的表面活性剂为遍及宽的pH范围内相当稳定的那些,例如,阴离子、阳离子、非离子或两性离子的表面活性剂。合适的表面活性剂更充分地描述于例如Agricola等人的美国专利号3,959,458;Haefele的美国专利号3,937,807;和Gieske等人的美国专利号4,051,234,其通过引用结合到本文中。
在某些实施方案中,本文可用的阴离子表面活性剂包括在烷基中具有10-18个碳原子的硫酸烷基酯的水溶性盐和具有10-18个碳原子的脂肪酸的磺化单甘油酯的水溶性盐。十二烷基硫酸钠、十二酰基肌氨酸钠和椰油单甘油酯磺酸钠为这种类型的阴离子表面活性剂的实例。也可利用阴离子表面活性剂的混合物。
在另一实施方案中,可用于本发明的阳离子表面活性剂可宽泛地定义为具有一个含有8-18个碳原子的长烷基链的脂族季铵化合物的衍生物,例如十二烷基三甲基氯化铵、十六烷基氯化吡啶、十六烷基三甲基溴化铵、二异丁基苯氧基乙基二甲基苄基氯化铵、椰油烷基三甲基亚硝酸铵、十六烷基氟化吡啶和它们的混合物。

可用于本发明的组合物的说明性非离子表面活性剂可宽泛地定义为通过氧化烯基团(亲水性性质)与有机疏水性化合物(其可为脂族或烷基芳族性质)缩合而生产的化合物。合适的非离子表面活性剂的实例包括但不限于普卢兰尼克(Pluronic)、烷基苯酚的聚氧化乙烯缩合物、衍生自使氧化乙烯与氧化丙烯和乙二胺的反应产物缩合的产物、脂族醇的氧化乙烯缩合物、长链叔胺氧化物、长链叔膦氧化物、长链二烷基亚砜和这些材料的混合物。

表面活性剂或相容的表面活性剂的混合物可按重量计以总组合物的0.1%-5.0%存在于本发明的组合物中,在另一实施方案中,为0.3%-3.0%,在另一实施方案中,为0.5%-2.0%。
调味剂:本发明的口腔护理组合物也可包括调味剂。用于本发明实践的调味剂包括但不限于精油以及各种调味的醛、酯、醇和类似的材料。精油的实例包括留兰香、胡椒薄荷、冬青、黄樟、丁香、鼠尾草、桉树、马郁兰、肉桂、柠檬、酸橙、葡萄柚和橙的油。还可用诸如薄荷醇、香芹酮和茴香脑等化学物质。某些实施方案采用胡椒薄荷和留兰香的油。
调味剂以0.1-5重量%和0.5-1.5重量%的浓度加入到口腔组合物中。在单个口腔护理组合物剂量(即,单一剂量)中,调味剂的剂量为0.001-0.05重量%,在另一实施方案中,为0.005-0.015重量%。
螯合剂:本发明的口腔护理组合物也可任选包括一种或多种能络合存在于细菌细胞壁的钙的螯合剂。该钙的结合减弱细菌细胞壁并且加强细菌溶解。
适于在本发明中用作螯合剂的另一组试剂为可溶性焦磷酸盐。用于本发明的组合物中的焦磷酸盐可为任何碱金属焦磷酸盐。在某些实施方案中,盐包括四碱金属焦磷酸盐、二碱金属二酸焦磷酸盐、三碱金属一酸焦磷酸盐和它们的混合物,其中所述碱金属为钠或钾。盐可按它们的水合和非水合形式两者使用。可用于本发明的组合物的有效量的焦磷酸盐通常足以提供至少1.0重量%焦磷酸盐离子,1.5重量%-6重量%,3.5重量%-6重量%的这些离子。

水:水也可存在于本发明的口腔组合物中。用于制备商品口腔组合物的水应为去离子的并且不含有机杂质。水通常构成组合物的余量,并且按重量计占口腔组合物的10%-90%、20%-60%或10%-30%。该量的水包括加入的游离的水加上连同其它材料(例如山梨糖醇或本发明的任何组分)被引入的量。
湿润剂:在口腔组合物的某些实施方案中,还期望加入湿润剂以防止组合物当暴露于空气时硬化。某些湿润剂也可为洁齿剂组合物赋予期望的甜味或香味。在一种实施方案中,湿润剂(基于纯的湿润剂)通常按重量计占洁齿剂组合物的15%-70%,或者在另一实施方案中,为30%-65%。合适的湿润剂包括可食用的多羟基醇,例如甘油、山梨糖醇、木糖醇、丙二醇以和其它多元醇和这些湿润剂的混合物。在某些实施方案中,可使用甘油和山梨糖醇的混合物作为本文的牙膏组合物的湿润剂组分。

喷雾干燥:使相对不溶的金属、金属盐或金属氧化物颗粒(例如,氧化锌粉末)、表面活性剂和涂布聚合物的悬浮液通过喷雾干燥机,以产生干燥的聚合物涂布的微聚集体。喷雾干燥为工业中已确立的技术,其可用于由湿的原料生产自由流动的干燥的颗粒产品。使用喷雾干燥技术的方法为本领域熟知。总的来说,生产的颗粒尺寸受到存在于湿进料中的主要颗粒尺寸的影响,并且通常在1 μm-50 μm范围,沿着主尺寸轴测量,主要尺寸通常为5微米-30微米;主要尺寸优选为10微米-25微米。特别优选的实施方案含有喷雾干燥的聚合物涂布的氧化锌颗粒,其通常为球形形状,并且主要尺寸为20微米。
此外,与大多数材料一样,所得到的颗粒尺寸将跨颗粒尺寸分布而变化。得到的颗粒尺寸分布通常为单峰,然而,也出现双峰或多峰分布,其中多于一种代表性尺寸类型存在于所得到的材料中。通常,代表总体的较大颗粒沿着主轴或直径大于5μm。优选微聚集体的尺寸在约5 μm-50 μm范围之间。

混合程序:
将HPMC E5、HPMC E50、PG和吐温80溶解于水中。随后将ZnO粉末加入到湿成分中,接着加入乙醇。将材料共混,直至观察到均匀混合物。
喷雾干燥程序
将所得到的湿制剂进料至Buchi B-290喷雾干燥器,设置入口温度=170℃,出口温度=99-100℃,抽吸=100%,空气流=40,进料速率=30% (7.2 mL/分钟)。
使用扫描电子显微镜观察得到的喷雾干燥的材料,其含有双峰颗粒分布,该分布由直径~20μm的聚合物涂布的大的球形ZnO颗粒和主要尺寸~5μm的干燥聚合物的形状不规则颗粒组成。
将喷雾干燥的材料的样品1-6的少量放置在玻璃载片上。将50 μL甘油/水(1:4)添加到每个载片。甘油/水的粘度为1.76 cp/mPa*s 。在显微镜(4倍放大)下观察颗粒。记录样品最快分解的时间以及分解完成到50%、75%、90%和99%的时间。时间记录于表3。

吸收方法
VITRO-SKIN®为有效模拟人皮肤的表面性质的测试基材。其含有优化的蛋白质和脂质组分两者,并且设计成具有与人皮肤类似的拓扑结构、pH、临界表面张力和离子强度。将Vitro-Skin切成正方形,以匹配HAP圆盘的尺寸(12.4×12.4 mm=½×½英寸)。样品用己烷漂洗三次,随后在1 mL的唾液中于37℃温育过夜(每次测量一式三份)。在去离子水中制备两种悬浮液,分别含有喷雾干燥的ZnO的1%悬浮液和ZnO粉末的0.5%悬浮液。使用Vitro-Skin将唾液从管中吸出。将ZnO悬浮液的2 mL等分试样加入到每个管中。将浆料从管中吸出,并且用5 mL水漂洗(涡流1×10秒)。用5 mL水(涡流1×10秒)再次漂洗管。将Vitro Skin 样品转移至新的管子,并且在1 mL HCl (1摩尔浓度)中涡流漂洗30秒。加入4 mL去离子水,让其放置2小时,偶尔使用涡流。随后通过原子光谱分析溶液,以测量锌的浓度。以微克/片的基准测量在样品上的ZnO吸收量,结果示于表4。
