本发明涉及组织蛋白酶H的用途。本发明的其它方面涉及用于筛选药物的方法、用于诊断疼痛易感性的方法以及用于治疗疼痛的方法。
在西方国家,慢性疼痛是未解决的重大健康问题,其逐渐损害数百万公民的健康和幸福。慢性疼痛严重困扰患有慢性疼痛的个人幸福并且往往伴随有或随后出现植物性症状(vegetativesigns),这常常导致抑郁症。慢性疼痛致使个体受苦并造成巨大程度的社会经济损失。现有药理学疼痛疗法就功效和安全性而言均普遍不令人满意。
鉴于与现有疼痛治疗技术相关的严重缺陷,极需新的选择用于治疗持续疼痛以及用于与慢性疼痛潜在发展有关的诊断和预后。尤其是鉴于对疼痛神经生物学的快速增进的了解与提供有效治疗且无现有治疗技术缺陷的未满足的临床需求之间的巨大鸿沟,有需要致力于发现新型镇痛药类的新靶标。
因此,本发明的目的是提供用于开发和提供调节疼痛的新型药物的新方法。
通过使用组织蛋白酶H或其功能片段或衍生物以鉴定调节疼痛并且特别是神经性疼痛的化合物来达到该目的。
本发明基于发明人以下意想不到的发现,首次证明在小鼠神经性疼痛模型中组织蛋白酶H的表达与疼痛易感性密切相关。
按照国际疼痛研究协会的定义,疼痛是与实际或潜在组织损伤有关、或依据此类损伤来描述的不愉快的感觉和情绪体验。疼痛通常是感受伤害的神经系统激活的结果,其特化为探测并编码(encode)组织损伤或潜在损伤。因此,疼痛是身体警报系统的一部分,以启动使身体的实际或潜在损伤最小化的反应。疼痛可为医学病症的原发症状或可为疾病状态的继发效应,通常无任何生物学意义。
疼痛可为急性或慢性的。急性疼痛是表明潜在或实际损伤的生理信号。其伴随组织破坏、感染、炎症或其它急性起因而发生,是身体损伤或机能障碍后对个体的警告。急性疼痛若治疗不当,可导致慢性疼痛。
慢性疼痛是具有不同起因、持续期、强度和特定症状的疾病状态。
慢性疼痛可为感受伤害起源的、炎性或神经性的。认为感受伤害的疼痛与躯体或内脏的疼痛-敏感性神经纤维的持续激活成相当比例。神经性疼痛是由外周或中枢神经元组织的任何种类损伤导致的疼痛;认为其通过外周神经系统、CNS或两者中的异常体觉过程而持续。(关于疼痛机制的综述,参见例如Scholz和Woolf,2002;Julius和Basbaum,2001,Woolf和Mannion,1999;Wood,J.D.,2000;Woolf和Salter,2000.)
慢性神经性疼痛因不同患者而异。最近的数据表明,对于个体所遭受的痛苦的量,个体的疼痛易感性起重要作用,即对于疼痛,特别是对于神经性疼痛的发展有着重要的遗传诱因。本发明基于本发明人在啮齿动物慢性疼痛模型中旨在鉴定疼痛易感性基因(即决定在给定的固定程度的组织损伤存在下感受的疼痛量的基因)的大量研究。发明人使用的啮齿动物模型和实验设置使得在不同个体之间具有以下实验条件:a)可精确控制神经损伤的性质和一致性;和b)可使遗传及环境的变化性最小化。
组织蛋白酶H(CTSH;依据theAtlasofGeneticsandCytogeneticsinOncologyandHaematology(http://atlasgeneticsoncology.org)可替代的名称为:ACC-4、ACC-5、CPSB、DKFZp686B24257、EC3.4.22.16、MGC1519、aleurain、小链(minichain))是一种溶酶体蛋白酶。
人组织蛋白酶H的基因座在染色体15q24-125上(参见例如AtlasofGeneticsandCytogeneticsinOncologyandHaematology)。该基因包含跨越超过23kb基因组序列的12个外显子。人前组织蛋白酶原H(preprocathepsinH)的核苷酸序列为例如由Fuchs和Gassen,1989,NucleicAcidsRes.17:9471-9471公布的。大鼠组织蛋白酶H的基因结构为例如由Ishidoh等,FEBSLetters,1989,第253卷,number1,2,103-107公布。人15号染色体上CTSH的基因组序列可例如在NCBI主页以登录号NG_009614.1(SEQIDNO.1)而检索到。

组织蛋白酶H是一种具有氨肽酶和较弱内肽酶功能的溶酶体蛋白酶。其属于C1(木瓜蛋白酶-样)半胱氨酸蛋白酶(C1(papain-like)cysteinproteases)家族。组织蛋白酶H蛋白作为335个氨基酸的前酶原合成并且蛋白水解加工成内体或溶酶体内的活性单链。除重链和轻链(一起命名为长链)之外,组织蛋白酶H还包含所谓的来源于前肽的小链“EPQNCSAT”。(关于组织蛋白酶H的结构,也可参见Turk等.,BiologicalChemistry,1997)。小链似乎参与组织蛋白酶H的氨肽酶活性并且在底物识别中起关键作用。缺乏小链的人组织蛋白酶H的重组形式显示为内肽酶(Valiljeva等,2003)。组织蛋白酶H的内肽酶底物为例如:Bz-Arg+NHNap、Bz-Arg+NH-Mec、Bz-Phe-Val-Arg+NHMec。其对于Pro-Gly+Phe和Pro-Arg+NHNap的底物具有二肽基肽酶活性。其对于以下底物具有氨肽酶活性:H-Arg-NH-Mec、Bz-Arg-NH-Mec、Bz-Arg-NH-Mec和H-Cit-NH-Mec(参见例如Rothe和Dodt,1992;Bz=苯甲酰基,NH-Mec=e-甲基香豆素基-7-酰胺;Cit=瓜氨酸)。天然存在的组织蛋白酶H抑制剂为半胱氨酸蛋白酶抑制剂(cystatin)、α2-巨球蛋白以及来自小鼠细胞毒淋巴细胞CTLA-2β的抗原。
组织蛋白酶H遍在表达并且在肾中表达水平最高。在一些癌组织中其表达可增加。组织蛋白酶主要位于内体-溶酶体区室,但也一定程度地分泌并且在血液中循环。
组织蛋白酶H的功能通常包括:蛋白酶活性例如水解酶活性,肽酶活性例如氨肽酶、二肽基肽酶、外肽酶或内肽酶活性,转酰酶活性(参见Koga等,1991);裂解上述列举的底物;裂解天然C5并且产生趋化因子C5a,加工疏水性表面活性剂-相关蛋白C,裂解和/或降解纤连蛋白和纤维蛋白原。更具体而言,其作为溶酶体半胱氨酸蛋白酶参与细胞内的蛋白质降解。其通过裂解天然C5而参与趋化因子C5a的产生。组织蛋白酶H参与疏水性表面活性剂-相关蛋白C的加工。此外,其似乎还参与不稳定动脉粥样斑块的发展,以及还可能参与早期动脉粥样硬化生成。
本发明的用途使得能够鉴定用于预防和/或治疗疼痛并且特别是神经性疼痛的新物质。本发明的用途包括鉴定具有所需特性(即降低疼痛感)的化合物以及鉴定具有不合乎需要特性(即增加疼痛感)的化合物。此外,本发明使得能够进一步表征已鉴定的可用于预防和/或治疗任何疾病或疾病状态的化合物。在此情况下,本发明可例如用于排除经鉴定具有非所需副作用(即增加疼痛感)的活性化合物:可例如针对它们对组织蛋白酶H的影响(蛋白质和/或核酸,表达和/或功能等)来表征(profile)给定疾病的候选化合物。
用于本发明不同方面的化合物/测试化合物/活性化合物可为任何生物或化学物质或天然产品提取物,它们可通过生化或分子生物学方法来纯化、部分纯化、合成或制备。
在本发明不同方面的意义上认为有效调节疼痛的化合物可为对以下方面有影响的任何物质:组织蛋白酶H的功能之一或细胞中组织蛋白酶H的量(蛋白质或核酸),对组织蛋白酶H的表达、翻译后修饰(例如N-糖基化或加工(例如裂解)、蛋白质折叠或活化。
为此,所述物质可调节组织蛋白酶H的任何功能(例如上文或下文列举的那些)。组织蛋白酶H的蛋白质活性可通过物质来调节,例如通过直接相互作用以及干扰组织蛋白酶H多肽/蛋白质或其片段来调节。所述物质还可调节组织蛋白酶H的表达,例如在转录(起始、延伸、加工等)、转录物稳定性、翻译水平上来调节。此外,其还可调节组织蛋白酶H的翻译后修饰、从无活性到活性形式的加工(将前原形式(prepro-form)裂解为活性蛋白质)、以及蛋白质折叠等。所述物质可直接或间接施加上述作用(间接地意即通过干扰(正向或负向)对组织蛋白酶H功能/蛋白质活性/表达等有影响的天然信号转导级联)。
组织蛋白酶H的功能包括上文列举的那些,例如蛋白酶活性;从一种或多种蛋白质底物的氨基端除去二肽的能力;与一种或多种蛋白质底物特异性相互作用的能力(蛋白质-蛋白质相互作用),例如上文列举的那些;裂解和/或活化一种或多种蛋白质底物的能力,例如上文列举的那些;加工蛋白质或肽底物的能力,例如上文列举的那些;被上文列举的抑制剂之一抑制的能力。
组织蛋白酶H的功能通常还包括组织蛋白酶H蛋白或核酸或其片段与其它分子(包括但不限于蛋白质、肽、核酸、合成分子)相互作用的能力,并且优选涉及其相互作用于及裂解蛋白质底物的能力。
根据本申请,酶底物理解为可由酶修饰的任何分子。本发明范围内天然存在的底物为对应于其在天然生理学或病理学环境(例如上文列举的那些)中存在的形式并且亦能被相应的酶修饰的分子。
对疼痛特别是神经性疼痛的调节可为降低或升高。
根据本相关发明组的一个方面,可使用组织蛋白酶H的片段或衍生物。片段可为蛋白质、多肽或多核酸片段。
蛋白质或多肽的片段是当与组织蛋白酶H的全长实例相比时,携带一个或多个末端(n-和/或c-末端)和/或一个、两个或多个内部氨基酸缺失的蛋白质或多肽,片段包括,例如如图7的描述中列举的结构域和/或片段(参见下文)。
组织蛋白酶H蛋白的功能片段为具有至少一种或多种全长蛋白功能特性(特别是上文列举的特性)的该蛋白的任何片段。
多核苷酸片段是当与全长基因组或编码序列相比时,携带一个或多个末端(5’-和/或3’-末端)和/或一个、两个或更多个内部核苷酸缺失的多核苷酸或寡核苷酸。组织蛋白酶H核酸的功能片段为具有至少一种或多种全长多核酸(mRNA、基因组或编码序列)功能特性的任何片段。

非人动物可为任何非人动物。优选啮齿类,例如大鼠或小鼠。
转基因动物为在其基因组中携带特意转入其中的外源DNA的动物。可按照标准程序将外源DNA引入动物基因组(参见例如TransgenicAnimalTechnologyALaboratoryHandboook.C.A.Pinkert编辑;AcademicPressInc.,SanDiego,California,1994(ISBN:0125571658)。
术语异源表达是指不同于在宿主生物体中正常基因表达的表达(涉及表达的基因的稳态水平、量、时机或组织分布,或涉及表达的基因类型(即所述基因通常完全不在该宿主中表达))。异源表达可为组成型或诱导型的。本领域熟知合适的诱导型表达系统(例如四环素诱导型系统等)。生物体可为细胞或非人动物。

敲除生物体(例如动物或细胞)是指其中基因的表达或功能部分或全部缺失的生物体,包括基因组敲除以及功能敲除、诱导型敲除以及组成型敲除。本领域熟知敲除生物体的产生以及可用于产生敲除生物体的细胞或动物。组织蛋白酶H-敲除小鼠的产生描述于Pham和Ley,1999中。
本发明再一方面涉及非人组织蛋白酶H敲除动物作为用于降低疼痛并且特别是降低神经性疼痛敏感性的模型系统的用途。
异源表达组织蛋白酶H或其功能片段的细胞用于鉴定调节疼痛,特别是神经性疼痛的化合物的用途是本发明的另一方面。
细胞可为任何原核或真核细胞,例如能够用核酸载体转染并表达报告基因的细胞。它们包括大体上的原代细胞和来自细胞培养物的细胞,例如真核细胞培养物,其包括来源于多细胞生物体和组织的细胞(例如HeLA、CHO、COS、SF9或3T3细胞)或单细胞生物体例如酵母(例如裂殖酵母(s.pombe)或酿酒酵母(s.cerevisiae))的细胞;或原核细胞培养物,优选毕赤酵母(Pichia)或大肠杆菌(E.coli)。可通过熟知技术(例如取血技术、组织穿刺技术或手术技术)获得来源于组织的细胞和样品。

此外,组织蛋白酶敲除细胞作为用于降低疼痛,特别是神经性疼痛敏感性的模型系统的用途是本相关发明组的另一方面。
按照本发明另一实施方案,与参考细胞(例如处于其未修饰状态的同样的细胞)相比,所述细胞可具有更高量的组织蛋白酶H。该细胞系统可用于模拟疼痛敏感性增强的状态,这是因为组织蛋白酶H的表达量与疼痛敏感性相关。
因此,本发明还涉及异源表达组织蛋白酶H或其功能片段的细胞作为用于增强疼痛敏感性,特别是神经性疼痛敏感性的模型系统的用途。
异源表达与组织蛋白酶H启动子和/或增强子表达上连接的报告基因的细胞用于鉴定或分析调节疼痛,特别是神经性疼痛的化合物的用途是本相关发明组的另一实施方案。
本发明以上方面基于本领域通常已知的典型报告基因测定。为此,将选择的启动子插入适于所选宿主细胞类型的表达载体中,所述启动子位于所选报告基因上游,这样,启动子若有活性则使报告基因表达。随后将构建体引入到所选宿主细胞中。本领域熟知合适的转化或转染方法以及用于细胞培养和报告基因表达检测的条件(参见例如以下所列标准文献)。本领域熟知合适的条件以及载体、报告基因和必要试剂(其也可市售)。
载体为环状或线性多核苷酸分子,例如DNA质粒、噬菌体或粘粒,多核苷酸片段(例如从其它载体切下或通过PCR扩增并插入到克隆载体中的多核苷酸片段)借助其可在合适的细胞或生物体中特异性扩增。表达载体使得在宿主细胞或生物体中能够异源表达目的基因(例如报告基因)。细胞或生物体类型主要取决于目的并且对其选择是在技术人员知识范围内。用于扩增核酸的合适的生物体例如主要是具有高繁殖率的单细胞生物体,例如细菌或酵母。合适的生物体亦可为从多细胞组织分离和培养的细胞,例如从各种各样生物体产生的细胞系(例如来自草地贪夜蛾(SpodopteraFrugiperda)的SF9细胞等)。合适的克隆载体为本领域所知并且其购自不同的生物技术供应商,例如RocheDiagnostics、NewEnglandBiolabs、Promega、Stratagene及其它更多。合适的细胞系例如购自美国典型培养物保藏中心(ATCC)。
为异源表达蛋白质或多肽,细胞可为能够用核酸载体转染并表达目的基因(例如报告基因)的任何原核或真核细胞。这些细胞包括大体上的原代细胞和来自细胞培养物的细胞,优选真核细胞培养物,其包括来源于多细胞生物体和组织的细胞(例如HEK293、RIN-5F、HeLA、CHO、COS、SF9或3T3细胞)或单细胞生物体例如酵母(例如裂殖酵母或酿酒酵母)的细胞;或原核细胞培养物,优选毕赤酵母或大肠杆菌。可通过熟知技术(例如取血技术、组织穿刺技术或手术技术)获得来源于组织的细胞和样品。

在本发明以上方面的情况中,对照载体可为包含报告基因或其功能片段的任何合适载体,但其中报告基因的表达不是由(功能性)组织蛋白酶H启动子驱动。这可例如意味着报告基因或其功能片段没有可操作地连接至功能性组织蛋白酶H启动子(即完全缺乏组织蛋白酶H启动子,包括无功能的组织蛋白酶H启动子或启动子片段或其中启动子和报告基因的连接不是功能性的)。另一可能性是报告基因或其功能片段可操作地连接至除组织蛋白酶H启动子之外的另一启动子(例如SV40或另一标准启动子)。还可将功能载体和对照载体转染到同一细胞中,但此情况下报告基因需要有差异。
可例如按照下述或本领域已知测定来进行符合以上用途的化合物的鉴定。
测定是监测生物学过程的任何类型的分析方法或系统。适宜地,在细胞或生化(体外)系统中重现代表生理代谢途径部分以及病理病况部分的分子级联和机制。因此,可根据其干扰或调节这些级联或机制的能力来测定潜在药物化合物的药理学活性。
对于在药物筛选,特别是新型药物化合物的高通量筛选中的应用,所述测定必须可重复,并还优选可升级(scable)且可靠。在本发明范围内,高通量筛选意指本发明方法用含几毫升甚至几纳升或甚至更少的极小体积的样品以极小规模地例如在96、386或1536孔板上实施。因此,在短时间内可分析极大量的样品。高通量筛选主要包括借助单个测定对大约500.000种不同化合物就某种能力进行筛选。所述测定优选适用于针对调节研究中的靶分子的活性的能力来高通量筛选化学物质。测定类型取决于例如所用靶分子的类型(例如多肽或多核苷酸)以及测定该靶分子活性所依据的“读出”即参数(参见下文)。
不同类型的所述测定通常为本领域所知并且可购自供应商。
用于不同目的的合适测定包括放射性同位素或荧光测定,例如荧光偏振测定(例如由Panvera提供的市售测定),或用于测量标记成员与非标记成员的相互作用(例如标记的蛋白质与其未标记蛋白质-配体的相互作用)的PackardBioScience(HTRF;ALPHAScreenTM)。
更多实例包括基于细胞的测定,其中细胞系稳定(诱导型或非诱导型;染色体的或附加型)或瞬时表达重组目的蛋白。这些测定包括例如报告基因测定,其中根据报告酶的活性来测量对某些启动子或信号转导级联成员的信号转导途径的调节,所述报告酶的表达在所述某些启动子的调控之下。对于此类型的测定,重组细胞系需构建成含有受限定的启动子调控的报告基因,所述启动子本身将被研究或受研究中的信号转导级联调控。合适的报告酶通常为本领域所知,其包括萤火虫萤光素酶、海肾(renilla)萤光素酶(例如由Packardreagents市售)、β-半乳糖苷酶。合适的细胞系视测定目的而定,但通常包括易于转染和易于培养的细胞系,例如HeLA、COS、CHO、NIH-3T3等。
为测定蛋白酶活性,已知典型的蛋白酶测定方式:例如用在底物的某一位置携带报告标签(例如发射冷光/荧光或其它信号的蛋白质/肽或实体)和在另一位置携带猝灭剂(实体(例如抑制报告标签信号发射的另一种肽,只要底物是完整/未裂解的即可))的底物;将底物与组织蛋白酶H在合适条件下孵育,以使底物裂解,从而导致可检测信号(例如光发射)因为猝灭剂与报告标签的分离而发射。
本领域熟知其它类型的测定和其它类型的“读出”。用于检测细胞中组织蛋白酶H活性的一种测定可例如从Rüttger等,BioTechniques,2006获得。
本发明测定涉及例如:

本文使用的术语“所取样品”是指取自/分离自一个或多个个体(人或非人动物)的身体的生物样品。生物学材料和生物样品包括例如细胞、组织或器官的制备物或部分(例如脑、血液、肝、脾、肾、心脏、血管等),优选来源于脊椎动物,并且更优选来源于哺乳动物(包括人)。还包括来自细胞培养物的细胞,优选真核细胞培养物,其包括来源于多细胞生物体和组织的细胞(例如HeLA、CHO、COS、SF9或3T3细胞)或单细胞生物体例如酵母(例如裂殖酵母或酿酒酵母)的细胞;或原核细胞培养物,优选毕赤酵母或大肠杆菌。可通过熟知技术(例如取血技术、组织穿刺技术或手术技术)获得来源于组织的细胞和样品。重组分子的制备和来自细胞或组织的天然存在的分子的纯化以及细胞或组织提取物的制备为本领域技术人员所熟知(亦参见例如下文列举的标准文献)。
术语“参考样品”是指取自具有已知给定疼痛表型的一个或多个个体的生物样品,或指体外生物样品(例如来源于体外细胞或组织培养物的样品(例如培养的细胞)),并且其在某些特性(例如其组织蛋白酶H活性、量或表达的水平)上对应于给定疼痛表型(例如高疼痛敏感性或低疼痛敏感性)。
本发明又一方面涉及用于检测组织蛋白酶H的工具(means)在用于通过分析取自待检个体身体的生物样品来测定个体中增强的疼痛敏感性(特别是神经性疼痛敏感性)中的用途。
用于检测组织蛋白酶H的工具可为能够特异性检测生物样品中存在的组织蛋白酶H多肽/蛋白质或核酸的任何工具。
检测组织蛋白酶H蛋白或多肽的工具可为能特异性检测野生型组织蛋白酶H蛋白/多肽的任何工具,并且还可为特异性检测具有与野生型多肽/蛋白质相比一个或多个关于大小或氨基酸序列的突变的组织蛋白酶H蛋白/多肽的工具。此类工具的优选实例为能够特异性检测组织蛋白酶H蛋白的抗体,例如用于免疫组织学或免疫组织化学技术(例如检测组织学组织切片中组织蛋白酶H蛋白或其某些突变或固定在合适载体例如膜、芯片、ELISA板等上的组织蛋白酶H蛋白)。
检测组织蛋白酶H核酸的工具可例如为检测组织蛋白酶HmRNA/cDNA或基因组DNA(野生型或还具有与野生型组织蛋白酶H核酸相比关于其长度或其核酸序列的一个或多个突变)的工具。所述工具可例如为特异性检测和/或定量组织蛋白酶HmRNA的工具,并优选包括或为特异性组织蛋白酶H核酸探针或引物组,所述核酸探针或引物组能够扩增组织蛋白酶HDNA,或例如用于PCR测序(用于检测核苷酸序列中的突变),或能够扩增组织蛋白酶HcDNA,例如用于RTPCR(用于检测和/或定量组织蛋白酶HmRNA的表达)。另一工具可例如为能够在标准条件下与组织蛋白酶HmRNA或cDNA特异性杂交的核酸探针,例如用于RNA印迹或芯片杂交技术。

本发明的其它方面涉及治疗方法,例如:
用于在正经受疼痛的受试者中治疗疼痛的方法,所述方法包括给予所述受试者治疗有效量的在所述受试者中降低组织蛋白酶H量或活性的组合物。这可为组织蛋白酶H的总量或总活性,或是在某种组织中的量或活性,例如在神经组织、在淋巴组织或免疫系统细胞(例如肥大细胞、巨噬细胞、嗜中性粒细胞、T-细胞(例如CD8+T-细胞)等)中的量或活性,其中治疗有效量包括足以改善个体疼痛感觉或敏感性(特别是关于神经性疼痛)的量。
本发明另一方面涉及用于降低受试者疼痛(并且特别是神经性疼痛)敏感性的方法,所述方法包括给予所述受试者治疗有效量的降低所述受试者中(例如在淋巴组织或神经组织或免疫系统细胞中)的组织蛋白酶H的量(例如表达、半衰期)或活性的组合物。
此外,本发明涉及用于调节非人雌性受试者的后代中疼痛(并且特别是神经性疼痛)敏感性的方法,所述方法包括将赋予受调节的组织蛋白酶H表达的核酸转移(例如电穿孔)到受精卵中,将所述受精卵转移到非人代孕母体中,并选出符合其组织蛋白酶H表达特性(与野生型受试者例如小鼠相比组织蛋白酶H表达降低或废除)的后代。
本发明另一方面涉及能够降低组织蛋白酶H活性和/或表达的用于治疗疼痛并且特别是神经性疼痛的化合物。
本领域已知组织蛋白酶H的抑制剂,例如E-64d(参见例如Rüttger等,BioTechniques,41:469-473,2006)以及Kirschke,H等,1995,ProteinProfile2:1581-1643)。
为生产药物,可将本发明组织蛋白酶H调节剂与合适的添加剂或辅助物质一起配制,所述添加剂或辅助物质例如生理缓冲溶液,例如氯化钠溶液、软化水;稳定剂,例如蛋白酶或核酸酶抑制剂,优选抑肽酶、ε-氨基己酸或胃酶抑素A;或多价螯合剂,例如EDTA;凝胶制剂,例如白凡士林、低粘度石蜡和/或黄蜡等,视给药类型而定。
合适的另外的添加剂为例如去污剂,例如TritonX-100或脱氧胆酸钠;还有多元醇,例如聚乙二醇或甘油;糖,例如蔗糖或葡萄糖;两性离子化合物,例如氨基酸(例如甘氨酸或特别是牛磺酸或甜菜碱)和/或蛋白质(例如牛血清白蛋白或人血清白蛋白)。优选去污剂、多元醇和/或两性离子化合物。
生理缓冲液优选具有约6.0-8.0的pH(特别是约6.8-7.8的pH,尤其是约7.4的pH)和/或约200-400毫渗克分子/升、优选约290-310毫渗透分子/升的同渗质量摩尔浓度。通常用合适的有机或无机缓冲液调节药物的pH,例如优选用磷酸盐缓冲液、tris缓冲液(三(羟甲基)氨基甲烷)、HEPES缓冲液([4-(2-羟乙基)哌嗪子基]乙磺酸)或MOPS缓冲液(3-吗啉代-1-丙磺酸)。相应缓冲液的选择通常视所需缓冲液摩尔浓度而定。磷酸盐缓冲液适用于例如注射溶液剂及输注溶液剂。
可用常规方式给予药物,例如借助口服剂型(例如片剂或胶囊剂);借助粘膜(例如鼻腔或口腔);以皮下植入放置形式;借助注射剂、输注剂或凝胶剂,其含有本发明药物。还可表面或局部给予药物,以便治疗上述特定关节病,若合适以脂质体复合物形式。此外,可借助透皮治疗系统(TTS)进行治疗,所述系统使得可在时间上控制药物释放。例如,可从EP0944398A1、EP0916336A1、EP0889723A1或EP0852493A1获知TTS。
若仅将相对少量的溶液剂或混悬剂(例如约1-约20ml)给予身体,则通常使用注射溶液剂。若给予大量溶液剂或混悬剂(例如1升或更多升),则通常使用输注溶液剂。因为与输注溶液剂相比,在注射溶液剂情况下仅给予几毫升,所以在注射时血液或组织液的pH及渗透压的小差异本身并不能被注意到,或仅就疼痛感觉方面在不显著的程度上其本身可被注意到。因此,通常不必在使用前稀释本发明制剂。然而,在给予相对大量的情况下,应该在临给药前将本发明制剂简单稀释到获得至少接近等渗溶液的程度。等渗溶液实例为0.9%浓度的氯化钠溶液。在输注情况下,可例如用无菌水进行稀释,同时可例如经由所谓分流术(bypass)进行给药。
根据本发明不同方面的一个优选实施方案,可作为分离的分子使用组织蛋白酶H、其衍生物或片段。
在本发明情况下,术语“分离的分子”(特别就组织蛋白酶H而言)是指从天然来源纯化的组织蛋白酶H多核苷酸或多肽以及纯化的重组分子(其中术语纯化的包括部分纯化以及完全纯化)。
重组多肽或多核苷酸分子的制备和从细胞或组织纯化天然存在的分子以及细胞或组织提取物的制备为本领域技术人员所熟知(亦参见例如以下所列标准文献)。
它们包括例如基于所公布的基因组或编码多核苷酸序列经由聚合酶链式反应(PCR)来扩增所需长度的多核苷酸,随后将所产生的多核苷酸克隆到宿主细胞中(参见例如以下所列标准文献)。

材料与方法
所用小鼠品系
使用5种不同的近交小鼠品系:AKR/J(AKR)、CBA/J(CBA)、C3H/HeJ(C3H)、C57BL/6J(B6)和C58/J(C58)。从Jackson实验室(BarHarbor,ME,USA)获得小鼠。已证明这些小鼠品系就若干体内疼痛测量值方面而言显著不同(Mogil等1999)。
总RNA分离
用PicoPureTMRNA分离试剂盒(Arcturus)按照制造商的说明书从DRG(背根神经节)分离总RNA。用2100生物分析仪和RNA6000NanoLabChipTM试剂盒(Agilent)评估RNA质量。
AffymetrixGeneChipTM微阵列
按照Baugh,L.R,Hill,A.A.,Brown,E.L.和Hunter,C.P.(2001)NucleicAcidsRes.29,e29用500ng总RNA与100pMT7-(dT)24寡聚体(GGCCAGTGAATTGTAATACGACTCACTATAGGGAGGCGG-dT24SEQIDNO:14),并遵循制造商的说明书用SuperScriptII逆转录酶来进行第一链cDNA的合成。合成双链cDNA,然后用苯酚-氯仿接着乙醇沉淀步骤来提取。用BioArray高产RNA转录标记试剂盒(BioArrayHighYieldRNATranscriptionLabelingkit)(Enzo)遵循制造商说明书用双链cDNA样品进行体外转录反应。在37℃将转录反应物孵育16小时。使用用于RNA清理的RNeasyTMMini试剂盒(Qiagen)方案来纯化cRNA,用分光光度计定量。用RNA片段化缓冲液(200mMTris-乙酸、500mMKOAc、150mMMgOAc、pH8.1)使生物素-标记的cRNA片段化。遵循制造商说明书进行小鼠基因组4302.0GeneChipsTM(Affymetrix)杂交及染色。用GeneChipTM3000扫描仪对微阵列扫描,输入扫描的数据并用Resolverv5.1表达数据分析软件(RosettaBiosoftware)分析。
L5脊椎神经横切和假手术程序
在麻醉小鼠中使左L5脊椎神经暴露,然后部分去除横突(transverseprocess)。在与L4脊椎神经分开后,横切L5脊椎神经。假手术(Shamsurgery)与L5脊椎神经横切手术相同,但不横切L5脊椎神经(参见DeLeo等2000)。
测定缩足阈值
用动态足底触觉测量仪(dynamicplantaraesthesiometer)评估缩足阈值(pawwithdrawalthreshold,PWT)(参见Szabo等2005)。在小鼠适应金属网地板隔离间后,将刺激物放置在动物的后爪下,由电动致动器驱动的直金属丝触及足底面,并且施加逐渐增大的力,直到动物移开脚爪(缩足阈值,PWT)。评估经过手术的同侧后爪和对侧后爪的PWT。每只动物仅使用一次。在所有动物实验中,遵守意识清醒动物的研究伦理指导方针,并由当地伦理委员会批准该程序。
相关性分析
为进行相关性分析,将每一只神经横切动物(Chung动物)的“疼痛表型”定义为C1-S1,其中C1=ln(同侧PWT/对侧PWT),S1=平均值同一品系的所有假手术动物ln(同侧PWT/对侧PWT)。
对每一只Chung动物定义两个差异转录调控的测量值,并且每一测量的基因基于其强度表达数据。对于各个基因及动物,采用“原始强度测量值”作为由Resolver表达数据分析软件(v5.1)计算的强度测量值。对特定基因和Chung动物,计算“log比率测量值”ln(C2/S2),其中C2=Chung表达强度,S2=平均值同一品系的所有假手术动物
假手术表达强度。
在计算相关性之前,过滤基因集合以排除表达低于噪声水平及无显著的Chung对比假手术调节的基因。符合条件的基因必须在至少60%的绝对倍数变化≥1.5的Chung动物中或至少20%的绝对倍数变化≥2.0的Chung动物中被调控。此外,相应的基因表达必须在至少5只由各自强度p-值<0.001定义的动物中为可检测(“存在”)的。
用R软件包(http://www.r-project.org/)来计算每一基因在疼痛表型分数与所定义的转录调节测量值之一之间的Pearson相关系数。基于这些,按照Storey等(2002)的方法产生统计显著性p-值及相应的假发现率(false-discoveryrate,FDR)。认为在“log比率测量值”或“强度测量值”下具有FDR<0.05的基因显著相关。

图1显示每一个体小鼠在L5DRG中其神经性疼痛表型(机械超敏感性,X-轴)和相应组织蛋白酶H的基因调控(log比率(Chung对比假手术对照),Y-轴)。根据所用品系对小鼠数据进行颜色编码。进行Pearson相关性分析,显示疼痛表型和组织蛋白酶H基因调控这两个参数显著正相关。这意味着对于个体小鼠,在经过神经性手术的Chung小鼠L5DRG中组织蛋白酶H表达越高,机械痛觉过敏越显著,其如行为测试所显示。
这种显著的相关性表明组织蛋白酶H基因表达对于诱导神经性疼痛表型的因果关系。

Claims (18)
1.组织蛋白酶H用于鉴定调节神经性疼痛的化合物的用途,其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
2.异源表达组织蛋白酶H的非人转基因动物用于鉴定或分析调节神经性疼痛的化合物的用途,其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关,其中所述用途不是诊断性且不是治疗性。
3.异源表达组织蛋白酶H的非人转基因动物作为用于增强的神经性疼痛敏感性的模型系统的用途,其中所述用途不是诊断性且不是治疗性。
4.非人组织蛋白酶H敲除动物用于鉴定或分析调节神经性疼痛的化合物的用途,其中较低的组织蛋白酶H活性与较低的疼痛敏感性呈正相关,其中所述用途不是诊断性且不是治疗性。
5.非人组织蛋白酶H敲除动物作为用于降低的神经性疼痛敏感性的模型系统的用途,其中所述用途不是诊断性且不是治疗性。

a)提供至少两份样品;
b)将含有组织蛋白酶H的一份样品与化合物接触,
c)在化合物存在下,测定组织蛋白酶H的活性,
d)在化合物不存在下,测定组织蛋白酶H的活性,和
e)比较c)与d)中的组织蛋白酶H的活性,
其中组织蛋白酶H表达与神经性疼痛表型呈正相关。
12.用于鉴定或分析调节和/或预防神经性疼痛的化合物的方法,所述方法包括:
a.将组织蛋白酶H蛋白与测试化合物接触;和
b.测定所述测试化合物是否调节组织蛋白酶H蛋白的活性,
其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
13.用于鉴定或分析调节和/或预防神经性疼痛的化合物的方法,所述方法包括:
a.将具有可检测量或活性的组织蛋白酶H的细胞与测试化合物接触;
b.测定所述测试化合物是否能够调节细胞中存在的组织蛋白酶H的量或活性,
其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
14.用于鉴定或分析调节和/或预防神经性疼痛的化合物的方法,所述方法包括:
a.在转录活性系统中将编码组织蛋白酶H蛋白的核酸与测试化合物接触;和
b.测定在所述化合物存在下在所述系统中存在的编码组织蛋白酶H蛋白的mRNA的量;和
c.测定所述化合物是否能够调节所述系统中存在的编码组织蛋白酶H蛋白的mRNA的量,
其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
15.用于鉴定或分析调节和/或预防神经性疼痛的化合物的方法,所述方法包括:
a)提供用包含可操作地连接到报告基因的功能性组织蛋白酶H基因启动子的核酸载体转染的细胞;
b)提供用包含没有可操作地连接到功能性组织蛋白酶H启动子的报告基因的对照载体转染的细胞;
c)测定在测试化合物存在下a)的细胞和b)的细胞的报告基因活性;
d)测定在测试化合物不存在下a)的细胞和b)的细胞的报告基因活性,
其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
16.用于鉴定或分析调节神经性疼痛的化合物的方法,所述方法包括:
a.选择调节组织蛋白酶H活性的化合物作为测试化合物;和
b.将所述测试化合物给予感觉疼痛的受试者,以测定疼痛是否被调节,
其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关,
其中所述方法不是诊断性且不是治疗性。
17.鉴定或分析在受治疗者中调节和/或预防神经性疼痛的化合物的方法,所述方法包括:
a.测定在一种或多种测试化合物存在下组织蛋白酶H的生物学活性,以鉴定一种或多种调节组织蛋白酶H生物学活性的调节化合物;和
b.测试一种或多种调节化合物降低受试者疼痛、疼痛感觉或疼痛敏感性的能力,
其中所述方法不是诊断性且不是治疗性,
其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
18.用于鉴定调节神经性疼痛的化合物的方法,包括测定组织蛋白酶H的表达,其中所述组织蛋白酶H表达与神经性疼痛表型呈正相关。
