Abstract

本发明公开了白皮松父系鉴定方法,包括如下步骤:(1)提取待测白皮松样品的DNA;(2)使用引物:Pb-F:5’-TGGCATTATGGATTCTCC-3’,Pb-R:5’-AATTCTGCACAGCCTCGTC-3’对DNA样品进行PCR扩增;(3)扩增产物进行测序,将测序结果与SEQ NO:H1-H10的序列进行比对以鉴别其父系。本发明通过对珍稀濒危植物白皮松叶绿体基因检测,能够利用单一基因对不同地理种源的进行有效鉴定,大幅度提高了白皮松父本的检测效率,弥补了现有叶绿体基因标记变异率低,需要的标记量大的不足。

白皮松父系鉴定方法
技术领域
本发明涉及基因检测方法,具体而言涉及白皮松父系鉴定方法。
背景技术
植物的种源鉴定、父本分析是物种保存、种子园设计的重要内容,旨在通过解读种子或个体的可能父本来源,判断产地以及花粉或种子散播方式。现在常规方法是利用大量的分子标记组合分析植物个体的基因型,推断子代可能的父本。在松科植物中主要采用叶绿体微卫星标记。一般认为叶绿体微卫星具有父系遗传、变异率高的特点,能够通过数个甚至十余个基因的组合鉴定个体的遗传特征,排除非亲本个体。白皮松(Pinusbungeana)是我国特产的珍稀濒危植物,因其树形优美而成为园林绿化的优良品种。为保存白皮松种质资源、合理规划种子园、满足亲本识别的要求,高变异叶绿体基因片段的筛选成为当务之急。
现有技术通常利用叶绿体微卫星(cpSSR)高变异的特点,植物个体的DNA通过PCR扩增多个cpSSR位点,然后把所有位点的基因组合在一起当作一个基因型,再寻找与该基因型相一致的基因组合,结合产地植物群体特征,确定种质来源与可能父本。可以看出,现有技术方案很大程度依赖与cpSSR变异的高低。cpSSR变异越高则父本识别效率越高,所需位点越少,成本越低。反之,则效率降低、成本增加。
发明内容
然而我们通过对白皮松叶绿体微卫星的研究表明,其变异率远较其它近缘种低,诸多微卫星位点没有变异,不能够用于种质鉴定与父系分析。因此,如何鉴别白皮松的父系需要采用其它方法来实现。
本发明的目的一方面是提供一种白皮松父系鉴定方法,为了实现本发明的目的,采用如下技术方案:
本发明一方面涉及白皮松父系鉴定方法,包括如下步骤:
(1)提取待测白皮松样品的DNA;
(2)使用引物:Pb-F:5’-TGGCATTATGGATTCTCC-3’,Pb-R:
5’-AATTCTGCACAGCCTCGTC-3’对DNA样品进行PCR扩增;
(3)扩增产物进行测序,将测序结果与SEQ NO:H1-H10的序列进行比对以鉴别其父系。
在本发明的一个优选实施方式中,所述的鉴定方法包括如下步骤:
(1)用CTAB法提取白皮松DNA;
(2)合成引物:Pb-F:5’-TGGCATTATGGATTCTCC-3’,Pb-R:
5’-AATTCTGCACAGCCTCGTC-3’
(3)采用步骤2中的引物对DNA样品进行PCR扩增:
PCR的反应体系(25μL):PCR-Mix 13.5μL;正反向引物各0.1μM;DNA模板50ng,ddH2O不足体积。PCR在MJ Research Inc.生产的PTC-200型热循环仪上进行,反应程序为:94℃预变性4min;94℃变性45sec;50.3℃退火45sec;72℃延伸1min;36个循环,72℃延伸5min;10℃保存;
(4)扩增产物进行测序,将测序结果与H1-H10的序列进行比对以鉴别其父系。
本发明另一方面还涉及SEQ NO:H1-H10或其互补序列在白皮松父系鉴定中的应用。
本发明通过对珍稀濒危植物白皮松叶绿体基因检测,能够利用单一基因对不同地理种源的进行有效鉴定,大幅度提高了白皮松父本的检测效率,弥补了现有叶绿体基因标记变异率低,需要的标记量大的不足。

具体实施方式
实施例1:
采集源于5个不同产地的15个皮松样本,通过如下步骤对其父系进行鉴定:
(1)用CTAB法提取白皮松DNA;
(2)合成引物:Pb-F:5’-TGGCATTATGGATTCTCC-3’,Pb-R:
5’-AATTCTGCACAGCCTCGTC-3’
(3)采用步骤2中的引物对DNA样品进行PCR扩增:

鉴定结果:
鉴定结果如图1所示,例如四川白皮松固定特殊的H2基因,秦岭南坡种源以H5为主,黄土高原群体以H1和H3的组合为主,该标记的高变异性表明其可用于个体识别与父系鉴定。
当理解的是,以上具体实施方式仅仅是举例性说明,对本领域普通技术人员来说,可以根据上述说明加以改进或变换,而所有这些改进和变换都应属于本发明所附权利要求的保护范围。

1.白皮松父系鉴定方法,包括如下步骤:
(1)提取待测白皮松样品的DNA;
(2)使用引物:Pb-F:5’-TGGCATTATGGATTCTCC-3’,Pb-R:5’-AATTCTGCACAGCCTCGTC-3’对DNA样品进行PCR扩增;
(3)扩增产物进行测序,将测序结果与SEQ NO:H1-H10的序列进行比对以鉴别其父系。
2.根据权利要求1所述的白皮松父系鉴定方法,所述的鉴定方法包括如下步骤:
(1)用CTAB法提取白皮松DNA;
(2)合成引物:Pb-F:5’-TGGCATTATGGATTCTCC-3’,Pb-R:5’-AATTCTGCACAGCCTCGTC-3’
(3)采用步骤2中的引物对DNA样品进行PCR扩增:
25μLPCR的反应体系:PCR-Mix13.5μL;正反向引物各0.1μM;DNA模板50ng,ddH20补足体积;PCR在MJ Research Inc.生产的PTC-200型热循环仪上进行,反应程序为:94℃预变性4min;94℃变性45sec;50.3℃退火45sec;72℃延伸1min;36个循环,72℃延伸5min;10℃保存;
(4)扩增产物进行测序,将测序结果与H1-H10的序列进行比对以鉴别其父系。
3.基因序列SEQ NO:H1-H10或其互补序列在白皮松父系鉴定中的应用。
