本发明涉及一种用于农业防毒害的核苷肽衍生物及其制备方法,本发明所述核苷肽衍生物(JSSH一12312),分子量为756.8,分子式为C30H45N8O11PS,本发明还公开了其结构式和制备方法。本发明是在胞嘧啶核苷肽结构中嘧啶氨基位上,通过与含有异硫氰酸酯的磷酸酯化合物进行缩合反应,在胞嘧啶核苷肽结构基础上引入硫脲和α一氨基磷酸酯基团。本发明的有益效果是:原料易得、价廉、适合大规模工业化生产,新型核苷肽衍生物相比胞嘧啶核苷肽有望具有更高活性,低毒,低残留的优点,更优的防治效果以及广谱的抗病毒性。

技术领域
本发明涉及用于农业防毒害的核苷肽衍生物及其制备方法。
背景技术
核苷肽属抗病毒类化合物,是由碱基通过糖苷键与戊糖缩合而成,核糖与碱基一般都是由糖的异头碳与嘧啶的N-1或嘌呤的N-9之间形成的β-N-糖苷键连接。在众多核苷肽化合物中,经发酵得到的胞嘧啶核苷肽型化合物在农业防毒害中有广泛的应用,尤其在防治番茄病毒病、烟草花叶病毒病、辣椒病毒病、大豆根腐病、苹果斑点落叶病、黄瓜白粉病,此外在防治油菜菌核病,其它作物病毒病、茎腐病、蔓枯病、白粉病等多种病害上也有推广应用。此化合物分子量为444[质谱法],分子式为C10H25N7O8,其结构式如下:

发明内容
本发明目的是通过对胞嘧啶核苷肽型化合物进行结构修饰,提供一种用于农业防毒害的核苷肽衍生物及其制备方法。
本发明所指用于农业防毒害的核苷肽衍生物(JSSH-12312),分子量为756.8,分子式为C30H45N8O11PS,化学名称为二异丙基(3-(1-((2R,3R,4S,5S,6S)-6-氨基甲酰-3,4-二羟基-5-((S)-3-羟基-2-(2-(甲氨基)乙酰氨基)丙氨基)四氢-2H-吡喃-2-基)-2-氧-1,2-二氢嘧啶-4-基)硫脲基)(苯基)磷酸甲酯,结构式为:

进一步的技术方案是,上述核苷肽衍生物,熔点为201℃,旋光度为[α]D25+47°。
本发明所指用于农业防毒害的核苷肽衍生物的制备方法,在含有异硫氰酸酯的磷酸酯化合物中,加入胞嘧啶核苷肽进行反应,制得核苷肽衍生物。
进一步的技术方案是,上述含有异硫氰酸酯的磷酸酯化合物包括含有异硫氰酸酯二异丙基的磷酸酯化合物。
进一步的技术方案是,在上述含有异硫氰酸酯的磷酸酯化合物中,加入上述胞嘧啶核苷肽进行缩合反应,制得核苷肽衍生物。
进一步的技术方案是,将上述含有异硫氰酸酯的磷酸酯化合物溶解在四氢呋喃(Tetrahydrofuran简称:THF)中,再加入上述胞嘧啶核苷肽,室温下搅拌过夜,蒸去溶剂,所得混合物经柱层析分离,制得核苷肽衍生物。
本发明是在上述胞嘧啶核苷肽结构中嘧啶氨基位上,通过与上述含有异硫氰酸酯的磷酸酯化合物进行缩合反应,在上述胞嘧啶核苷肽结构基础上引入硫脲和α-氨基磷酸酯基团。
本发明的有益效果是:原料易得、价廉、适合大规模工业化生产,这类新的核苷肽衍生物相比胞嘧啶核苷肽有望具有更高活性,低毒,低残留的优点,更优的防治效果以及广谱的抗病毒性,可用于农业防毒害。

具体实施方式
下面结合实施例对本发明进行更进一步详细说明,但不是对本发明的限定。
实施例
本发明的制备方法之一是将异硫氰酸酯二异丙基的磷酸酯化合物1mmol溶解在5毫升四氢呋喃中,在室温下搅拌,再将胞嘧啶核苷肽1.1mmol分批加入至反应混合物中,在室温下搅拌过夜,蒸去溶剂,所得混合物经柱层析分离,得到产物核苷肽衍生物JSSH-12312,上述核苷肽衍生物JSSH-12312,熔点为201℃,旋光度为[α]D25+47°。
在上述实施例中仅仅选用了异硫氰酸酯二异丙基的磷酸酯化合物作为含有异硫氰酸酯的磷酸酯化合物,本发明还包括其它含有异硫氰酸酯的磷酸酯化合物。
本发明原理是在胞嘧啶核苷肽结构中嘧啶氨基位上,通过与含有异硫氰酸酯的磷酸酯化合物进行缩合反应,在胞嘧啶核苷肽结构基础上引入硫脲和α-氨基磷酸酯基团。
尽管这里参照本发明的解释性实施例对本发明进行了描述,但是,应该理解,本领域技术人员可以设计出很多其他的修改和实施方式,这些修改和实施方式将落在本申请公开的原则范围和精神之内。更具体地说,在本申请公开、附图和权利要求的范围内,可以对主题组合布局的组成部件和/或布局进行多种变型和改进。除了对组成部件和/或布局进行的变型和改进外,对于本领域技术人员来说,本发明的原理也将是明显的。

1.用于农业防毒害的核苷肽衍生物,其特征在于:分子量为756.8,分子式为C30H45N8O11PS,化学名称为二异丙基(3-(1-((2R,3R,4S,5S,6S)-6-氨基甲酰-3,4-二羟基-5-((S)-3-羟基-2-(2-(甲氨基)乙酰氨基)丙氨基)四氢-2H-吡喃-2-基)-2-氧-1,2-二氢嘧啶-4-基)硫脲基)(苯基)磷酸甲酯,结构式为:

所述核苷肽衍生物熔点为201℃,旋光度为[α]D25+47°。
3.权利要求1所述的用于农业防毒害的核苷肽衍生物的制备方法,其特征在于:在含有异硫氰酸酯的磷酸酯化合物中,加入胞嘧啶核苷肽进行反应,制得核苷肽衍生物。
4.权利要求3所述的用于农业防毒害的核苷肽衍生物的制备方法,其特征在于:所述含有异硫氰酸酯的磷酸酯化合物包括含有异硫氰酸酯二异丙基的磷酸酯化合物。
5.根据权利要求4所述的用于农业防毒害的核苷肽衍生物的制备方法,其特征在于:在所述含有异硫氰酸酯的磷酸酯化合物中,加入所述胞嘧啶核苷肽进行缩合反应,制得核苷肽衍生物。
6.根据权利要求5所述的用于农业防毒害的核苷肽衍生物的制备方法,其特征在于:将所述含有异硫氰酸酯的磷酸酯化合物溶解在四氢呋喃中,再加所述胞嘧啶核苷肽,室温下搅拌过夜,蒸去溶剂,所得混合物经柱层析分离,制得核苷肽衍生物。
7.根据权利要求5或6所述的用于农业防毒害的核苷肽衍生物的制备方法,其特征在于:在所述胞嘧啶核苷肽结构中嘧啶氨基位上,通过与所述含有异硫氰酸酯的磷酸酯化合物进行缩合反应,在所述胞嘧啶核苷肽结构基础上引入硫脲和α-氨基磷酸酯基团。
