本发明是有关于一种核酸切割复合物,包括:一纳米粒;一核酸切割试剂;以及一多核苷酸链,是专一性辨识一标的核酸的序列,其具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的序列配对使该多核苷酸链呈发夹式。本发明还提供了有关上述核酸切割复合物的使用方法。
Description
技术领域
本发明是关于一种核酸切割复合物及其使用方法,尤其指一种对标的核酸具有高度专一性辨识的核酸切割复合物及其使用方法。
背景技术
在核酸切割技术发展的初期,化学合成的切割试剂在核酸上作用的切割位置是随机发生而无法操控的,但是随着技术的提升,近年来出现具序列专一性的核酸切割工具如限制酶,故已经可以部份解决上述问题,进而能够在如质体重组、基因转殖、基因图谱分析等医学、生物基因工程技术中大幅应用,以提升相关技术的良率。
然而,前述限制酶对于序列的辨识长度大部分约为4至8个碱基对,但此长度的序列辨识,在一长片断DNA序列中往往出现在多个位置,而无法专一的在特定位置进行切割,因此已经难以负荷日趋提升的医学、生物基因工程技术的需求。因此,目前相当需要能够发展出一种核酸切割工具,得以针对细胞内或外的核酸进行切割,而不会随意攻击非特定的序列,最好可以同时针对多个标的核酸或基因进行专一性切割,且能克服限制酶易受序列、长度限制的缺点,以及需要不同的缓冲液环境等困扰,将更能提升医学、生物基因工程技术。
发明内容
本发明的目的在于提供一种核酸切割复合物。
本发明的又一目的在于提供一种上述核酸切割复合物的使用方法。
为实现上述目的,本发明提供的核酸切割复合物,包括:
一纳米粒;
一核酸切割试剂;以及
一多核苷酸链,为专一性辨识一标的核酸的序列,其具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的部分序列配对使该多核苷酸链呈发夹式。
所述的核酸切割复合物,其中该纳米粒的平均粒径介于1至100纳米的范围。
所述的核酸切割复合物,其中该多核苷酸链的长度介于10至30个核苷酸的范围。
所述的核酸切割复合物,其中该多核苷酸链中该第一端与该第二端的序列配对长度为3至10对碱基对。

提供一核酸切割组成物及一标的核酸,该核酸切割组成物包括一纳米粒、一核酸切割试剂以及一多核苷酸链,其中该多核苷酸链具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的部分序列配对使该多核苷酸链呈发夹式;
将该核酸切割组成物与该标的核酸接触,使该核酸切割组成物的该多核苷酸链专一性辨识该标的核酸的序列;以及
活化该核酸切割组成物的该核酸切割试剂,使其截断该标的核酸的序列。
所述的使用方法,其中该核酸切割试剂为一光激发型核酸切割试剂。
所述的使用方法,其中活化该光激发型核酸切割试剂的方式,是提供可见光、紫外光或近红外光。
所述的使用方法,其中该纳米粒的平均粒径介于1至100纳米的范围。

本发明的核酸切割复合物及其使用方法得以突破以往因为限制酶在较长核酸序列中识别能力不足的问题,提高核酸切割技术应用的实用性,且各种切割序列仅需单一种缓冲液,并且其激发切割能使用光学设计控制同步性、作用位置、进行动态等。并且在使用时可以搭配显影技术追踪核酸切割复合物所在的位置,其应用性相当广泛,本发明的核酸切割复合物及其使用方法具备以下的优点:
(1)可同时针对多个目标基因进行专一性的切割作用;
(2)相对于限制酶酵素比较不易受序列限制;
(3)较小的球体体积使其容易进入细胞及其细胞核进行切割调控;
(4)对于标的核酸可产生永久性的破坏;
(5)可整合影像追踪及光调控切割的机制;
(6)可通过光学系统进行切割位置及切割动能等的精确控制;
(7)经由发夹式设计可大幅提升其专一性及切割效能并减少切割化学物质被降解;
(8)激发光源为生物体较不吸收的近红外光。

具体实施方式
本发明通过活化复合物中的核酸切割试剂,可针对标的核酸序列进行专一性的切割,不会攻击标的核酸序列之外的核酸,故不会损害其它非标的核酸。
本发明的核酸切割复合物包括:一纳米粒;一核酸切割试剂;以及一多核苷酸链(polynucleotide chain),为专一性辨识一标的核酸的序列,其具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的部份序列配对使该多核苷酸链呈发夹式。
本发明上述核酸切割复合物中,该多核苷酸链的长度没有特殊限制,可接近10至30个核苷酸的范围,具体例如15、20、25、35个核苷酸等。此外,多核苷酸链主要包含三个区段,其中第一区段的一端即是连接该纳米粒的第一端,另一端则连接第二区段;第三区段的一端即是连接该核酸切割试剂的第二端,其另一端则连接第二区段,因此连接顺序依次是纳米粒-第一区段-第二区段-第三区段-核酸切割试剂,其中第三区段的序列会与第一区段的部份序列互相配对,故使此多核苷酸链呈发夹式。此外,由于第一区段与第三区段配对的序列长度会影响该多核苷酸链展开进而专一性辨识标的核酸的动作,因此第一区段与第三区段配对的序列长度需要适中,例如3至10对碱基对的配对。举例而言,第一区段的序列长度大致上可为1至25个核苷酸,第二区段的序列长度大致上可为5至80个核苷酸,第三区段的序列长度大致上可为1至25个核苷酸。较佳而言,第一区段的序列长度大致上为6至20个核苷酸,第二区段的序列长度大致上可为15至70个核苷酸,第三区段的序列长度大致上可为6至20个核苷酸。
于此所述的「专一性辨识」一词,意指根据华森-克立克碱基配对(Watson-Crick base pairing)规则、非华森-克立克碱基配对(non-Watson-Crick base pairing)规则(如虎格斯汀碱基配对(Hoogsteen basepairing)、摇摆式碱基配对(wobble base pairing))等规则。本发明的多核苷酸链各个核苷酸中的碱基,是依据虎格斯汀碱基配对(Hoogsteen basepairing)规则或非虎格斯汀碱基配对(Non-Hoogsteen base pairing)规则,与标的核酸相对应位置的各个核苷酸中的碱基配对而生成氢键。举例而言,若本发明的多核苷酸链中具有嘌呤(purines)与嘧啶(pyrimidines)此种碱基时,于标的双股脱氧核醣核酸上与其对应的碱基可为嘌呤如腺嘌呤(adenine)、鸟嘌呤(guanine)及其经修饰后的衍生物等。若本发明的多核苷酸链中只有嘧啶(pyrimidines)此种碱基时,于标的双股脱氧核醣核酸上与其对应的碱基可为嘌呤如腺嘌呤(adenine)与质子化的鸟嘌呤(guanine)。由此可知,若本发明的多核苷酸链无法与某一核酸配对产生氢键,则无法完成此辨识过程,故称为专一性辨识。
此外,由于本发明的多核苷酸链可根据前述配对规则与标的核酸互相配对,因此标的核酸可为单股核酸或双股核酸、核糖核酸或脱氧核糖核酸、天然核酸或人工合成核酸等。具体例如由天然物中纯化、聚合酶连锁反应(polymerase chain reaction,PCR)、化学合成或其它公知方式所获得的单股核醣核酸或双股脱氧核糖核酸皆可为标的核酸。因此,经过本发明多核苷酸链专一性辨识后的标的核酸,可形成出双股螺旋核酸或三股螺旋核酸,若为三股螺旋核酸,多核苷酸链的配对结合主要发生在双股螺旋标的核酸的主沟槽(major groove),且亦表示本发明的多核苷酸链的作用同于具有三股形成能力的寡核苷酸链(triplex-forming oligonucleotide,TFO)。
另一方面,标的核酸的序列长度亦没有特别限制,一般其中被多核苷酸专一性辨识的区域大致的序列长度约可为10至30个核苷酸,较佳约可为11至25个核苷酸。由于本发明的多核苷酸链必须要能专一性辨识标的核酸,因此两者的序列中必须要有部份是互补,即是必须要能更互相配对,如此方能达到辨识的功能。另外,假使多核苷酸链上专一性辨识标的核酸的区块在第一端与第二端的第二区段,由于核酸切割试剂连接在多核苷酸链的第二端的第三区段,因此核酸切割试剂作用的位置则会距离辨识区块数个核苷酸远。由此点特色,可预定切割在标的核酸的位置。
另外,于此所述的「切割」、「截断」等词,意指利用外力或作用使得连续连结的核酸分子上两相邻核苷酸之间产生间隙(gap)或股裂(nick),致使被切割或截断的核酸序列无法达到其原有功能。
本发明的核酸切割复合物中,单一纳米粒上一般可连接30至100条多核苷酸链,且不限于只连接一种多核苷酸链,亦可同时连接其它种多核苷酸链,达到专一性辨识标的核酸其它位置或其它对应的标的核酸的目地。因此,倘若核酸切割复合物中的纳米粒上有多种不同的多核苷酸链时,即可针对单一或多种不同的标的核酸进行专一性辨识,而在核酸切割试剂经过活化后可执行切割标的核酸的动作。
此外,该纳米粒的种类、形状、粒径及材质等没有特别限定,可依应用所需或核酸切割复合物的制备方法而为选择适合的纳米粒。对于纳米粒的材质,举例可为金属,如铟、钛、铜、金、银、铁、钴、铝、钯、铂、锌、锡、铬、镍等;磁性物质,具有顺磁性的物质,如氧化铁、氧化镍等,可经由外加磁场达到吸引或排斥的效果,以利操控或回收本发明的核酸切割复合物;半导体,如硫化镉、硒化镉、经过硫化锌包覆的硒化镉或硫化镉等;无机物质,如硅、二氧化硅等;有机聚合物,如乳酸-甘醇酸共聚物(poly(lactide-co-glycolide),PLGA)、聚乙二醇(polyethylene glycol,PEG)等;或前述材料的组合。对于纳米粒的种类,举例可为纳米棒;等向性纳米粒(isotropic nanoparticle),如实心球体纳米粒或中空球体纳米粒;非等向性纳米粒(anti-isotropic nanoparticle),如非等向性锥形、四边形或菱形纳米粒;高树枝状分子(dendrimer);以及复合型纳米粒,如内核-外壳纳米粒(core-shell nanoparticle,如SiO2Au)。对于纳米粒的平均粒径,一般可接近1至100纳米的范围,较佳约为10至30纳米的范围。
上述核酸切割复合物中,该多核苷酸链与该纳米粒的连接方式没有特别限制。举例而言,该多核苷酸链的该第一端与该纳米粒可由官能基(如硫醚基(thioether))、单体(乙二醇)、聚合物(如聚乙二醇)等连接。另一方面,该核酸切割试剂与该多核苷酸链的连接方式亦没有特别限制。举例而言,可通过修饰该多核苷酸链的该第二端形成胺基,并由此胺基与该核酸切割试剂共价连接。
此外,对于该核酸切割试剂而言,其种类也没有特别限制,举例可为光激发型核酸切割试剂,具体如苯甲醛(hydrazone)、氮基原黄素(azidoproflavine)、迭氮基苯乙酮(azidophenacyl)、迭氮基(azido)衍生物、玫瑰树碱(ellipticine)衍生物、靛花青(Indocyanine Green,ICG)衍生物等,其激发光举例可为可见光(Visible spectrum)、紫外光(UV)或近红外光(Near-IR)。本发明所使用的核酸切割试剂是属于对称型靛花青(carbocyanine)一族的靛花青(ICG)的衍生物-近红外多次甲基菁染料(NIR cyanine dyes,cypate),其激发光源是近红外光。研究指出,生物体内的水、红血球或血色素在800nm附近的吸收相对最小,对生物体有较深的穿透性,可作为热疗法的光源及深部组织非侵入性的监测(Ralph Weissleder.A clearer vision for in vivoimaging.Nature biotechnology 19;316-317(2001))。由此可知,在核酸切割试剂尚未活化时,由于发夹式多核苷酸链的结构使得核酸切割试剂的激发能量被纳米棒表面等离子体共振光学所吸收而产生衰减现象(quenchingeffect),因此无法进行核酸切割动作,只有在发夹式多核苷酸链辨识到特定的序列,展开成直线状时,该核酸切割试剂远离纳米棒表面等离子体共振吸收作用而被激发,经过激发后,才可针对标的核酸进行切割动作。此外,本发明的核酸切割试剂需要与搭配标的核酸的种类来挑选,例如欲切割的标的核酸为脱氧核醣核酸时,则须选择能够切割脱氧核醣核酸的核酸切割试剂。
本发明提供的上述核酸切割复合物的使用方法,核酸切割复合物在缺乏标的核酸时,即使核酸切割试剂经过光照或其它方式活化,但因多核苷酸链依旧维持在发夹式,此时核酸切割试剂与纳米粒的距离相当接近,因此核酸切割试剂的活化能量会被纳米粒的表面等离子体共振光学所吸收而被衰减(quenching),该核酸切割试剂不会外露而随意被激发,故该核酸切割试剂可以安全地暴露在非标的核酸序列中而呈现不活化状态。唯有在发夹式多核苷酸链遇到标的核酸时,由于会与标的核酸进行专一性辨识,因此原本呈发夹式的多核苷酸链会解开而与标的核酸配对,此时核酸切割试剂才会外露,当适合波长的光源激发时,便可执行切割动作达到永久性地抑制标的核酸表现。除此,发夹式多核苷酸链的结构能有效保护光激发的核酸切割试剂在生物体中被降解,强化其专一性与切割效能。
本发明的核酸切割复合物的使用方法,包括以下步骤:提供一核酸切割组成物及一标的核酸,该核酸切割组成物包括一纳米粒、一核酸切割试剂以及一多核苷酸链,其中该多核苷酸链具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的部份序列配对使该多核苷酸链呈发夹式;将该核酸切割组成物与该标的核酸接触,使该核酸切割组成物的该多核苷酸链专一性辨识该标的核酸的序列;以及活化该核酸切割组成物的该核酸切割试剂,使其截断该标的核酸的序列。
举例而言,当本发明的使用方法于试管溶液中进行时,该核酸切割复合物则如同限制酶的功能,可以辨识分布于溶液中的特定序列并达到切割的目的,后续可以结合使用接合酶(ligase),达到基因或质体重组建构。另外,若本发明的使用方法同样于试管细胞中进行时,将该核酸切割复合物分散于陪养基中,其自然(此指可无需电场或高压气体压力等外力)会进入细胞中针对特定辨识序列进行切割。若细胞不含有此特定辨识序列,核酸切割复合物便无法达到细胞特定基因序列标定的作用,如此可以达到如筛选并处理具有特定基因序列突变的癌细胞的目的。由上述可知,本发明的核酸切割复合物及其使用方法,在应用上并无特别的限制,只要有需要进行核酸切割的相关应用则可使用,例如基因图谱分析、体外核酸切割、体外核酸检测、基因或质体重组、亲源鉴定等。此即表示本发明的核酸切割复合物及其使用方法,可以在标的核酸被切割前或切割后与本领域通常技术来搭配、组合、置换使用。
另一方面,本发明的核酸切割复合物也可与医药上可接受的受体组合使用而形成医药组成物。若成为医药组成物,核酸切割组成物的剂量则须为有效量,即对标的核酸具高专一性切割作用所需的剂量,亦即需考虑药物动力学等药学原理与核酸切割复合物在穿过细胞膜时所造成的剂量损失。另外,医药上可接受的受体例如为甘露糖醇(mannitol)、葡萄糖、微晶质纤维素(microcrystalline cellulose)、脱脂奶粉、聚乙烯(polyvinylprrolidone)、淀粉或其组合。医药组成物的投药方式可依情况而定,举例如经喷剂吸入、局部(topically)给予、鼻腔吸收、口服、肠胃外(parentally)投予、药物储槽的殖入、显微注射(microinjection)、基因枪(genegun)转殖或其组合,其中,肠胃外投予可包括经皮下、皮内、静脉内、肌肉内、关节内、动脉内、滑液内或胸骨内的注射或注入方式。
由上述可知,本发明可依照不同标的核酸来进行设计并置换核酸切割复合物中的发夹式多核苷酸链,能符合客制化(customize)的需求,即针对不同的标的核酸制出相对应的核酸切割复合物。而还可由光学方式集中特殊波长的光子于特定部位以局限基因切割的范围。
以下由特定的具体实施例说明本发明的实施方式,熟习此技术的人士可由本说明书所揭示的内容轻易地了解本发明的其它优点与功效。本发明亦可由其它不同的具体实施例加以施行或应用,本说明书中的各项细节亦可基于不同观点与应用,在不悖离本发明的精神下进行各种修饰与变更。
本发明的实施例中的附图均为简化的示意图。惟该些附图标仅显示与本发明有关的组件,其所显示的组件非为实际实施时的态样,其实际实施时的组件数目、形状等比例为一选择性的设计,且其组件布局型态可能更复杂。
实施例一核酸切割复合物的制备
参考图1A至1C,其为本发明核酸切割复合物的制作流程。首先,如图1A所示,提供一纳米粒10及发夹式多核苷酸链20。在本实施例中,纳米粒10为金纳米棒,此金纳米棒的准备方法可依种子间接成长法(又称为植晶法)。0.5mM四氯金酸(HAuCl4)水溶液中加入1.822g溴化十六烷三甲基铵(CTAB)及60μL 100mM硝酸银(AgNO3),充分混合搅拌。在剧烈搅拌中,加入300μL 100mM维他命C(Ascorbic acid),此时Au3+被还原成Au+。保持剧烈搅拌,经过五秒后,立即加入20μL 100mM氢硼酸钠(NaBH4),此时Au+被还原成零价的金原子,成核与长晶开始进行;于两小时后反应完成。硝酸银的银离子(Ag+)与CTAB的溴离子(Br-)形成溴化银(AgBr)固体选择性地沉积吸附在{110}晶格面上并使其长晶速度缓慢,所以产物为纳米金棒而非金球,其长宽比(aspect ratio)为3.5到4(J.P.Juste et al.,Electric-field-directed growth of gold nanorods in aqueous surfactant solutions.Adv.Funct.Mater 14;571-579(2004))最后将制备好的金纳米棒用连续波长400至1100nm扫瞄。
接着,准备发夹式多核苷酸链20,其核酸序列举例可为序列表的SEQID NO.1及SEQ ID NO.2,但序列不限于此,可根据需要来加以设计,其中SEQ ID NO.1的5’端及3’端有5个碱基配对,SEQ ID NO.2的5’端及3’端有6个碱基配对,此部份的碱基配对属于整体多发夹式多核酸链的主干部(Stem),其余未配对的部份则属于主干间的环部(Loop),在发夹式多核苷酸链20的5’端及3’端分别修饰成硫醇基(SH,thiol)及胺基(NH2,amino),且前述官能基与核苷酸之间则以6个碳原子做连接,并由MDbio,Inc.(Taiwan,Taipei)合成。序列的设计可以参考以下文献:Philippe Simon et al.,Targeting DNA with triplex-forming oligonucleotides to modify gene sequence.Biochimie 90:1109-1116(2008)。
由于使用金纳米棒作为本发明的纳米粒10,故可采用本领域已知方法结合金纳米棒及发夹式多核苷酸链20。举例而言,将金纳米棒及发夹式多核苷酸链20以摩尔数比1∶100混合,且将混合液均匀混合16小时,使发夹式多核苷酸链20得由其上的硫醇基氧化成硫醚基后而可与金纳米棒以共价键结连结。以10000rcf的转速离心去掉上清液,去除没结合上金纳米棒的发夹式多核苷酸链再利用1ml的二次去离子重复此步骤,最后以相同的转速离心去除废液,并回溶至0.5ml二次去离子水中。
其后,参考图1B,提供核酸切割试剂30,以便与发夹式多核苷酸链20的另一端3’端连结。对于核酸切割试剂30及多核苷酸链20的的连结方式,可根据本领域已知方法。举例而言,在本实验例中使用Cypate作为核酸切割试剂30,并以125μM的浓度,将Cypate及已与金纳米棒10连接的发夹式多核苷酸链20,以1∶100的反应摩尔数比均匀混合作用16小时,之后以10000rcf的转速离心并去除上清液,以二次去离子水重复此步骤,最后回溶于50ul的二次去离子水中。
最后制得的核酸切割复合物1的结构可如图1C所示,其中Cypate与发夹式多核苷酸链20的连结如图2所示。
实施例二核酸切割复合物的使用方法
参考图3A至3D,其为本发明核酸切割复合物的使用方法。
首先,如图3A所示,提供一核酸切割组成物1及一标的核酸40。此标的核酸40可于试管溶液、细胞中,此核酸切割组成物1包括一纳米粒10、一核酸切割试剂30以及一多核苷酸链30,其中该多核苷酸链20具有一第一端及一相对的第二端,该第一端连接该纳米粒10,该第二端连接该核酸切割试剂30,且该第一端与该第二端的部分序列配对使该多核苷酸链20呈发夹式。
如图3B所示,均匀混合核酸切割组成物1及标的核酸40,使核酸切割组成物1与标的核酸40在溶液或细胞中接触,因此核酸切割组成物1的多核苷酸链20便会专一性辨识标的核酸40的部份序列(如图上的黑色圆点)。由于标的核酸40为双股螺旋结构,故专一性辨识进行时,多核苷酸链20会展开并与标的核酸40上受辨识区域形成三股螺旋结构。
如图3C所示,若使用光激发型核酸切割试剂作为核酸切割试剂30时,则可以选择适合波长的光源活化核酸切割组成物1的核酸切割试剂30,例如本实施例使用近红外光进行活化,此时受到活化的核酸切割试剂30则会呈现激发状态,对标的核酸40的碱基对或邻近的碱基对进行切割作用。
最后,如图3D所示,切割完标的核酸40的切割试剂失去活性而呈现未激发状态,并使标的核酸40失去功能。
另一方面,如图3E所示,其余未与标的核酸40辨识的多核苷酸链30,其所连接的核酸切割试剂30,经过光照后则会自我分解而失去活性。
试验例一试管中的专一性辨识试验
首先,建构出试验载体。以全长的Easy载体,建构出含有发夹式多核苷酸链可辨识的位置,其命名为pST3试验载体。另一方面,设计两组(a,b)可辨识pST3试验载体的发夹式多核苷酸链与两组(c,d)组成相同顺序不同的争夺序列(scramble sequence)的发夹式多核苷酸链。
而后,将前述载体与这四组发夹式多核苷酸链,以1∶1000(图4的栏3,5,7,9)与1∶10000(图4的栏4,6,8,10)的穆尔数比相混合,置于37℃的培养箱反应,最后利用12%非变性之聚丙烯酰胺胶体(polyacrylamide gel),利用电泳移动测定法(Electrophoretic mobility shift assay,EMSA)来观察发夹式多核苷酸链是否可以专一性的辨识载体上相对应的位置。
实验结果如图4所示,其中栏1为分子量标记(marker),栏2为载体,栏3,4与5,6为质体与争夺的发夹式多核苷酸链(c,d)形成的复合物,图4的栏7,8与9,10为质体与可辨识pST3试验载体的发夹式多核苷酸链(a,b)形成的复合物。由图4的栏3,4与5,6可知,因为争夺的发夹式多核苷酸链序列无法辨识质体上的辨识序列,因此不会形成三股。而图4的栏7,8与9,10可知,因为可辨识pST3试验载体的发夹式多核苷酸链可以辨试质体上的辨识序列而形成三股。此实验验证本发明设计的序列是可以专一性的辨识到目标基因的序列。
试验例二试管中核酸切割试剂的DNA切割试验
首先,准备试验例一的pST3试验载体。将前述载体与核酸切割试剂(Cypate),以1∶1000的穆尔数比相混合。图5的栏2表示质体与限制酶酵素作用后,形成的直链质体(Form III plasmid)。图5的栏3,4,7表示质体本身在经过照光(栏3)与不照光(栏4,7)10分钟后都不会因光而被切,所以维持超螺旋的结构(Form I plasmid)。图5的栏5,6表示核酸切割试剂(Cypate)在胶体中的位置。图5的栏8,9表示载体与核酸切割试剂形成混合物后在经过照光(栏8)与不照光(栏9)5分钟后,有照光的质体”部分”被切割,形成缺口圆的质体(Form II plasmid)。图5的栏10,11表示载体与核酸切割试剂形成混合物后在经过照光(栏10)与不照光(栏11)10分钟后,有照光的质体”全部”被切割,均形成缺口圆的质体(Form II plasmid)。由此实验得知,我们合成的核酸切割试剂(Cypate)是可以达到DNA切割的效果。
上述实施例仅为了方便说明而举例而已,本发明所主张的权利范围自应以申请的权利要求范围所述为准,而非仅限于上述实施例。
Claims (18)
1.一种核酸切割复合物,包括:
一纳米粒;
一核酸切割试剂;以及
一多核苷酸链,为专一性辨识一标的核酸的序列,其具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的部分序列配对使该多核苷酸链呈发夹式。
2.如权利要求1所述的核酸切割复合物,其中,该纳米粒的平均粒径介于1至100纳米的范围。
3.如权利要求1所述的核酸切割复合物,其中,该多核苷酸链的长度介于10至30个核苷酸的范围。
4.如权利要求1所述的核酸切割复合物,其中,该多核苷酸链中该第一端与该第二端的序列配对长度为3至10对碱基对。
5.如权利要求1所述的核酸切割复合物,其中,该多核苷酸链的该第一端与该纳米粒以一硫醚基连接。
6.如权利要求1所述的核酸切割复合物,其中,该核酸切割试剂为一光激发型核酸切割试剂。
7.如权利要求6所述的核酸切割复合物,其中,该光激发型核酸切割试剂的激发光为可见光、紫外光或近红外光。
8.如权利要求1所述的核酸切割复合物,其中,该标的核酸为一单股核醣核酸或一双股脱氧核糖核酸。
9.如权利要求1所述的核酸切割复合物,其中,该纳米粒为一金纳米棒或内核-外壳纳米粒。
10.一种核酸切割复合物的使用方法,包括以下步骤:
提供一核酸切割组成物及一标的核酸,该核酸切割组成物包括一纳米粒、一核酸切割试剂以及一多核苷酸链,其中该多核苷酸链具有一第一端及一相对的第二端,该第一端连接该纳米粒,该第二端连接该核酸切割试剂,且该第一端与该第二端的部分序列配对使该多核苷酸链呈发夹式;
将该核酸切割组成物与该标的核酸接触,使该核酸切割组成物的该多核苷酸链专一性辨识该标的核酸的序列;以及
活化该核酸切割组成物的该核酸切割试剂,使其截断该标的核酸的序列。
11.如权利要求10所述的使用方法,其中,该核酸切割试剂为一光激发型核酸切割试剂。
12.如权利要求11所述的使用方法,其中,活化该光激发型核酸切割试剂的方式,是提供可见光、紫外光或近红外光。
13.如权利要求10所述的使用方法,其中,该纳米粒的平均粒径介于1至100纳米的范围。
14.如权利要求10所述的使用方法,其中,该多核苷酸链的长度介于10至30个核苷酸的范围。
15.如权利要求10所述的使用方法,其中,该多核苷酸链中该第一端与该第二端的序列配对长度为3至10对碱基对。
16.如权利要求10所述的使用方法,其中,该多核苷酸链的该第一端与该纳米粒以一硫醚基连接。
17.如权利要求10所述的使用方法,其中,该标的核酸为一单股核醣核酸或一双股脱氧核糖核酸。
18.如权利要求10所述的使用方法,其中,该纳米粒为一金纳米棒或内核-外壳纳米粒。
