Abstract
本发明开发了能够由DAO酶缺损小鼠与其他疾病模型小鼠的交配实验中生产的多个动物中迅速筛选Dao-/-的纯合体、并对多个样品中所含的D-氨基酸迅速进行定量测定的评价方法。本发明提供一种试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞造成的影响的评价方法,所述评价方法包括以下步骤:准备Dao1-/-小鼠等的步骤、将前述Dao1-/-小鼠等的活体组织等暴露于前述试验条件下的步骤、和对将前述Dao1-/-小鼠等的活体组织等暴露于前述试验条件下造成的影响进行分析的步骤。
Description
技术领域
本发明涉及试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞造成的影响的评价方法、用于实施该评价筛选方法的评价系统、和使用该评价系统的医药品和/或化妆品候补物质。
背景技术
除甘氨酸以外的全部氨基酸均存在D型和L型这2种光学异构体。L-氨基酸用于生物的蛋白质合成,蛋白质中所含的氨基酸大部分是L-氨基酸。与此相对,D-氨基酸存在于部分低等生物的生理活性肽中,很多都是经由翻译后修饰的过程而被生物合成。因此,构成蛋白质或肽的氨基酸主要是L-氨基酸,D-氨基酸是例外的存在。
D-氨基酸是细菌的细胞壁的肽聚糖的构成成分。此外,关于不构成肽的游离的D-氨基酸,一直以来报道存在于水栖动物、昆虫等低等动物中。但是,有一段时期相信存在于高等动物中的氨基酸为L型、D型仅微量存在(非专利文献1)。

但是,关于包括人在内的哺乳动物类中的D-氨基酸的存在和其作用,由于近年来的以光学拆分法为首的分析方法的进步而终于得到阐明(非专利文献2)。通过使用了抗D-天门冬氨酸抗体的双重染色法等,从而阐明了D-天门冬氨酸在大鼠脑垂体中局部存在于产生催乳素的细胞中。此外,通过对大鼠脑垂体来源的细胞株中合成、分泌催乳素的细胞给予D-天门冬氨酸,从而催乳素分泌出现用量依赖性增加。根据以上事实可以认为,在产生催乳素的细胞中D-天门冬氨酸控制着催乳素的分泌(非专利文献3)。

另一方面,据报道,在大鼠精巢的静脉中总是检测到比其他静脉血更高浓度的D-天门冬氨酸,而且通过对从大鼠精巢分离、纯化得到的Leydig细胞给予D-天门冬氨酸,睾酮的合成及分泌得到用量依赖性促进(非专利文献4)。

据报道,D-丝氨酸选择性地刺激推测与精神分裂症有关的NMDA受体的甘氨酸结合位点,通过增强介由谷氨酸的该受体发挥的作用,从而促进神经传导(非专利文献5)。实际上报道了通过投与D-丝氨酸来改善精神分裂症,以及精神分裂症患者血清中的D-丝氨酸浓度低于健康人。

作为与皮肤科学相关的认识,有报道称D-天门冬氨酸存在于像晶状体那样的不易发生蛋白质代谢的组织中(非专利文献6)。此外,有报道称D-天门冬氨酸在皮肤中存在于老年受试者的受日晒的皮肤的弹性纤维中,但不存在于没有受日晒的皮肤的弹性纤维中,所以暗示了UV暴露与皮肤的弹性纤维中的D-天门冬氨酸形成是强烈相关的(非专利文献7)。

然而,在探索包括人在内的哺乳类中的D-氨基酸的存在及作用的过程中,较大障碍是D-氨基酸快速分解。在D-氨基酸的分解中,首先,D-氨基酸通过D-氨基酸氧化酶(EC 1.4.3.3,以下称为“DAO酶”。)而发生氧化脱氨基化,转变成对应的α-酮酸。然后α-酮酸通过转氨酶而转变成对应的L-氨基酸。DAO酶是仅特异性地氧化D-氨基酸的酶,在肾脏及其他脏器中表达(非专利文献8)。

DAO酶在小鼠中由第5染色体上的Dao1基因编码,已报道了ddY小鼠中的该基因的错义突变体(非专利文献9)。在该突变基因(Daoc或DaoG181R)中,第181位的Gly残基被置换成Arg残基,产生丧失了酶活性的蛋白质。因此,失去DAO酶活性的表型进行隐性遗传。在隐性纯合体的个体(以下称为“DAO酶缺损小鼠”。)中D-丙氨酸及D-丝氨酸的血中浓度上升至5~8倍,表现出共济失调(ataxia)及定型行为(stereotypic behavior)(非专利文献10)。关于DAO酶缺损小鼠与其他疾病模型小鼠的交配实验未有报道。

发明内容
发明要解决的问题
需要从DAO酶缺损小鼠与其他疾病模型小鼠的交配实验中生产的多个动物中迅速筛选DaoG181R/DaoG181R的纯合体。此外,由于有时D-氨基酸的存在量只有L-氨基酸的存在量的约1%以下,所以需要从大量的L-氨基酸中分离并检测出微量的D-氨基酸。进而,需要迅速地对多个小鼠个体来源的样品、多个不同组织来源的样品、多个暴露于试验条件下的样品中所含的D-氨基酸进行定量测定。
用于解决问题的方案
本发明提供试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞造成的影响的评价方法。前述评价筛选方法包括以下步骤:(1)准备具有Dao1+/+的基因型的小鼠、和具有Dao1-/-的基因型的小鼠的步骤;(2)将前述具有Dao1+/+的基因型的小鼠、和前述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞暴露于前述试验条件下的步骤;和(3)对将前述具有Dao1+/+的基因型的小鼠、和前述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞暴露于前述试验条件下所造成的影响进行分析的步骤。
本发明的评价方法中,前述步骤(1)有时通过Dao1+/+、+/-和/或-/-的基因型的判定方法筛选前述具有Dao1+/+的基因型的小鼠、和前述具有Dao1-/-的基因型的小鼠。前述判定方法有时包括以下步骤:对Dao1+/+、+/-和/或-/-的基因型的小鼠进行个体识别的步骤;从前述经个体识别的小鼠个体中分别提取染色体DNA的步骤;对前述提取的染色体DNA中包含第7外显子的区域进行扩增从而获得扩增DNA片段的步骤;将前述扩增DNA片段用HpaII限制酶进行消化的步骤;以及,对前述扩增DNA片段的限制酶消化产物进行分析的步骤。
本发明的评价方法中获得前述扩增DNA片段的步骤有时包括:使用由序列号1及2中列举的核苷酸序列构成的寡核苷酸引物进行扩增。
本发明的评价方法中,前述具有Dao1+/+的基因型的小鼠与具有Dao1-/-的基因型的小鼠,对于其他基因中的至少1个而言,有时等位基因的组合是相同的。
本发明的评价方法中,前述其他基因的等位基因的组合有时包括Hr-/-。
前述步骤(3)有时包括:在前述步骤(2)之前,测定前述具有Dao1+/+的基因型的小鼠、和前述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞中的D-氨基酸含量;在前述步骤(2)之后,在暴露于前述试验条件下后,测定前述具有Dao1+/+的基因型的小鼠、和前述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞中的D-氨基酸含量;以及,对前述步骤(2)之前测定的D-氨基酸含量、和前述步骤(2)之后测定的D-氨基酸含量进行比较。

本发明的评价方法中,前述D-氨基酸含量有时通过使用识别光学异构体的单克隆抗体的免疫学方法进行测定。
通过本发明的评价方法来评价试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞造成的影响的D-氨基酸有时为D-脯氨酸。
前述测定D-氨基酸含量的活体组织或该活体组织来源的培养组织细胞有时为选自由真皮、表皮、肾脏、胰腺、精巢、肾上腺、小脑、脑垂体及血清组成的组中的1种或2种以上的活体组织或该活体组织来源的培养组织细胞。
通过本发明的评价方法来评价试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞造成的影响的D-氨基酸为D-脯氨酸,该D-脯氨酸有时通过使用光学拆分柱的柱色谱法而测定,前述活体组织为真皮或表皮。
本发明提供用于实施本发明的评价方法的评价系统。前述评价系统包括:具有Dao1+/+及Hr-/-的基因型的小鼠、具有Dao1-/-及Hr-/-的基因型的小鼠、由序列号1及2中列举的核苷酸序列构成的寡核苷酸引物、和识别脯氨酸的光学异构体的光学拆分柱。
本发明提供一种医药品和/或化妆品的候补物质的筛选方法,其特征在于,通过本发明的评价系统来评价医药品和/或化妆品的候补物质。
本说明书中记载为Dao1的基因是称为D-氨基酸氧化酶1的小鼠的基因,美国Jackson研究所的Mouse Genome Informatics Project的主页(http://www.informatics.jax.org/javawi2/servlet/WIFetch?page=markerDetail&key=7803)中公开了详细说明。作为Dao1基因的突变,已知有Dao1G181R(http://www.informatics.jax.org/searches/allele_report.cgi?_Marker_key=7803&int:_Set_key=847160)。本发明中的Dao1-的基因型包括Dao1G181R,但并不限定于此,是指任一D-氨基酸氧化酶1的酶活性缺失突变等位基因的基因型。本发明中的Dao1+的基因型是指如下等位基因的基因型,所述等位基因对于D-氨基酸氧化酶1酶而言,与野生型、即Dao1基因所编码的蛋白质的第181位的氨基酸残基为甘氨酸的酶相同、或者实质上相同,D-氨基酸的分解以与野生型相同或者实质上相同的程度进行。
本发明中的具有Dao1+及-的基因型的小鼠有时具有其他任一基因的等位基因。前述其他基因有时纯合体具有特异性表型,有时杂合体具有特异性表型。因此,本发明中,有时在对于前述其他基因中至少1个基因而言等位基因的组合相同的条件下,对具有Dao1+/+的基因型的小鼠和具有Dao1-/-的基因型的小鼠,进行试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞中的D-氨基酸含量造成的影响的评价。前述试验条件造成的影响有时是指前述试验条件对Dao1基因型的作用的差异造成的影响,所述作用的差异是Dao1基因型对前述其他基因中至少1个基因的等位基因的第1组合的表型性状和该等位基因的第2组合的表型性状的作用的差异。
前述其他基因除了表示疾病模型的表型的情况以外,也可以是参与D-氨基酸的合成或分解的代谢途径的基因、或与老化、免疫、应激反应、营养、运动、感觉、记忆、行动、血液循环、消化、排泄、生殖及其他小鼠的各脏器或全身的健康有关的任意基因。前述其他基因有时是包括但不仅限于下述疾病模型小鼠的原因基因(causal gene):例如肥胖小鼠(Lepob/Lepob)、胸腺依赖性免疫缺陷小鼠(Foxn1nu/Foxn1nu)、快速老化小鼠(Senescence Accelerated Mouse、SAMP1/TaSlc、SAMP6/TaSlc、SAMP8/TaSlc和/或SAMP10/TaSlc)关节炎自然发病小鼠(Laq1MRL/Laq1MRL)及无毛小鼠(Hrhr/Hrhr)。根据约翰斯·霍普金斯大学的Online Inheritance of Man的主页,认为Hr基因的产物是参与体毛形成的转录因子(http://www.ncbi.nlm.nih.gov/entrez/dispomim.cgi?id=602302)。前述Mouse Genome Informatics Project的主页报道了,在小鼠中,除了Hrhr那样的因MLV原病毒的插入导致的自发突变体以外,还有因转基因的插入及其他偶然性制成的突变体和基因敲除小鼠等若干等位基因(http://www.informatics.jax.org/imsr/fetch?page=imsrSummary&op:gsymname=%3D&gsymname=Hr&gsymnameBreadth=C)。本发明中的Hr-的基因型包括Hrhr,但并不限定于此,是指任意的由于Hr基因产物的功能缺损而导致的Hrhr的纯合体的成体中没有体毛的基因型。
本发明中,在前述其他基因中至少1个基因的等位基因的组合相同的条件下,进行试验条件对D-氨基酸含量造成的影响的评价时,在本发明的评价方法中的步骤(1)中,除了Dao1+/+、+/-和/或-/-基因型的判定方法以外,有时还包括对于前述其他基因中至少1个基因的基因型进行判定。
在本发明的评价方法中的步骤(1)通过Dao1+/+、+/-和/或-/-基因型的判定方法进行筛选的情况下,小鼠的个体识别通过对小鼠的饲养笼和/或小鼠的身体进行标记来实施。小鼠的身体的标记可以采用耳朵的打孔、高频及其他标记用芯片在体内的植入及其他本领域技术人员公知的任意方法进行。
本发明的评价方法的步骤(1)中的Dao1+/+、+/-和/或-/-基因型的判定可以按照任意步骤进行。Dao1基因型的判定通过从各小鼠个体中回收少量的活体组织及其他生物学材料,并对其中所含的Dao1基因或其基因产物进行分析来进行。优选的Dao1基因型的判定方法是能够辨别编码Dao1基因产物的第181位的氨基酸残基的Dao1基因序列的方法。从小鼠个体回收染色体DNA、和Dao1基因型的分析可以采用本领域技术人员公知的任意方法。Dao1基因型的分析有时采用PCR(Polymerase Chain Reaction,聚合酶链反应)法、SMAP(Smart Amplification Process)法、LAMP(Loop-mediated isothermal amplification,环介导恒温扩增)法等基因扩增法。Dao1基因的不同基因型的辨别有时通过特定的限制酶对染色体DNA或扩增的DNA有无切断、以及寡核苷酸引物的不同有无引起扩增来进行。前述特定的限制酶的切断的有无和/或扩增的有无可以如下进行:分离扩增的DNA、或者分离限制酶消化后的染色体DNA后,使用能够检测前述特定的限制酶的切断位点有无切断的探针并通过southern印迹法获得与该探针反应的电泳带图案。优选的Dao1基因的不同基因型的辨别是以下说明的基因扩增法。
本发明的评价方法中利用基因扩增法来辨别Dao1基因的不同基因型时,利用了限制酶HpaII识别Dao1基因的突变位点的特点。图1是本发明的Dao1基因型的判定方法相关的核苷酸序列的比对图。图1表示Dao1基因的染色体DNA序列(Dao1_genomic)中从Dao1基因的转录起始点起第15111位碱基与第15460位碱基之间的野生型序列(序列表的序列号4)、包括Dao1G181R突变体中的点突变(从转录起始点起第15223位的鸟嘌呤置换成腺嘌呤。)的第7外显子部分的cDNA的野生型序列(序列表的序列号3的第625位~726位的碱基)、本发明的实施例中使用的基因扩增用引物(正向引物(序列号1)及反向引物(序列号2))、和限制酶HpaII的识别序列的比对结果。另外,序列表中序列号5列举了反向引物序列的反向互补序列(反向引物*)。图2是本发明的Dao1基因型的判定方法中采用的DNA区域的HpaII限制酶图谱(restriction map)。野生型染色体DNA的第7外显子中存在3处HpaII切断位点(向下的箭头)。其中5’最末端侧的HpaII识别序列(CCGG)在Dao1G181R突变型小鼠染色体DNA中变成CCAG,HpaII无法识别(*)。第7外显子区域通过位于第6内含子中的正向引物(Forward)和位于第7内含子中的反向引物(Reverse)而扩增。用HpaII消化野生型小鼠基因组DNA来源的扩增产物(野生型)和前述突变型小鼠基因组DNA来源的扩增产物(Dao1G181R)时,分别在三处及两处被切断。
本发明的评价方法中的D-氨基酸含量的测定可以采用本领域技术人员公知的任意方法来实施。例如,预先用邻苯二甲醛(OPA)、N-叔丁氧羰基-L-半胱氨酸(Boc-L-Cys)及其他修饰试剂将D-及L-氨基酸立体异构特异性地衍生,然后,用ODS-80TsQA那样的分析柱用100mM的醋酸盐缓冲液(pH6.0)和乙腈的混合液进行梯度洗脱并分离的方法,该方法可以用于同时测定天门冬氨酸、丝氨酸及丙氨酸的D型及L型。此外,预先用4-氟-7-硝基-2,1,3-苯并氧杂噁二唑(NBD-F)那样的荧光试剂将D-及L-氨基酸衍生,然后,用ODS-80TsQA、MightysilRP-18GP等那样的分析柱立体异构非特异性地分离各氨基酸后,用Pirkle型手性固定相柱(例如Sumichiral OA-2500S或R)进行光学拆分而进行立体异构特异性分离的方法,该方法可以用于脯氨酸、亮氨酸及其他氨基酸的微量测定(浜濑健司及财津洁、分析化学、53卷、677-690(2004))。本说明书中的光学拆分柱系统是指至少使用了光学拆分柱的分离分析系统,有时也包括利用光学拆分柱以外的分析柱的分离分析。作为其替代,可以通过使用了识别氨基酸的光学异构体的单克隆抗体、例如与D-亮氨酸、D-天门冬氨酸等特异性结合的单克隆抗体的免疫学方法对D-氨基酸进行定量(日本特愿2008-27650说明书)。
本发明中的试验条件是指对待测动物全身或局部进行的物理、化学和/或生物学处理条件。前述物理处理包括:包含紫外线及红外线的光线或电磁波、声音、振动及包括失重的加速度、温度、利用温水或冷水的水浴、干燥或湿润等,但并不限定于这些。化学处理包括氢离子、无机物质和/或有机物质的适用,但并不限定于这些。生物学处理包括:食饵、水等的摄取、明暗周期、笼面积、笼的种类、同一笼中饲养的动物个体数量及其他饲养环境、药物的投与等,但并不限定于这些。本发明中的试验条件有时为前述任一处理或其组合,有时连续和/或间断地实施各处理。
本发明的评价方法中的D-氨基酸含量的测定中,“在前述步骤(2)之后,在暴露于前述试验条件下后,测定前述具有Dao1+/+的基因型的小鼠和前述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞中的D-氨基酸含量”包括:在前述试验条件下的暴露结束后测定D-氨基酸含量的情况;和在前述试验条件下暴露的过程中测定D-氨基酸含量的情况。
根据本发明的评价方法,可以评价前述试验条件对来源于具有Dao1+/+的基因型的小鼠和具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞的各种特性造成的影响。可以评价前述试验条件对下述特性造成的影响:D-氨基酸含量的变动;代谢途径中与D-氨基酸相关的其他物质、例如L-氨基酸、α酮酸等的含量的变动;与这些物质的代谢、消化吸收、分解排泄相关的消化器官、肝脏、肾脏、循环器官等的生理和/或病理特性,以及与包括对花粉、室内尘埃及其他的过敏、特应性皮炎、皮肤及其他脏器的移植免疫在内但并不限定于这些的免疫相关的特性的变动;与感染微生物或与微生物的共存相关的特性的变动;行动、记忆、感觉及其他神经生物学特性的变动;与癌和/或细胞增殖的亢进和/或抑制相关的特性的变动;与皮肤及其他脏器的老化相关的特性,例如与皱纹、脱毛等相关的特性;与保湿性、屏障特性、及其他皮肤的健康和/或美容相关的特性。

具体实施方式
以下,对本发明进行详细说明。本发明的技术范围已通过权利要求书进行了限定,本发明的实施例只是例示。
实施例1
Dao1+/+、+/-和/或-/-基因型的判定方法的开发
Dao1G181R是序列号3中列举的Dao1基因的cDNA的核苷酸序列中第661位的鸟嘌呤置换成腺嘌呤的突变。因此,在野生型中成为限制酶HpaII的切断序列(C↓CGG),与此相对,在Dao1G181R突变体中变成CCAG而不被切断。这里,包含突变位点的第625-726位的核苷酸包含于第7外显子中(GenBank登记号NM_010018.2),所以在染色体DNA中也能够通过HpaII切断位点的有无来识别野生型或突变型。
对断乳后的小鼠进行个体识别后,用市售的哺乳类基因组DNA小量提取试剂盒(Miniprep Kit)(Sigma、G1N70-1 KT)从各小鼠个体的尾巴中提取染色体DNA并进行纯化。作为正向引物,使用位于第6内含子中的由序列号1的核苷酸序列构成的寡核苷酸,作为反向引物,使用位于第7内含子中的由序列号2的核苷酸序列构成的寡核苷酸。用市售的反应混合液(PromegaKK.、M7122),在(1)94℃、4分钟、1次、(2)94℃、30秒钟、55℃、30秒钟、72℃、30秒钟、40次、(3)72℃、10分钟、1次、(4)4℃、保存的热循环设定下对小鼠染色体DNA进行扩增。对于PCR反应产物,用市售的试剂盒(QIAGEN、28104)进行纯化,在HpaII(TOYOBO、HPA201)存在下,在37℃下孵育3小时,进行限制酶处理。通过70℃、5分钟的加热使限制酶失活后,通过市售的电泳微芯片(安捷伦、2100 Bioanalyzer)分析DNA片段的长度。
图3-1是由已根据Dao1酶活性确认了的Dao1+/+纯合体小鼠(第1泳道)、Dao1G181R/G181R纯合体小鼠(第4泳道)及Dao1+/G181RF1代杂合体小鼠(第2及3泳道)的染色体DNA来源的扩增产物的HpaII分解片段的电泳图案。由图3-1可知,由Dao1+基因的染色体DNA获得95bp的DNA片段,由Dao1G181R基因的染色体DNA获得107bp的DNA片段,所以野生型纯合体、突变型纯合体及杂合体被清楚辨别。图3-2是对无毛小鼠(Hrhr/Hrhr、Dao1+/+)与Dao1酶缺损小鼠(Hr+/+、Dao1G181R/G181R)交配的F2代中具有无毛表型的12只F2代的小鼠个体进行Dao1基因判定的结果。表1是对具有无毛表型的138只F2代的小鼠个体的Dao1基因型及性别进行整理得到的表。

如上所述,根据本判定方法,能够快速判定多个小鼠个体的Dao1基因型。
实施例2
脯氨酸及4-羟基脯氨酸的光学异构体的定量分析方法的开发
皮肤胶原中含有很多脯氨酸及4-羟基脯氨酸。因此,开发了对于脯氨酸及4-羟基脯氨酸这两者能够同时分离全部光学异构体并进行定量分析的方法。
图4-1表示脯氨酸及4-羟基脯氨酸的光学异构体。脯氨酸的光学异构体只有L-型和D-型这2种,但4-羟基脯氨酸的光学异构体除了L-型和D-型的区别以外,还有反式和顺式的区别,总计有4种。首先,用荧光试剂NBD-F将氨基酸衍生而进行荧光标记。然后,如图4-2所示,用第1柱进行反相分离色谱,检测反式-4-羟基脯氨酸、顺式-4-羟基脯氨酸及脯氨酸的各峰。然后,通过柱转换阀收集前述各峰的级分,并导入到第2柱中进行光学拆分色谱。
图5的1D的波形图是对使用了流速40μL/分钟的溶剂的整体(monolithic)ODS柱的洗脱图谱照射470nm的激发光以530nm的荧光发光进行检测得到的图。2D的波形图是自动检测1D的洗脱图谱的反式-4-羟基脯氨酸、顺式-4-羟基脯氨酸及脯氨酸的各峰并进行阀切换而仅将各峰的级分导入到QN-2-AX柱中进行光学拆分色谱得到的结果。
图6-1的波形图是Dao1基因的野生型纯合体小鼠(Dao1+/+)血清中的L-型及D-型脯氨酸的光学拆分柱的洗脱图谱,图6-2的波形图是Dao1酶活性缺损型纯合体小鼠(Dao1G181R/G181R)血清中的L-型及D-型脯氨酸的光学拆分柱的洗脱图谱。D-脯氨酸在Dao1野生型纯合体小鼠中基本未检测到,但在Dao1酶活性缺损型纯合体小鼠中被明显检测到。
图7-1是实施例1中辨别的无毛小鼠与Dao1酶活性缺损小鼠交配的F2代的无毛小鼠中基因型被判定为Dao1+/+及Dao1G181R/G181R的个体的各种脏器中的D-脯氨酸存在量的比较结果,图7-2是真皮中的D-脯氨酸存在量的比较结果。关于图7-1的图表的纵轴的单位,血清为pmol/μL,其他组织为pmol/mg。图7-2的图表的纵轴的单位为nmol/μgDNA。均表示各5只小鼠的脏器的测定值的平均和标准误差(SE)。除血清以外,在脑垂体、肾上腺、胰腺及真皮中,在Dao1酶活性缺损小鼠的情况下,也存在比野生型小鼠多数倍以上的D-脯氨酸。此外,在具有Dao1酶活性的野生型小鼠的小脑、肾脏及肝脏中D-脯氨酸几乎未被检测到,但在Dao1酶活性缺损小鼠中D-脯氨酸被明显检测到。Dao1酶活性缺损小鼠的精巢中的D-脯氨酸量与野生型小鼠相比仅多少许,没有明显差别。此次首次阐明了在皮肤中Dao1酶活性缺损小鼠中D-氨基酸含量也多于野生型。从此明确了紫外线照射、老化对皮肤中的D-氨基酸含量的影响。另外,在这次的分析中,关于D-4-羟基脯氨酸的含量,顺式-异构体及反式-异构体在全部组织中均为检测限以下。
实施例3
Dao1酶缺损小鼠中的肿瘤增殖
用添加有10%胎牛血清(Irvine Scientific、批次#300A80601)的DMEM(Sigma)培养基,在5%CO2、37℃的加湿条件下培养Swiss Webster Sarcoma 180株的肉瘤细胞。调制1x107个/mL的悬浮液,通过皮内注射各移植0.05mL到Dao1酶缺损小鼠或野生型小鼠的右后肢足肉垫中。移植后每周用游标卡尺测量肿瘤的长径、短径及厚度,通过以下的式子计算出肿瘤体积。
肿瘤体积(mm3)=长径(mm)×短径(mm)×(厚度(mm)-3)
这里厚度是将原来的足的厚度设为3mm并以与该3mm的差作为肿瘤的厚度。
结果如图8所示。图8是对Dao1酶缺损小鼠和对照小鼠移植后经过1、2及3周时的肿瘤体积的变化进行比较的柱形图。对照小鼠中移植后1周肿瘤体积已经为60mm3,3周增加至2倍,与此相对,在Dao1酶缺损小鼠中,移植后1周肿瘤体积仅为约20mm3,2周时减少至5mm3左右,3周时完全消失。该结果表明,在Dao1酶缺损小鼠具有抑制移植后的肿瘤的增殖并使肿瘤消失的活性。
这次建立的无毛小鼠与Dao1酶缺损小鼠的交配实验体系,可以期待今后广泛用于阐明D-氨基酸对红斑狼疮、硬皮病、皮肌炎、干燥综合征、结节性多动脉炎、白塞氏病、风湿性关节炎等胶原病或胶原病的近缘性疾病、糖尿病、日光性皮炎、接触性皮炎、褥疮、老人斑、暗淡、皱纹、松弛等其他皮肤外观问题所发挥的作用。
Claims (13)
1.一种试验条件对小鼠的活体组织或该活体组织来源的培养组织细胞造成的影响的评价方法,其特征在于,包括以下步骤:
(1)准备具有Dao1+/+的基因型的小鼠和具有Dao1-/-的基因型的小鼠的步骤、
(2)将所述具有Dao1+/+的基因型的小鼠和所述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞暴露于试验条件下的步骤、和
(3)对将所述具有Dao1+/+的基因型的小鼠和所述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞暴露于所述试验条件下造成的影响进行分析的步骤。
2.根据权利要求1所述的评价方法,其特征在于,所述步骤(1)包括通过Dao1+/+、+/-和/或-/-基因型的判定方法来筛选所述具有Dao1+/+的基因型的小鼠和所述具有Dao1-/-的基因型的小鼠,所述判定方法包括以下步骤:对Dao1+/+、+/-和/或-/-的基因型的小鼠进行个体识别的步骤、从所述经个体识别的各个小鼠个体提取染色体DNA的步骤、对所述提取的染色体DNA中包含第7外显子的区域进行扩增从而获得扩增DNA片段的步骤、将所述扩增DNA片段用HpaII限制酶进行消化的步骤、和对所述扩增DNA片段的限制酶消化产物进行分析的步骤。
3.根据权利要求2所述的评价方法,其特征在于,所述获得扩增DNA片段的步骤包括使用由序列号1及2中列举的核苷酸序列构成的寡核苷酸引物进行扩增。
4.根据权利要求1~3中任一项所述的评价方法,其特征在于,所述具有Dao1+/+的基因型的小鼠和具有Dao1-/-的基因型的小鼠,对于其他基因中至少1个基因而言,等位基因的组合是相同的。
5.根据权利要求4所述的评价方法,其特征在于,所述其他基因的等位基因的组合包括Hr-/-。
6.根据权利要求1~5中任一项所述的评价方法,所述步骤(3)包括:在所述步骤(2)之前,测定所述具有Dao1+/+的基因型的小鼠和所述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞中的D-氨基酸含量;在所述步骤(2)之后,在暴露于所述试验条件下后,测定所述具有Dao1+/+的基因型的小鼠和所述具有Dao1-/-的基因型的小鼠的活体组织或该活体组织来源的培养组织细胞中的D-氨基酸含量;以及,对在所述步骤(2)之前测定的D-氨基酸含量和所述步骤(2)之后测定的D-氨基酸含量进行比较。
7.根据权利要求6所述的评价方法,其特征在于,所述D-氨基酸含量通过使用光学拆分柱系统的柱色谱法进行测定。
8.根据权利要求6所述的评价方法,其特征在于,所述D-氨基酸含量通过使用识别光学异构体的单克隆抗体的免疫学方法进行测定。
9.根据权利要求6~8中任一项所述的评价方法,其特征在于,所述D-氨基酸为D-脯氨酸。
10.根据权利要求6~9中任一项所述的评价方法,其特征在于,所述测定D-氨基酸含量的活体组织或该活体组织来源的培养组织细胞是选自由真皮、表皮、肾脏、胰腺、精巢、肾上腺、小脑、脑垂体及血清构成的组中的1种或2种以上活体组织或该活体组织来源的培养组织细胞。
11.根据权利要求10所述的评价方法,其特征在于,所述D-氨基酸为D-脯氨酸,该D-脯氨酸通过使用光学拆分柱的柱色谱法来测定,所述活体组织为真皮或表皮。
12.一种用于实施权利要求11所述的评价方法的评价系统,其特征在于,其包括:具有Dao1+/+及Hr-/-的基因型的小鼠、具有Dao1-/-及Hr-/-的基因型的小鼠、由序列号1及2中列举的核苷酸序列构成的寡核苷酸引物、和识别脯氨酸的光学异构体的光学拆分柱系统。
13.一种医药品和/或化妆品的候补物质的筛选方法,其特征在于,通过权利要求12所述的评价系统来评价医药品和/或化妆品的候补物质。
