Abstract

一种壳聚糖-酪朊酸钠共混膜的制备方法,是以壳聚糖、酪朊酸钠作为膜材料,通过控制酪朊酸钠的含量制备不同性能的壳聚糖-酪朊酸钠共混膜。酪朊酸钠的引入对壳聚糖膜的力学强度、吸水率、水蒸汽透过率有明显的改善。该共混膜可以用作分离膜,亦可用作缓释膜和药物的包裹膜。

壳聚糖-酪朊酸钠共混膜的制备方法
技术领域
本发明涉及一种以壳聚糖、酪朊酸钠共混膜的制备方法。
背景技术
酪朊酸钠(sodium casetnate,简称EC),又称为酪蛋白酸钠、酪蛋白钠、干酪素钠,是酪蛋白的一种衍生物。酪朊酸钠具有很好的乳化、热稳定、增稠、起泡、保湿、成膜等特性,易被人体吸收。根据GB2760-1996规定,酪蛋白酸钠可用于各类食品,并按生产需要适量使用。由于酪朊酸钠从牛乳分离制得,含有人体所需的全部必需氨基酸,营养价值高,除了具有良好的乳化、增稠作用外,还可用作蛋白质的增强剂,被广泛用于蛋白饮料、肉制品、麦片、奶粉、果冻等食品以及营养保健品、特殊病人食品、方便食品、药用辅料、生物制品、饲料等方面,近年来日益受到食品加工企业的高度重视。
壳聚糖作为性能优良的天然高分子材料,其独特的分子结构,具备良好的抑菌抗菌性、吸湿保湿性、气体选择渗透性、阻湿性、可食性、生物可解降解性等特性,可制成超滤膜、渗透膜、食品保鲜膜等应用于食品、医药等诸多领域,从而使得壳聚糖膜工业具有巨大的应用价值。但由于单纯的壳聚糖膜在各方面性能上的不足,又制约了它的应用。因此有人将壳聚糖进行改性或加入其他成分制成共混膜,从而改良壳聚糖膜的性能,研究表明:壳聚糖与醋酸纤维共混制成渗透蒸发膜,其性能优于壳聚糖膜,可用来分离50%~95%的乙醇-水溶液。壳聚糖还可与聚乙烯醇共混制成渗透膜,可以保证高的分离因子,又具有高的渗透通量。壳聚糖与对羟基苯甲酸乙酯制成的涂膜剂有较好的保鲜效果,可明显抑制其失重和纤维素含量的增加。由此可见,通过改良壳聚糖的成膜性能,对提高壳聚糖膜的应用范围有着重要的意义。
发明内容
本发明的目的在于提供一种可用作分离膜、缓释膜和药物包裹膜的共混膜的制备方法。由于酪朊酸钠分子因缺乏二级结构、其肽链呈任意卷曲,容易形成分子间氢键、静电引力、疏水键等而成膜,因此该共混膜可由壳聚糖溶液和酪朊酸钠按不同比例通过共混交联制得。
本发明的制备方法如下:
常温常压下,将5g壳聚糖溶于100ml、1%稀醋酸溶液中,配成溶液,将1g酪朊酸钠溶于20ml纯净水中。将上述两种溶液按一定比例混合,用机械搅拌法使之完全共混,将共混液过滤、减压脱泡后在玻璃板上流延成膜,于烘箱中25~35℃烘干。将上述玻璃板置于1.5~2.5%的NaOH溶液中浸泡至膜自然脱落,取出膜,用蒸馏水充分洗涤至中性,再将其固定于有机玻璃板上,置烘箱中烘干,取下,即得共混膜。
本发明有如下优点:
1、与壳聚糖-聚乙烯醇共混膜相比,壳聚糖-酪朊酸钠共混膜的力学强度明显上升。
2、随酪朊酸钠的加入量的不同,在一定范围内改善壳聚糖膜的抗水性能。
3、改善壳聚糖膜的水蒸汽透过率性能。
具体实施方式
本发明以壳聚糖和酪朊酸钠作为膜材料,根据共混膜中壳聚糖的质量分数100%,95%,90%,85%制备成A、B、C、D四种不同比例的壳聚糖-酪朊酸钠共混膜,用生物显微镜观察不同比例制备的壳聚糖/酪朊酸钠共混膜表面的结构和形态;测定共混膜力学性能,吸水率、透光率和水蒸汽透过率等技术指标进行比较。
结果表明,酪朊酸钠的引入对壳聚糖膜的力学强度、吸水率、水蒸汽透过率有明显的改善。力学强度:A<B<C<D,吸水率:A>B>C,D>B,水蒸汽透过率:A>B,C>D,因此可根据实际应用的需求,通过控制酪朊酸钠的含量制备不同性能的壳聚糖-酪朊酸钠共混膜。
实施例1
常温常压下,将5g壳聚糖溶于100ml、1%稀醋酸溶液中,配成溶液,将1g酪朊酸钠溶于20ml纯净水中。取90ml上述壳聚糖溶液及10ml上述酪朊酸钠溶液混合,用机械搅拌法使之完全共混,将共混液过滤、减压脱泡后在玻璃板上流延成膜,于烘箱中25~35℃烘干。将上述玻璃板置于1.5~2.5%的NaOH溶液中浸泡至膜自然脱落,取出膜,用蒸馏水充分洗涤至中性,再将其固定于有机玻璃板上,置烘箱中烘干,取下,即得壳聚糖含量为90%共混膜。测得该共混膜的抗张强度为:6184×10N/cm,吸水率为:71.9%,水蒸汽透过率第1天为:0.034%,第2天为:0.026%,第3天为0.022%。

Claims (2)

1.一种壳聚糖-酪朊酸钠共混膜的制备方法,其特征是在常温常压下,将5g壳聚糖溶于100ml、1%稀醋酸溶液中,配成溶液,将1g酪朊酸钠溶于20ml纯净水中,将上述两种溶液混合,用机械搅拌法使之完全共混,将共混液过滤、减压脱泡后在玻璃板上流延成膜,于烘箱中25~35℃烘干,将上述玻璃板置于1.5~2.5%的NaOH溶液中浸泡至膜自然脱落,取出膜,用蒸馏水充分洗涤至中性,再将其固定于有机玻璃板上,置烘箱中烘干,取下,即得共混膜。
2.根据权利要求1所述的壳聚糖-酪朊酸钠共混膜的制备方法,其特征在于,可以用作分离膜,亦可用作缓释膜和药物的包裹膜。
