Abstract

本发明涉及生物技术领域动物细胞培养用天然血清替代物,其特征是:提取物来源于禽蛋、其中包括新鲜禽蛋,和未成形胚胎;家畜胎盘以及驯化昆虫幼虫,三种提取物可单独应用也可混合应用。本发明所涉及的天然血清替代物来源广泛、成本低廉,适合常规细胞培养尤其是在工业发酵中应用。

技术领域
本发明是生物技术领域的一种细胞培养用血清替代物。
背景技术
自上世纪初细胞培养基出现以来,细胞培养技术已经成为生物技术和生物工程领域不可或缺的基础性技术,同时对培养基的要求也在不断提高。目前的培养基主要由基础培养基和添加剂构成,基础培养基主要由平衡盐、蛋白质水解物、维生素和能量成分构成;添加剂主要是各种动物血清(司徒振强,吴军正细胞培养世界图书出版公司北京2004;44-61)。
细胞学研究早期人们也曾经采用组织浸出液进行细胞培养,由于组织中含有细胞生长抑制因子,效果并不稳定,而血清中含有抗体和补体等免疫分子会对细胞构成影响,而且血清蛋白质成分负责和目标蛋白可能存在交叉,而且受到人畜共患病的影响,人们不断在摸索各种血清的替代产品,比如各种细胞因子的组合,或者利用植物源生物活性物质,但是这些技术要么过于昂贵、要么细胞生长效率较低,难以完全替代血清产品。因此我们希望开发一种新的血清替代产品,并结合对基础培养成分的调整建立一套新的细胞培养体系,目前国内和国际上尚未发现类似的报道。
发明内容
本发明涉及生物技术领域动物细胞培养用天然血清替代物,其特征是:提取物来源于禽蛋、其中包括新鲜禽蛋,和未成形胚胎;家畜胎盘以及驯化昆虫幼虫,三种提取物可单独应用也可混合应用。
本发明所涉及的天然血清替代物中,禽蛋提取物提取自禽蛋卵黄或者未成形胚胎,其中首选鸡卵卵黄或者未成形胚胎;家畜胎盘首选但不限于牛羊胎盘;驯化昆虫幼虫包括蝇蛆,黄粉虫,菜青虫,家蚕,首选但不限于无菌蝇蛆。
实施实例
1、受精禽蛋于30到42摄氏度孵育0到72小时,优选但不限于24到48小时。无菌提取淡黄或者胚胎,加入适量预冷平衡盐溶液制备匀浆混匀。
2、动物胎盘无菌取出后,立刻加入适量预冷平衡盐溶液制备匀浆;或者立刻加入液氮或者干冰中冻结,粉碎加入适量预冷平衡盐溶液混匀
3、化蛹前期昆虫幼虫立刻加入适量预冷平衡盐溶液制备匀浆,或者压榨后加入适量预冷平衡盐溶液制备匀浆。
4、后期处理方式
低温高速离心,取出提取液中组织碎片、颗粒物和脂类物质。
微孔滤膜过滤除菌
以标准血清为参照测定提取物相对生长活性
本发明所涉及的血清代用品来源广泛,成本低廉,利用自制基础培养成分进一步降低了成本,培养适合工程细胞尤其是昆虫细胞的工业化培养,具有一定的经济价值、实用价值和社会价值。
具体实施实例
下面举例说明本发明所涉及培养基的配置过程,其内容不构成对本发明所要求各项权利的限制。
实施实例1
1、鸡受精卵37摄氏度孵育48小时,取出卵黄。
2、加入等体积预冷D-Hinkers溶液,低温匀浆
3、低温4000g到6000g离心10分钟,匀浆分为三层,提取中层清液
4、0.22微米微孔滤膜正压过滤,收集滤液,低温保存或冻干保存,滤液既为添加剂。
5、将滤液按照5%、10%、20%、30%的体积比例添加到RPMI1640培养基中混合,并以添加10%标准胎牛血清的RPMI1640培养基作为参照,以Hela细胞作为检验对象,测定添加剂活性和添加比例。
实施实例2
1、无菌取出牛胎盘,加入适量生理盐水冲洗后备用。也但冷冻后低温粉碎。
2、加入等体积预冷D-Hinkers溶液,低温匀浆
3、低温4000g到6000g离心10分钟,匀浆分为三层,提取中层清液
4、0.22微米微孔滤膜正压过滤,收集滤液,低温保存或冻干保存,滤液既为添加剂。
5、将滤液按照5%、10%、20%、30%的体积比例添加到RPMI1640培养基中混合,并以添加10%标准胎牛血清的RPMI1640培养基作为参照,以Hela细胞作为检验对象,测定添加剂活性和添加比例。

1、收集生长正常的无菌蝇蛆,加入适量生理盐水冲洗后备用。
