Abstract

技术领域
本发明涉及一种酶耐温性能的检测装置和方法,同时涉及一种用于实现蒸汽冲击下酶活鉴别方法的检测装置。
背景技术
酶是由各种微生物产生的,具有各自不同的特性(如底物的亲和性,最佳温度和pH值)。为了检测某种酶的活性,需要找到一种酶的分析方法,该方法能够测定不同来源的该种酶的活性,但需要对测定条件进行修改。
饲料工业中所使用的酶最主要是木聚糖酶,β-葡聚糖酶和植酸酶。其它酶(α-淀粉酶、α-半乳糖昔酶、纤维二糖酶、纤维素酶、果胶酶、脂肪酸和蛋白酶)使用较少。
饲料酶活性的检测方法没有统一的标准,而且酶活性的体外分析法并不能用来评价酶在体内的作用。酶活的分析方法主要是用于产品的质量的控制,说明目前使用的产品质量情况。应该在酶的体外和体内评定法之间建立某种联系,以便能够通过体外的方法来预测酶在体内发挥的作用。但到目前为止,酶产品中还没有一个酶的生产和销售厂家能够提出一种预测酶在体内的功能的方法。
饲料酶的耐热稳定性都较差,特别是湿热下,如饲料颗粒料的生产过程中用蒸汽进行调质(85~90℃)及挤压时,粉酶酶活几乎完全失活。
储存稳定性对于某些酶产品是必须考虑的。有些霉菌产纤维素酶的能力很强,但是产品的储存稳定性很差。即使在冬季(环境温度不超过15℃)存放3个月,活力损失也要超过50.0%。显然,这种情况无论是对于生产企业还是用户都要引起重视。
在申请号为95193609.3的发明专利中,公开了一种检测分枝杆菌属细菌存在的方法以及其所用的试剂盒和抗体,它是关于检测和鉴别存在于人和动物的生物样品中的分枝杆菌属和细胞的一种诊断测定法。该测定法是基于用免疫学方法检测来源于分枝杆菌属细菌的一种或多种抗原。为了检测抗体抗原反映,可以用酶或荧光染料标记这些特异性抗体,或者使抗体吸附于乳胶颗粒或其他合适的标记物。可以用ELISA进行诊断测定,在实际测定之前,需要或者不需要预行浓缩分枝杆菌属细菌抗原。
在专利号为200410053801.0的发明专利中,公开一种溶菌酶检测试剂及其制备方法。该发明溶菌酶检测试剂含有灭活微球菌和明胶。该检测试剂采用新的灭活微球菌的方法,同时利用明胶作为分散菌体的介质和稳定剂,制备高度稳定的菌悬液作为溶菌酶检测试剂。可用于各种检测需要,能用于全自动生化分析,实现溶菌酶活性的批量、自动化检测。
在专利号为88108455.7的发明专利种,公开了一种免疫检测方法,包括使用一种或更好是两种酶放大步骤以提高检测速度或灵敏度。在第一步中,以与样品中分析物浓度相关的量产生第一酶。在第二步中,第一酶作用于第一底物以暴露第一产物的抗原决定基。在第三步中,使第一产物与抗体反应。测定此免疫反应的程度,最好是通过酶信号系统进行测定。描述了用β-半乳糖苷酶作为第一酶检测异羟基毛地黄毒苷的两种方法和检测17α-羟基黄体酮的一种方法。
在专利号为02817900.5的发明专利中,公开了细胞内蛋白酶检测系统。在一个实施方案中,该系统包括嵌合蛋白质,其包括共价连接成分:1)至少一种可选地掩蔽信号蛋白质;2)至少一种蛋白酶特异的切割位点;3)至少一种可检测的氨基酸序列。该发明有广谱的应用,包括用于细胞和组织内新蛋白酶抑制剂的检测。
目前,饲料颗粒料占饲料总量的70%,由于制粒过程中的高温阶段,严重影响了饲料酶的作用效果。如饲料酶在添加前能判断其能否耐温,将极大地提高饲料酶的应用,故急需开发饲料酶耐温性检测装置。
发明内容
本发明的目的是提供一种酶耐温性能的鉴别方法,用于实现蒸汽冲击后,快速比较酶活保留的判别装置和判别方法,适用于各种需要判别耐温性能的酶。
一种蒸汽冲击下酶活检测装置,该装置包括蒸汽发生室、样品池、蒸汽冲击室、比色系统四部分,所述的蒸汽发生室包括一容器(1)、加热装置(2)、所述的样品池(3)、筛网(4)、蒸汽冲击室包括孔板(5)、套管(6)、比色系统包括一测试片(塑料瓶)(7)、试管(8)、比色架(9)、所述的孔板(5)放置在蒸汽发生室的容器(1)上,套管(6)放在孔板上,样品池放在孔板上的套管内。
2、孔板(5)为单孔或多孔或筛网,主要作用为支撑样品池。
3、样品池为蒸笼形,其中筛网选用40目以上。
4、样品池为蒸笼形,其中筛网选优选60目以上。
5、套管为管底有单孔或多孔,该孔可调节大小或关闭。通过调节孔大小来调节套管内的温度。
6、将样品置于样品池中,铺平后置于孔板上对准孔眼,让蒸汽冲击样品。
6、比色系统,包括测试片1,测试片2,试管。比色过程为:①将测试片1加入试管,加纯净水溶解;②将测试样品置于样品池中,经蒸汽冲击后放入含测试片1的试管中溶解;③加入测试片2,进行比色,比较蒸汽冲击前后及冲击时间长短对酶活的影响。

实施例
取耐高温包被微丸酶样品、粉酶样品,分别置于样品池中,铺平后将样品池置于孔板上,对准孔眼放置,然后置套管于孔板上,调节套管下端的孔大小来调节套管内的温度,控制温度为90-93℃。
样品经蒸汽冲击后,进行比色:①将测试片1加入试管,加纯净水溶解;②将测试样品置于样品池中,经蒸汽冲击后放入含测试片1的试管中溶解;③加入测试片2,进行比色,比较蒸汽冲击前后及冲击时间长短对酶活的影响。

Claims (5)
1.一种蒸汽冲击下酶活检测装置,其特征在于,该装置包括蒸汽发生室、样品池、蒸汽冲击室、比色系统四部分,所述的蒸汽发生室包括一容器(1)、加热装置(2),加热装置(2)设置在容器(1)的下方并对容器(1)加热,样品池(3)的下部设置有筛网(4),蒸汽冲击室包括孔板(5)、套管(6),比色系统包括一测试片、试管(8)、比色架(9),所述的孔板(5)放置在蒸汽发生室的容器(1)上,套管(6)放在孔板上,样品池放在孔板上的套管内,样品池对准孔板上的孔眼放置。
2.权利要求1所述的检测装置,其特征在于所述孔板为单孔或多孔。
3.权利要求1所述的检测装置,其特征在于所述样品池为蒸笼形,其中筛网选用40目以上。
4.权利要求3所述的检测装置,其特征在于所述样品池为蒸笼形,其中筛网选用80目以上。
5.权利要求1所述的检测装置,其特征在于所述套管为管底有单孔或多孔,该孔可调节大小或关闭。
