技术领域
本发明涉及用于处理化学浆(chemical pulp)中的树脂问题(pitch problem)的酶组合物和方法。
背景技术
木材除了它的主要组分纤维素、半纤维素和木质素之外包含约1至10%的树脂(pitch)或提取物(extractive)。树脂的主要组分是脂肪酸、甘油三酯、甾醇、甾基酯(steryl ester)和树脂酸(resin acid),例如枞酸(abietic acid)。由于粘于辊(roller)并在纸材料产生点或孔,树脂在造纸机中产生问题。
已使用了多种酶法处理树脂问题。WO 00/53843公开了甾基酯酶制备物及其在造纸中水解树脂的甾基酯部分的用途。
美国专利No.6,066,486公开了一种酶制备物,其包含源自莓实假单胞菌(Pseudomonas fragi)的胆固醇酯酶,及其水解浆树脂(pulp resin)的用途。
JP 2000080581公开了某些过氧化物酶用于在制浆或造纸方法过程中分解枞酸的用途。
X.Zhang;Pulp & Paper Canada,101:3(2000),第59-62页公开了关于漆酶去除溶解的和胶质的物质的能力的研究。

发明内容
本发明涉及通过用脂肪酶和过氧化物源的组合处理化学浆工艺用水来处理化学浆中的树脂问题的组合物和方法。虽然不局限于任一种操作理论,但认为脂肪酶和过氧化物的添加导致过酸(peracid)的形成,所述过酸反过来通过环氧化(expoxidation)反应将不饱和脂肪酸和树脂酸氧化。所得的产物是亲水的并且容易从浆中洗涤,由此减少与化学浆相关的树脂问题。
在另一个实施方案中,本发明涉及通过用脂肪酶、过氧化物源和有机酸处理化学浆工艺用水来处理化学浆中的树脂问题的组合物和方法。
发明详述
“造纸方法”涉及一种方法,其中将化学浆悬浮于水中,与多种添加剂混合然后传到设备中用于进一步加工,例如在该设备中将纸、纸板(cardboard)、棉纸(tissue)、纸巾(towel)等成形、挤压和干燥。
术语“纸材料”涉及能够由浆制成的产物,如纸、挂面纸板(linerboard)、波纹纸板(corrugated paperboard)、棉纸(tissue)、纸巾(towel)、瓦楞纸箱(corrugatedcontainer)或箱子(box)。
术语“造纸浆”或“浆”指任何化学浆,其可用于生产纸材料。
术语“化学浆”涉及化学浆(如Kraft浆或亚硫酸盐浆)或半化学浆(SCP)。化学浆通常通过碱法蒸煮(alkaline cooking)制备,由此将大部分木质素和一些半纤维素组分除去。在Kraft制浆或硫酸盐蒸煮中,通常将硫化钠或氢氧化钠用作主要的蒸煮化学品。在这样的浆中,作为碱法蒸煮的结果,树脂的甘油三酯部分将被水解为脂肪酸和甘油。
在本发明的用途和方法的具体实施方案中,化学浆是Kraft浆或亚硫酸盐浆。在具体的实施方案中,所述Kraft浆是漂白的Kraft浆,例如软材漂白的(softwood bleached)Kraft(SWBK,也称为NBKP(Nadel Holz Bleached Kraft浆)),硬材漂白的(hardwood bleached)Kraft(HWBK,也称为LBKP(Laub HolzBleached Kraft浆和))或它们的混合物。
待处理的Kraft浆可为漂白的Kraft浆,其可由软材漂白的Kraft(SWBK,也称为NBKP(Nadel Holz Bleached Kraft浆)),硬材漂白的Kraft(HWBK,也称为LBKP(Laub Holz Bleached Kraft浆和))或这些的混合物组成。
待用于本发明的方法的浆是化学浆的悬浮液。待用于本发明的方法的浆可包括至少10%,至少20%,至少30%,至少40%,至少50%,至少60%,至少70%,至少80%,至少90%,至少95%,或100%的化学浆(如Kraft浆或亚硫酸盐浆)。化学浆的百分比在1-100%的范围内。在具体的实施方案中,化学浆的百分比(如Kraft浆或亚硫酸盐浆)在1-99%,2-98%,3-97%,4-96%,5-95%,6-94%,7-93%,8-92%,9-91%,10-90%,15-85%,20-80%,25-75%,30-70%,40-60%或45-55%的范围内。
根据本发明,用脂肪酶和过氧化物源的组合处理化学浆工艺用水。所述化学浆工艺用水含有导致树脂问题的组分(pitch causing component),如脂肪酸、甘油三酯、甾醇、甾基酯和/或树脂酸(例如,枞酸)。本发明的方法具体可用于在制浆或造纸方法的过程中减少构成树脂的化合物,从而例如避免树脂问题。
可使用任何合适的脂肪酶。脂肪酶包括分类为EC 3.1.1.3的酶。参考theRecommendations(1992)of the Nomenclature Committee of the InternationalUnion of Biochemistry and Molecular Biology,Academic Press Inc.,1992。

在优选的实施方案中,脂肪酶源自曲霉属的菌株、无色杆菌属的菌株、芽孢杆菌属的菌株、念珠菌属的菌株、色杆菌属的菌株、镰孢属的菌株、腐质霉属的菌株,Hyphozyma的菌株、假单胞菌属的菌株、根毛霉属的菌株、根霉属的菌株,或嗜热霉属的菌株。
优选的脂肪酶包括美国专利No.6,074,863和WO 02/055679中所述的脂肪酶。优选的商品化的(commercial)脂肪酶包括Resinase A2X和Resinase HT(Novozymes A/S)。在另一个优选的实施方案中,脂肪酶是南极念珠菌脂肪酶A(CALA)或南极念珠菌脂肪酶B(CALB)(可由Novozymes A/S得到)
如本文中使用的,“过氧化物源”或“过氧化氢源”涉及过氧化氢本身或能够产生过氧化物的组分。可在脂肪酶处理方法的一开始或过程中加入过氧化氢源,例如以约0.001-100mM,尤其是0.01-50mM的浓度加入。过氧化氢的一个来源包括过氧化氢的前体,例如过硼酸盐或过碳酸盐。过氧化氢的另一个来源包括能够将分子氧和有机或无机底物分别转变为过氧化氢和氧化底物的酶。这些酶只产生低水平的过氧化氢。能够产生过氧化氢的酶的实例包括,但不限于葡糖氧化酶、尿酸氧化酶、半乳糖氧化酶、醇氧化酶、胺氧化酶、氨基酸氧化酶和胆固醇氧化酶。
虽然不局限于任一种操作理论,但认为脂肪酶和过氧化物的添加导致过酸(peracid)的形成,所述过酸反过来通过环氧化反应将不饱和脂肪酸和树脂酸和其它树脂组分氧化。所得的产物是亲水的并且容易从浆中洗涤,由此减少或消除树脂问题。
也可将有机酸加入浆,从而例如增强树脂处理。有机酸指包含至少一个酸性基团的任何有机物质。有机酸的实施例是乙酸、丁酸和亚油酸。有机酸的浓度优选为0.001-500mM。
在纸和浆加工的情况下,根据本发明的方法可在任何浆生产阶段进行。可将酶加入任何保持罐(holding tank),例如加入储存浆的容器(pulp storingcontainer)(贮料池(storage chest))、贮料塔(storage tower)、混合池(mixing chest)或计量池(metering chest)。优选在浆漂白方法之前或之后或者在浆漂白阶段之间施加酶处理。可将酶加入源自漂白的循环工艺用水(白水(white water))。在Kraft制浆方法的具体实施方案中,在粗浆(brown-stock)洗涤的过程中加入酶。
在本上下文中,术语“工艺用水”可包括作为原料加入造纸方法的水;由制造纸材料的方法的任何步骤产生的中间水产物;以及作为该方法的输出或副产物的废水。在具体的实施方案中,工艺用水是/已经/正在/或意欲进行循环或再循环,即在方法的另一步骤中再利用。术语“水”则是指任何含水介质、溶液、悬浮液,例如普通自来水,和与多种造纸方法中通常使用的佐剂和添加剂混合的自来水。在具体实施方案中,工艺用水具有低含量的固体(干)物质,例如低于20%,18%,16%,14%,12%,10%,8%,7%,6%,5%,4%,3%,20%或低于1%的干物质。
可在纸和浆加工中的常规条件下进行本发明的方法。工艺条件将为施加的酶、反应时间和给定条件的函数。
应该以有效量添加酶和过氧化物。术语“有效量”是指足以实现减少或抑制树脂组分的理想效果的量,如通过将所述组分降解或转变为可更容易地从浆或浆工艺用水中除去的形式。
在具体实施方案中,脂肪酶的剂量为每吨纸浆约0.1mg酶蛋白至约100.000mg(每种酶的)酶蛋白。
酶处理可在例如0.5-10%干物质的稠度(consistency)进行。在具体的实施方案中,稠度为0.5-45;0.5-40;0.5-35;0.5-30;0.5-25;0.5-20;0.5-15;0.5-10;0.5-8;0.5-6;或0.5-5%干物质的范围。
酶处理可在约10至约100℃的温度进行。温度的其它实例为如下范围(所有都是“从大约”和“至大约”):20-100、30-100、35-100、37-100、40-100、50-100、60-100、70-100、10-90、10-80、10-70、10-60和30-60℃,以及本文所示的上限值和下限值的任何组合。温度可为约20至90℃,或20至95℃,优选约40至70℃,或40至75℃。
酶处理可在约2至约12的pH进行。pH的其它实例为如下范围(所有都是“从大约”和“至大约”):3-12,4-12,5-12,6-12,7-12,8-12,9-12,2-11,2-10,2-9,2-8,4-10,5-8以及本文所示的上限值和下限值的任何组合。pH范围可为约2至11,优选在约3-9的范围。
酶处理的合适持续时间可为几秒至几小时的范围,例如约30秒至约48小时,或约1分钟至约24小时,或约1分钟至约18小时,或约1分钟至约12小时,或约1分钟至5小时,或约1分钟至约2小时,或约1分钟至约1小时,或约1分钟至约30分钟。反应时间可为约10分钟至3小时,10分钟至10小时,优选15分钟至1小时,或15分钟至2小时。
除酶和过氧化物处理之外,可将多种添加剂用于本发明的方法或用途。表面活性剂和/或分散剂常常存在于和/或添加至造纸浆。因而本发明的方法和用途可在常规用于造纸浆的阴离子的、非离子的、阳离子的和/或两性离子的表面活性剂和/或分散剂的存在下进行。阴离子表面活性剂的实例为烷基、取代的烷基或芳基的羧化物(carboxylate)、硫酸盐、磺酸盐或的磷酸盐。脂肪酸是烷基-羧化物的实例。非离子表面活性剂的实例是聚氧乙烯化合物,如醇乙氧基化物、丙氧基化物或混合的乙氧基-/丙氧基化物、聚甘油和其它多元醇(polyol),以及一些嵌段共聚物。阳离子表面活性剂的实例是水溶性阳离子聚合物,如硫酸季铵(quartenary ammonium sulphate)和一些胺,例如表氯醇/二甲胺聚合物(EPI-DMA)及其交联的溶液,聚氯化二烯丙基二甲铵(polydiallyl dimethylammonium chloride)(DADMAC),DADMAC/丙烯酰胺共聚物,和紫罗烯(ionene)聚合物,如美国专利号5,681,862和5,575,993中公开的那些。两性离子的或两性表面活性剂的实例是甜菜碱、甘氨酸酯(glycinate)、氨基丙酸酯(aminopropionate)、亚氨基丙酸酯(imino propionate)和多种咪唑啉(imidazolin)衍生物。还可以使用美国专利号5,256,252中公开的聚合物。
还根据本发明,可将如上所述的表面活性剂,包括它们的任何组合用于造纸方法。所述组合物中每种表面活性剂的量可达组合物的约8至约40%(w/w)。在具体实施方案中,每种表面活性剂的量为约10至约38,或约12至约36,或约14至约34,或约16至约34,或约18至约34,或约20至约34,或约22至约34,或约24至约34,或约26至约34,或约28至约32%(w/w)。
可理解的是术语酶,以及本文提及的多种酶和酶分类(enzyme class)包括野生型酶,以及其保留所述活性的任何变体。所述变体可通过重组技术产生。野生型酶也可通过重组技术产生或通过从天然来源分离和纯化来产生。
在具体实施方案中,所述的酶是明确的(well-defined),意思是只存在一种主要的酶组分。这可以通过例如在适当的大小排阻柱上的分级来进行推断。所述明确的,或纯化的,或高度纯化的酶可按照本领域所公知的和/或与所述的特定酶有关的出版物中描述的来获得。
术语“与...一起应用”(或“与...一起使用”)意思是在本发明的方法的相同步骤或另一步骤中可应用附加的酶。与用脂肪酶和过氧化物源处理造纸浆或工艺用水的步骤相比,其它工艺步骤可处于造纸方法中的上游或下游。
在具体实施方案中,附加的酶是具有如下酶活性的酶:蛋白酶、木聚糖酶、角质酶、氧化还原酶、纤维素酶、内切葡聚糖酶、淀粉酶、甘露聚糖酶、甾基酯酶,和/或胆固醇酯酶活性。氧化还原酶的实例是具有漆酶和/或过氧化物酶活性的酶。
术语方法的“步骤”意思是至少一个步骤,且其可为一个、两个、三个、四个、五个或甚至更多工艺步骤。因此,可将脂肪酶和过氧化物源应用于至少一个工艺步骤,并且还可将附加的酶应用于至少一个工艺步骤,该步骤与使用脂肪酶和过氧化物源的步骤相比可以是相同或不同的工艺步骤。
术语“酶制备物”意思是含有至少一种脂肪酶的产物。除酶活性之外,所述制备物优选含有至少一种佐剂。纸和浆工业的酶制备物中使用的佐剂的实例为缓冲剂、聚合物、表面活性剂和稳定剂。
可将具有蛋白酶、木聚糖酶、角质酶、氧化还原酶、纤维素酶、内切葡聚糖酶、淀粉酶、甘露聚糖酶、甾基酯酶,和/或胆固醇酯酶活性的任何酶在本发明的用途和方法中用作附加的酶。下面列出了这些附加的酶的一些非限制性实例。以大写字母写出的酶是可由Novozymes A/S,Krogshoejvej 36,DK-2880 Bagsvaerd,Denmark得到的商品化的酶。可使用本领域公知用于所述酶的方法(包括引用的文件中提及的方法)来分析任何这些附加的酶的活性。
角质酶的实例是源自如下的那些:特异腐质霉(US 5,827,719);镰孢属的菌株,例如大刀粉红镰孢,或尤其是豌豆腐皮镰孢(WO 90/09446;WO 94/14964,WO 94/03578)。角质酶还可源自丝核菌属的菌株,例如立枯丝核菌,或链格孢属的菌株,例如A.brassicicola(WO 94/03578),或其变体,如WO 00/34450或WO 01/92502中所述的那些。
蛋白酶的实例是ALCALASE,ESPERASE,SAVINASE,NEUTRASE和DURAZYM蛋白酶。其它蛋白酶源自拟诺卡氏菌属(Nocardiopsis)、曲霉属、根霉属、嗜碱芽孢杆菌(Bacillus alcalophilus)、蜡状芽孢杆菌(B.cereus)、纳豆芽孢杆菌(B.natto)、普通芽孢杆菌(B.vulgatus)、蕈状芽孢杆菌(B.mycoide)和来自芽孢杆菌属的枯草杆菌蛋白酶(subtilisin),特别是来自拟诺卡氏菌属的菌种和达松维尔拟诺卡氏菌(Nocardiopsis dassonvillei)的蛋白酶,如WO88/03947中公开的那些,及其突变体,例如WO 91/00345和EP 415296中公开的那些。
淀粉酶的实例是BAN、AQUAZYM、TERMAMYL和AQUAZYM Ultra淀粉酶。木聚糖酶的实例是PULPZYME HC半纤维素酶。内切葡聚糖酶的实例是NOVOZYM 613,342,和476酶产物。
甘露聚糖酶的实例是 et al,J.Biotechnol.29(1993),229-242中描述的里氏木霉内切-β-甘露聚糖酶。
甾基酯酶、过氧化物酶、漆酶和胆固醇酯酶的实例在本文背景技术部分提及的文件中公开。氧化还原酶的其它实例是EP 730641;WO 01/98469;EP719337;EP 765394;EP 767836;EP 763115和EP 788547中公开的过氧化物酶和漆酶。在本上下文中,当提及需要或受益于受体、增强子、介质和/或激活剂的存在的氧化还原酶时,如果所述的化合物并非已经存在,应认为包括这些化合物。增强子和介质的实例公开于EP 705327;WO 98/56899;EP677102;EP 781328和EP 707637中。如果需要的话,可通过将氧化还原酶酶系统(例如漆酶或过氧化物酶酶系统)限定为所述酶与其受体,任选地还与所述酶的增强子和/或介质的组合来进行区别。

实施例
实施例1.通过南极念珠菌脂肪酶B(CALB)将Kraft浆树脂脱树脂(deresination)
Dammar树脂(resin)由Fluka获得并用作模型树脂以模拟Kraft浆树脂。TAED也由Fluka获得。将75mg的Dammar树脂置于烧瓶内和加75mL的DI水。加入根据表1中所示条件的化学品或酶。在环境温度摇动(stir)过夜。通过UV-vis在600nm测定溶液的浊度。

将样品混合过夜后,观察到用脂肪酶(CALB)和过氧化物处理的样品变为奶状乳液,而对照样品仍然澄清如水并含有沉积在瓶底或瓶壁的树脂。浊度结果清楚地显示过氧化物与脂肪酶的组合可导致dammar树脂的更好的乳化。由过氧化物和漂白激活剂(TAED)原位产生的过乙酸也是非常有效的,但不如过氧化物和脂肪酶的组合那么有效。
实施例2.通过Resinase A2X和Resinase HT将Kraft浆树脂脱树脂。
Claims (16)
1.用于减少化学浆中的树脂问题的方法,其包括用脂肪酶和过氧化物源以有效减少树脂问题的量处理化学浆。
2.权利要求1的方法,其中所述处理在漂白阶段之前、之后或之间进行。
3.权利要求1的方法,其中所述化学浆是Kraft浆。
4.权利要求1的方法,其中所述化学浆是亚硫酸盐浆。
5.权利要求1的方法,其中所述化学浆是漂白的Kraft浆。
6.权利要求1的方法,其还包括由所述浆制备纸、挂面纸板、波纹纸板、棉纸、纸巾、瓦楞纸箱或箱子的步骤。
7.权利要求1的方法,其中所述脂肪酶源自曲霉属的菌株、无色杆菌属的菌株、芽孢杆菌属的菌株、念珠菌属的菌株、色杆菌属的菌株、镰孢属的菌株、腐质霉属的菌株,Hyphozyma的菌株、假单胞菌属的菌株、根毛霉属的菌株、根霉属的菌株,或嗜热霉属的菌株。
8.权利要求1的方法,其中所述脂肪酶源自念珠菌属的菌株。
9.权利要求1的方法,其中所述脂肪酶源自南极念珠菌脂肪酶A或南极念珠菌脂肪酶B的菌株。
10.权利要求1的方法,其包括用脂肪酶、过氧化物源和有机酸处理化学浆。
11.权利要求1的方法,其中过氧化物源是过氧化氢。
12.权利要求1的方法,其中过氧化物源是过氧化氢的前体。
13.权利要求12的方法,其中所述过氧化氢的前体是过硼酸盐或过碳酸盐。
14.权利要求1的方法,其中所述过氧化物源是将分子氧和有机或无机底物转变为过氧化氢的酶。
15.脂肪酶和过氧化物源用于减少树脂问题的用途。
16.脂肪酶和过氧化物源用于减少化学浆中的树脂问题的用途。
