本发明一般地涉及免疫学和药物,更特殊地涉及产生抗HIV-1免疫应答的方法。
背景信息
获得性免疫缺陷综合征(AIDS)的发病率已达到流行性,特别是在非洲、亚洲和加勒比海的第三世界国家中。AIDS的致病因子是人免疫缺陷病毒(HIV-1)。HIV-1是灵长类慢病毒,据信在五十年代从非洲的猴群交叉至人类中。
在人类中,HIV-1感染两种主要细胞类型:巨噬细胞和T淋巴细胞。在一典型的传递事件中,亲巨噬细胞毒株从一个人的体液转移到另一人的皮肤或粘膜的巨噬细胞。最初,病毒可在巨噬细胞中复制而不产生临床疾病。随着感染进程,由于病毒的快速突变率产生不同的毒株,这至少是由于反转录酶可靠性低所致。最终,产生亲T细胞毒株,其导致免疫系统的耗竭,及已知为AIDS的临床表象。
这一进程典型地需6-10年或更长,其部分取决于宿主免疫系统因素。但是,亲T细胞HIV-1毒株可以直接传递,导致更快的感染过程。随着疾病的进程,宿主系统置换T细胞群不能象病毒感染对T细胞群的耗竭那样快,导致T细胞水平的灾难性疾病。其结果是AIDS患者典型地由于感染而死亡,但这些感染在健康个体中通常最坏也仅导致中等水平的疾病。
尽管投入了数十亿美元用于研究,但是治疗此疾病仅取得了中等进展。最初,治疗限于治疗免疫低下患者中产生的机会性感染。后来,鉴别了能干扰HIV-1复制的核苷类似物如AZT,ddC和ddI。这些药物特别是在联合时延长了某些AIDS患者的生命。最近,一类称为蛋白酶抑制剂的药物已被证明在抑制HIV-1复制时更有效。当与核苷类似物联合应用时,蛋白酶抑制剂将某些患者的病毒滴度将至检测不到的水平。最近,美国的AIDS死亡率从流行开始首次下降,这据信至少部分是这种治疗方案的结果,并伴随着劝阻高度危险性行为的教育计划。
这些治疗方案的希望已被后续出现的对多药物混合物的抗性而降低。另外,尽管在用蛋白酶抑制剂和核苷类似物联合治疗的某些患者中病毒的循环水平下降,但是这种治疗不能从这些患者中消除病毒。据信在甚至没有可检测的循环病毒的患者中仍存在病毒库,例如在淋巴系统和巨噬细胞中。另外,即使不存在循环病毒,由于已被HIV-1感染的细胞的融合性质仍可能发生病毒的传播。最后,药物混合物不能在全部患者中均产生阳性结果,因此似乎不能有效抗全部病毒毒株。
多药物疗法所需的麻烦的剂量方案也产生了患者服从的问题。随着治疗的患者存活更长的时间,这些服从问题会不断增加,导致这种患者可能最终存在产生和传播抗这些药物混合物的HIV-1毒株的途径。
另外,尽管美国的一些患者可以负担得起这种昂贵的联合治疗,但是这种药物的费用远远超过许多第三世界国家国民的年收入。这一点由于HIV-1传播在第三世界快速增加而特别令人警觉。
很明显,优选的方案应是防止HIV-1传播,疫苗是实现这一目的的符合逻辑的选择。例如,已使用疫苗防止或降低各种病毒性疾病如脊髓灰质炎、麻疹、天花和流感的严重性。另外,疫苗可刺激已被病毒感染的个体的免疫系统。例如,狂犬病疫苗仅在传播事件如被感染的动物咬后给予。
已进行了各种尝试来开发增加人对HIV-1抗性的疫苗,但是目前这些方案均受到严重限制。例如,由部分HIV-1蛋白组成疫苗或使用灭活的HIV-1病毒的疫苗已产生,但是HIV-1在病毒复制过程中迅速突变,产生新变种的速度快于免疫系统反应的速度。其结果是,针对特定HIV-1蛋白或毒株所刺激的免疫应答对感染过程中产生的变种可能无效。另外,用HIV-1表面蛋白的部分接种可刺激可能促进而不是防止HIV-1感染的抗体的产生。由活的但复制缺陷的病毒组成的减毒的疫苗也已被提议,但是存在用这种HIV-1病毒感染个体特别是健康人体的公平性考虑。因此,仍存在对能提供抗HIV-1的免疫应答但不携带所述危险及与HIV-1的使用相关的限制的疫苗来作为接种剂的需要。
开发动物模型以研究HIV-1感染和可能的疫苗策略的一个途径是产生猴免疫缺陷病毒(SIV)和HIV-1之间的重组病毒,或称SHIV,其能感染低等灵长类。SHIV模型的体内行为已证实反转录病毒病理学和宿主应答的许多方面,这些方面包括证实宿主范围,细胞致病性和中和抗体的特异性由包膜序列决定(Luciw等,美国科学院院报92:7490-7494(1995))。用作活减毒疫苗,SHIV已被证明有效保护弥猴抗SIV的粘膜攻击(Baba等,科学267:1820-1825(1995);Quesda-Rolander等,AIDS Res.Hum.Retro.12:993-999(1996))。

本领域中需要能在灵长类特别是人中产生抗HIV-1免疫应答的非致病性慢病毒。
发明概述
本发明满足了这一需要并提供了另外的优势。在一个实施方案中,提供了一种嵌合反转录病毒基因组,其包括CAEV和HIV-1的元件。这一嵌合反转录病毒称为CHIV,这一反转录病毒基因组包括CAEV的调节序列和赋予CHIV非致病性的其他CAEV编码序列,以及HIV-1 env基因。其他反转录病毒基因可由CAEV或HIV-1提供。CHIV必须能在至少一种哺乳动物宿主细胞中繁殖。

本发明的详细描述
本文所述嵌合CAEV/HIV-1反转录病毒(“CHIV”)是开发活的减毒AIDS疫苗的一个新途径,CHIV具有CAEV的核心及HIV-1的免疫原性。开发CHIV的优势是原则上任何所需的HIV-1 env序列可取代CAEV env基因以产生更强的和更特异的免疫应答。

如本文所用,术语“表达控制区段”是指能指导重组CHIV反转录病毒转录和翻译的多核苷酸区段。这种区段包括启动子、增强子、polyA信号、剪接供体和受体位点、翻译起始位点和翻译终止位点。典型地,用于转录起始和终止的表达控制区段包括反转录病毒的长末端重复(LTR)区,优选地得自CAEV或HIV-1。
如本文所用,术语“样品”当用于本发明的诊断方法中时,是指组织样品或体液样品,如血(其可是全血、血浆或血清)或尿,其得自待测试CHIV或HIV-1感染的个体。获得这种样品的方法是本领域公知的和常规的。
如本文所用,术语“TCID50”是指“组织培养物感染剂量50%”,即感染50%培养细胞群所需的感染因子的量。
如本文所用,术语“治疗有效量”是指足以在进行治疗的个体中刺激免疫应答的免疫原的量。
如本文所用,术语“疫苗”是指一种含有免疫原的组合物,当其给予个体时,能刺激免疫应答。具体地,本发明的疫苗含有CHIV免疫原,其可给予例如人,其中刺激与抗HIV-1交叉反应的抗CHIV免疫应答。因此,CHIV免疫原可用作刺激抗HIV-1免疫应答的替代免疫原。
嵌合CHIV反转录病毒的构建
9.7kb Co克隆是目前唯一的CAEV感染性DNA克隆(Pyper等,病毒学杂志58(2):665-670(1986))。CAEV-Co的核酸序列以GenBank登录号M33677可得(也参见Saltarelli等,病毒学179:347-364(1990);Knowles等,病毒学杂志65:5744-5750(1991))。但是,原则上CAEV或HIV-1的任何变种均可用作本发明的嵌合体的材料来源。根据Hamming距离测量,这类变种优选地与已知CAEV或HIV-1序列呈小于15%的序列差异百分率(Faulkner等,TIBS13:321-322(1988))。尽管由于遗传密码的简并性,这一序列差异百分率在CAEV或HIV-1的分离株中核苷酸水平上测得,但是可合成在核苷酸水平具有大于15%序列差异百分率但仍编码类似蛋白质的多核苷酸。因此,可使用这样的多核苷酸,其编码能在重组病毒中履行相应的CAEV或HIV-1反转录病毒蛋白的功能并可由与已知CAEV或HIV-1多核苷酸的序列差异百分率小于15%的多核苷酸编码的蛋白。
类似地,嵌合体的HIV-1组分可从任何HIV-1毒株中分离。在具体的实施方案中,HIV-1 env编码区由两种毒株获得:SF162(亲巨噬细胞的)或SF33(亲T细胞系的)。也可使用与本文所述的趋异的env或其他HIV-1编码区,包括可介导巨噬细胞和T细胞感染的编码区。如上所述,趋异的HIV-1基因和已知的HIV-1基因的序列差异百分率小于15%,或编码可由序列差异百分率小于15%的多核苷酸编码的并能在重组病毒中履行所编码的蛋白的功能的蛋白。另外,这种趋异性env蛋白优选能介导对人T细胞、人巨噬细胞或两者的感染。
用于制备本文所述的嵌合CHIV病毒的技术包括多核苷酸操作和增殖的标准技术。原则上,可用任何技术产生嵌合多核苷酸,包括化学合成重叠的寡核苷酸,随后延伸和连接;用具有互补5’延伸的能在随后的PCR中组合的寡核苷酸对各个起始材料的所需部分进行PCR;以及更传统的限制酶消化以分离所需的区段,随后用所需的接头连接。另外,已发现病毒构建体的未连接互补部分共转染进宿主细胞可在宿主细胞中产生完整的可复制病毒。
得到的多核苷酸构建体可用标准技术繁殖。例如,嵌合病毒基因组可被插入细菌载体中并在细菌宿主细胞中繁殖,或者病毒可在哺乳动物宿主细胞中表达并且所得的病毒原液可系列传代,或者包含嵌合病毒基因组的DNA构建体可用如长PCR的技术体外繁殖。病毒也可在动物如弥猴中繁殖并从血样中回收。优选地,多核苷酸构建体的繁殖方式为能保持所产生的构建体的完整性,所述完整性是指多核苷酸序列及嵌合反转录病毒的整体结构。繁殖多核苷酸构建体的优选方式因此是在一重组缺陷的优选为recA-的细菌宿主菌株中的复制性细菌载体,例如一种质粒。这抑制了在嵌合反转录病毒构建体的LTR区中发现的直接重复之间的重组。如果在recA-细菌宿主菌株中确实发生了重组,可使用现有技术中已知的其他重组缺陷菌株来繁殖这种质粒构建体。
原型CAEV/HIV嵌合体由将HIV-1的tat,vpu,env和rev基因分子取代进CAEV-Co的9.7kb感染性克隆中,该感染性克隆保留CAEV的gag,pol,vif和LTR功能。图3A-C中示出的置换策略的原理是除去CAEV的env基因以及env和rev基因的起始位点,同时用一终止密码子提前终止CAEV tat基因。该弱反式激活CAEV tat基因的缺失或终止密码子终止在体内或体外对CAEV复制速率均无影响(Harmache等,病毒学杂志69:5445-5454(1995))。这一选择很大程度上是基于需要保持CAEV vif基因的活性,该基因是功能性复制所需的(Harmache等,病毒学杂志69:5445-5454(1995))并在其终止位点和tat起始位点之间具有8bp不同框重叠。
这一CHIV构建体基于来自HIV-1的env和rev基因而履行活性,因为这两个基因是CAEV复制所必须的。观察到CAEV基因组含有在位置、稳定性和构型方面与HIV-1相似的稳定茎环结构的rev应答元件(RRE),这一观察是rev在CAEV和HIV-1中的角色类似的支持证据。因此,这一CHIV构建体经经验测试。如果需要,可进行更复杂的工程以将CAEV rev基因掺入嵌合体。位置也与3’HIV-1 LTR重叠的非必须nef基因在这一CHIV构建体中缺失。
另外的编码区也可掺入CHIV构建体中。例如,选择性或检测性标记可包括在CHIV构建体中以选择或检测表达细胞。也可包括编码免疫刺激分子的基因以增加感染细胞的抗原性,从而刺激更强的免疫应答。
另外,尽管本发明涉及能复制的CHIV构建体和病毒原种的应用,但是复制缺陷的CHIV构建体和病毒原种也属于本发明的范围。当导入个体时,这种构建体产生流产感染。复制缺陷的CHIV构建体可如上所述制备,但是缺少至少一种病毒繁殖所需的功能性编码区。例如,病毒构建体可在关键的基因中具有一缺失或突变,而所用宿主细胞可提供该缺失的功能以产生复制缺陷的病毒原种。另外,CHIV构建体可缺少pol基因,其功能可由宿主细胞提供。宿主细胞可随后产生感染性病毒颗粒,掺入宿主细胞产生的pol蛋白,在病毒颗粒感染另一细胞后,其可使得缺陷性RNA反转录病毒基因组反转录成DNA。一旦不能复制的病毒基因组整合进感染细胞的基因组并表达后,病毒抗原可由感染的细胞产生,但是缺少pol蛋白将阻止随后的生产性感染。还可以通过在宿主细胞中不使用嵌合多核苷酸而独立地表达CAEV和HIV-1蛋白的编码序列,而产生含有CAEV和HIV-1蛋白的嵌合反转录颗粒。
得到的CHIV构建体可通过任何技术导入哺乳动物宿主细胞。可使用的技术的例子包括转染,如磷酸钙转染、脂转染、在人工病毒包膜中包囊化、电穿孔、刮或擦法和bolistic。一旦含有构建体的病毒颗粒产生后,在表达合适的表达受体的细胞中也可采用病毒感染。
本发明所用的哺乳动物宿主细胞包括能表达导入的CHIV构建体的细胞,以及能经产生的CHIV病毒颗粒进行感染的细胞。宿主细胞可建立成细胞系或原代分离物。例如,CHIV构建体可被导入由本领域已知技术制备的原代山羊滑膜(GSM)细胞,或人横纹肌肉瘤细胞系,例如细胞系RD(ATCC#CCL-136)或RD-5(Luciw等,美国科学院院报92:7490-7494(1995))。可采用的哺乳动物宿主细胞的进一步例子包括由本领域已知技术制备的原代人和弥猴PBMC和巨噬细胞,和CD4+细胞系HUT78(Marchalonis等,细胞免疫学77(1):161-75(1983);ATCC#TIB-161)和CEMX174(Sei等,细胞免疫学125(1):1-13(1990))。
可分析宿主细胞以检测导入的构建体的功能。可进行的分析包括:(a)检测gp120(通过gp120抗原捕获ELISA)和CAEV gag蛋白p28(通过用多克隆山羊血清的Western印迹)的产生的免疫分析;(b)GSM或RD-5基因组DNA的CAEV gag和HIV-1 env PCR以检测原病毒DNA序列;(c)总细胞RNA的CAEV gag和HIV-1 env RT-PCR。
完整CHIV构建体的存活性通过转染进合适的宿主细胞,例如GSM细胞,或人横纹肌肉瘤细胞系RD-5(Luciw等,美国科学院院报92:7490-7494(1995))而测试。经反转录酶(RT)分析和CAEV和HIV-RT-PCR监测上清。所产生的嵌合体的宿主细胞范围可通过接种入和恒河弥猴PBMC和巨噬细胞以及HUT78和CEMX174细胞系,随后测试病毒复制(RT-PCR和RT分析)而分析。HIV-1 env和CAEV gag蛋白的产生可经gp120抗原捕获ELISA和CAEV p28 Western印迹而分析。基于衍生包膜基因的HIV-1毒株的嗜性可预测CHIV嵌合体的嗜性。
例如,预测CHIVSF33在HUT78和CEMX174细胞系中复制,这两个细胞系支持亲T细胞系HIV-1毒株的表达。预计CHIVSF162嵌合体在人、弥猴和GSM细胞培养物中生长。嵌合基因在培养物中的表达经RT分析和CAEV gag和HIV-1 env的RT-PCR而评价。另外,可进行RT-PCR产物的测序以证实表达的基因的性质。经显微镜观察检查培养物的合胞体形成或细胞病变。合胞体是巨大的多核细胞,其特征是活性反转录病毒感染。细胞病变可通过合胞体形成以及细胞质液泡化或细胞死亡而确定。希望的是CHIV在靶细胞中体外形成合胞体或使之发生细胞病变,因为没有宿主免疫系统对抗感染。优选地CHIV在大部分这种细胞中产生这种细胞病变效应。
病毒蛋白产生通过如转染实验中的免疫学方法监测,并且如果需要,经用例如35S-甲硫氨酸标记的培养物的免疫沉淀而监测。
如果在不存在蛋白时检测原病毒DNA和细胞RNA,一或多种基因功能的异常剪接的可能性通过Northern印迹而分析。在嵌合体中不经意活化的隐蔽剪接位点可通过标准技术如定点诱变或诱变PCR而除去。当需改进所需剪接位点的利用时,可经类似的技术导入强共有剪接位点。或者,可将内部核糖体进入位点导入构建体以直接促进所需蛋白的翻译。
如果在细胞中检测到蛋白,但是没有形成成熟的病毒颗粒,可能存在组装问题。在HIV-1组装中,新生的包膜分子被定向到十四烷基化的基质蛋白(MA)位点,这是在有蹄动物慢病毒组装中未知的机制(Dorfman等,病毒学杂志68(3):1689-1696(1994))。但是,由于缺少十四烷基化所致的不相容性可通过提供来自另一种病毒的十四烷基化的膜结合功能域而校正(Wills等,病毒学杂志65(7):3804-3812(1991))。
最后,如果未检测到完整的嵌合DNA,可用报道基因如细菌氯霉素乙酰转移酶(CAT)评价CAEV-Co中的tat基因的作用(Saltarelli等,病毒学197(1):35-44(1993))。CAT基因可置于CAEV-Co LTR启动子的控制下,并评价具有或不具有CAEV tat基因的构建体在宿主细胞中的表达。如果CAT在宿主细胞中表达而CHIV不表达,则可将额外的CAEV基因掺入CHIV以使得病毒复制。
基于这一诊断途径的结果,进行图3A-C所示的对构建体的修饰。来自CAEV和/或HIV-1的额外的编码区可掺入嵌合体以获得功能更好的反转录病毒。负面影响嵌合体的编码区可被删除。
结果得到一种嵌合反转录病毒,其:包括来自HIV-1毒株的至少包膜基因;包括足够的CAEV序列以弱化重组病毒在灵长类中的生物学作用以赋予CHIV非病原性;具有来自CAEV或HIV-1的病毒基因组的其余部分;能在宿主细胞中体外复制;产生抗HIV-1的免疫应答。
含有CHIV病毒颗粒的病毒原种可用标准技术从一表达宿主细胞产生。例如,来自表达CHIV的3H-尿苷标记的宿主细胞的培养物上清可进行蔗糖密度梯度离心,反转录病毒颗粒的位置可通过收集梯度组分并分析放射性和反转录酶活性而确定。更直接地,表达CHIV的培养物可在130000xg离心而沉淀颗粒,如果需要得到的沉淀可洗涤并再沉淀,并重悬于合适的溶液中。
当病毒颗粒需导入灵长类时,病毒应在衍生自该灵长类的宿主细胞中传代以避免对宿主细胞抗原的非特异性免疫应答。例如,当病毒颗粒需导入人中时,病毒优选在人PBMC中繁殖以避免对不经意包装进病毒颗粒的非人宿主细胞抗原的免疫应答。
可使用动物模型以在用于人类之前进一步评价CHIV。可在动物例如弥猴中进行体外感染以确定CHIV是否是病原性的,以及是否产生针对HIV-1包膜的HIV-1毒株特异性免疫应答。动物可用如上所述产生的病毒接种物转输。可定期收集血样以确定是否发生感染,以及是否发生动物血细胞计数的病原性改变。
当CHIV免疫原包含含有CHIV病毒基因组的多核苷酸时,该多核苷酸可通过本领域制备多核苷酸疫苗已知的技术制备。例如,美国专利5,561,064,5,703,055,5,580,859和5,589,466可用于制备适合给予哺乳动物的多核苷酸以产生免疫应答。
本发明的疫苗除CHIV免疫原外还优选地含有药物可接受的载体,药物可接受的载体是本领域公知的,包括溶液如生理学缓冲的盐水或其他溶剂或载体如乙二醇、甘油、油例如橄榄油,或可注射的有机酯。如果需要,本发明的疫苗可含有是佐剂的药物可接受的载体。佐剂如磷酸铝、氢氧化铝、弗氏完全或不完全佐剂,QS21(美国专利5,889,176所述,Rovinski等),或其他合适佐剂是本领域已知的和可商购的(Ribi免疫化学研究公司,Hamilton,MT)。加入佐剂通常降低刺激免疫应答所需的CHIV免疫原的量。
药物可接受的载体也可含有作用为例如稳定CHIV免疫原或增加其吸收的生理学可接受的化合物,这种生理学可接受的化合物包括例如,碳水化合物如葡萄糖、蔗糖或右旋糖;抗氧化剂,例如抗坏血酸或谷胱甘肽;螯合剂;低分子量蛋白质;或其他稳定剂或赋型剂。本领域技术人员应理解药物可接受的化合物包括生理学可接受的化合物的选择取决于,例如,疫苗的给药途径和CHIV免疫原的特定生理学-化学特征。
CHIV免疫原可以本身是免疫原性的,可以体内产生免疫原性分子,或可附着于载体分子如牛血清白蛋白或惰性载体,从而CHIV免疫原-载体复合物可刺激免疫应答(例如参见,Harlow and Lane,抗体实验手册,冷泉港实验室出版社,1988)。例如,当疫苗含有CHIV病毒颗粒时,病毒是免疫原性的并可在接种个体中刺激免疫应答。
接种个体以刺激免疫应答的方法也是公知的(Harlow and Lane,文献同上,1988)。例如免疫原可以经静脉内、皮内、皮下、口服、肛门,阴道、鼻内或经皮给予。疫苗考虑希望的给药途径而配制。另外,有利的是给予一或多次加强免疫,给予加强免疫的需要和这种加强免疫的时间可通过测量例如接种个体的血清中的抗HIV-1抗体的存在和滴度而确定。
虽然本文描述了将CHIV免疫原给予个体,但应理解的是抗HIV-1的免疫应答可体内或来自体内而刺激。例如,希望的是来自体内刺激抗HIV-1的免疫应答,其中待治疗的个体具有AIDS或被HIV-1感染。对于来自体内刺激,从个体中取出淋巴细胞,并在培养中免疫,同时可扩增淋巴细胞群,随后这些刺激的、扩增的免疫细胞可输入个体,由此提供治疗优势。这种疗法可以是姑息治疗并可以增加AIDS患者的生活质量。
构成治疗有效量的CHIV免疫原的量可根据如下情况而变化,例如免疫应答的刺激是体内还是来自体内,给药是第一次给药还是加强给药,佐剂是否与免疫原一起给予,以及,当体内给药时,根据给药途径和患者体重而变化。确定免疫原的治疗有效量的方法是常规的和本领域公知的(参见Powell and Newman,疫苗设计:亚基和佐剂途径(Plenum Publ.Corp.;1994))。
例如,当CHIV病毒颗粒给予猴子时,颗粒CHIV免疫原的治疗有效量的范围可以是约0.1-1×108TCID50,通常为约7×102-7×104TCID50。当免疫原是包含单一完整CHIV反转录病毒基因组的多核苷酸时,治疗有效量的范围可以是约0.05微克/千克-50毫克/千克,通常是约0.005-5毫克/千克。
可从接受CHIV免疫原的个体取样并测试确定是否该个体已产生抗HIV-1的抗体。检测样品中存在的抗原-抗体相互作用的方法是本领域公知的并包括,例如,使用可检测标记的抗原或使用可检测标记的第二抗体,其是特异地与来自特定哺乳动物物种的特定类别的抗体如IgG,IgA,IgM,IgD或IgE结合的抗体(例如,参见Greenand David,美国专利4,376,110,1983年8月授权,引入本文作参考)。例如,如果样品是来自人的血清或血浆样品,第二抗体可以是山羊抗人IgG。这种第二抗体可用公知方法制备或可商购。可检测标记CHIV抗原或抗体的方法也是本领域已知的。
下述实施例意在描述而非限制本发明。
实施例1 产生嵌合CAEV/HIV-1反转录病毒
实验设计基于感染性克隆CAEV-Co,其中取代进HIV-1的env功能。图3A-C示出了将衍生自毒株SF162(Cheng-Mayer等,病毒学杂志64(8):4012-4020(1990))和SF33(York-Higgins等,病毒学杂志64(8):4012-4020(1990))的HIV-1 env功能取代进CAEV-Co的详细步骤。为制备CAEV/HIV-1(CHIV)构建体,通过缺失表达所需的tat/env/rev基因,然后用HIV-1的tat/env/rev基因置换而修饰CAEV的5′9.4kb HindIII片段。这一修饰如图3A,步骤1-5所示。
分别含有9.4kb 5′和0.4kb 3′CAEV-Co区段(Pyper等,病毒学杂志58(2):665-670(1986))的pUC18和pGem2载体得自J.Clements博士(Johns Hopkins大学)并转化进recA-细菌宿主DH5α。用标准SDS-NaOH裂解制备选择的克隆,高盐沉淀以除去细菌DNA和碎片,异丙醇沉淀。重溶的质粒DNA用RNase和蛋白酶K处理后进行苯酚氯仿抽提,乙醇沉淀和重悬于TE缓冲液。两个质粒均进行限制消化和部分测序以证实。
图3A中示作构建体1的pUC 18中的5′CAEV-Co 9.4kb插入片段在5745和8097具有独特的Spl 1位点。Spl 1-Spl 1片段含有大部分CAEV tat基因以及env和rev基因的起始位点和大部分编码区。这一片段可通过用Spl 1消化构建体1并重新连接切割的质粒而缺失,以形成图3A中示作构建体2的单Spl 1插入位点5′CAEV质粒。3′远端env和rev基因的未切割区(包括env的Tm区)不具有翻译起始位点,因此不应被翻译。另外,CAEV tat基因在取代HIV-1 tat序列的同时被提前终止(如下所述)。
两种HIV-1盒插入片段,每个均含完整的HIV-1 tat,vpu,env和rev基因来自质粒构建体p33/3′SX和p162/3′SX(Luciw等,美国科学院院报92:7490-7494(1995)),分别含有SF33(York-Higgins等,病毒学杂志64(8):4012-4020(1990))和SF162(Cheng-Mayer等,病毒学杂志64(9):4390-4398(1990))的env基因,在图3A中示作构建体3。这些质粒构建成HIV-1 tat/vpu/env/rev插入片段侧翼为5′Sph1和3′XhoI位点。切割这些Sph1-Xho1片段并连接特异的终端接头序列,该序列两端为Spl1位点,在HIV-1构建体中不存在该位点。图3C示出了DNA接界序列。
在构建5′CAEV-HIV构建体的最后步骤,将末端为Spl 1位点的HIV-1插入片段4连接进用Spl 1切割的插入载体2,这一操作的效率可通过首先用小牛碱性磷酸酶(CIP)除去插入载体2的5′突出磷酸基团以防止载体的自我闭合而保证。
如图3B所示,通过将来自最终5′构建体的HindIII片段与含有3′CAEV LTR的0.4kb CAEV片段6的HindIII片段连接而组装完整的CAEV-HIV克隆。或者,这两个片段可一起导入GSM细胞,在该细胞中两个片段可自身连接。这一连接的最终结果是CAEV-HIV″CHIV″病毒构建体7。
在5′CAEV/HIV和3′HIV/CAEV接界的Spl 1DNA接头的细节见图3C所示。5′CAEV-HIV接界显示出CAEV vif和tat基因的8bp不同框重叠(5688-5693),其需要在接头区设计CAEV tat基因的“框内”终止密码子(TGA)。在其下示出从起始位点至终止密码子的截短的CAEV tat基因的DNA和21个氨基酸序列。p162/3′SX质粒中的Sph 1限制位点(5814)与162至HIV-1 tat起始位点后的位置的连续HIV-1序列一起示出。在5′Spl 1接头中还设计有一独特的Cla 1位点(ATCGAT),其在CAEV质粒或HIV-1插入片段中均不存在(与Not 1位点一起应用,见下述)。
在图3C中还示出了3′HIV-CAEV接界DNA序列,以及HIV-1 Xho 1位点和接头/CAEV Spl 1位点,和HIV-1 env基因终止位点(8801)。3′Xho 1-Spl 1接头还包括独特的Not 1位点(GCGGCCGC),该位点在CAEV克隆和HIV-1插入片段中不存在。该Not 1位点与5′接头中的Cla 1位点使得能以5′Cla 1-3′Not 1方向特异方式置换这一HIV-1 DNA盒。

实施例2 重组反转录病毒在宿主细胞中的表达
完整CHIV构建体的活性经转染进GSM细胞或人横纹肌肉瘤细胞系RD-5而测试(Luciw等,美国科学院院报92:7490-7494(1995))。经CAEV gag和HIV-1 env RT-PCR和RT分析测试转染的空白转染的培养物上清中病毒的产生。转染细胞进行如下的逐步分析以检测构建体的功能,所述分析包括:(a)免疫分析,以检测gp120(经gp120抗原捕获ELISA)和CAEV gag蛋白p28(经使用多克隆山羊血清的Western印迹)的产生;(b)GSM或RD-5基因组DNA的CAEV gag和HIV-1 env PCR以检测原病毒DNA序列;(c)总细胞RNA的CAEV gag和HIV-1 env RT-PCR。
图2A-B示出两个包含CAEV-Co基因组的CAEV片段,pUC 18中的9.4kb 5′片段和pGem2中的0.4kb 3′片段,当转染进GSM细胞中时将产生活病毒。GSM细胞用(1)未连接的5′和3′片段,或连接的DNA,未用(2)或用(3)小牛碱性磷酸酶处理,而脂转染胺转染,如图2A所示。如培养物上清的CAEV gag RT-PCR所测定,所有三种方式均产生病毒(图2B)。因此,在用嵌合体的5′和3′片段转染GSM时,预连接可能不是必需的。空白转染的GSM培养物,泳道(4),(5)和(6)是PCR阴性。
方法:CAEV-Co 9.4kb(5′克隆)和0.4kb(3′克隆)质粒分别用HindIII消化,片段用凝胶纯化以获得制备量。连接片段,较小的片段用或不用磷酸酶处理,或不经连接而转染。用脂转染胺(BRL)根据厂商规定的条件转染进GSM细胞中。12小时后,在Optimem/脂转染胺/DNA试剂中,GSM培养物在DMEM中洗若干次,并返回标准组织培养条件,即含5%胎牛血清的DMEM,37℃,5%CO2。
转染后6天,用triazol(BRL)提取上清等份,进行CAEV gag巢式RT-PCR,扩增一173bp序列。接受两个片段的转染细胞,无论这是磷酸酶处理的或连接的片段,均从提取的上清中产生扩增的173bp片段。
类似地将CHIV构建体转染进GSM或RD-5细胞中,分析培养物上清中重组嵌合病毒的产生。
实施例3 CHIV宿主范围的确定
如Luciw等所述(Luciw等,美国科学院院报92:7490-7494(1995))确定活嵌合体的宿主细胞范围,即用CHIV病毒原种接种下述细胞培养物,该病毒原种产自如上所述转染的细胞或其后续的传代并且如果需要将其浓缩,所述的细胞培养物为人和弥猴PBMC和巨噬细胞,GSM细胞,CD4+细胞系HUT78和CEMX174。可用病毒原种的系列稀释液接种培养物,所用病毒量典型地约为0.1-1×106TCID50。嵌合基因在培养物中的表达经如在转染实验中的RT分析和CAEV gag和HIV-1 env RT-PCR评价。另外,将RT-PCR产物进行测序以证实表达的基因的性质。病毒蛋白的产生经在转染实验中所述的免疫学方法监测,并且如果需要,经放射标记的培养物上清或裂解物的免疫沉淀而监测,放射标记例如可用35S-甲硫氨酸标记。经显微镜观察检查培养物的合胞体形成或细胞病变。
CHIV在弥猴PBMC中的体外繁殖:从来自两个首次做实验的弥猴的10毫升血中制备PBMC,并在6孔微滴板(Falcon)中铺板,3×106个细胞/孔于RPMI,10%胎牛血清和10微克/毫升PHA中。24小时后,每孔加入0.5毫升病毒接种物或培养基。4天后收获培养物,用Trizol分离和提取细胞及上清,用于巢式gag RT-PCR。
CHIV在人PBMC中的体外繁殖:从CAEV和HIV-1阴性人供体经Ficoll密度离心制备PBMC,将PBMC加入24孔微滴板(Falcon3047)中铺板,0.8×106个细胞/孔于RPMI,10%人AB血清和10微克/毫升PHA中。24小时后,加入200微升病毒接种物。4天后收获培养物,离心分离细胞及上清。细胞在PBS,pH7.4中洗3次。提取基因组DNA,细胞RNA和上清RNA,用上述巢式CAEV gag引物进行PCR和RT-PCR。
检测由病毒传代产生的反转录酶活性:来自PCR(-)供体的PHA刺激的人PBMC在24孔微滴板上铺板,用CHIV病毒接种物接种(105TCID50/孔),同时,将CHIV病毒接种物的无细胞等份置于CO2温箱中。在第0、2和6天收集培养物上清等份,进行RT分析(Cheng-Mayer等,病毒学64(9):4390-4398(1990))。简要地,将15微升无细胞上清加入两个96孔板的每孔中,并与50微升32P-TTP以及含有50mM Tris,pH7.8,75mM KCl,2mM DTT,5mM MgCl2,5μg/ml poly-A模板,6.2μg/ml寡dT 12-18引物,0.05% NP40,0.5mMEGTA和10μCi/ml32P的混合物混合。在恒湿箱中于37℃保温板1.5-2小时,然后将每孔中的10微升点至CD81滤纸上,空气干燥30分钟。然后滤纸用1×SSC洗5次,用乙醇洗1次。用Phosphor ImagerTM定量测定放射性掺入。
实施例4 制备CHIV病毒原种
来自3H-尿苷标记的产CHIV宿主细胞培养物的上清进行蔗糖密度梯度离心,通过从梯度收集组分并分析放射性和反转录酶活性而确定反转录病毒颗粒的位置。或者,培养物可以是未标记的,并可用免疫学方法鉴别含病毒蛋白质的组分。
实施例5 用于CHIV感染的动物模型
进行弥猴的体内感染以确定CHIV是否是病原性的,以及在CHIV感染时是否产生HIV-1毒株特异性免疫应答。致病性可由升高的病毒血症水平得以证实,其可产生Coomb阳性溶血性贫血或血小板减少。致病性也可由临床迹象如体重减轻或周围淋巴结增大证实。具有这种致病性的CHIV可重新工程化,用其他的CAEV序列取代CHIV中的相应HIV-1序列。
方法:经股静脉向健康成年弥猴转输10毫升含约7×102-7×104TCID50,优选约7×103TCID50的CHIV病毒接种物。以约4周的间隔从弥猴获得血收集物,以分离PBMC和进行RT-PCR。如上所述进行细胞RNA的巢式gag和env RT-PCR。
实施例6 用CHIV疫苗接种个体
这一实施例描述给予个体CHIV疫苗以刺激免疫应答的方法。
如上所述通过接种宿主细胞而制备CHIV病毒。收获细胞上清,4℃、150000xg离心以沉淀CHIV病毒颗粒,以获得含50%甘油作为稀释剂的约2×108pfu/ml无菌悬液(参见Graham等,感染性疾病杂志166:244-252(1992))。用无菌分叉针将50微升CHIV悬液皮内接种至活病毒受体的单处皮肤部位。也可用灭活的或减毒的CHIV制备物或其他CHIV免疫原,例如含有CHIV病毒基因组的DNA构建体进行接种。在接种部位使用无菌透明绷带,然后在形成痂后去掉。
在给予CHIV免疫原后第14、28和54天取血样,确定抗体滴度(体液应答)或T辅助细胞或胞毒性T细胞免疫应答(细胞应答)。检查体液和细胞应答的方法是本领域熟知的(例如参见Egan等,感染性疾病杂志171:1623-1627(1995);还参见Harlow和Lane,文献同上,1988)。如果需要,在初次接种后第56天给予第二次(加强)免疫。在初次接种后第70、90、160、180、270和355天进行额外的体液和细胞应答评价。如果需要,在第365天或随后需要时给予第3次(加强)接种。个体中HIV-1 env和CAEV gag蛋白的产生可从血样中经gp120抗原捕获ELISA和CAEV p28 Western印迹而分析。
实施例7 在用CHIV接种的人中检测中和抗体
这一实施例描述了用于证实CHIV感染的个体产生针对CHIV抗原的免疫应答的方法。
血样和试剂
收集人和山羊血样并将血清组分用于免疫学测试。ELISA和Western印迹分析中所用的测试血清抗体水平的抗原包括粗CHIV和CAEV,以及购自Intracel(Shepreth UK)的重组HIV-1 gp120和p24。
CHIV病毒如上所述从表达宿主细胞培养物如GSM或RD-5细胞产生。收集CHIV感染的培养物的培养基并在600xg离心澄清。粗CHIV制备物等份可在-70℃冷冻储存,粗CHIV制备物可用于分离纯化的CHIV或可用于制备纯化的蛋白质。
HIV-1 ELISA和Western印迹分析
验证的HIV-1和Western印迹诊断试剂盒购自Organon Teknika(Cambridge UK),如厂商建议进行分析。
用重组HIV-1抗原、gp120和p24和HRP缀合的兔抗山羊和山羊抗人第二抗体(Zymed Laboratories Inc.)如前所述(Douvas andTakehana,AIDS Res.Hum.Retrovir.10:253-262(1994);Crow等,细胞免疫学121:99-112(1989))进行ELISA和Western印迹分析。
HIV-1病毒中和分析
在48孔组织培养板中用以1×106个细胞/毫升浓度维持在生长培养基中的PHA活化的正常供体PBMC作为感染靶而进行病毒中和分析。各种稀释度的HIVIGTM,热激活的(56℃,30分钟)血清或纯化的IgG与10或50TCID50的病毒在37℃保温30分钟。
血清和HIVIGTM的阴性对照分别包括来自未被HIV-1感染的和未患系统性风湿病的热激活的血清,和IVIGTM。HIVIGTM是来自9个处于HIV-1感染早期阶段的血清阳性供体的合并血清的中和IgG制备物,IVIGTM是来自正常供体的IgG,它们均得自北美生物制品公司(Miami FL)。或者,可使用其他来源的HIV-1中和抗体和对照抗体。TCID50用病毒滴定的常规方法确定。
血清或IgG与病毒保温后,向靶细胞中加入血清/IgG-病毒混合物,共重复三份。在37℃保温过夜后,1000rpm离心板5分钟,并完全更换培养基。这一洗涤步骤重复3次以保证血清/IgG-病毒接种物的完全除去。在培养第4天,更换培养基,在第7天收获上清定量p24核心抗原。通过比较每个血清/IgG处理的组织培养孔中的平均p24核心抗原浓度与特异阴性对照的该浓度而确定抑制百分比。
平行地,制备对照以测定p24核心抗原量的任何非特异降低,这可能是由于血清或HIVIGTM在洗涤步骤中未完全除去所致。在血清/IgG-病毒混合物加入靶细胞时,每种血清,IVIGTM或HIVIGTM不经预先与病毒保温也加入靶细胞中,这些孔与实验培养物平行处理。培养7天后,每孔的上清与阴性对照的上清1∶1混合,定量p24抗原浓度。低于阴性对照值50%的测量值被视为p24抗原的非特异抑制,其可能是由于存在残余p24抗体所致。
尽管本发明已参照上述实施例进行了描述,应理解的是在不偏离本发明的精神的范围内可进行各种修改。因此,本发明仅限于下述权利要求书。所有引述的对比文献均引入本文作参考。
Claims (5)
1、包含CHIV免疫原和药物可接受载体的疫苗,其中所述CHIV免疫原包括一种多核苷酸或包含该多核苷酸的纯化的反转录病毒,其中所述多核苷酸包含嵌合CHIV反转录病毒基因组,该基因组包含HIV-1包膜基因,山羊关节炎脑炎病毒的病毒基因组的一或多个区段,所述的一或多个区段足以使由该嵌合反转录病毒基因组在哺乳动物宿主细胞中表达产生的反转录病毒颗粒对于灵长类是无致病性的,其中该嵌合基因组包含能指导嵌合反转录病毒基因组在哺乳动物宿主细胞中表达的得自山羊关节炎脑炎病毒的包括5′和3′LTR区域在内的表达控制区段,并且由该反转录病毒基因组在宿主细胞中表达产生的反转录病毒颗粒能产生抗HIV-1的免疫应答。
2、权利要求1的疫苗,其中该CHIV免疫原是包含所述多核苷酸的纯化的反转录病毒。
3、权利要求1中限定的CHIV免疫原在制备用于预防或治疗HIV感染的药物中的应用。
4、体外与治疗有效量的权利要求1中限定的CHIV免疫原接触的淋巴细胞在制备用于预防或治疗HIV感染的药物中的应用。
5、产生病毒制备物的体外方法,包括将一种多核苷酸导入能表达该反转录病毒基因组的分离的哺乳动物宿主细胞,在培养基中培养该宿主细胞以产生病毒颗粒,以及从培养物培养基中纯化病毒颗粒,其中所述多核苷酸包含嵌合CHIV反转录病毒基因组,该基因组包含HIV-1包膜基因,山羊关节炎脑炎病毒的病毒基因组的一或多个区段,所述的一或多个区段足以使由该嵌合反转录病毒基因组在哺乳动物宿主细胞中表达产生的反转录病毒颗粒对于灵长类是无致病性的,其中该嵌合基因组包含能指导嵌合反转录病毒基因组在哺乳动物宿主细胞中表达的得自山羊关节炎脑炎病毒的包括5′和3′LTR区域在内的表达控制区段,并且由该反转录病毒基因组在宿主细胞中表达产生的反转录病毒颗粒能产生抗HIV-1的免疫应答。
