Abstract

Moisturising composition comprises a Simarouba plant extract, amino acids and ceramides. The composition may also contain an unsaturated fatty acid or ester. The composition preferably contains (by weight) 5-20% Simarouba extract, 55-85% amino acids, 3-10% ceramides and 0-15% unsaturated fatty acid or ester. The amino acids are especially selected from L-lysine, L-citrulline, L-threonine and L-histidine. The ceramides are especially plant (e.g. wheat) glycoceramides. The unsaturated fatty acid or ester is especially selected from linoleic acid, oleic acid, tocopheryl linoleate or oleate, and 1-12C alkyl esters, e.g. decyl oleate or ethyl linoleate (vitamin F).

  La présente invention concerne un complexe à base d'extrait de Simarouba ayant un effet favorisant et entretenant l'hydratation de l'épiderme, ainsi que des compositions cosmétiques pour les soins de la peau contenant ce complexe. 



  La plante Simarouba est une plante de la famille des Simaroubacées, parmi laquelle on peut citer les plantes Simarouba amara Aubl., Simarouba glauca, Simarouba versicolor et Simarouba excelsa. Un extrait de la plante Simarouba amara Aubl. par exemple, qui est un arbre assez répandu en Guyane française, et en particulier l'écorce de cette plante, est utilisée en médecine indigène pour traiter différents symptômes tels que la dysenterie, l'insuffisance de tonicité de l'intestin et les insomnies. Par ailleurs, l'utilisation en cosmétique d'extraits de Simarouba amara a été décrite dans la demande de brevet français publiée sous le No FR-A1 2 699 080 au nom de LVMH RECHERCHE.

   Cette utilisation vise en particulier l'atténuation des taches pigmentaires cutanées et le renforcement de la fonction protectrice de la peau, et elle a pour origine la mise en évidence de l'action positive d'un extrait de cette plante sur la différenciation des kératinocytes, conduisant notamment à l'amélioration qualitative de la couche cornée. 



  Or, les présents inventeurs ont constaté qu'un extrait de la plante Simarouba, associé à des acides aminés et des  substances lipidiques telles que des céramides, exerce en plus et de manière inattendue des effets de renforcement de l'hydratation des couches supérieures de l'épiderme. 



  Ainsi, le complexe hydratant, premier objet de cette invention, comporte comme ingrédients actifs un extrait de la plante Simarouba en association avec des acides aminés et des céramides. 



  Un second objet de cette invention consiste en une composition cosmétique pour les soins de la peau contenant le complexe hydratant ci-dessus, ainsi qu'un excipient acceptable du point de vue cosmétique. 



  Plus particulièrement, le complexe selon l'invention peut comporter de 5 à 20% en poids d'extrait de Simarouba, de 55 à 85% d'acides aminés, de 3 à 10% de céramides, et éventuellement jusqu'à 15% d'acide gras insaturé, de préférence sous forme d'ester. 



  Selon des variantes de l'invention, les acides aminés peuvent être choisis parmi la L-lysine, la L-citrulline, la L-thréonine et la L-histidine. Quant aux céramides, ils peuvent être constitués par des glycocéramides, en particulier d'origine végétale, par exemple de blé. Enfin, l'acide gras insaturé optionnel peut être l'acide linoléique ou l'acide oléique, de préférence sous forme d'ester avec l' alpha -tocophérol, ou bien un ester d'alkyle comprenant de 1 à 12 atomes de carbones, tel que l'oléate de décyle ou le linoléate d'éthyle ou vitamine F. 



  Ainsi, à titre d'exemples, on mentionnera les trois compositions du complexe différentes suivantes:

  Le complexe selon l'invention a, comme mentionné précédemment, pour effet principal de restaurer et de préser ver l'hydratation des couches supérieures de l'épiderme. 



  Cet effet est le résultat d'une triple action du complexe: 



  a) Il facilite la fixation de l'eau dans les cellules du Stratum corneum, les cornéocytes. 



  b) Il améliore la structure de l'enveloppe de ces cellules cornées; en particulier, il favorise l'épaississement de cette enveloppe. 



  c) Il renforce la qualité du ciment intercellulaire qui, à l'état normal, possède une structure lipidique lamellaire. 



  On comprend ainsi que ce complexe renforce considérablement la barrière cutanée au niveau du Stratum corneum, qui constitue la partie superficielle de l'épiderme au contact des agressions extérieures. En effet, il agit d'une part au niveau des cellules elles-mêmes, en leur permettant de restaurer et de préserver l'intégrité de leur enveloppe, évitant ainsi leur déshydratation par perte d'eau normalement retenue à l'intérieur des cellules par les acides aminés, et, d'autre part, au niveau de l'espace situé entre ces cellules, en renforçant le ciment intercellulaire, ce qui permet de limiter considérablement la perte de l'eau présente dans les couches plus profondes de la peau. 


 - Mise en évidence du pouvoir hydratant du complexe: 
 



  Pour étudier l'action "in vivo" du complexe selon l'invention, on a préparé la formulation suivante, dans laquelle le complexe est dispersé dans du Solucire< TM > (exci pient commercialisé par la société ROC, Colombes, France), et additionné d' huile de germe de blé, pour des raisons de dispersion des composants actifs

 - Mode de préparation: 
 



  Les glycocéramides et le linoléate d'éthyle sont dispersés dans l'huile de germe de blé sous agitation pendant 1 heure. On ajoute au Solucire< TM > chaud (40 DEG C) la phase obtenue précédemment, sous agitation au moyen d'un agitateur Raynerie, puis progressivement on ajoute à l'ensemble l'eau déminéralisée contenant le conservateur, les acides aminés et l'extrait de Simarouba. On homogénise sous forte agitation. 


 - Tests: 
 



  Le produit a été testé sur 5 volontaires (V1 à V5) de sexe féminin, présentant une peau très sèche, d'âge moyen 29 ans, selon la protocole suivant. 



  Sur une plage de 9 cm<2> de l'avant-bras, une zone d'application est déterminée par marquage avec un feutre. Une noisette de produit correspondant à une dose normale d'application y est appliquée, puis l'application est renouvelée matin et soir pendant les 4 jours suivants. 



  Quatre heures après la première application et 4 heures après la dernière application, des mesures de constante diélectrique de la peau sont réalisées avec "un skin test" commercialisé pour ce type de mesure par la société Dermatec, Paris (France). 



  Six mesures sont réalisées, et la moyenne et l'écarttype sont calculés. L'hydratation Ht de la peau est comparée à l'hydratation initiale Hi, et l'activité (A) est calculée selon la relation

  Les résultats obtenus sont présentés dans le tableau ci-après, plus particulièrement pour les mesures effectuées 4 heures après la première application (A4heures), respectivement après un traitement de 5 jours (A5jours) avec applications biquotidiennes. Les valeurs de ces mesures sont exprimées en unité arbitraire.

  Il ressort des résultats obtenus que 4 heures après l'application de la formulation testée, contenant le complexe selon l'invention, on peut constater chez 4 des 5 sujets soumis au test une augmentation significative de l'hydratation. Après un traitement de 5 jours, la valeur de l'hydratation cutanée est augmentée clairement et de manière significative chez tous les sujets testés. 



  Le complexe selon l'invention peut donc être utilisé avantageusement pour la formulation de différentes compositions de soins cosmétiques, par exemple sous la forme d'une émulsion telle que crème hydratante ou fluide hydratant, de maquillage traitant, de fond de teint hydratant, etc. Des exemples de telles compositions sont décrits ci-après.

  Cette émulsion fluide contient des excipients pour phase grasse et phase aqueuse classiques. Elle est préparée de la façon suivante: 



  On émulsionne la phase grasse dans environ 50 g de l'excipient aqueux contenant la glycérine et le propylène glycol, en présence de l'agent tensio-actif. on incorpore ensuite sous agitation l'amidon de maïs et le complexe hydratant, puis on disperse les pigments et les parfums. On complète enfin avec le reste de l'excipient aqueux.

  Cette crème est une émulsion huile dans eau comprenant des excipients bien connus de l'homme de l'art. Elle est préparée de manière classique, suivant un procédé analogue à celui décrit ci-dessus pour la préparation de l'émulsion fluide, en commençant par émulsionner la phase grasse dans environ 40 g de l'excipient aqueux. On incorpore ensuite le complexe C3 sous agitation, et on complète enfin par les ingrédients restant. 



  
 



  The present invention relates to a complex based on Simarouba extract having an effect which promotes and maintains hydration of the epidermis, as well as cosmetic compositions for skin care containing this complex.



  The Simarouba plant is a plant of the Simaroubaceae family, among which the plants Simarouba amara Aubl., Simarouba glauca, Simarouba versicolor and Simarouba excelsa can be mentioned. An extract from the plant Simarouba amara Aubl. for example, which is a fairly widespread tree in French Guiana, and in particular the bark of this plant, is used in native medicine to treat various symptoms such as dysentery, insufficient intestinal tone and insomnia. Furthermore, the use in cosmetics of Simarouba amara extracts has been described in the French patent application published under the number FR-A1 2 699 080 in the name of LVMH RECHERCHE.

   This use aims in particular attenuation of the pigmentary spots on the skin and the strengthening of the protective function of the skin, and it originates from the demonstration of the positive action of an extract of this plant on the differentiation of keratinocytes, leading in particular to the qualitative improvement of the stratum corneum.



  However, the present inventors have found that an extract of the Simarouba plant, associated with amino acids and lipid substances such as ceramides, exerts in addition and unexpectedly effects of strengthening the hydration of the upper layers of the 'epidermis.



  Thus, the hydrating complex, the first object of this invention, comprises as active ingredients an extract of the Simarouba plant in association with amino acids and ceramides.



  A second object of this invention consists of a cosmetic composition for skin care containing the above hydrating complex, as well as an excipient acceptable from the cosmetic point of view.



  More particularly, the complex according to the invention can comprise from 5 to 20% by weight of Simarouba extract, from 55 to 85% of amino acids, from 3 to 10% of ceramides, and optionally up to 15% of unsaturated fatty acid, preferably in the form of an ester.



  According to variants of the invention, the amino acids can be chosen from L-lysine, L-citrulline, L-threonine and L-histidine. As for ceramides, they can consist of glycoceramides, in particular of vegetable origin, for example wheat. Finally, the optional unsaturated fatty acid may be linoleic acid or oleic acid, preferably in the form of an ester with alpha -tocopherol, or else an alkyl ester comprising from 1 to 12 carbon atoms, such as decyl oleate or ethyl linoleate or vitamin F.



  Thus, by way of examples, the following three different complex compositions will be mentioned:

  The complex according to the invention has, as mentioned previously, the main effect of restoring and preserving the hydration of the upper layers of the epidermis.



  This effect is the result of a triple action of the complex:



  a) It facilitates the fixation of water in the cells of the Stratum corneum, the corneocytes.



  b) It improves the structure of the envelope of these horny cells; in particular, it promotes the thickening of this envelope.



  c) It strengthens the quality of the intercellular cement which, in the normal state, has a lamellar lipid structure.



  It is thus understood that this complex considerably strengthens the skin barrier at the level of the Stratum corneum, which constitutes the superficial part of the epidermis in contact with external aggressions. Indeed, it acts on the one hand at the level of the cells themselves, allowing them to restore and preserve the integrity of their envelope, thus avoiding their dehydration by loss of water normally retained inside the cells by amino acids, and, on the other hand, at the level of the space between these cells, by strengthening the intercellular cement, which considerably limits the loss of water present in the deeper layers of the skin.


 - Demonstration of the hydrating power of the complex:
 



  To study the "in vivo" action of the complex according to the invention, the following formulation was prepared, in which the complex is dispersed in Solucire <TM> (excipient sold by the company ROC, Colombes, France), and added with wheat germ oil, for reasons of dispersion of the active components

 - Method of preparation:
 



  The glycoceramides and the ethyl linoleate are dispersed in the wheat germ oil with stirring for 1 hour. The phase obtained above is added to the hot Solucire <TM> (40 DEG C), with stirring using a Raynerie agitator, then gradually the demineralized water containing the preservative, the amino acids and the extract from Simarouba. Homogenize with vigorous stirring.


 - Tests:
 



  The product was tested on 5 volunteers (V1 to V5) of female sex, with very dry skin, average age 29 years old, according to the following protocol.



  On a range of 9 cm <2> of the forearm, an application area is determined by marking with a felt-tip pen. A small amount of product corresponding to a normal dose of application is applied to it, then the application is repeated morning and evening for the next 4 days.



  Four hours after the first application and 4 hours after the last application, dielectric constant measurements of the skin are carried out with a "skin test" marketed for this type of measurement by the company Dermatec, Paris (France).



  Six measurements are made, and the mean and standard deviation are calculated. The hydration Ht of the skin is compared to the initial hydration Hi, and the activity (A) is calculated according to the relation

  The results obtained are presented in the table below, more particularly for the measurements carried out 4 hours after the first application (A4 hours), respectively after a treatment of 5 days (A5 days) with twice daily applications. The values of these measurements are expressed in arbitrary units.

  It appears from the results obtained that 4 hours after the application of the tested formulation, containing the complex according to the invention, one can note in 4 of the 5 subjects subjected to the test a significant increase in hydration. After a 5-day treatment, the value of skin hydration is clearly and significantly increased in all the subjects tested.



  The complex according to the invention can therefore be advantageously used for the formulation of various cosmetic care compositions, for example in the form of an emulsion such as a moisturizing cream or hydrating fluid, treating makeup, moisturizing foundation, etc. Examples of such compositions are described below.

  This fluid emulsion contains conventional fatty phase and aqueous phase excipients. It is prepared as follows:



  The fatty phase is emulsified in about 50 g of the aqueous excipient containing the glycerin and the propylene glycol, in the presence of the surfactant. the corn starch and the hydrating complex are then incorporated with stirring, and the pigments and fragrances are dispersed. Finally, complete with the rest of the aqueous excipient.

  This cream is an oil-in-water emulsion comprising excipients well known to those skilled in the art. It is prepared in a conventional manner, according to a process analogous to that described above for the preparation of the fluid emulsion, starting by emulsifying the fatty phase in approximately 40 g of the aqueous excipient. The C3 complex is then incorporated with stirring, and finally the remaining ingredients are completed.


    

Claims (10)

1. Complexe hydratant comportant comme ingrédients actifs un extrait de plante Simarouba en association avec des acides aminés et des céramides.     1. Hydrating complex comprising as active ingredients an extract of Simarouba plant in association with amino acids and ceramides. 2. Complexe selon la revendication 1, caractérisé en ce qu'il comporte en plus un acide gras insaturé ou un ester de celui-ci. 2. Complex according to claim 1, characterized in that it additionally comprises an unsaturated fatty acid or an ester thereof. 3. Complexe selon la revendication 1 ou la revendication 2, caractérisé en ce qu'il comporte de 5 à 20% en poids d'extrait de Simarouba, de 55 à 85% d'acides aminés, de 3 à 10% de céramides, et éventuellement jusqu'à 15% d'acide gras insaturé ou d'ester de celui-ci. 3.
