Gemäss der vorliegenden Erfindung gelangt man zu den erfindungsgemässen Verbindungen, indem man in einem entsprechenden 8-CH2-Y-Derivat, worin Y für einen austauschbaren Rest steht. Die erfindungsgemässe Umsetzung kann analog zu bekannten Methoden für die Herstellung ähnlicher Verbindungen erfolgen.
Der austauschbare Rest Y steht z.B. für Halogen, wie Chlor oder Brom oder für einen Rest -O-SO2-R, worin R niederes Alkyl oder gegebenenfalls substituiertes Phenyl bedeutet.
Die Umsetzung zwischen den Verbindungen der Formeln II und IV wird zweckmässig in einem Lösungsmittel durchgeführt. Besonders geeignet sind unter den Reaktionsbedingungen inerte, aprotische, polare Lösungsmittel, beispielsweise Amide organischer Carbonsäuren wie Dimethylformamid oder aber auch Hexamethylphosphorsäuretrisamid oder Acetonitril, gegebenenfalls im Gemisch mit einem geringen Anteil Wasser.
Man arbeitet vorzugsweise bei erhöhterTemperatur. beispielsweise zwischen etwa 50 bis 100 .
Die Reaktion wird zweckmässig unter Sauerstoffausschluss, beispielsweise unter Stickstoffatmosphäre vorgenommen.
Man setzt mit Vorteil einen Überschuss an Verbindung der Formel III ein, beispielsweise etwa 2 bis 10 Mol Verbindung der Formel 111 pro Mol Verbindung der Formel II.

Die Verbindungen der Formel II, worin Rt für Wasserstoff steht, können auch hergestellt werden, indem man auf an sich bekannte Weise in einem 6-Alkyl-ergolin-8ss-carbonsäu- realkylester das Stickstoff in Stellung 1 z.B. mit einer Benzoxycarbonylgruppe schützt, die 2-Methylgruppe einführt und die Schutzgruppe entfernt, wobei die 6-Alkyl-2-methyl ergolin-8-carbonsäure entsteht, die in die gewünschte Verbindung der Formel II überführt wird.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die erfindungsgemässen Verbindungen in freier Form bzw. in Form von Additionssalzen mit physiologisch verträglichen Säuren, weisen im Tierversuch interessante pharmakodynamische Eigenschaften auf. Sie können als Heilmittel verwendet werden.
Die erfindungsgemässen Verbindungen bewirken im Beobachtungstest ab 3 mg/kg p.o. (bei der Maus) oder mit 30 mg/kg i.p. (bei der Ratte) eine Verlängerung des Wachzustandes sowie eine erhöhte Reaktivität auf externe Reize.
Ausserdem verstärken sie in vitro bei 0,1 mg/ml den elektrisch induzierten Acetylcholin-Release aus Hippocampusschnitten der Ratte (Serotoninagonistische Wirkung).
Aufgrund dieser Aktivitäten können die erfindungsge mässen Verbindungen für die Indikation senile Demenz Anwendung finden.
Ferner bewirken sie im Versuch des über 48 Stunden kontinuierlich abgeleiteten EEG bei der Ratte (H. Kleinlogel et al., Waking and Sleeping 1980, 4, S.77-85), mit Dosen von 3 bis 30 mg/kg p.o., eine Hemmung des Paradoxalschlafes ohne darauf folgende Zunahme (rebound effect).
Aufgrund dieser Wirkung können sie als Antidepressiva verwendet werden.
Die erfindungsgemässen Verbindungen antagonisieren überdies in vitro bei 1 mg/l die Apomorphinwirkung auf den elektrisch stimulierten Acetylcholin-Release aus Striatumschnitten der Ratte [Methode nach R. Markstein. J. Neural Transmission 51, 39-59(1981)].
Im Striatum bewirken sie bei 10 mg/kg p.o. eine starke Erhöhung an beiden Dopamin-Metaboliten 3,4-Dihydroxyphenylessigsäure und Homovanillinsäure (DOPAC und HVA) [für die quantitative Bestimmung der Parameter siehe F. Karoum et al., Europ. J. Pharmacol. 44, 311-318 (1977) und H.R. Bürki et al., Psychopharmacology 57, 227-237 (1978)]. Gleichzeitig wird bei derselben Dosis die Aktivität der Tyrosinhydroxylase in vivo erhöht (Methode nach A. Carlson et al.. Naunyn-Schmiedeberg's Arch. Pharmacol.
275. 163-168;. (Diese Eigenschaften sprechen für eine dopa minantagonistische Wirkung.) Aufgrund dieser Aktivitäten können die erfindungsgemässen Verbindungen als Neuroleptika, insbesondere für die Indikation Schizophrenie Anwendung finden.
Ferner verstärken sie bei 0,01-0,1 mg/kg i.v. oder If) mg ski p.o. die '4C-Deoxyglucose-Aufnahme in bestimmten Gehirnarealen, insbesondere den lateralen Habenulakernen (Methode nach L. Sokoloff, Journal of Cerebral Blood Flow and Metabolism 1981, 1,7-36, H.E. Savaki et al., Brain Research 1982,233, 347 und J. McCulloch et al., Journal of Cerebral Blood Flow and Metabolism 1981, 1, 133-136).
Aufgrund dieser Aktivitäten können sie für die Indikation zerebrale Insuffizienz Anwendung finden.
Überdies bewirken sie vasokonstriktorische Aktivität, welche in vitro an Spinalstreifen der Arteria carotis externa des Hundes [E. Müller-Schweinitzer, Naunyn-Schmiedeberg's Arch. Pharmacol., 292, 113-118(1976)] mit Dosen ab 10 nM/Liter gezeigt werden kann.
Aufgrund dieser Wirkung können die erfindungsgemässen Verbindungen zur Behandlung der Migräne verwendet werden.
Für die oben genannten Anwendungen variiert die zu verwendende Dosis selbstverständlich je nach verwendeter Sub stanz, Art der Administration und der gewünschten Behandlung. Im allgemeinen werden aber befriedigende Resultate mit Dosen von ungefähr 0,1 bis 10 mg/kg Körpergewicht erreicht; die Administration kann mit einer Dosis täglich vorgenommen werden oder nötigenfalls in mehreren Teildosen erfolgen. Für grössere Säugetiere liegt die Tagesdosis im Bereich von etwa 1 bis 100 mg der Substanz, geeignete Dosierungsformen für z.B. orale Anwendungen enthalten im allgemeinen ungefähr 0,3 bis 50 mg wirksame Substanz neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Gegenstand der Erfindung sind auch pharmazeutische Zusammensetzungen, die die erfindungsgemässen Verbindungen enthalten. Für ihre Herstellung können die in der Pharmazie gebräuchlichen Hilfs- und Trägerstoffe verwendet werden.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Die in diesen Beispielen aufgeführten Verbindungen können analog dem nachfolgenden. für die Herstellung des 7.6-Dimethyl-88-(2-pyridyl-thiomethyl)-9 1 O-didehydroergo- lins beschriebenen Verfahrens hergestellt werden.
2,6-Dimethyl-8ss-(2-pyridyl-thiomethyl)-9,10-didehydroer- golin a) 2-( 1.3-Dithian-2-yl)-lysergolmesylat
Zu einer Lösung von 4,2 g (33,9 mM) Dithian in 100 ml Chloroform werden innerhalb 30 Minuten bei -30" 2,9 ml (35,6 mM) Sulfurylchlorid in 15 ml Chloroform zugetropft.
Nachdem die Temperatur auf 20 gestiegen ist, wird 30 Minuten bei 20 gerührt. Bei - 100 werden 7,5 g (22,6 mM) Lysergolmesylat in 100 ml Chloroform zugegeben. Nachdem die Temperatur des Reaktionsgemisches auf 20 gestiegen ist, wird 30 Minuten bei dieser Temperatur gerührt. Zur Aufarbeitung wird mit Eis/Wasser versetzt, mit 2 N Sodalösung neutralisiert und mit Methylenchlorid, enthaltend 10% Isopropanol, extrahiert. Die vereinigten organischen Phasen werden mit Natriumsulfat getrocknet, filtriert und eingedampft. Es resultieren 1 log Rohprodukt, die an Kieselgel mit Methylenchlorid, enthaltend 2% Methanol, chromatographiert werden. wobei 4.3 g der Titelverbindung nebst 2,7 g Lysergolmesylat eluiert werden.
b) 2-Methyl-lysergolmesylat
100 ml einer Suspension von Raney-Nickel in Wasser wird 4mal mit je 100 ml Dimethylformamid/Aceton (1:4) gewa- schen. Der gewaschene Raney-Nickel wird unter Argon mit 115 ml Dimethylformamid/Aceton (1:4) versetzt. Zur gerührten Suspension wird bei Zimmertemperatur eine Lösung von 4,3 g (9,6 mM) 2-(1,3-Dithian-2-yl)-lysergolme- sylat in 80 ml Dimethylformamid/Aceton (1:4) gegossen und 2% Stunden bei Zimmertemperatur gerührt.
Anschliessend wird filtriert und der Filterrückstand 2mal mitje 115 ml Dimethylformamid/Aceton (1:2) gewaschen. Nach dem Eindampfen des Filtrates und Trocknen am Hochvakuum resultieren 2 g der rohen Titelverbindung, die an 90 g Kiegelgel mit Methylenchlorid, enthaltend 2% Methanol, chromatographiert werden. Es werden 1,3 g der Titelverbindung als beiger Schaum eluiert.
c) 2,6-Dimethyl-8(2-pyridyl-thiomethyl)-9 1 0-didehydro- ergolin
Eine Lösung von 1.3 g (3,75 mM) 2-Methyl-lysergolmesylat und 1 g (9 mM) 2-Mercapto-pyridin in 20 ml Dimethylformamid wird mit 4,5 ml 2 N Natronlauge versetzt und 18 Stunden bei Zimmertemperatur gerührt. Der Ansatz wird mit Wasser versetzt und mit Methylenchlorid. enthaltend 10% Isopropanol, extrahiert. Die vereinigten organischen Phasen werden mit Natriumsulfat getrocknet, filtriert, eingedampft und am Hochvakuum getrocknet. Das Rohprodukt wird an 120 g Kieselgel mit Chloroform, enthaltend 2% Methanol und 0,1% konz. NH3, chromatographiert. Es resultieren 1,3 g der Titelverbindung. Das Hydrochlorid der Titelverbindung kristallisiert aus Äthanol beim Einengen.
