Also preferred are the compounds in which Y 'is hydrogen and Y2 is methyl, in particular those having the R configuration on the C atom bearing the radicals Y' and Y2; as well as the compounds in which Z is optionally m-chlorinated or m-trifluoromethylated phenyl or phenoxymethyl, in particular those with an R configuration on the C atom bearing the group Zl and the compounds in which Rl is hydrogen and R2 and R are carbamoyl, 2-carbomethoxy or 2-Carbäthoxy- I-methylvinyl are.
The alkyl, alkoxy and alkanoyl radicals mentioned here are preferably lower radicals having 1 to 6, in particular 1 to 4, carbon atoms. These residues can be straight-chain or branched. Examples of lower alkyl and lower alkoxy radicals are methyl, ethyl, propyl, isopropyl, n-butyl and isobutyl or methoxy, ethoxy, propoxy, isopropoxy, n-butoxy and
Isobutoxy. Alkanoyloxy radicals are preferably derived from lower alkane carboxylic acids, such as formic acid, acetic acid,
Propionic acid and butyric acid. Halogen is preferred
Chlorine or bromine.
The compounds of formula I form acid addition salts with acids, which are also the subject of the invention.
Examples of such salts are salts with physiologically compatible mineral acids, such as hydrochloric acid, hydrobromic acid,
Sulfuric acid, phosphoric acid; or with organic acids, such as methanesulfonic acid, acetic acid, propionic acid, citric acid, oxalic acid, succinic acid, malic acid, fumaric acid,
Phenylacetic acid or salicylic acid. Carboxylic acids of formula I can be present as salts. Examples of such salts are alkali metal, alkaline earth metal, ammonium and alkyl ammonium salts such as Na, K, Ca, trimethyl ammonium and ethanol ammonium salts.
The compounds of formula I contain at least one asymmetric carbon atom and can thus be present as optically active enantiomers, as diastereomers or as race mates.
The compounds of formula I can be obtained according to the invention by reacting an amine
formula
EMI2.1
wherein m, Y ', Y2, Zl and Z2 have the meaning given above, with a compound of the formula HC (O) X, in which X is hydrogen or alkoxycarbonyl, and if desired converting a compound of the formula I obtained into a salt.
The reaction of an amine of the formula II with a compound of the formula HC (O) X is advantageously carried out in a solvent, preferably an aromatic hydrocarbon, such as benzene or toluene, and at room temperature or at elevated temperature, preferably by azeotropic distillation of the reaction mixture.
The compounds of formula II can be obtained by a) an epoxide of the formula
EMI2.2
or a p-keto halide of the formula Zl-C (O} CH2-Hal III-2 with an amine of the formula
EMI2.3
implements, or b) a compound of one of the formulas
EMI2.4
reduced, with the symbols Y ', Y2, Zl, Z2 and m in the formulas above having the meaning given earlier.
The reaction of a compound of formula III-1 or III-2 with a compound of formula IV can be carried out in an inert organic solvent such as an alkanol, e.g.
Ethanol. The reaction temperature is not critical, it can be between room temperature and the reflux temperature of the reaction mixture.
The reduction of a compound of formula V, VI and VII can be conveniently carried out with a complex metal hydride such as NaBH4 in an inert organic solvent such as an alkanol e.g. Methanol, be carried out at temperatures of 20-30 "C.
A suitable complex metal hydride for the reduction of the compounds V is NaBH4. The compounds VI are expediently reduced with LiAIH4.
The oxazolidines of the formula I and the physiologically tolerable salts thereof can be used as active ingredients in pharmaceutical preparations for the treatment of obesity and / or diabetes mellitus, in particular of the above adult diabetic. In animal experiments, increased catabolism, especially fats, was observed when compounds of the formulas were administered.
The compounds were also observed to stimulate the formation of brown adipose tissue in rats and obes hyperglycemic mice. Defects in brown adipose tissue are known to play an essential role in the development of obesity. In obes hyperglycemic mice, the compounds have a pronounced antidiabetic effect in that they have a hypoglycemic effect and reduce glycosuria. These compounds have little effect on cardiac activity and circulation. The dosage can be 0.5-1000 mg, preferably 2-200 mg per day for an adult, depending on the potency of the individual compounds and the individual needs of the patient, the dose being administered as a single dose or in multiple doses throughout the day can be.
Furthermore, an increase in the body protein content and a decrease in the fat content could be demonstrated with compounds of the formula I in animal experiments. These compounds therefore lead to an increase in lean body mass at the expense of fat. Therefore, they can first be used in human medicine to treat conditions associated with increased protein breakdown, e.g. for convalescence after surgery. The doses of administration are in the same range as in the treatment of obesity and / or diabetes mellitus.
The above compounds can also be used in the diet of fattening animals such as cattle, pigs, sheep and poultry. The doses and forms of administration can be the same as for vitamins. These compounds can also be used as feed additives in doses of 0.01-100 mg / kg depending on the substance, species and age.
The pharmaceutical preparations contain the active ingredient together with a compatible pharmaceutical, organic or inorganic carrier material, such as e.g. Water, gelatin, gum arabic, lactose, starch, magnesium stearate, talc, vegetable oils, polyalkylene glycols, petroleum jelly and the like. The pharmaceutical preparations are preferably administered orally, e.g. administered in the form of tablets, capsules, pills, powders, granules, solutions, syrups, suspensions, elixirs and the like. Administration can also be parenteral, e.g. in the form of sterile solutions, suspensions or emulsions. The pharmaceutical preparations can be sterilized and / or contain components such as preservatives, stabilizers, wetting agents, emulsifiers, salts to vary the osmotic pressure, and buffer substances.
The activity of compounds of formula I is clear from the following test results: 1) Effect on deti oxygen consumption
Male albino rats weighing 160-180 g were placed in metabolic cages after fasting for 24 hours.
The cages were ventilated with 6 liters of room air / minute, which was equilibrated at a dew point of 11 "C. After re-equilibration, samples were collected from the exhaust air for periods of 14 minutes each and the oxygen and CO2 content was analyzed After an adaptation time of 4 hours, the animals divided into groups of 6 received either placebo (5% gum arabic) or the test substance (suspended in 5% gum arabic) per os. The determinations were then carried out for 12 hours.
The table shows the percentage of the mean oxygen consumption after medication during the first 3 hours and the entire test period (12 hours) of the oxygen consumption during the adjustment period, taking into account corrections for changes in the placebo group.
