REVENDICATIONS
1. Procédé de préparation d'ester méthylique d'a-L-aspartyl-Lphénylalanine, caractérisé en ce qu'il consiste à faire réagir l'ester méthylique d'a-L-N-formylaspartyl-L-phénylalanine avec l'acide hydroxylamine-O-sulfonique.
2. Procédé selon la revendication 1, caractérisé en ce qu'on utilise l'acide hydroxylamine-O-sulfonique à raison de 0,5 à 1,5 mol par mole d'ester méthylique d'a-L-N-formylaspartyl-L-phénylalanine.
3. Procédé selon les revendications 1 et 2, caractérisé en ce qu'on effectue la réaction dans un alcool aliphatique aqueux comportant 1 à 5 atomes de carbone.
4. Procédé selon les revendications 1 et 2, caractérisé en ce qu'on effectue la réaction dans l'acétonitrile.
5. Procédé selon les revendications I à 4, caractérisé en ce qu'on utilise les solvants organiques aqueux dans les proportions de 50 à 95% par rapport à l'eau.
6. Procédé selon les revendications 1 à 5, caractérisé en ce qu'on améne la réaction à la température de 30 à SOC.
La présente invention concerne la préparation de l'édulcorant aspartame: L'ester méthylique d'a-L-aspartyl-L-phénylalanine, et en particulier un procédé pour éliminer le radical formyl de protection dudit composé.
L'emploi de l'acide formique dans le blocage de la fonction aamino des acides aminés et peptides est bien connu et posséde de nombreux avantages: généralement, ces N-formylaminopeptides cristallisent bien, leur synthése ne provoque pas de racémisation, s'agissant d'un radical de petite taille, on n'entraîne pas de ballast volumineux au cours de la synthèse, enfin, de nombreuses possibilités existent pour son élimination.
Dans le cas particulier de l'aspartame, s'y ajoute encore un autre avantage, soit le fait que la séparation des isomères a et B - qui se forment inévitablement au cours de la synthèse - est simple et pra- tiquement quantitative sous forme de N-formylaspartame.
Pour le déblocage du radical formyl, différentes méthodes sont connues. Ainsi, la saponification avec des acides forts comme décrite - entre autres - dans le brevet français N" 2320934. Toutefois, le désavantage de cette méthode subsiste, étant donné que d'autres liaisons fragiles (ester, peptide) sont aussi attaquées, même si on utilise l'acide dans des proportions sensiblement équimolaires, et que l'on travaille à des conditions douces. Ainsi, la séparation du produit final est laborieuse, entraîne différentes purifications, qui diminuent considérablement le rendement final.
Selon le brevet américain N" 4021418, on élimine la protection en faisant réagir le N-formylaspartame avec des sels d'hydroxylamine qu'on utilise dans un excès molaire de 50 à 600% - favorablement à 400% - sous la forme d'un mélange des sels d'un acide fort et d'un acide faible. Cette méthode est sélective, les réactions secondaires sont moindres mais, à cause de la grande quantité de sels,
I'isolation de l'aspartame devient compliquée.
La titulaire a découvert qu'on peut éliminer le groupe de blocage de façon simple et avec un rendement avantageux en faisant réagir le
N-formyl-a-aspartame avec l'acide hydroxylamine-O-sulfonique. La quantité nécessaire de ce réactif est surprenante: pour 1 mol de Nformyl-a-aspartame, elle se situe entre 0,5 et 1,5 mol. En effet, cet ester réunit favorablement le mode d'action des deux méthodes susmentionnées. Ainsi, les réactions secondaires sont peu importantes,
I'isolation du produit désiré est simple, sa pureté est remarquable.
Avantageusement, on effectue ladite réaction dans un mélange d'un solvant organique et d'eau. Les solvants organiques préférés sont les alcools aliphatiques comportant 1 à 5 atomes de carbone ou l'acétonitrile. Les alcools particulièrement favorables sont l'iso- propanol et le butanol tertiaire. Le rapport approprié du solvant organique à l'eau se situe entre 50 et 95%.
La température du mélange agit sur la vitesse de réaction de façon habituelle; elle est tenue entre 30 et SOC, en général. Toutefois, on obtient de meilleurs rendements en effectuant la réaction entre 50 et 60 C, pendant 8 à 15 h.
Une forme de mise en oeuvre du procédé particulièrement avantageuse consiste à faire réagir le N-formyl-a-aspartame avec l'acide hydroxylamine-O-sulfonique dans une suspension d'un solvant organique aqueux, de filtrer à la fin la solution trouble, de concentrer les eaux mères sous vide - à la moitié du volume initial - à une température inférieure à 30 C, de diluer ce concentrat avec de l'eau, puis de précipiter l'aspartame en ramenant le pH de la solution ainsi obtenue de 0,8 à 1,2 à la valeur de 4,8 à 5,0 avec une base.
Cette neutralisation peut s'effectuer avec des bases inorganiques ou organiques couramment employées, telles que NaOH, KOH, NH40H, triéthylamine, pyridine, etc.
L'aspartame ainsi obtenu peut être purifié selon les méthodes usuelles, telles que recristallisation ou délayage ou par formation d'un sel et reprécipitation de la base.
La présente invention est illustrée plus en détail par les exemples ci-après qui ne la limitent aucunement.
Exemples:
1. On mélange 32,2 g (0,1 mol) d'ester méthylique de N-formyl a-L-aspartyl-L-phénylalanine et 12,5 g d'acide hydroxylamine-Osulfonique à 90% (0,1 mol) et l'on y ajoute 170 ml d'isopropanol à 80%. On brasse le mélange réactionnel 15 h à 50"C, puis on soumet un échantillon pour une analyse chromatographique sur couche mince (CCM). Selon le résultat obtenu, I'échantillon contient encore 3-5% de produit non déformylé, 5% d'acide 5-phényl-1,4-dicétopipérazinyl-2-acétique, diverses impuretés de l'ordre de 5%; le reste est transformé en aspartame.
Pour l'isoler, on concentre le mélange à 100 g, sous vide, à une température inférieure à 30 C, ajoute 100 ml d'eau (pH de la solution: 1,08), traite au charbon, puis on règle le pH avec NH40H à 25% jusqu'à 4,8-5,0 où l'aspartame précipite. On refroidit ce précipité 2 h à 0-59C, on le filtre et sèche. On obtient ainsi 22,9 g d'aspartame; rendement: 78%.
2. On fait réagir 9,6 g (0,03 mol) d'ester méthylique de Nformyl-a-L-aspartyl-L-phénylalanine avec 2,0 g d'acide hydroxylamine-O-sulfonique à 90% (0,016 mol) dans 30 ml d'isopropanol à 50% pendant 12h à 50"C. La CCM démontre, à part l'aspartame, 10% de N-formylaspartame, plus au total 3% d'impuretés.
3. On brasse à 50" C 19,2 g (0,06 mol) d'ester méthylique de N- formyl-a-L-aspartyl-L-phénylalanine et 6,8 g d'acide hydroxylamine-O-sulfonique à 90% (0,054 mol) dans 100 ml de butanol tertiaire à 60% pendant 8 h. Selon l'analyse par CCM, la déformylation est à 90%; mis à part 10% de N-formylaspartame, les impuretés sont imperceptibles.
4. On chauffe à 50-C 32,2 g (0,1 mol) d'ester méthylique de Nformyl-a-L-aspartyl-L-phénylalanine et 15,3 g d'acide hydroxylamine-O-sulfonique à 90% (0,135 mol) dans 120 ml d'acétonitrile à 90% pendant 15 h. On obtient ainsi une suspension; on filtre les sels formés. La détermination par CCM, effectuée sur les eaux mères, dé montre - à part l'aspartame - la présence de 4% d'acide 5-phényl- 1,4-dicétopipérazinyl-2-acètique, 8% de N-formylaspartame, et des impuretés diverses de 5%. Après concentration des eaux mères, on dilue avec 100 ml d'eau, puis on précipite l'aspartame en ramenant le pH à la valeur de 4,8-5,0 avec une solution de NaOH à 30%. L'aspartame ainsi obtenu pèse 22,5 g et correspond à un rendement de 76,4%.

CLAIMS
1. Process for the preparation of a-L-aspartyl-Lphenylalanine methyl ester, characterized in that it consists in reacting the a-LN-formylaspartyl-L-phenylalanine methyl ester with hydroxylamine acid O-sulfonic.
2. Method according to claim 1, characterized in that hydroxylamine-O-sulfonic acid is used in an amount of 0.5 to 1.5 mol per mole of a-LN-formylaspartyl-L- methyl ester phenylalanine.
3. Method according to claims 1 and 2, characterized in that the reaction is carried out in an aqueous aliphatic alcohol having 1 to 5 carbon atoms.
4. Method according to claims 1 and 2, characterized in that the reaction is carried out in acetonitrile.
5. Method according to claims I to 4, characterized in that the aqueous organic solvents are used in the proportions of 50 to 95% relative to water.

The present invention relates to the preparation of the aspartame sweetener: The methyl ester of a-L-aspartyl-L-phenylalanine, and in particular a process for eliminating the formyl protecting radical of said compound.
The use of formic acid in blocking the amino function of amino acids and peptides is well known and has many advantages: generally, these N-formylaminopeptides crystallize well, their synthesis does not cause racemization, as regards 'a radical of small size, one does not involve bulky ballast during the synthesis, finally, many possibilities exist for its elimination.
In the particular case of aspartame, there is yet another advantage, that the separation of the isomers a and B - which inevitably form during synthesis - is simple and practically quantitative in the form of N-formylaspartame.
Different methods are known for unblocking the formyl radical. Thus, saponification with strong acids as described - among others - in French patent N "2320934. However, the disadvantage of this method remains, given that other fragile bonds (ester, peptide) are also attacked, even if the acid is used in substantially equimolar proportions, and that the work is carried out under mild conditions, thus the separation of the final product is laborious, results in various purifications, which considerably reduce the final yield.
According to US Patent No. 4021418, the protection is eliminated by reacting N-formylaspartame with hydroxylamine salts which are used in a molar excess of 50 to 600% - favorably 400% - in the form of a mixture of the salts of a strong acid and a weak acid. This method is selective, the side reactions are less but, because of the large quantity of salts,

Advantageously, said reaction is carried out in a mixture of an organic solvent and water. The preferred organic solvents are aliphatic alcohols having 1 to 5 carbon atoms or acetonitrile. Particularly favorable alcohols are isopropanol and tertiary butanol. The appropriate ratio of organic solvent to water is between 50 and 95%.
The temperature of the mixture acts on the reaction rate in the usual way; it is held between 30 and SOC, in general. However, better yields are obtained by carrying out the reaction between 50 and 60 C, for 8 to 15 h.
A particularly advantageous embodiment of the process consists in reacting N-formyl-a-aspartame with hydroxylamine-O-sulfonic acid in a suspension of an aqueous organic solvent, at the end filtering the cloudy solution, to concentrate the mother liquors under vacuum - to half the initial volume - at a temperature below 30 C, to dilute this concentrate with water, then to precipitate the aspartame by reducing the pH of the solution thus obtained from 0 , 8 to 1.2 to the value of 4.8 to 5.0 with a base.
This neutralization can be carried out with commonly used inorganic or organic bases, such as NaOH, KOH, NH40H, triethylamine, pyridine, etc.
The aspartame thus obtained can be purified according to the usual methods, such as recrystallization or dilution or by formation of a salt and reprecipitation of the base.
The present invention is illustrated in more detail by the following examples which do not limit it in any way.
Examples:
1. 32.2 g (0.1 mol) of methyl ester of N-formyl aL-aspartyl-L-phenylalanine and 12.5 g of hydroxylamine-Osulfonic acid at 90% (0.1 mol) are mixed and 170 ml of 80% isopropanol are added thereto. The reaction mixture is stirred for 15 h at 50 ° C., then a sample is submitted for a thin layer chromatographic analysis (TLC). Depending on the result obtained, the sample still contains 3-5% of non-deformylated product, 5% of 5-phenyl-1,4-diketopiperazinyl-2-acetic acid, various impurities of the order of 5%; the rest is transformed into aspartame.
To isolate it, the mixture is concentrated to 100 g, under vacuum, at a temperature below 30 C, added 100 ml of water (solution pH: 1.08), treated with charcoal, then the pH is adjusted with NH40H at 25% up to 4.8-5.0 where the aspartame precipitates. This precipitate is cooled for 2 h to 0-59C, filtered and dried. 22.9 g of aspartame are thus obtained; yield: 78%.
2. 9.6 g (0.03 mol) of Nformyl-aL-aspartyl-L-phenylalanine methyl ester are reacted with 2.0 g of hydroxylamine-O-sulfonic acid at 90% (0.016 mol) in 30 ml of 50% isopropanol for 12 hours at 50 ° C. The TLC demonstrates, apart from aspartame, 10% of N-formylaspartame, plus a total of 3% of impurities.
3. 19.2 g (0.06 mol) of methyl ester of N-formyl-aL-aspartyl-L-phenylalanine and 6.8 g of hydroxylamine-O-sulfonic acid at 90% are stirred at 50 ° C. (0.054 mol) in 100 ml of tertiary butanol at 60% for 8 hours According to the analysis by TLC, the deformylation is at 90%, apart from 10% of N-formylaspartame, the impurities are imperceptible.
4. 32.2 g (0.1 mol) of Nformyl-aL-aspartyl-L-phenylalanine methyl ester and 15.3 g of hydroxylamine-O-sulfonic acid at 90% (0.135) are heated to 50 ° C. mol) in 120 ml of 90% acetonitrile for 15 h. A suspension is thus obtained; the salts formed are filtered. The determination by TLC, carried out on the mother liquors, shows - apart from aspartame - the presence of 4% of 5-phenyl-1,4-diketopiperazinyl-2-acetic acid, 8% of N-formylaspartame, and miscellaneous impurities of 5%. After concentrating the mother liquors, dilute with 100 ml of water, then precipitate the aspartame by bringing the pH to the value of 4.8-5.0 with a 30% NaOH solution. The aspartame thus obtained weighs 22.5 g and corresponds to a yield of 76.4%.
