Das erfindungsgemässe Verfahren kann nach an sich zur Alkylierung des Indol-Stickstoffs üblichen Methoden durchgeführt werden. Beispielsweise kann die Verbindung der Formel II mit Verbindungen der Formel III,

worin X für den Säurerest eines reaktionsfähigen Esters, vorzugsweise für Halogen oder einen organischen Sulfonsäurerest steht, auf an sich bekannte Weise umgesetzt werden.
Die Umsetzung erfolgt vorzugsweise in einem inerten organischen Lösungsmittel, bei Temperaturen zwischen ca. -40 und + 100"C, in Gegenwart einer Base.
Die erhaltene Verbindung der Formel I kann auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden.
Die freie Base kann in bekannter Weise in Säureadditionssalze umgewandelt werden und umgekehrt.
Die Ausgangsverbindung der Formel II ist bekannt.
Die Verbindung der Formel I fällt unter den allgemeinen Schutzumfang der Deutschen Offenlegungsschrift 26 56 344, ist aber bisher noch nicht spezifisch beschrieben worden.
Die Verbindung der Formel I und ihre Säureadditionssalze zeichnen sich im Tierversuch im Vergleich zu den bekannten Verbindungen der Deutschen Offenlegungsschrift 2656344 durch ein besonders interessantes pharmakologisches Wirkungsprofil aus und können daher als Heilmittel verwendet werden.
Insbesondere zeigen diese Verbindungen eine dopaminrezeptorenstimulierende Wirkung. Die dopaminergen Eigenschaften konnten an Ratten, bei denen durch eine 6-Hydroxydopamin-Injektion in die substantia nigra eine unilaterale Verletzung der nigro-neostriatalen Dopaminbahn erzeugt wurde, mit Dosen zwischen etwa 0,05 bis 3 mg/kg festgestellt werden [Methode nach U. Ungerstedt, Acta physiol. scand.
Suppl. 367, 69-93 (1973)1. Nach Verabreichung des Wirkstoffes war eine deutliche Aktivierung dadurch erkennbar, dass die Ratten in Richtung der nicht denervierten Seite rotierten.
Die Verbindungen können aufgrund ihrer dopaminergen Eigenschaften zur Behandlung von Parkinsonismus Anwendung finden.
Ferner besitzen die Verbindungen eine Prolaktinsekretionshemmende Wirkung. So hemmen sie bei der Ratte die Implantationen nach subkutaner Applikation von Dosen zwischen 0,03 und 3 mg/kg [Lit.: Experientia 34, 1330(1978)1 und bei der männlichen Ratte die Prolaktinsekretion mit Dosen von 0,05 bis 5 mg/kg p.o. [Lit.: Flückiger et al., Postgraduate Med. Journal, 52 Suppl. 1,57(1976)].
Die Verbindungen können aufgrund ihrer prolaktinsekre
tionshemmenden Wirkung zur Behandlung der Hyperprolactinämie Anwendung finden.
Die Verbindung der Formel I bewirkte im o.e. Versuch nach Ungerstedt 1608 Umdrehungen nach 3 mg/kg p.o. Im o.e. Implantationen-Versuch und im Versuch bei der männlichen Ratte weist sie eine EDso von 0,08 mg/kg s.c. bzw.
0,22 mg/kg s.c. auf. Ferner blieb sie im Infusionsversuch bei der Katze ohne nennenswerten Einfluss auf den Blutdruck.
Diese Ergebnisse deuten auf eine besonders selektive dopaminerge Wirkung.
Für die oben genannten Anwendungen variiert die zu verwendende Dosis selbstverständlich je nach Art der Administration und der gewünschten Behandlung. Im allgemeinen werden aber befriedigende Resultate mit Dosen von ungefähr 0,1 bis 10 mg/kg Körpergewicht erreicht; die Administration kann mit einer Dosis täglich vorgenommen werden oder nötigenfalls in mehreren Teildosen erfolgen. Für grössere Säugetiere liegt die Tagesdosis im Bereich von etwa 0,5 bis 100 mg der Substanz, geeignete Dosierungsformen für z.B. orale Anwendungen enthalten im allgemeinen ungefähr 0,3 bis 10 mg wirksame Substanz neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Als Heilmittel kann die Verbindung der Formel I bzw. ihre physiologisch verträglichen Salze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Stoffen verabreicht werden.
Das nachfolgende Beispiel erläutert die Erfindung. Temperaturangaben erfolgen in Celsiusgraden und sind unkorrigiert.
Beispiel
N,N-Diäthyl-N' -[(8a)- 1 -isopropyl-6-methyl-ergolin-8-yl]- sulfamid
Zu einem Gemisch von 50 mg (0,31 mmol) Eisentrichlorid, 11,28 ml (120 mmol) Tertiärbutylalkohol und 40 ml flüssigem Ammoniak wurden unter Rühren und Trockeneiskühlung portionenweise insgesamt 2,53 g (110 mA) Natriummetall und anschliessend 7,53 g (20 mmol) N,N-Diäthyl-N'-[(8a)-6methyl-ergolin-8-yl]-sulfamid gegeben. Unter Rückfluss wurden innert 4 Stunden 11,0 ml (10 mmol) Isopropyljodid zugetropft und das Reaktionsgemisch anschliessend 3 Stunden unter Rückfluss weitergerührt.
Über Nacht liess man unter Rühren das Ammoniak abdampfen und das Reaktionsgemisch auf Zimmertemperatur aufwärmen. Nach Verteilung zwischen 110 ml 2 M wässeriger Ammoniumsulfat-Lösung und Methylenchlorid, Nachextrahieren, Waschen und Trocknen mit Natriumsulfat, Eindampfen der organischen Phasen und Chromatographie an 170 g Kieselgel mit Methylenchlorid/Methanol/konz.
wässerigem Ammoniak (98/1,8/0,2) wurde aus 2-Propanol kristallisiert.

To a mixture of 50 mg (0.31 mmol) of iron trichloride, 11.28 ml (120 mmol) of tertiary butyl alcohol and 40 ml of liquid ammonia, a total of 2.53 g (110 mA) of sodium metal and then 7.53 g were added in portions with stirring and dry ice cooling (20 mmol) of N, N-diethyl-N '- [(8a) -6methyl-ergolin-8-yl] sulfamide. 11.0 ml (10 mmol) of isopropyl iodide were added dropwise under reflux and the reaction mixture was subsequently stirred under reflux for 3 hours.
The ammonia was allowed to evaporate overnight with stirring and the reaction mixture was warmed to room temperature.
