PATENTANSPRÜCHE
1. Verfahren zur Gewinnung von Silymarin, wobei man die Früchte von Silybum marianum von einem grossen Teil fetten Öles durch kaltes Abpressen befreit, die abgepresste Masse zerkleinert, den Pressrückstand mit Äthylacetat extrahiert, den Äthylacetatextrakt eindampft und aufarbeitet, dadurch gekennzeichnet, dass man:
a) den Pressrückstand mehrmals mit wasserfreiem Äthylacetat extrahiert und die Extrakte vom Rückstand abtrennt, b) entweder den Eindampfungsrückstand der vereinigten Äthylacetatextrakte a) mit Petroläther mehrmals durch festflüssig-Extraktion entfettet, oder b') den Eindampfungsrückstand der vereinigten Äthylacetatextrakte a) in Methanol auflöst und die Lösung mit Petroläther mehrmals in flüssig-flüssig-Extraktion entfettet, c) entweder den entfetteten Rückstand b) in Methanol weitgehend auflöst und die Lösung von den ungelösten Anteilen abtrennt, oder c') die entfettete methanolische Lösung b') vom Petrol äther abtrennt, d) der methanolischen Lösung c) oderc')jeweils ein chlorsubstituiertes niederes Alkan als Lösungsmittel und Wasser zusetzt, ausschüttelt, die wässrig-methanolische Lösung abtrennt und aufbewahrt,
e) die Phase des chlorierten Lösungsmittels mehrmals mit Methanol und Wasser unter Rühren durchmischt und abdekantiert, f) die vereinigten wässrig-methanolischen Phasen eindampft, g) den Eindampfungsrückstand in wenig Methanol suspendiert, anschliessend die Suspension in Wasser einrührt und nach Zentrifugieren den Niederschlag mehrmals mit Wasser nachwäscht, und h) den Niederschlag im Vakuum trocknet, mahlt und erneut im Vakuum trocknet.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man: a) den Pressrückstand 2- bis 4mal, vorzugsweise 3mal, je 1/2 bis I l/2, vorzugsweise 1 Stunde lang mit wasserfreiem Äthylacetat bei 65 bis 76"C, vorzugsweise 75"C, im Gewichtsverhältnis 1:8 bis 1:12, vorzugsweise 1:10 unter kräftigem Rühren extrahiert und die extrahierte Droge abtrennt, b) entweder die vereinigten Äthylacetatextrakte bei 40-55"C, vorzugsweise 50"C, im Vakuum bei 15 mbar eindampft, den Eindampfungsrückstand a) durch mehrmalige 10- bis 20minütige, vorzugsweise 1 Sminütige fest-flüssig Extraktion mit Petroläther im Siedebereich 60-70"C im Gewichtsverhältnis 1:15 bis 1:25, vorzugsweise 1:
:20, entfettet, oder b') den Eindampfungsrückstand a) in der 6- bis lOfachen, vorzugsweise 8fachen Gewichtsmenge Methanol auflöst, die ungelöst verbleibenden Bestandteile abtrennt, durch dreimalige 10- bis 20minütige, vorzugsweise 1 Sminütige flüssigflüssig Extraktion mit Petroläther im Siedebereich 60-70"C im Volumenverhältnis 1:1,5 bis 1:2,5, vorzugsweise 1:
:2, entfettet, c) entweder den entfetteten Rückstand b) in der 6- bis 1 fachen, vorzugsweise 8fachen Gewichtsmenge Methanol auflöst und die ungelöst verbleibenden Bestandteile abtrennt, oder c) die entfettete methanolische Lösung b') vom Petrol äther abtrennt, d) der methanolischen Lösung c) oder c') Chloroform und Wasser in jeweils der gleichen Volumenmenge zusetzt und das Gemisch ca. 15 Minuten kräftig rührt und abdekantiert, e) die Chloroformphase fünfmal mit Methanol und Wasser im Volumenverhältnis 1:1:
:1 je ca. 15 Minuten unter kräftigem Rühren mischt und anschliessend die Chloroformphase abtrennt, f) die vereinigten wässrig-methanolischen Phasen bei 30-50"C, vorzugsweise 40"C, im Vakuum bei 15 mbar eindampft, g) den eingedampften Rückstand in Methanol im Gewichtsverhältnis 1:4 bis 1:6, vorzugsweise 1:
:5, suspendiert, und die Suspension in die 8- bis 12fache, vorzugsweise 10fach Volumenmenge Wasser einrührt, sowie nach Zentrifugieren den Niederschlag mehrfach mit Wasser nachwäscht, h) den Niederschlag 20 bis 30, vorzugsweise 24 Stunden, bei 30 bis 50"C, vorzugsweise 40"C, im Vakuum bei 10 mbar trocknet, anschliessend vermahlt und das vermahlene Produkt ebenfalls bei 30 bis 50"C, vorzugsweise 40"C, im Vakuum bei 10 mbar nachtrocknet.
3. Silymarin mit mindestens 90 Gew.%iger Reinheit, erhalten nach dem Verfahren gemäss Anspruch 1.
4. Lebertherapeutikum enthaltend Silymarin nach Anspruch 3 und einen pharmazeutischen Träger und/oder Verdünnungsmittel.
Die Erfindung betrifft ein Verfahren zur Gewinnung von reinem Silymarin durch Extraktion von Früchten der Mariendistel (Silybum marianum Gaertn.), das nach dem Verfahren erhaltene Silymarin sowie ein das Silymarin enthaltendes Lebertherapeutikum.
Unter der Bezeichnung Silymarin versteht man nach dem Stand der Technik die Silymaringruppe, d.h. die Polyhyxdroxyphenylchromanone (Silymarin 1 bis Silymarin IV), die bereits aus den Früchten der Mariendistel in 70 bis 80%iger Reinheit - je nach Droge - isoliert werden konnten (DE-AS 1 923 082).
Silymarin 1 bis Silymarin III sind Isomere der Summenformel C25H22OI0 und in der Literatur als Silybin, Silydianin und Silychristin struktur-chemisch charakterisiert worden.

Silymarin IV ist Taxifolin (DE-AS 1 923 082).
Silymarin ist ein wertvolles Lebertherapeutikum, und das Silymaringemisch ist aufgrund eines Synergismus wirksamer als die Summe der Wirkungen der einzelnen Isomeren (DE-AS 1 923 082).
Aus der DE-AS 1 923 082 ist ein Verfahren zur Gewinnung von Silymarin (Silymarin I-IV) bekannt, bei dem die getrockneten Früchte von Silybum marianum Gaertn.
mechanisch ausgepresst werden, um sie von der Hauptmenge des fetten Öles zu befreien. Durch erschöpfende Extraktion des Pressrückstandes mit Äthylacetat, Eindampfen der Äthylacetatlösung, Aufnahme des Trockenrückstandes in wasserhaltigem Methanol, Entfettung durch multiplikate gleichförmige Verteilung im Gegenstrom, und Eindampfen der fettfreien methanolischen Lösung im Vakuum zur Trockne erhält man ein 70-80%iges Polyhydroxyphenylchromanongemisch (Silymarin I-IV). Das Verhältnis Silymarin I-III liegt etwa bei 3:1:1; der Gehalt an Silymarin IV, Taxifolin, beträgt etwa 2-3% des Gesamtanteils Silymarin.
Als weiterer Stand der Technik ist die DE-OS 2020407 zu
nennen. Das Verfahren dieser OLS beschäftigt sich mit der Isolierung von Silymarin ohne Vorentfettung, durch direkte Gesamtextraktion mittels eines Lösungsmittelgemisches Alkohol-Wasser-niedere Fettsäureester oder Kohlenwasserstoff und Abtrennung der Fettanteile durch anschliessendes Abdestillieren von 10-40% des Volumens der erhaltenen Extraktlösung, anschliessendes Stehenlassen und Entfernen der oben auf der Lösung abgesetzten Fette . Dieses Verfahren führt laut Anmeldung nur zu einem 40%igen Silymarinextrakt; weiter empfiehlt die Patentanmeldung eine Umkristallisation mit Methanol, ohne weitere Angaben über etwaige Verfahrensmassnahmen zu machen. Bei einer Nacharbeitung war dieses Verfahren nicht reproduzierbar, und im
Endprodukt verblieben ca. 13% ursprüngliches Fett.
Ein weiteres Verfahren ist aus der DE-OS 2017 789 bekannt. Die Endprodukte dieses Verfahrens sollen 40 bis 70% Silymarin enthalten, und die Ausbeute an Endprodukt beträgt 0,8 bis maximal 2,4% (bei 40%igem Silymarin). Eine Nacharbeitung zeigte, dass dieses Verfahren ebenfalls nicht reproduzierbar war.
Weiterhin existiert zum Stand der Technik das französische Patent Nr.72.14042, das sich mit der Herstellung eines Extraktes aus Silybum marianum beschäftigt, ohne eine Vorentfettung vorzunehmen. Es wird angegeben, dass das Endprodukt ca. 82% an Polyhydroxyphenylchromonen hat; von 82% sollen 50% Silymarine darstellen. Es sei dazu angemerkt, dass das Silymarin einer anderen Verbindungsklasse zugeordnet ist, und zwar den Polyhydroxyphenylchromanonen, nicht aber den Polyhydroxyphenylchromonen. Auch dieses Verfahren ist nicht reproduzierbar.
Es ist bisher nicht gelungen, das Verfahren der DE-AS
1 923 082 wesentlich zu verbessern. Einesteils liegen die Schwierigkeiten in der Abtrennung der Wirkstoffe aus der Droge unter Bewahrung des vorhandenen Silymaringemisches und andernteils in der Abtrennung desselben von ca.
30% fetten Ölen und schwer zu entfernenden Begleitstoffen.
Diese Schwierigkeiten wurden durch die DE-AS 1 923 082 nur teilweise überwunden.
Da Silymarin ein wertvolles Lebertherapeutikum ist, besteht ein Bedarf an einem möglichst hochkonzentrierten Silymarin, das über der aus dem Stand der Technik bisher bekannten Konzentration liegt.
Der vorliegenden Erfindung liegt die Aufgabe zugrunde, ein neues Verfahren zur Gewinnung von reinem Silymarin (Silymarin I-IV), das weitgehend frei von Begleitstoffen ist und das alle natürlichen Isomeren enthält, zu schaffen.
Dieses neue Verfahren soll auf einfachem Weg zu einem besonders reinen Produkt und höherer Ausbeute führen.
Es wurde überraschenderweise ein Verfahren gefunden, das die Nachteile der bekannten Methoden vermeidet und das aufwirtschaftlichem Weg zu reinem Silymarin (Silymarin I-IV) (Reinheit 90-96%) führt.
Das erfindungsgemässe Verfahren zur Gewinnung von Silymarin, wobei man die Früchte von Silybum marianum von einem grossen Teil fetten Öles durch kaltes Abpressen befreit, die abgepresste Masse zerkleinert, den Pressrückstand mit Äthylacetat extrahiert, den Äthylacetatextrakt eindampft und aufarbeitet, ist dadurch gekennzeichnet, dass man:
a) den Pressrückstand mehrmals mit wasserfreiem Äthylacetat extrahiert und die Extrakte vom Rückstand abtrennt, b) entweder den Eindampfungsrückstand der vereinigten Äthylacetatextrakte a) mit Petroläther mehrmals durch festflüssig- Extraktion entfettet, oder b') den Eindampfungsrückstand der vereinigten Äthylacetatextrakte a) in Methanol auflöst und die Lösung mit Petroläther mehrmals in flüssig-flüssig-Extraktion entfettet, c) entweder den entfetteten Rückstand b) in Methanol weitgehend auflöst und die Lösung von den ungelösten Anteilen abtrennt, oder c') die entfettete methanolische Lösung b') vom Petrol äther abtrennt, d) der methanolischen Lösung c) oder c')jeweils ein chlorsubstituiertes niederes Alkan als Lösungsmittel und Wasser zusetzt, ausschüttelt, die wässrig-methanolische Lösung abtrennt und aufbewahrt,
e) die Phase des chlorierten Lösungsmittels mehrmals mit Methanol und Wasser unter Rühren durchmischt und abdekantiert, f) die vereinigten wässrig-methanolischen Phasen eindampft, g) den Eindampfungsrückstand in wenig Methnol suspendiert, anschliessend die Suspension in Wasser einrührt und nach Zentrifugieren den Niederschlag mehrmals mit Wasser nachwäscht, und h) den Niederschlag im Vakuum trocknet, mahlt und erneut im Vakuum trocknet.
Eine bevorzugte Ausführungsform des erfindungsgemässen Verfahrens ist dadurch gekennzeichnet, dass man a) den Pressrückstand 2- bis 4mal, vorzugsweise 3mal, je 1 bis 11/2, vorzugsweise 1 Stunde lang mit wasserfreiem Äthylacetat bei 65 bis 76"C, vorzugsweise 75"C, im Gewichtsverhältnis 1:8 bis 1:12, vorzugsweise 1:10 unter kräftigem Rühren extrahiert und die extrahierte Droge abtrennt, b) entweder die vereinigten Äthylacetatextrakte bei 40-55"C, vorzugsweise 50"C, im Vakuum bei 15 mbar eindampft, den Eindampfungsrückstand a) durch dreimalige 10bis 20minütige, vorzugsweise 15minütige fest-flüssig-Extraktion mit Petroläther im Siedebereich 60-70"C im Gewichtsverhältnis 1:15 bis 1:25, vorzugsweise 1:
:20, entfettet, oder b') den Eindampfungsrückstand a) in der 6- bis 10fachen, vorzugsweise 8fachen Gewichtsmenge Methanol auflöst, die ungelöst verbleibenden Bestandteile abtrennt, durch dreimalige 10- bis 20minütige, vorzugsweise 1 Sminütige flüssigflüssig- Extraktion mit Petroläther im Siedebereich 60-70"C im Volumenverhältnis 1:1,5 bis 1:2,5, vorzugsweise 1:

:5, suspendiert, und die Suspension in die 8- bis 12fache, vorzugsweise 10fach Volumenmenge Wasser einrührt, sowie nach Zentrifugieren den Niederschlag mehrfach mit Wasser nachwäscht, h) den Niederschlag 20 bis 30, vorzugsweise 24 Stunden, bei 30 bis 50"C, vorzugsweise 40"C, im Vakuum bei 10 mbar trocknet, anschliessend vermahlt und das vermahlene Produkt ebenfalls bei 30 bis 50"C, vorzugsweise 40"C, im Vakuum bei 10 mbar nachtrocknet.
Die auf diesem Wege isolierten Produkte verfügen über einen Silymarin-(Silymarin I-IV)-Gehalt von 90-96 Gew.%, bezogen auf wasserfreie Substanz und können in einem Lebertherapeutikum gemäss Anspruch 4 verwendet werden.
Verfahrenstechnisch sind die einzelnen Schritte und die spezifischen Massnahmen so gewählt, dass das in der Droge vorliegende Wirkstoff-Gemisch praktisch vollständig isoliert wird. Weicht man von diesen Verfahrensschritten ab, so würde die eine oder andere Verbindung ganz oder teilweise verlorengehen. Diese Schwierigkeit wächst mit dem Anreicherungsgrad dieses Wirkstoffgemisches, insbesondere bei den fortgeschrittenen Verfahrensstufen, da die gewünschten Wirkstoffe teilweise als Einzelverbindungen oder als Gesamtgemisch mit den abzutrennenden Begleitstoffen sich der Gewinnung entziehen würden.
Das erfindungsgemässe Verfahren berücksichtigt diese Punkte, so dass es zu einem reinen Produkt in optimaler Ausbeute unter weitgehender Erhaltung des Isomerenverhältnisses, das in der Droge vorliegt, führt.
Überraschenderweise kommt man nur durch die ausgesuchte Kombination der Verfahrensmassnahmen, der Lösungsmittel bzw. Lösungsmittelkombinationen und der physikalischen bzw. chemischen Bedingungen zu dem vorteilhaften Produkt.
Durch das kalte Pressen der Früchte des Silybum marianum verhindert man einen chemischen Abbau oder Polymerisation des Silymarins, und man vermeidet inniges Emulgieren des Silymarins und der Begleitstoffe in dem fettigen Öl, was sich für die nachfolgende Trennung nachteilig auswirken würde. Durch das Pressen wird die Droge teilweise entfettet. Etwa 25-30% des Öls werden ausgepresst.
Die Feinvermahlungsmassnahme ermöglicht eine raschere nachfolgende Extraktion, weil das Extraktionsmittel leichter in die Drogenpartikel eindringen kann.
Es ist in diesem Verfahren entscheidend, dass die Äthylacetatextraktion mit wasserfreiem Äthylacetat durchgeführt wird. Der Gehalt an Wasser kann maximal 0,5% betragen. Ist der Gehalt an Wasser über 0,5%, dann lassen sich bei den nachfolgenden Verfahrensschritten die Begleitsubstanzen nicht mehr trennen. Dieser überraschende Effekt ist in Kom- bination mit den anderen Verfahrensmassnahmen massgebend für den Erfolg des Verfahrens, nämlich die Gewinnung von reinem Silymarin.
Das Verhältnis Droge/Lösungsmittel bei der Äthylacetatextraktion von 1:8 bis 1:12, vorzugsweise 1:10, ist so gewählt, dass es ohne unnötige Überschüsse die Auflösung des Silymarins spezifisch und schonend gewährleistet. Die zwei- bis viermalige, vorzugsweise dreimalige, Extraktion erlaubt die vollständige Gewinnung des Silymarins aus der Droge. Die Temperatur von 60-80 C, vorzugsweise 75"C, ist günstig für eine gute Lösung der Wirkstoffe und führt gleichzeitig nicht zu Zersetzungserscheinungen der hier vorhandenen chemischen Verbindungen.
Das Ausrühren mit einem Intensivrührer bringt eine mechanische Auflockerung der Droge mit sich, was die Extraktion begünstigt. Die gewählte kurze Zeit bei jeder der mehreren Extraktionen von 1/2 bis 11/2 Stunden, vorzugsweise 1 Stunde, erlaubt es trotzdem, erschöpfend zu extrahieren.
Auf schonende Weise werden die vereinigten Äthylacetatextrakte bei niedriger Temperatur im Vakuum eingedampft.
Die nachfolgende mehrmalige fest-flüssig-Entfettung des Rückstandes mit Petroläther bei einem relativ hohen Verhältnis von Rückstand zu Petroläther, von z.B. ca. 1:20 hat den Vorteil, dass einerseits in kurzer Zeit und ohne grossen apparativen Aufwand erschöpfend das fettige Öl abgetrennt wird. Wahlweise kann man die Eindampfungsrückstände in der ca. Sfachen Gewichtsmenge Methanol auflösen, die ungelösten Bestandteile abtrennen und anschliessend eine mehrmalige flüssig-flüssig-Extraktion mit Petroläther im Verhältnis von ca. 1:2 vornehmen. Die Ergebnisse sind praktisch gleich.
Methanol, als verhältnismässig billiges Lösungsmittel, eignet sich zum Auflösen des entfetteten Rückstandes besonders gut wegen seines grossen Solvatationsvermögens. Das Silymarin (Silymarin I-IV) löst sich relativ gut darin. Das hohe Verhältnis Rückstand/Lösungsmittel ist vorteilhaft, um das Silymarin (Silymarin I-IV) vollständig aufzunehmen. Im Rückstand bleibt kein Silymarin zurück.
Neu und besonders erfinderisch ist der Einsatz von chlorhaltigen organischen Lösungsmitteln, insbesondere von chlorsubstituierten niederen Alkanen, wie Chloroform, Met hylenchlorid,1,2-Dichloräthan, vorzugsweise Chloroform, für die Abtrennung von noch verbleibenden Begleitstoffen vom Silymarin (Silymarin 1-1V), ohne dass das Silymarin insgesamt einen Verlust erleidet und wobei auch keine Isomeren verlorengehen. Dabei erfolgt die Abtrennung von weiteren lipophilen Begleitstoffen, die bei der Entfettungsstufe mit dem Öl und Petroläther nicht ausgeschieden wurden, sowie weiterer weniger polarer Begleitstoffe, die zwar zunächst in Methanol aufgelöst wurden, aber sich in einem Verteilungsphasensystem entsprechend verhalten.
Das Lösungsgleichgewicht - Zustandsdiagramm - des gewählten Lösungsmittelsystems - sowohl in der Art wie in der Menge -, in diesem Fall Methanol-Wasser-Chloroform 1:1:1, ist in Gegenwart von Silymarin (Silymarin I-IV) und etwaigen noch vorhandenen Begleitstoffen besonders geeignet, da gute Trennungen in zwei Phasen mit diesem Lösungsmittelsystem erreicht werden. Das Entmischungsgebiet des Zustandsdiagrammes erfasst die angegebenen Lösungsmittelverhältnisse.
Der Verteilungskoeffizient des Silymarins (Silymarin I bis Silymarin IV) zwischen beiden Phasen ist sehr hoch. Nach ausgeführten sechs Ausschüttelungen im System Methanol/ Chloroform/Wasser 1/1/1 (v/v/v) ergibt sich ein Verhältnis von Silymarin in den vereinigten wässrig-methanolischen Phasen zu dem in der Chloroformphase verbleibenden Silymaria wie 13:1.
Dadurch ist das Silymarin (Silymarin I-IV) aus der methanolischen Phase praktisch vollständig zu gewinnen.
Erfindungsgemäss konnte überraschenderweise erreicht werden, dass die Silymarine I bis IV auch als Einzelverbindungen anteilmässig in der wässrig-methanolischen Phase verbleiben, so dass das Isomerenverhältnis aufrechterhalten bleibt.
Die wässrig-methanolische Phase wird schonend bei vorteilhaft niedriger Temperatur und unter Vakuum zur Trockne eingedampft und der Rückstand in Wasser zwecks Auswaschens mehrere Male suspendiert. Dieser Auswaschvorgang hat sich als vorteilhaft erwiesen, um die bei normaler Filtration auftretenden Okkludierungen von Unreinheiten im Produkt zu beseitigen.
Gegenüber den Verfahren des Standes der Technik hat das erfindungsgemässe den Vorteil, dass es auf einfachem Wege zu einem 90-96%igen, reinen Silymarin (Silymarin I-IV) bei einer Ausbeute von - 1,7% führt. Nur die erfindungsgemässe Kombination der gewählten Verfahrensmassnahmen mit Einsatz von wasserfreiem Äthylacetat und einem mit Chlor substituierten Alkan, wie Chloroform, führt überraschenderweise zu der Gewinnung von weitgehend reinem Silymarin aus den Früchten von Silybum marianum.
Der Effekt der wasserfreien Äthylacetat-Extraktion der Droge ist gegenüber dem Stand der Technik als besonders erfinderisch hervorzuheben. Hiermit bleiben hydrophile Substanzen der Droge im Rückstand. Bei den nachfolgenden Verfahrensstufen, wie Verteilung in Lösungsmitteln, sind normalerweise diese hydrophilen Begleitstoffe weitgehend vom Silymarin nicht mehr zu trennen.
Weiterhin ist besonders überraschend, die Verteilungswirkung von Lösungsmitteln eines chlorsubstituierten niederen Alkan, insbesondere Chloroform, in der zweiten Phase des Verfahrens wegen der spezifischen Selektivität, wobei praktisch alle noch bei dieser Verfahrensstufe vorhandenen Begleitstoffe entfernt werden, so dass Silymarin in der methan nolischen Lösung weitgehend rein bleibt.
Beispiele für die Gewinnung von Silymarin (Silymarin I-IV)
Beispiel I
300 kg feinvermahlene, kalt abgepresste Mariendistelfrüchte werden mit 30001 wasserfreiem Äthylacetat versetzt und auf 75"C erhitzt. Bei dieser Temperatur wird die
Mischung mit einem Intensivrührer eine Stunde kräftig durchgerührt. Danach wird filtriert und der Drogenrückstand noch zweimal, wie beschrieben, mit Äthylacetat extrahiert.
Die vereinigten, geklärten Äthylacetat-Lösungen werden bei 50"C im Vakuum bei 15 mbar zur Trockne eingedampft.
Zur Entfettung wird der Rückstand bei Raumtemperatur dreimal mit je 3001 Petroläther 60/70 15 min. ausgerührt und danach im Vakuum vom Lösungsmittel befreit. Das auf diese Weise erhaltene Rohprodukt wird in der vierfachen Menge Methanol aufgenommen, wobei der dabei nicht in Lösung gehende Feststoff abzentrifugiert, zweimal mit je der Hälfte der erwähnten Menge Methanol nachgewaschen und schliesslich - da er praktisch kein Silymarin enthält - abgetrennt wird.
Die vereinigten methanolischen Lösungen werden zunächst mit Chloroform, danach mit Wasser im Volumenverhältnis 1:1:1 versetzt und in Dekantationsbehälter überführt.
Nachdem die Mischung 15 min. kräftig durchgerührt worden und die Phasentrennung erreicht ist, wird die untere Phase (Chloroformphase) abgetrennt und noch fünfmal ebenfalls im Volumenverhältnis 1:1:1 mit Methanol-Wasser je 15 min. extrahiert. Danach wird die Chloroformphase abgetrennt.
Die vereinigten wässrig-methanolischen Phasen werden bei 50"C im Vakuum bei 15 mbar eingedampft und der Rückstand wird in der Sfachen Menge Methanol suspendiert. Die Suspension wird in dünnem Strahl zu der 10fachen Volumenmenge Wasser unter Rühren zugegeben und danach 15 min.
lang kräftig nachgerührt. Danach wird zentrifugiert und das Zentrifugat mehrfach mit Wasser unter Suspendierung nachgewaschen. Das Produkt wird 24 Stunden bei 40"C im Vakuumtrockenschrank getrocknet, vermahlen und weitere 48 Stunden bei 40"C nachgetrocknet.
Es resultiert ein hellbeige-farbenes Pulver, das über einen Silymaringehalt von 90-96% verfügt (bestimmt nach der Dinitrophenylhydrazin-Methode (DNPH-Methode) analog zur Bestimmung von Silymarin in Mariendistelfrüchte, DAB VIII Ausgabe 1968,2. Nachtrag 1975, Seite 198, sowie nach quantitativer Auswertung der Remissionsortskurven.) Der Identitätsnachweis erfolgte über Dünnschichtchromatografie.
Das Verhältnis der einzelnen Isomeren zueinander wurde durch die quantitative Auswertung der Remissionsorts kurven bestimmt (siehe Fig. 1).
Die Ausbeute beträgt 4,9 kg - 1,7% - bezogen auf trockene
Droge.
Beispiel 2

Die methanolische Lösung wird zunächst mit Chloroform, danach mit Wasser im Volumenverhältnis 1:1:1 versetzt und in Dekantationsbehälter überführt.

Das Verfahren führt zu einem 90-96%igen Silymarin der gleichen Beschaffenheit wie im Beispiel 1.
Beispiel 3
Die gleichen Verfahrensmassnahmen mit dem Einsatz von Methylenchlorid anstelle von Chloroform führen zu einem 90-94%igen Silymarin.
Die Ausbeute beträgt 1,7%.
Beispiel 4
Die gleichen Verfahrensmassnahmen mit dem Einsatz von 1,2-Dichloräthan anstelle von Chloroform führen zu einem 90-93%igen Silymarin.

According to the state of the art, the term silymarin means the silymarin group, i.e. the polyhyxdroxyphenylchromanones (silymarin 1 to silymarin IV), which could be isolated from the fruits of milk thistle in 70 to 80% purity - depending on the drug - (DE-AS 1 923 082).
Silymarin 1 to Silymarin III are isomers of the empirical formula C25H22OI0 and have been structurally characterized in the literature as silybin, silydianin and silychristine.

DE-OS 2020407 is another state of the art

Another method is known from DE-OS 2017 789. The end products of this process should contain 40 to 70% silymarin, and the yield of the end product is 0.8 to a maximum of 2.4% (with 40% silymarin). A rework showed that this method was also not reproducible.
There is also prior art French patent No. 72.14042, which deals with the production of an extract from Silybum marianum without pre-degreasing. The final product is said to have approximately 82% polyhydroxyphenyl chromones; of 82% are said to represent 50% silymarine. It should be noted that the silymarin is assigned to a different class of compounds, namely the polyhydroxyphenylchromanones, but not the polyhydroxyphenylchromones. This process is also not reproducible.
So far, the method of DE-AS has not been successful
1 923 082 to be significantly improved. On the one hand, the difficulties lie in the separation of the active substances from the drug while preserving the existing silymarin mixture, and on the other hand in the separation of the approx.
30% fatty oils and difficult to remove accompanying substances.
These difficulties were only partially overcome by DE-AS 1 923 082.
Since silymarin is a valuable liver therapeutic agent, there is a need for a highly concentrated silymarin that is above the concentration known from the prior art.
The present invention has for its object to provide a new process for the production of pure silymarin (Silymarin I-IV), which is largely free of accompanying substances and which contains all natural isomers.
This new process should lead to a particularly pure product and a higher yield in a simple way.
Surprisingly, a process was found which avoids the disadvantages of the known methods and which leads to pure silymarin (silymarin I-IV) (purity 90-96%) in an economical manner.

In terms of process technology, the individual steps and the specific measures are selected so that the active substance mixture present in the drug is practically completely isolated. If one deviates from these procedural steps, one or the other connection would be lost in whole or in part. This difficulty increases with the degree of enrichment of this mixture of active substances, in particular in the advanced process stages, since the desired active substances would in some cases elude extraction as individual compounds or as a total mixture with the accompanying substances to be separated off.
The method according to the invention takes these points into account, so that it leads to a pure product in optimal yield while largely maintaining the isomer ratio that is present in the drug.
Surprisingly, the advantageous product can only be obtained by the selected combination of the process measures, the solvents or solvent combinations and the physical or chemical conditions.
The cold pressing of the fruits of the Silybum marianum prevents chemical degradation or polymerization of the silymarin, and avoids intimate emulsification of the silymarin and the accompanying substances in the fatty oil, which would have an adverse effect on the subsequent separation. The drug is partially degreased by pressing. About 25-30% of the oil is squeezed out.
The fine grinding measure enables a quicker subsequent extraction because the extractant can penetrate the drug particles more easily.
It is crucial in this process that the ethyl acetate extraction be carried out with anhydrous ethyl acetate. The water content can be a maximum of 0.5%. If the water content is above 0.5%, the accompanying substances can no longer be separated in the subsequent process steps. In combination with the other process measures, this surprising effect is decisive for the success of the process, namely the extraction of pure silymarin.
The ratio of drug / solvent in the ethyl acetate extraction from 1: 8 to 1:12, preferably 1:10, is selected so that it ensures the dissolution of the silymarin specifically and gently without unnecessary excesses. The extraction two to four times, preferably three times, allows the silymarin to be completely extracted from the drug. The temperature of 60-80 C, preferably 75 "C, is favorable for a good solution of the active ingredients and at the same time does not lead to any signs of decomposition of the chemical compounds present here.
Stirring with an intensive stirrer mechanically loosens the drug, which favors the extraction. The chosen short time for each of the several extractions from 1/2 to 11/2 hours, preferably 1 hour, nevertheless allows exhaustive extraction.
The combined ethyl acetate extracts are gently evaporated in vacuo at low temperature.
The subsequent repeated solid-liquid degreasing of the residue with petroleum ether at a relatively high ratio of residue to petroleum ether, e.g. approx. 1:20 has the advantage that, on the one hand, the greasy oil is exhaustively separated in a short time and without great expenditure on equipment. You can either dissolve the evaporation residues in about five times the amount by weight of methanol, separate the undissolved constituents and then carry out repeated liquid-liquid extraction with petroleum ether in a ratio of about 1: 2. The results are practically the same.
Methanol, as a relatively cheap solvent, is particularly suitable for dissolving the degreased residue because of its great solvation capacity. The silymarin (silymarin I-IV) dissolves in it relatively well. The high residue / solvent ratio is beneficial to fully absorb the silymarin (silymarin I-IV). No silymarin is left behind.
New and particularly inventive is the use of chlorine-containing organic solvents, in particular chlorine-substituted lower alkanes, such as chloroform, methylene chloride, 1,2-dichloroethane, preferably chloroform, for the removal of any accompanying substances from silymarin (silymarin 1-1V) without that the silymarin suffers a total loss and no isomers are lost. This involves the separation of other lipophilic accompanying substances that were not eliminated in the degreasing stage with the oil and petroleum ether, as well as other less polar accompanying substances that were initially dissolved in methanol, but behave accordingly in a distribution phase system.
The solution balance - state diagram - of the selected solvent system - both in terms of type and quantity - in this case methanol-water-chloroform 1: 1: 1, is special in the presence of silymarin (silymarin I-IV) and any accompanying substances that are still present suitable because good separations are achieved in two phases with this solvent system. The separation area of the state diagram records the specified solvent ratios.
The distribution coefficient of silymarin (Silymarin I to Silymarin IV) between the two phases is very high. After six shakes in the methanol / chloroform / water 1/1/1 (v / v / v) system, the ratio of silymarin in the combined aqueous-methanolic phases to the silymaria remaining in the chloroform phase is 13: 1.
As a result, the silymarin (silymarin I-IV) can be obtained almost completely from the methanolic phase.
According to the invention, it was surprisingly achieved that the silymarines I to IV also remain as individual compounds in proportion in the aqueous-methanolic phase, so that the isomer ratio is maintained.
The aqueous-methanolic phase is gently evaporated to dryness at an advantageously low temperature and under vacuum and the residue is suspended in water several times for washing out. This washing-out process has proven to be advantageous in order to eliminate the occlusions of impurities in the product that occur during normal filtration.
Compared to the methods of the prior art, the advantage of the invention is that it leads in a simple way to a 90-96% pure silymarin (silymarin I-IV) with a yield of - 1.7%. Only the combination of the selected process measures according to the invention with the use of anhydrous ethyl acetate and an alkane substituted with chlorine, such as chloroform, surprisingly leads to the production of largely pure silymarin from the fruits of Silybum marianum.
The effect of the anhydrous ethyl acetate extraction of the drug should be emphasized as particularly inventive compared to the prior art. This leaves the drug's hydrophilic substances behind. In the subsequent process stages, such as distribution in solvents, these hydrophilic accompanying substances are normally largely no longer separable from the silymarin.
Furthermore, it is particularly surprising that the distribution effect of solvents of a chlorine-substituted lower alkane, in particular chloroform, in the second phase of the process because of the specific selectivity, with practically all accompanying substances still present in this process step being removed, so that silymarin in the methanolic solution is largely pure remains.
