PATENTANSPRÜCHE
1. 1,2,3,4,9,10-Hexahydro-5,8-dihydroxy-2, 3-dimethy len-1,4-epoxyanthracen und dessen di-Methyläther.
2. Verfahren zur Herstellung von 1,2,3,4,9,1 0-Hexahydro- 5, 8-dihydroxy-2,3-dimethylen- 1 4-epoxyanthracen, dadurch gekennzeichnet, dass man 2,3,5,6-Tetramethylen-7-oxabicy- clo[2.2. 1]heptan mit p-Benzochinon zu 1,2,3,4-8a,9,10,10a- Octahydro-2,3-dimethylen- 1 ,4-epoxyanthracen-5,8-dion umsetzt und letzteres in Gegenwart katalytischer Mengen einer Säure oder einer Base isomerisiert.
3. Verfahren zur Herstellung von 1,2,3,4,9,10-Hexahy- dro-S,8-dimethoxy-2,3-dimethylen-1 ,4-epoxyanthracen, dadurch gekennzeichnet, dass man 2,3,5,6-Tetramethylen-7 oxabicyclo[2.2. 1] heptan mit p-Benzochinon zu 1,2,3,4,8a,9, 10,10a-Octahydro-2,3-dimethylen-1,4-epoxy- anthracen-S,8-dion umsetzt, letzteres in Gegenwart katalytischer Mengen einer Säure oder einer Base zu 1,2,3,4,9,10-He xahydro-5,8-dihydroxy-2,3-dimethylen 1 ,4-epoxyanthracen isomerisiert und dieses methyliert.
Auf Grund ihrer beachtlichen Aktivität gegen verschiedene Tumorarten haben die Anthracycline in letzter Zeit erhöhte Bedeutung gewonnen. Verschiedene Arbeitsgruppen haben sich insbesondere mit dem Aufbau des Anthracyclinon-Ringsystems durch Diensynthese beschäftigt, doch blieben an mancher Stelle in Bezug auf Ausbeuten und Zahl der Reaktionsschritte in der Totalsynthese noch Wünsche offen (S. Blechert, Nachr. Chem. Tech. Lab. 27, 7, (1979)).
Die vorliegende Erfindung betrifft einen neuen synthetischen Zugang zu Anthracyclinonen, den Aglyconen der Anthracycline.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man 2,3,5,6-Tetramethylen-7-oxabicyclo- [2.2. l]heptan (I) mit p-Benzochinon zu 1 ,2,3,4,8a,9, 10, 10a Octahydro-2,3-dimethylen-1 ,4-epoxy-anthracen-5,8-dion (II) umsetzt, letzteres in Gegenwart katalytischer Mengen einer Säure oder Base zu 1,2,3,4,9,1 0-Hexahydro-5,8-dihydroxy- 2,3-dimethylen-1 ,4-epoxyanthracen (V) isomerisiert und dieses gewünschtenfalls zu 1,2,3,4,9,10-Hexahydro-5,8-dimeth- oxy-2,3-dimethylen - 1 ,4-epoxyanthracen (VI) methyliert.
Die erfindungsgemässe Umsetzung findet in an sich bekannter Weise unter den Bedingungen einer Diels-Alder-Addition bei Einsatz äquimolarer Mengen des Diens und des Dienophils statt. Sie verläuft mit hoher Ausbeute zum steriscl richtigen Produkt.
Die Isomerisierung des erhaltenen 1,2,3,4,8a,9,10,10a-Oc- tahydro-2,3 -dimethylen- 1 ,4-epoxyanthracen-5, 8-dions (II) zur entsprechenden 5,8-DihydroxyverbindungV und deren Verätherung zum 1,2,3,4,9,1 O-Hexahydro-5,8-dimethoxy- 2,3-dimethylen-1 ,4-epoxyanthracen (VI) kann in an sich bekannter Weise durch Behandlung mit einer geringen Menge einer Säure oder Base, z.B. Kieselsäure, und anschliessende Methylierung, z.B. mit Methyljodid, erfolgen.
Beide Reaktionen können mit nahezu quantitativer Ausbeute auch in einem Zuge durchgeführt werden, indem man die Verbindung II z.B. in Gegenwart von Kaliumcarbonat mit Methyljodid behandelt.
Die Verbindungen V und VI können wie folgt in bekannte Verbindungen umgewandelt werden:
Die Verbindungen können in beliebiger Reihenfolge im Ring B aromatisiert und einer weiteren Diels-Alder-Addition unterworfen werden. Die Dehydrierung kann z.B. mit einem entsprechend substituierten Chinon, wie 2,3-Dichlor-5,6-dicyano-1,4-benzochinon oder Chloranil, in einem inerten Lösungsmittel durchgeführt werden, während als geeignetes Dienophil zum weiteren Aufbau des polycyclischen Systems Acrylsäureester, vorzugsweise Acrylsäuremethylester, oder Methylvinylketon verwendet werden können. Man erhält somit entweder zunächst eine Verbindung XIII, die dann durch Diels-Alder-Addition in eine Verbindung XIV übergeführt wird, oder eine Verbindung VIII, die dann zur Verbindung XIV dehydriert werden kann.

Eine 5, 12-Epoxyverbindung XIV kann durch Behandlung mit einer Säure, z.B. Trifluoressigsäure, in einem inerten organischen Lösungsmittel, wie Chloroform oder Benzol, unter Offnung des Epoxyrings in ein Gemisch der entsprechenden 5- und 12-Hydroxyverbindungen übergeführt werden. Arbeitet man dabei in Gegenwart von Acetanhydrid so erhält man ein Gemisch aus entsprechenden 5- und 12-Acetoxyverbindungen. Die Gemische können chromatographisch aufgetrennt werden, was jedoch für den weiteren Syntheseverlauf nicht notwendig ist, denn durch Behandlung mit T1(OAc)3.
11/2 H2O als Acetylierungsmittel bei Raumtemperatur in Gegenwart von Acetanhydrid erhält man aus dem Gemisch der 5- und 12-Hydroxyverbindungen die 5,12-Diacetoxyverbindung XVI. Andererseits kann das Gemisch der 5- und 12-Hydroxyverbindungen zu dem Chinon der Formel SV oxydiert werden, z.B. mit PbO2 oder Luftsauerstoff in saurem Medium, vorzugsweise Essigsäure, bei Zimmertemperatur, das dann reduktiv zur 5, 12-Diacetoxyverbindung XVI acetyliert werden kann. Dies kann z.B. durch kurzfristiges Erhitzen mit Zinkstaub in Acetanhydrid bis zum Rückfluss geschehen.

Ein Gemisch aus 2 g (13,7 mMol) 2,3,5, 6-Tetramethylen 7-oxabicyclo[2.2.13heptan,1,47 g (13,7 mMol) frisch subli miertem p-Benzochinon und 20 ml Chloroform wurde 5
Stunden unter Stickstoff auf 80 C erwärmt. Nach dem Ab kühlen wurde zur Trockne eingeengt und aus Aceton/Meth anol anol (4:1) umkristallisiert.
Es wurden 3,28 g (95%) 1,2,3,4,8a,

5,8 -dion, Smp. 147 C (Zers.), in Form gelber Kristalle er halten.
Zu einem Gemisch aus 2 g (7,9 mMol) 1,2,3,4,8a, 9,10,1 0a-Octahydro -2,3- dimethylen -1,4- epoxyanthracen 5,8-dion,10,9 g (79 mMol) getrocknetem Kaliumcarbonat und 100 ml wasserfreiem Aceton wurden unter Stickstoff tropfenweise 11,21 g (79 mMol) Methyljodid zugegeben. Es wurde 15 Stunden auf 80 C erwärmt, filtriert, der Rückstand 1 eingeengt, mit 80 ml Chloroform aufgenommen und die Lö sung mit Wasser (3 x 40 ml) gewaschen.
Nach Trocknen über Magnesiumsulfat, Einengen und Umkristallsation aus

The process according to the invention is characterized in that 2,3,5,6-tetramethylene-7-oxabicyclo- [2.2. l] heptane (I) with p-benzoquinone to 1, 2,3,4,8a, 9, 10, 10a octahydro-2,3-dimethylene-1, 4-epoxy-anthracene-5,8-dione (II) implemented, the latter isomerized in the presence of catalytic amounts of an acid or base to 1,2,3,4,9,1 0-hexahydro-5,8-dihydroxy-2,3-dimethylene-1, 4-epoxyanthracene (V) and this if desired, methylated to 1,2,3,4,9,10-hexahydro-5,8-dimethoxy-2,3-dimethylene-1,4-epoxyanthracene (VI).
The reaction according to the invention takes place in a manner known per se under the conditions of a Diels-Alder addition using equimolar amounts of the diene and the dienophile. It proceeds with high yield to the sterically correct product.
The isomerization of the 1,2,3,4,8a, 9,10,10a-octahydro-2,3-dimethylene-1, 4-epoxyanthracen-5, 8-dione (II) obtained to the corresponding 5.8- Dihydroxy compound V and their etherification to 1,2,3,4,9,1 O-hexahydro-5,8-dimethoxy-2,3-dimethylene-1,4-epoxyanthracene (VI) can be treated in a manner known per se by treatment with a small amount of an acid or base, e.g. Silica, and subsequent methylation, e.g. with methyl iodide.
Both reactions can also be carried out in one go with almost quantitative yield by using compound II e.g. treated with methyl iodide in the presence of potassium carbonate.

Dehydration can e.g. be carried out with an appropriately substituted quinone, such as 2,3-dichloro-5,6-dicyano-1,4-benzoquinone or chloranil, in an inert solvent, while as a suitable dienophile for the further construction of the polycyclic system, acrylic acid esters, preferably methyl acrylate, or Methyl vinyl ketone can be used.
