Description
The object of the present invention is to provide a method for producing a preparation which works better than the previously known.
This object is achieved according to the invention in a process of the type mentioned at the outset in that the acidic filtrate forming the starting material repeatedly undergoes alkaline hydrolysis and the filtrate obtained after the precipitation of the precipitate is acidified and concentrated again and then neutralized and evaporated dry, with the remaining stock being co-evaporated hydrated ethanol extracted and the extract obtained in this way from the excess ballast substance extracted separately with a mixture of ethanol and other organic solvents and the water layer obtained separated, concentrated and divided into chromatography columns.
The final preparation obtained by the process according to the invention differs from that described in the prior art with regard to the chemical composition and the experimentally confirmed strong activity against neoplasms.
It is expedient if peat with 8 to 30 percent ash content and a 30 to 70 percent decomposition stage at a pH of 3.5 to 7.0 is used as the starting material for obtaining the hydrolyzate present. It is also advantageous if the selective removal of bases and acids is carried out by means of ion exchange.
The present invention also relates to the preparation produced by the method according to the invention.
The preparation obtained investigated “in vitro” on lym-phoidal cells, type SL 2, which were taken from mice, showed the activity at a concentration of 0.5 μg / ml. The above-mentioned preparation had been studied in animals for over 10 years in terms of toxicity and the activity counteracting the neoplasms.
In oncostatic examinations, transplantable neoplasms of the Crocker and Ehrlich types were used. Under the action of the preparation, the "immovable" neoplasms detached from the pathologically unchanged tissue and made it possible to perform an operation. No acute or chronic toxicity was found.
The form of application of the preparation depends on the type of neoplasm disease and on the stage of development of the neoplasm; it can be applied in the form of solutions orally or parenterally (i.m.) as well as by means of envelopes and as an ointment.
The peat preparation obtained, which has an activity counteracting the neoplasms, has no negative side effect during use, on the contrary, an improvement in the blood count and an increase in appetite and weight are observed.
I. Example: 1 kg of air-dried peat is crushed and extracted with a water solution of 0.1 to 8 percent sodium hydroxide (81) for 36 hours at room temperature with constant mixing. The alkaline extract is centrifuged to remove the undisassembled parts of the plant, then acidified to about pH = 1 using hydrochloric acid and left to stand for 12 hours. The separated precipitate is filtered, while the clear filtrate is alkalized with sodium hydroxide to pH = 9 and filtered again.
The filtrate is acidified accordingly with hydrochloric acid and concentrated under reduced pressure to about Ys of the volume, neutralized with sodium hydroxide, evaporated to dryness and extracted with hydrated ethanol. The alcoholic-aqueous extract is freed from the lubricant and oil inventory and the inorganic salts and then condensed until the salt exits. After the salt which has separated out is filtered, the filtrate obtained is extracted with a mixture of ethanol and other organic solvents. The inorganic salt that continues to separate is filtered and the water filtrate is concentrated to Vi its volume, then centrifuged.
