PATENTANSPRÜCHE
1. Arzneimittel für die Behandlung von Krebs und bösartigen Neoplasmen, dadurch gekennzeichnet, dass es als aktive Komponente eine Platin-Koordinationsverbindung der Strukturformel

enthält, worin X und Y gleich oder verschieden Halogen oder Pseudohalogen und A und B gleich oder verschieden Ammoniak oder geradkettige oder C-substituierte, verzweigte aliphatische Amingruppen, die mit dem Platin über ihre N-Atome koordiniert sind, wobei jede dieser Gruppen die Formel CnR2n+1NH2 (1ru) aufweist, worin n eine Zahl von 3-9 und R gleich oder verschieden Wasserstoff, gegebenenfalls substituiertes Alkyl, Aryl, Alkaryl, Aralkyl, Halogen, Pseudohalogen, Hydroxy, Carbonyl, Formyl, Nitro, Amido, Amino, Sulfon- oder Carbonsäure oder ein Salz davon bedeuten.
2. Arzneimittel nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel I Halogen Chlor, Brom, Jod und Fluor und Pseudohalogen Cyanid, Cyanat, Thiocyanat, und Azid umfassen.
3. Arzneimittel nach Anspruch 1, dadurch gekennzeichnet, dass in der Formel II sämtliche R Wasserstoff bedeuten.
4. Arzneimittel nach Anspruch 1, dadurch gekennzeichnet, dass R in der Formel II für Niederalkyl oder eine die Löslichkeit fördernde Gruppe steht.
5. Arzneimittel nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass das Platin in der Formel I als Pt4+ vorliegt.
6. Arzneimittel nach einem der vorangehenden Ansprüche, dadurch gekennzeichnet, dass in der Formel I A und B 1 -Aminohexan, l-Aminoheptan oder l-Aminooctan bedeuten.
Gegenstand der vorliegenden Erfindung ist das im Patentanspruch 1 definierte, eine Platin-Koordinationsverbindung der Formel I als aktive Komponente enthaltende Arzneimittel für die chemotherapeutische Behandlung von Krebs und bösartigen Neoplasmen.
In der Formel I bedeuten X und Y beide vorzugsweise Chlor, können aber auch ein anderes Halogen oder Pseudohalogen, wie Cyanat, Thiocyanat und Azid sein, und falls für A und B aliphatische Amingruppen der Formel CnR2n+ 1NH2 (II) stehen, bedeuten vorzugsweise sämtliche R Wasserstoff.
Das Platin liegt vorzugsweise als Po4+ vor, so dass die Verbindung der Formel I einen neutralen Komplex mit je zwei Hydroxyl- und Halogen-Lganden darstellt.
Obschon andere R-Gruonen als Wasserstoff normalerweise nicht bevorzugt werden. können diese auch Niederalkvl, wie Methyl oder Athvl oder ene die Löslichkeit fördernde Gruppe, wie eine Sulfonsäuregruppe, umfassen. Die Löslichkeit fördernden Gruppen als Substituenten, wie eine Carbonsaure, Sulfonsäure, Carbonsäuresalz, Sulfonsäuresalz, z. B. als Natrium-, Kalium- oder Lithiumsalz, sind manchmal dort erwünscht, wo die klinischen Bedingungen eine hohe Löslichkeit verlangen.
Die Bezeichnung Halogen umfasst Chlor, Brom, Jod und Fluor, und Pseudohalogen z.B. ein Cyanid, Cyanat, Thiocyanat oder Azid.
Geeignete Aminverbindungen für die Gruppen A und B in der Formel I sind l-Aminohexan, l-Aminoheptan und 1 Aminooctan.
Im folgenden werden Testergebnisse dargelegt unter Verwendung von trans-Dihydroxo-Komplexen von Pt(IV), worin in der Formel I X und Y beide Chlor und A und B dasselbe Molekül, wie im vorliegenden Falle Ammoniak, sind:

Der therapeutische Index (TI) ist somit grösser als 11,25.
Herstellungsmethoden
Nach einer bevorzugten Methode zur Herstellung von trans-Dihydroxo-cis-dichloro-diamin (bzw. -diamin, wenn A = B = NH3 ist) Pt(IV)-Komplexen aus dem entsprechenden Pt(II)-Komplex werden 20 g PtCI2(n-CnH2n+ 1NH2)2 in
50 ml heissem Wasser aufgeschwemmt und 100 ml 30%iges
Gew./Vol. wässriges Wasserstoffperoxid zugefügt. Die
Suspension wird während 30 Minuten gekocht, auf Zimmer temperatur abgekühlt, über Nacht gekühlt und dann fil triert. Das erhaltene rohe Produkt wird mit einem Minimum an Wasser und Äthanol gewaschen und dann an der Luft ge trocknet. Das rohe Produkt mit einer Ausbeute von 69% wird aus 15 ml 30%igem Gew.lVol. H202 in 280 rni Wasser umkristallisiert und im Vakuum bei 50 "C über Nacht ge trocknet.
Ausbeute 35%.
Herstellung von n-Alkylaminplatin IV-Komplexen RtCl2(0H)2 (n-RNH2)2] worin
R = C3H - Propyl C4H,, - Butyl
CH3OC3H6 - 3-Methoxypropyl C1H1 7 - Octyl ist.
Herstellung von cis-PtCl2(n-C3H7NH2)2 nach der von Dhara in Indian Journal of Chemistry , Bd. 8, S. 143 (1970) beschriebenen Methode
50 g K2[PtCl4] wurden in 500 ml Wasser gelöst, mit Ak tivkohle gerührt und mit einem Filter der Porosität 4 fil triert. Zum Filtrat wurden 79 g Kaliumjodid mit 200 ml
Wasser zugefügt und während 5 M[inuten, vor Zugabe von
21,5 ml n-Propylamin, gerührt. Das Rühren des Gemisches wurde dann bei Zimmertemperatur während zwei Stunden fortgesetzt, es wurde filtriert und das erhaltene [PtI2(n C3HNH2)2j wurde mit Wasser, dann mit Athanol gewa-
schen und im Vakuum getrocknet. Man erhielt 63,9 g mit einer 94%igen Ausbeute.
Dieses Produkt wurde in 200 ml Wasser bei 40 "C aufgeschwemmt, und es wurde eine wässrige Lösung von 38 g Silbernitrat in 100 ml Wasser zugefügt und dann während zwei Stunden gerührt. Der Niederschlag von AgI wurde abfiltriert, mit Wasser gewaschen, und das Filtrat wurde auf den Überschuss von Silber durch Zugabe eines NaCl-Kristalls geprüft. Es wurde dann 50 ml konzentrierte Chlorwasserstoffsäure zur silberfreien Lösung zugefügt und bei 40 "C während zwei Stunden gerührt. Die Aufschwemmung des rohen Produktes wurde über Nacht gekühlt, dann wurde filtriert und der Rückstand mit wässrigem Äthanol gewaschen und im Vakuum getrocknet. Das rohe Produkt wurde aus 500 ml N,N-Dimethylformamid durch Zugabe von einem Liter von 0,1N Chlorwasserstoffsäure, umkristallisiert.
Man erhielt als Ertrag 36,7 g mit einer 80%igen Ausbeute. IR v Pt-Cl 320 cm1.
Herstellung von PtCl2(OH)2(n-C3H7NH2)2
13 g cis-PtCl2(n-C3H7NH2)2 wurden in 25 ml Wasser aufgeschlämmt und während einer halben Stunde mit 50 ml einer 37%igen Wasserstoffperoxidlösung aufgekocht. Die Aufschwemmung änderte die Farbe von hell- zu dunkelgelb, sie wurde auf 0 "C über Nacht abkühlen gelassen, das Pro dukt wurde abfiltriert, mit Wasser und Äthanol gewaschen und im Vakuum getrocknet. Der Ertrag betrug 11,6 g, das ist in 79%iger Ausbeute.

Die IR-Ergebnisse zeigen den Oxidationszustand des Platins an. Sämtliche geprüfte Komplexe sind Pt(1V), für welche die typische Pt-Cl-Streckfrequenz im Bereiche von 330 bis 350 cm- 1 liegt.
Das Ausgangsmaterial, d. h. cis PtCl2(n-CnH2n+lNH2)2 ist Pt(IE) und weist eine typische Pt Cl-Streckfrequenz: von etwa 220 cm- auf.

Der n-Octylamin-PtII-Komplex wurde mittels einer verschiedenartigen Methode hergestellt. Es wurden 25 g K2PtCI4 in 250 ml Wasser gelöst und durch Aktivkohle mittels eines Filters der Porosität 4 filtriert; zum Filtrat wurden 22,5 ml n-Octylamin zugefügt und während zwei Stunden gerührt. Man erhielt einen weissen Niederschlag, der abfiltriert, mit Wasser, Äthanol, ferner mit Äther gewaschen und im Vakuum getrocknet wurde. Das erhaltene rohe Produkt wurde aus DMF/0,lN HCl umkristallisiert. Die Ausbeute betrug 80%.

Das PtCl4a2-Derivat wurde als Vergleichssubstanz präpariert. Ausbeute 76%. v Pt-CI = 320 scharf, 345 cm

Die folgende Tabelle stellt übersichtlich Ergebnisse unter Verwendung der oben angeführten trans-Dihydroxy-Komplexe dar und zeigt einen therapeutischen Index (TI) von grösser als 11,12 an. Wie schon bei früheren Ergebnissen war der Tumor ein ADJ/PC6, der Träger war Erdnussöl, und es wurde intraperitoneal an Balb-c weisse Mäuse verabreicht.

The present invention relates to the medicament for the chemotherapeutic treatment of cancer and malignant neoplasms, which contains a platinum coordination compound of the formula I as active component and is defined in claim 1.
In formula I, X and Y are both preferably chlorine, but can also be another halogen or pseudohalogen, such as cyanate, thiocyanate and azide, and if A and B are aliphatic amine groups of the formula CnR2n + 1NH2 (II), preferably all R are Hydrogen.
The platinum is preferably present as Po4 +, so that the compound of the formula I is a neutral complex with two hydroxyl and halogen ligands each.

The IR results indicate the oxidation state of the platinum. All complexes tested are Pt (1V), for which the typical Pt-Cl stretching frequency is in the range from 330 to 350 cm-1.

The n-octylamine-PtII complex was prepared using a variety of methods. 25 g of K2PtCl4 were dissolved in 250 ml of water and filtered through activated carbon using a porosity 4 filter; 22.5 ml of n-octylamine were added to the filtrate and the mixture was stirred for two hours.
